Pleurotus tuber-regium has been used as fodder and traditionally used as medicinal supplement. The aim of this study was to screen in vitro proximate biochemical composition and nutritional potentiality of P. tuber-regium extract for further in vivo analysis of impact of the extract on serum lipid and testosterone profile of rats. Total cholesterol, HDL cholesterol, LDL cholesterol were measured photometrically. Testosterone was measured by chemiluminescence immune assay. P. tuber-regium extract contains different biochemicals such as proteins, carbohydrates, tannins, flavonoids, phenols etc. and showed highly quantified calorific value (297.89±1.92 calori/100g). Acute toxicity test showed no mortality and toxic behavioural symptoms. Significant decrease in total cholesterol (75.52±0.39mg/dL), LDL cholesterol (34.58±1.69mg/dL) and triglyceride (81.31±1.25) were observed in rat group treated with 200mg/kg dose of extract. 400mg/kg dose of extract showed higher efficacy than 200mg/kg dose of extract and significantly decreased total cholesterol (72.25±1.11mg/dL), LDL cholesterol (26.37±1.21mg/dL) and triglyceride (69.42±0.72). The extract showed testosterone enhance efficacy. 400mg/kg dose of extract showed high level testosterone (178.96±0.68 ng/dL) enhance efficacy than 200mg/kg dose (174.64±0.64ng/dL). Due to hypocholesterolaemia and testosterone elevating properties of P. tuber-regium extract, it can be said that, mushroom P. tuber-regium can be consumed as healthy diet supplement to maintain cholesterol level and to maintain good male reproductive health.

The aim of this work is to analyze the relationship between the level of professional stress and the concentration of total cholesterol, HDL cholesterol, LDL cholesterol and triglycerides in the serum of security employees. . The study included 415 security officers (exposed group) divided into four sub-groups and 150 administrative workers (control group). By applying a standardized questionnaire, the level of professional stress in their workplaces was calculated. To determine the serum lipid concentration in the all subjects, venous blood samples were taken in the morning after a twelve-hour food abstinence Security officers are exposed to a high level of professional stress, where the level of stress index depends on the type of the job they are engaged in. The highest values of the total index of professional stress, total cholesterol, LDL cholesterol and triglycerides were registered with the employees who transport money and those carrying firearms. The lowest total index of professional stress, total cholesterol, LDL cholesterol and triglycerides were registered with property security officers and persons who do not carry firearms.  The correlation between the level of stress index at work, an increase in total cholesterol, LDL cholesterol, triglycerides and HDL cholesterol levels is statistically significant. We believe that there is a significant correlation between the level of the overall stress index at work and the risk of atherosclerosis in security officers.

Drug-eluting stents (DES) have significantly improved restenosis rates as compared to bare metal stents (BMS), however, an increased risk in late stent thrombosis (LST) has been observed due to delayed healing and incomplete endothelialization. These adverse events have been associated to the high level of anti-proliferative drugs eluted from 1st generation DES, as well as non-erodible polymers that remain intact. The purpose of this study was to fist determine optimal surface modification in vitro and to then assess trends in endothelial coverage and recovery of the modified non-polymeric DES in a rabbit iliac stent model.

