Comparison of the effects of single probiotic strains Lactobacillus casei Zhang and Bifidobacterium animalis ssp. lactis Probio-M8 and their combination on volatile and nonvolatile metabolomic profiles of yogurt

To find out the effect of probiotic strains on insulin resistance induced by glutamate-induced obesity in rats. Obesity was induced by neonatal administration of monosodium glutamate (4 mg/g,s.c.) at 2, 4, 6, 8, 10-day life. The introduction of probiotics started at the end of 4 week after birth and continued intermittently for 2-week courses. After 4 months, the rats of all groups glucose and insulin levels and insulin resistance index HOMA were analyzed. Neonatal administration of monosodium glutamate leads to the development of insulin resistance in rats 4 months of age. The periodic introduction of the combined probiotics prevented the development insulin resistance in rats. The most significant effect was revealed in the group of animals treated with the combined probiotic strains Lactobacillus casei IMVB-7280, Bifidobacterium animalis VKL, Bifidobacterium animalis VKB. The results suggest the effectiveness of the use of probiotic strains of bifidobacteria and to prevent the development of insulin resistance.

The aim of the present work was to investigate the potential prebiotic action of Goji berry powder on selected probiotic bacteria grown in a nutritive synthetic substrate and in simulated gastric and intestinal juices. Different probiotic strains of Bifidobacterium and Lactobacillus were grown in these substrates with or without the addition of encapsulated goji berry extracts of different polysaccharide and polyphenol contents. The results proved that the addition of the extracts promoted the proliferation of probiotic strains and, in particular, increased the number of bacterial colonies of Bifidobacterium animalis subsp. lactis (Bb12), Bifidobacterium longum (Bb46), and Lactobacillus casei by 2, 0.26, and 1.34 (log cfu/mL), respectively. Furthermore, the prebiotic effect seems to be correlated to Goji berry polysaccharides and/or polyphenols, higher contents of which (under the tested concentrations) could increase the stress tolerance of B. lactis and B. longum in a simulated gastrointestinal environment. According to the findings of the present research, it can be suggested that the Goji berry encapsulated extracts could be used as prebiotic additives in food or nutraceuticals, in order to stimulate growth or protect the viability of probiotic strains of Bifidobacterium and Lactobacillus.

The objective of the study was determining the prophylactic effect of Lactobacillus casei IMV B-7280, L. acidophilus IMV B-7279, L. delbrueckii subsp. bulgaricus IMV B-7281, Bifidobacterium animalis VKL and B. animalis VKB probiotic strains on experimental vaginitis in BALB/c mice induced by Staphylococcus aureus 8325-4. The infection with S. aureus 8325-4 caused an imbalance of microbiota in the vagina and intestine, as evidenced by an increase in the number of opportunistic microorganisms and a decrease in the amount of lactobacilli and bifidobacteria. L. casei IMV B-7280, B. animalis VKL and B. animalis VKB probiotic strains altered the microbiota spectrum of the vagina and intestine of Staphylococcus-infected mice: the amount of Lactobacillus and bifidobacteria increased with the reduction of the number of opportunistic microorganisms. Also under the influence of these strains, the normalization of the microbiota spectrum typical for vagina and intestine was observed in different periods of observation – in the intestines of mice the number of coliform bacteria increased, the number of microscopic fungi, streptococci and staphylococci decreased; in the vagina, the number of coliform bacteria and microscopic fungi decreased, the number of streptococci normalized. Rapid elimination of S. aureus 8325-4 from the vagina and prevention of the spread of infection to the intestine were observed after use of probiotics. Preventive effect of L. acidophilus IMV B-7279 and L. delbrueckii subsp. bulgaricus IMV B-7281 for bacterial vaginitis in mice was less effective. So, the target probiotic strains L. casei IMV B-7280, B. animalis VKL and B. animalis VKB are promising for the creation of highly effective novel probiotic drugs that can be used for directed prevention of infectious and inflammatory diseases of the genitourinary system caused by pathogenic and opportunistic microorganisms.

