Abstract Background: Dyslipidemia is a major cause of atherosclerosis and atherosclerosis induced conditions. Atorvastatin is an effective drug for dyslipidemia and reduce the risk of cardiovascular morbidity and mortality. Ezetimibe is used as an adjunct to statins hypercholesterolemia. Garlic is known for the hypolipidemic effect in traditional medicine. There are very limited studies comparing the additive effects of Allium sativum on atorvastatin and ezetimibe. Aims: To compare the additive hypolipidemic effects of Allium sativum with atorvastatin and ezetimibe. Setting and Design: The experimental study was done in Department of Pharmacology and Biochemistry, Burdwan Medical College, Burdwan from February 2014 - October 2015. Material and Methods: Dyslipidemia rat by (induced by atherogenic diet) were randomized into five groups of six rats in each and each cage was labelled for identification of different groups and treated with drugs (atorvastatin, ezetimibe, garlic homogenate, atorvastatin + garlic homogenate, ezetimibe + garlic homogenate) for twelve weeks and assessment of lipid profiles were done. Change of parameters checked for any significant difference by appropriate statistical tests. Results: Significant TC (Total Cholesterol) & TG (Triglyceride) concentrations reduction were maximum among ezetimibe group (51% and 47%) respectively. LDL (Low Density Lipoprotein) & VLDL (Very Low-Density Lipoprotein) concentrations reduction were maximum (62% and 26%) among combination of atorvastatin and garlic group when compared to other treatment groups. HDL (High Density Lipoprotein) concentration was maximally increased (31%) among combination ezetimibe and garlic group which was also statistically significant. Conclusion: Garlic have significant hypolipidemic effect when used in combination with atorvastatin and ezetimibe.

Adlay (Coix lachryma-jobi L. var. ma-yuen Stapf) is a cereal food for humans and has been also used as a superior medical herb substance and functional food for traditional treatment of diabetes in China. However, its scientific basis as a functional food is still unclear. The purpose of this study was to investigate the effect of dietary dehulled adlay on plasma lipid and glucose concentrations in diabetic rats. The diabetic male Sprague-Dawley (SD) rats, induced by injection of streptozotocin (60 mg/kg subcutaneously), were fed a cholesterol-rich diet (0.5% cholesterol) containing corn starch or dehulled adlay for four weeks. After completion of the experimental period, the abdominal adipose tissue and liver of rats were excised and weighed, and the plasma glucose, triglyceride, and lipoprotein cholesterol concentrations were assayed. The results showed that diabetic rats fed a dehulled adlay diet exhibited a greater adipose tissue weight (9.36 ± 3.43 vs. 5.39 ± 3.04 g, p < 0.05) and a reduced food intake (39.3 ± 5.9 vs. 61.0 ± 11.7 g/day, p < 0.05) when compared with animals fed a cornstarch diet. Significantly decreased plasma glucose (261.6 ± 96.6 vs. 422.1 ± 125.4 mg/dL, p < 0.05), total cholesterol (289.4 ± 140.6 vs. 627.3 ± 230.5 mg/dL, p < 0.05), and triglyceride (52.3 ± 14.4 vs. 96.5 ± 36.6 mg/dL, p < 0.05) levels were observed in rats fed the dehulled adlay diet. In addition, the ingestion of dehulled adlay appears to significantly decrease plasma low-density lipoprotein (LDL) plus very low-density lipoprotein (VLDL) cholesterol concentrations. Rats fed a dehulled adlay diet showed an increase in fecal weight and cholesterol contents of stools. Although a significantly decreased plasma thiobarbituric reactive substances (TBARS) value was observed in diabetic rats fed the dehulled adlay diet (6.2 ± 3.4 vs. 11.0 ± 3.8 nmol malondialdehyde (MDA)/mL, p < 0.05), no significant difference in the hepatic TBARS value was observed between the two dietary groups. Results from the present study suggest that dehulled adlay exhibited not only a hypolipidemic effect but also displayed a hypoglycemic ability in diabetic rats, indicating that dehulled adlay may play an important role in the regulation of plasma lipid and glucose metabolisms in diabetic rats induced by streptozotocin.