AbstrakPenyakit kardiovaskuler merupakan penyebab kematian nomor satu secara global. Salah satu penyakit kardiovaskuler itu adalah Sindrom Koroner Akut (SKA) yang merupakan keadaan gawat darurat dari Penyakit Jantung Koroner (PJK). Salah satu faktor risiko SKA adalah perubahan dari kadar fraksi lipid yaitu kolesterol total, kolesterol LDL, kolesterol HDL, dan trigliserida yang dikaitkan dengan pembentukan plak aterosklerosis. Penelitian ini bertujuan untuk mengetahui gambaran profil lipid pada pasien SKA di rumah sakit khusus jantung Sumatera Barat tahun 2011-2012. Telah dilakukan penelitian deskriptif dengan bentuk cross sectional study dan pendekatan retrospective menggunakan data rekam medik mengenai hasil pemeriksaan profil lipid (kolesterol total, HDL, LDL, dan trigliserida) di rumah sakit khusus jantung Sumatera Barat untuk mengetahui gambaran profil lipid pada pasien SKA tahun 2011-2012. Hasil penelitian ini menemukan 98 kasus SKA. hasil ini menunjukkan bahwa pasien SKA dengan kadar kolesterol total tinggi adalah 44 orang (44,9%) dan normal sebanyak 54 orang (55,1%), pasien SKA dengan kadar kolesterol HDL rendah adalah 63 orang (64,3%) dan normal sebanyak 35 orang (35,6%), pasien SKA dengan kadar kolesterol LDL tinggi adalah 44 orang (44,9%) dan normal sebanyak 54 orang (55,1%), dan pasien SKA dengan kadar trigliserida tinggi adalah 21 orang (21,4%) dan normal sebanyak 77 orang (78,6%). Kejadian SKA terbanyak adalah STEMI sebanyak 51 kasus (52%), kemudian NSTEMI sebanyak 24 kasus (24,5%) dan yang paling sedikit adalah angina pektoris tak stabil sebanyak 23 kasus (23,5%) frekuensi umur terbanyak dari pasien SKA adalah 40-59 tahun, jenis kelamin terbanyak dari pasien SKA adalah laki-laki sekitar 74,5%. One of the cardiovascular disease is Acute Coronary Syndrome (ACS) which is a state of emergency from Coronary Heart Disease (CHD). One of the risk factors for ACS is a change in the levels of lipid fractions such as total cholesterol, LDL Cholesterol, HDL Cholesterol and triglycerides which are associated with the formation of atherosclerotic plaque. This study aims to determine description of lipid profile in patients with acute coronary syndrome at the heart hospital of west sumatera 2011-2012. Descriptive research has been conducted with a a cross-sectional study and a retrospective approach using medical record about the result of lipid profile test (total cholesterol, HDL, LDL, and triglycerides) at the heart hospital of West Sumatera to describe the lipid profile in patients with ACS in 2011-2012. The results of this study found 98 cases of ACS. It showed that ACS patients with high total cholesterol levels are 44 people (44,9%) and normal are 54 people (55,1%), acute coronary syndrome patients with low levels of HDL cholesterol are 63 people (64,3%) and normal are 35 people (35,6%), acute coronary syndrome patients with high levels of LDL cholesterol are 44 people (44,9%) and normal are 54 people (55,1%), acute coronary syndrome patients with high levels of triglyceride are 21 people (21,4%) and normal are 77 people (78,6%), the highest incidence of SKA is STEMI with 51 cases (52%), then NSTEMI with 24 cases (24,5%) and the lowest incidence is unstable angina pectoris with 23 cases (23,5%), most age of the patients of ACS are 40-59 years old, the most gender of ACS are male about 74,5%. Research shows that the number of ACS patients who have a high level of total cholesterol are less than normal, low levels of HDL cholesterol are more than normal, high level of LDL cholesterol are less than normal, high level of triglyceride are less than normal, most incidens of ACS is STEMI then NSTEMI and the lowest is unstable angina pectoris, most age of ACS patients are 40-59 years, and most of gender are men.Keywords:acute myocardial infarction, total cholesterol, LDL cholesterol, HDL cholesterol, triglyceride

Introduction: A systematic review and meta-analysis of clinical trials was undertaken to evaluate the effect of diacerein intake on cardiometabolic profiles in patients with type 2 diabetes mellitus (T2DM). Methods: Electronic databases such as PubMed, EMBASE, Scopus, Web of Science, Google Scholar, and Cochrane Central Register of Controlled Trials were searched from inception to 31 July 2019. Statistical heterogeneity was evaluated using Cochran’s Q test and I-square (I2 ) statistic. Data were pooled using random-effect models and weighted mean difference (WMD). Results: From 1,733 citations, seven clinical trials were eligible for inclusion and meta-analysis. A significant reduction in hemoglobin A1c (HbA1c) (WMD -0.73; 95%CI -1.25 to -0.21; P= 0.006; I2 = 72.2%) and body mass index (BMI) (WMD -0.55; 95%CI -1.03 to -0.07; P= 0.026; I2 = 9.5%) were identified. However, no significant effect of diacerein intake was identified on fasting blood sugar (FBS) (WMD - 9.00; 95%CI -22.57 to 4.57; P= 0.194; I2 = 60.5%), homeostatic model assessment for insulin resistance (HOMA-IR) (WMD 0.39; 95%CI 0.95 to 1.73; P= 0.569; I2 = 2.2%), body weight (WMD -0.54; 95%CI -1.10 to 0.02; P= 0.059), triglycerides (WMD -0.56; 95%CI -24.16 to 23.03; P= 0.963; I2 = 0.0%), total-cholesterol (WMD -0.21; 95%CI -12.19 to 11.78; P= 0.973; I2 = 0.0%), HDL-cholesterol (WMD -0.96; 95%CI -2.85 to 0.93; P= 0.321; I2 = 0.0%), and LDL-cholesterol levels (WMD -0.09; 95%CI -8.43 to 8.25; P= 0.983; I2 = 37.8%). Conclusion: Diacerein intake may reduce HbA1c and BMI; however, no evidence of effect was observed for FBS, HOMA-IR, body weight, triglycerides, total-cholesterol, HDL-cholesterol or LDL-cholesterol.