Background: Diabetes mellitus is one of the most widespread endocrine disorders in female and its complications are increasing all over the world, leading to life threatening medical problems like cardiovascular diseases, stroke and end stage renal diseases. A correlation between hyperlipidemia and type 2 diabetes mellitus has been identified. The study was carried out to observe the correlation of serum low density lipoprotein cholesterol (LDL-C) and high density lipoprotein cholesterol (HDL-C) level with type 2 diabetes mellitus in adult female subjects.Method: This cross sectional study was conducted in the Department of Physiology, Dhaka Medical College, Dhaka, during the period of January 2011 to December 2011. Total sixty female subjects were selected with age ranging from 30 to 50 years. Among them 30 female subjects with diabetes mellitus were included from out-patient department of Endocrinology, Dhaka Medical College Hospital, Dhaka as study group (B) and 30 apparently healthy females were taken as control group (A) for comparison. Estimation of serum fasting serum LDL-C and HDL-C levels was done by enzymatic method in the department of Physiology, Dhaka Medical College Dhaka in both groups. Fasting serum insulin level was measured by ELISA method in the laboratory of National Institute of ENT, Dhaka and fasting blood glucose was estimated by glucose oxidase method in the department of Physiology, Dhaka Medical College in both groups. Data were analyzed by Unpaired Student’s- test and Pearson’s correlation co-efficient (r) test as applicable.Results: The value of fasting serum LDL-C level was significantly higher in study subjects than those of control. Again, fasting serum HDL-C level was significantly lower in study subjects in comparison to controls. In study subjects fasting serum LDL showed positive correlation and fasting serum HDL-C levels showed negative correlation with fasting blood glucose and serum insulin level.Conclusion: Present study reveals that serum insulin and blood glucose level have positive relationship with low density lipoprotein cholesterol (LDL-C) and negative relationship with high density lipoprotein cholesterol (HDL-C) levels.J Dhaka Medical College, Vol. 26, No.2, October, 2017, Page 140-147


Dyslipidemia is a metabolic disorder that is characterized with an elevation in the cholesterol serum levels that can be treated with various hypolipidemic drugs like rosuvastatin. The present study was undertaken to determine and evaluate the hypolipidemic effect of milk thistle seeds extract in comparison with rosuvastatin and the combination of both for the treatment of dyslipidemia in rats. Also its effect on blood glucose levels on experimentally induced dyslipidemic rats. In vivo studies were conducted on wister albino laboratory rats, in which 49 rats were induced to be dyslipidemic by a daily intragastric administration of cholesterol (2 g/kg). The induction of dyslipidemia was evaluated by comparing these rats with a negative control group that was composed of 10 healthy rats. Then, after one month dyslipidemia was induced in 49 rats that were divided into 6 groups, as the following; positive control group (n=9) received cholesterol (2 g/kg) for another one month, and the other five groups each of 8 rats continued to receive cholesterol (2 g/kg) for one month along with therapy as; rosuvastatin low dose (RL) group received 10 mg/kg, rosuvastatin high dose (RH) group received 20 mg/kg, milk thistle (MT) group received 7.15 mg/kg, (RL+MT) group received a combination of 10 mg/kg of rosuvastatin and 7.15 mg/kg of milk thistle, and (RH+MT) group received a combination of 20 mg/kg of rosuvastatin and 7.15 mg/kg of milk thistle. The statistical results of biochemical analysis showed that all the studied therapeutic protocols whether given alone; RL, RH, and MT or in a combination; RL+MT and RH+MT led to a significant (p≤0.05) hypolipidemic effect that reduced the total cholesterol (TC), triglyceride (TG), low density lipoprotein (LDL) and very low density lipoprotein (VLDL) and increased the high density lipoprotein (HDL) cholesterol levels. In conclusion, all therapeutic protocols were effective in treating dyslipidemia, as they all reduced the TC, TG, LDL, and VLDL, and increased the HDL cholesterol significantly (p≤0.05). Furthermore, we found that milk thistle can be used in the management of dyslipidemia, as it has a hypolipidemic effect. Also, the addition of milk thistle to rosuvastatin therapy reduced the risk of developing diabetes mellitus (DM), as it has a glucose modulating activity either when it was given alone or in combination with rosuvastatin. Moreover, the combination of milk thistle and rosuvastatin was of a great benefit, as it gave an intensive goal of therapy than each one alone in altering all lipid profile parameters.