AbstractFork-head box protein M1 (FoxM1) is a transcriptional factor which plays critical roles in cancer development and progression. However, the general regulatory mechanism of FoxM1 is still limited. STMN1 is a microtubule-binding protein which can inhibit the assembly of microtubule dimer or promote depolymerization of microtubules. It was reported as a major responsive factor of paclitaxel resistance for clinical chemotherapy of tumor patients. But the function of abnormally high level of STMN1 and its regulation mechanism in cancer cells remain unclear. In this study, we used public database and tissue microarrays to analyze the expression pattern of FoxM1 and STMN1 and found a strong positive correlation between FoxM1 and STMN1 in multiple types of cancer. Lentivirus-mediated FoxM1/STMN1-knockdown cell lines were established to study the function of FoxM1/STMN1 by performing cell viability assay, plate clone formation assay, soft agar assay in vitro and xenograft mouse model in vivo. Our results showed that FoxM1 promotes cell proliferation by upregulating STMN1. Further ChIP assay showed that FoxM1 upregulates STMN1 in a transcriptional level. Prognostic analysis showed that a high level of FoxM1 and STMN1 is related to poor prognosis in solid tumors. Moreover, a high co-expression of FoxM1 and STMN1 has a more significant correlation with poor prognosis. Our findings suggest that a general FoxM1-STMN1 axis contributes to cell proliferation and tumorigenesis in hepatocellular carcinoma, gastric cancer and colorectal cancer. The combination of FoxM1 and STMN1 can be a more precise biomarker for prognostic prediction.

Abstract DHR38 is a member of the steroid receptor superfamily in Drosophila homologous to the vertebrate NGFI-B-type orphan receptors. In addition to binding to specific response elements as a monomer, DHR38 interacts with the USP component of the ecdysone receptor complex in vitro, in yeast and in a cell line, suggesting that DHR38 might modulate ecdysone-triggered signals in the fly. We characterized the molecular structure and expression of the Dhr38 gene and initiated an in vivo analysis of its function(s) in development. The Dhr38 transcription unit spans more than 40 kb in length, includes four introns, and produces at least four mRNA isoforms differentially expressed in development; two of these are greatly enriched in the pupal stage and encode nested polypeptides. We characterized four alleles of Dhr38: a P-element enchancer trap line, l(2)02306, which shows exclusively epidermal staining in the late larval, pre-pupal and pupal stages, and three EMS-induced alleles. Dhr38 alleles cause localized fragility and rupturing of the adult cuticle, demonstrating that Dhr38 plays an important role in late stages of epidermal metamorphosis.

AbstractBrian metastasis, which is diagnosed in 30% of triple-negative breast cancer (TNBC) patients with metastasis, causes poor survival outcomes. Growing evidence has characterized miRNAs involving in breast cancer brain metastasis; however, currently, there is a lack of prognostic plasma-based indicator for brain metastasis. In this study, high level of miR-211 can act as brain metastatic prognostic marker in vivo. High miR-211 drives early and specific brain colonization through enhancing trans-blood–brain barrier (BBB) migration, BBB adherence, and stemness properties of tumor cells and causes poor survival in vivo. SOX11 and NGN2 are the downstream targets of miR-211 and negatively regulate miR-211-mediated TNBC brain metastasis in vitro and in vivo. Most importantly, high miR-211 is correlated with poor survival and brain metastasis in TNBC patients. Our findings suggest that miR-211 may be used as an indicator for TNBC brain metastasis.

Abstract Background Hepatocellular carcinoma (HCC) has an extremely poor prognosis due to the development of chemoresistance, coupled with inherently increased stemness properties. Long non-coding RNAs (LncRNAs) are key regulators for tumor cell stemness and chemosensitivity. Currently the relevance between LINC00680 and tumor progression was still largely unknown, with only one study showing its significance in glioblastoma. The study herein was aimed at identifying the role of LINC00680 in the regulation HCC stemness and chemosensitivity. Methods QRT-PCR was used to detect the expression of LINC00680, miR-568 and AKT3 in tissue specimen and cell lines. Gain- or loss-of function assays were applied to access the function of LINC00680 in HCC cells, including cell proliferation and stemness properties. HCC stemness and chemosensitivity were determined by sphere formation, cell viability and colony formation. Luciferase reporter, RNA immunoprecipitation (RIP), and RNA pull-down assays were performed to examine the interaction between LINC00680 and miR-568 as well as that between miR-568 and AKT3. A nude mouse xenograft model was established for the in vivo study. Results We found that LINC00680 was remarkably upregulated in HCC tissues. Patients with high level of LINC00680 had poorer prognosis. LINC00680 overexpression significantly enhanced HCC cell stemness and decreased in vitro and in vivo chemosensitivity to 5-fluorouracil (5-Fu), whereas LINC00680 knockdown led to opposite results. Mechanism study revealed that LINC00680 regulated HCC stemness and chemosensitivity through sponging miR-568, thereby expediting the expression of AKT3, which further activated its downstream signaling molecules, including mTOR, elF4EBP1, and p70S6K. Conclusion LINC00680 promotes HCC stemness properties and decreases chemosensitivity through sponging miR-568 to activate AKT3, suggesting that LINC00680 might be a potentially important HCC diagnosis marker and therapeutic target.