Hypothesis: Oxidized low density lipoprotein (Ox-LDL) has properties that profoundly affect cardiovascular function. We hypothesized that Ox-LDL is likely to be formed in the left ventricular blood (LVB) when the heart is subjected to ischemic conditions and the ejection fraction (EF) is low. We speculated whether “stagnation” of LDL in the LV could result in increased formation of Ox-LDL. Objective: We studied whether there is an increased level of Ox-LDL in the LVB as opposed to peripheral blood (PB), and whether its presence correlated with the EF. Also we examined whether a higher level of Ox-LDL negatively correlated with the activity of paraoxonase 1 (PON 1). Methods: Following the Institutional Review Board (IRB) approval, 62 HF patients were enrolled in the study. All patients underwent pre-operative transthoracic echocardiographic assessment of ventricular function. Left ventricular ejection fractions were determined using the Simpsons bi-plane technique. 2ml of LVB and 5ml of PB samples were taken before coronary artery bypass surgery, or a surgery with replacement of mitral, aortic or tricuspid valve. Blood level of Ox-LDL was determined by ELISA (Mercodia), and PON 1 activity was determined by the rate of conversion of its substrate p-nitrophenyl acetate into p-nitrophenol. Results: The result showed significant increase in Ox-LDL in LVB as compared to PB (p=0.032) in HF subjects even when EF was near normal. There was no significant increase in subjects with lower EF. In contrast, Ox-LDL levels increased in the PB of subjects with lower EF and reached those of LVB. We also noticed that there was a statistically significant negative correlation between EF and Ox-LDL levels in both LVB and PB (p < 0.05). The activity of PON1, an antioxidant enzyme that protects LDL from oxidation showed decreased levels both in LV blood as well as in PB with decreased EF. It was observed that there was a statistically significant difference in PON1 levels between LV and PB of subjects having EF>60% (p = 0.03). Conclusions: In conclusion the results suggest that there might be oxidative stress associated with LVB even when the EF is not compromised. In contrast, the increase in PB Ox-LDL with poor EF might suggest that the low blood flow to peripheral tissues and end organs also might contribute to increased oxidative stress. The results also might suggest that persistent oxidative stress could have affected the clearance mechanisms of Ox-LDL.

Abstract Background Chronic kidney disease (CKD), including nephrotic syndrome, is a major cause of cardiovascular morbidity and mortality. The literature indicates that CKD is associated with profound lipid disorders due to the dysregulation of lipoprotein metabolism which progresses kidney disease. The objective of this study is to evaluate the protective effects of curcumin on dyslipidaemia associated with adenine-induced chronic kidney disease in rats. Methods Male SD rats (n = 29) were divided into 5 groups for 24 days: normal control (n = 5, normal diet), CKD control (n = 6, 0.75% w/w adenine-supplemented diet), CUR 50 (n = 6, 50 mg/kg/day curcumin + 0.75% w/w adenine-supplemented diet), CUR 100 (n = 6, 100 mg/kg/day curcumin + 0.75% w/w adenine-supplemented diet), and CUR 150 (n = 6, 150 mg/kg/day curcumin + 0.75% w/w adenine-supplemented diet). The serum and tissue lipid profile, as well as the kidney function test, were measured using commercial diagnostic kits. Results The marked rise in total cholesterol, low-density lipoprotein (LDL) cholesterol, very low-density lipoprotein (VLDL) cholesterol, triglycerides and free fatty acids in serum, as well as hepatic cholesterol, triglyceride and free fatty acids of CKD control rats were significantly protected by curcumin co-treatment (at the dose of 50, 100 and 150 mg/kg). Furthermore, curcumin significantly increased the serum high-density lipoprotein (HDL) cholesterol compared to the CKD control rats but did not attenuate the CKD-induced weight retardation. Mathematical computational analysis revealed that curcumin significantly reduced indicators for the risk of atherosclerotic lesions (atherogenic index) and coronary atherogenesis (coronary risk index). In addition, curcumin improved kidney function as shown by the reduction in proteinuria and improvement in creatinine clearance. Conclusion The results provide new scientific evidence for the use of curcumin in CKD-associated dyslipidaemia and substantiates the traditional use of curcumin in preventing kidney damage.

Abstract An immunochemical technic has been found to be specific, efficient and reproducible in removing lipoproteins of density less than 1.063 gm./ml. from measured drops of human serum. Estimations of microcapillary columns of immunoprecipitate compare favorably with ultracentrifugal analysis of Sf 0-400 lipoproteins. An equation derived from these data converts millimeters of lipoprotein precipitate into milligrams lipoprotein Sf 0-400 per 100 ml. serum. A highly significant difference was obtained between mean lipoprotein precipitate levels of a clinically healthy population and those of a group of patients with coronary artery disease or diabetes mellitus.

The lectin-like, oxidized low-density lipoprotein (ox-LDL) receptor-1 (LOX-1)/ox-LDL system contributes to atherosclerosis and may be involved in cartilage degeneration. The purpose of this study was to determine whether the LOX-1/ox-LDL system contributes to age-related osteoarthritis (OA) in vivo, using LOX-1 knockout (LOX-1 KO) mice. Knee cartilage from 6, 12, and 18-month old (n = 10/group) C57Bl/6 wild-type (WT) and LOX-1 KO mice was evaluated by determining the Osteoarthritis Research Society International (OARSI) score of Safranin-O stained samples. The prevalence of knee OA in both mouse strains was also investigated. Expression levels of LOX-1, ox-LDL, runt-related transcription factor-2 (Runx2), type-X collagen (COL X), and matrix metalloproteinase-13 (MMP-13) in the articular chondrocytes were analyzed immunohistologically. No significant difference was observed in the mean scores of WT (2.00±0.61) and LOX-1 KO mice (2.00±0.49) at 6 months of age (P=1.00, n=10). At 12 and 18 months of age, the mean scores of LOX-1 KO mice (3.75±0.93 and 5.50±0.78) were significantly lower than those of WT mice (5.25±1.14 and 9.00±1.01; P<0.001 in both cases; n=10). The prevalence of OA in LOX-1 KO mice was lower than that in WT mice at 12 and 18 months of age (40 vs 70%, 70 vs 90%, respectively; n=10). The expression levels of Runx2, COL X, and MMP-13 in articular chondrocytes significantly decreased in LOX-1 KO, mice compared with those in WT mice. The study indicated that the LOX-1/ox-LDL system in chondrocytes plays a role in the pathogenesis of age-related knee OA, which is potentially a target for preventing OA progression.

ABSTRACT: Obesity has been increasing in cats andis associated with metabolic and cardiovascular diseases. The association of these alterations can trigger the onset of metabolic syndrome (MS). Therefore, this study aimed to analyze the serum levels of glucose, fructosamine, cholesterol, triglycerides, high-density lipoprotein, low-density lipoprotein, very low-density lipoprotein (VLDL), andalanine aminotransferase of cats and to identify the possibility of MS,as well as to evaluate changes in arterial pressure. Thirty-seven cats were classified by ECC and morphometric measurements, namely, 15 obese, 12 overweight, and 10 controls. Nocat manifested MS. Only VLDL had a statistically significant difference (P<0.05) between groups. Therefore, obesity may not be associated with arterial hypertension, and more studies are needed to evaluate the metabolic alterations in overweight and obese cats.

Previously, we demonstrated that the hydrophobic surfactant Pluronic L-81 (L-81) inhibits the intestinal formation and transport of chylomicrons (CM) but not of very low-density lipoprotein-sized (VLDL) particles. The present study was undertaken to determine whether infusion of egg lecithin results mainly in secretion of VLDL by the small intestine and whether L-81 has any effect on their formation and secretion. Intestinal fistula rats were infused intraduodenally at a rate of 3 ml/h with a lipid emulsion containing 20 mM egg lecithin and 19 mM sodium taurocholate for 8 h. This was then followed by another 8 h of infusion of a similar lipid emulsion but with 0.5 mg/h of L-81 added. Lymphatic lipid output was measured, and lymph lipoproteins were sized by use of electron microscopy. Whether L-81 was present or not, no significant difference was detected in the lymphatic triglyceride, phospholipid, or cholesterol outputs. Based on agarose gel electrophoresis, sizing of intestinal lymph lipoproteins, and also the determination of lipid in the intestinal lymph CM and VLDL as separated by ultracentrifugation, VLDL were the major lipoproteins present in lymph during the infusion of egg lecithin. Thus, intraduodenal infusion of egg lecithin in the rat results mainly in the transport of VLDL and is not affected by the administration of L-81. The results suggest that CM and VLDL are assembled separately by the enterocytes and indicate the usefulness of L-81 in further investigating the pathways and regulation of intestinal lipoprotein synthesis, assembly, and secretion.

Diabetes mellitus is a complex multifarious metabolic disorder characterized by chronic hyperglycemia that requires non-stop treatment with multiple factors that will reduce the risk of long-term complications and drug side effects. Streptozotocin is well known for its selective pancreatic islet β-cell cytotoxicity used to induce diabetes mellitus in animals. It also interferes with cellular metabolic oxidative mechanisms. Phytomedicine has been in existence since the primitive age and has proven to be effective in treating this condition. This study was aimed at evaluating the effect of Gangaria de flush (Gdf) herbal mixture on lipid profile in streptozotocin (STZ) (60mg/kg) induced diabetic rat. The diabetic rats were treated with Gangaria de flush (0.3ml/kg body weight) and Glibenclamide (5mg/kg body weight) for 7days. The effects of both Gangaria de flush herbal mixture and glibenclamide were analyzed. Gangaria de flush showed a reduction in the lipid profile with a significant percentage reduction of 45.9% for the total cholesterol, 65.7% for the total triglyceride, 63.5% for the low-density lipoprotein, 65.8% for the very low-density lipoprotein as against the negative control and a 7.6% significant increase in the high density lipoprotein. Gangaria de flush showed a significant (p<0.05) positive effect on the lipid profile. Hence, it can be suggested that Gangaria de flush herbal mixture has a significant effect in maintaining the lipid level.