AbstractOral-nasal mucosal immunity plays a crucial role in protecting the body against bacterial and viral invasion. Safe probiotic products have been used to enhance human immunity and oral health. In this study, we verified the beneficial effects of mixed viable probiotic tablets, consisting of Lactobacillus salivarius subsp. salicinius AP-32, Bifidobacterium animalis subsp. lactis CP-9, and Lactobacillus paracasei ET-66, and heat-killed probiotic tablets, consisting of L. salivarius subsp. salicinius AP-32 and L. paracasei ET-66, on oral immunity among 45 healthy participants. Participants were randomly divided into viable probiotic, heat-killed probiotic, and placebo groups. The administration of treatment lasted for 4 weeks. Saliva samples were collected at Weeks 0, 2, 4, and 6, and Lactobacillus, Bifidobacterium and Streptococcus mutans populations and IgA concentration were measured. IgA concentrations, levels of TGF-beta and IL-10 in PBMCs cells were quantified by ELISA method. Results showed that salivary IgA levels were significantly increased on administration of both the viable (119.30 ± 12.63%, ***P < 0.001) and heat-killed (116.78 ± 12.28%, ***P < 0.001) probiotics for 4 weeks. Among three probiotic strains, AP-32 would effectively increase the levels of TGF-beta and IL-10 in PBMCs. The oral pathogen Streptococcus mutans was significantly reduced on viable probiotic tablet administration (49.60 ± 31.01%, ***P < 0.001). The in vitro antibacterial test confirmed that viable probiotics effectively limited the survival rate of oral pathogens. Thus, this clinical pilot study demonstrated that oral probiotic tablets both in viable form or heat-killed form could exert beneficial effects on oral immunity via IL-10, TGB-beta mediated IgA secretion. The effective dosage of viable probiotic content in the oral tablet was 109 CFUs/g and the heat-killed oral tablet was 1 × 1010 cells/g.

Abstract Bovine mastitis is one of the major diseases which directly affects the milk production performance and it causes huge economic losses in the dairy industry. Bacterial infection is the main risk factor of bovine mastitis and the antibiotic therapy is the primary choice to control the disease. However, persistence use of antibiotic increases the incidence of bacterial resistance and traces of antibiotic residues in animal products. Lactobacillus casei Zhang is one of the probiotics with multiple biological functions, which has certain bacteriostatic effect on pathogenic microorganism. The purpose of this study was to explore the effect of Lactobacillus casei Zhang (L. casei Zhang) on the prevention of E. coli-induced milk-blood barrier damage. Bovine mammary epithelial cells (BMECs) were used to establish a milk-blood model and Control group (PBS), E. coli group, and L. casei Zhang pretreatment plus E. coli group were set up respectively. The results showed that: L. casei Zhang could significantly reduce the increase of LDH release caused by E. coli treatment (P&lt; 0.05). And it can also significantly reduce the decrease of transmembrane resistance of monolayer cells caused by E. coli treatment (P&lt; 0.05). In addition, L. casei Zhang could significantly reduce the expression of tight junction proteins ZO-1, Claudin-1, Claudin-4 and Occludin (P &lt; 0.05). In conclusion, L. casei Zhang could effectively improve the damage of the blood-milk barrier caused by E. coli and could protect BMECs during bacterial infection.

Os objetivos deste trabalho foram avaliar a resistência de espécies probióticas de Lactobacillus na presença de sais de cura e testar sua sensibilidade frente a diferentes antimicrobianos para o desenvolvimento de meio de cultura seletivo. As culturas Lactobacillus casei, Lactobacillus paracasei e Lactobacillus rhamnosus foram semeadas em ágar MRS contendo concentrações de 0% a 3% de cloreto de sódio e 0 a 200 ppm de nitrito de sódio. O efeito do uso concomitante dos sais de cura foi verificado utilizando-se 3% de cloreto de sódio e 200 ppm de nitrito de sódio. As bactérias probióticas e a cultura starter Pediococcus pentosaceus foram testadas frente a 20 discos de antimicrobianos pela técnica de disco-difusão. O crescimento dos probióticos não apresentou alteração nas diferentes concentrações de cloreto de sódio, assim como nas concentrações de até 200 ppm de nitrito de sódio. Verificou-se resistência ao uso simultâneo de cloreto e nitrito de sódio. Entre os antimicrobianos testados, a tetraciclina apresentou resultados satisfatórios para a inibição dos probióticos permitindo o crescimento isolado da cultura starter. Os probióticos apresentaram desenvolvimento satisfatório na presença dos sais de cura, demonstrando potencial para sua utilização em produtos cárneos fermentados com ação probiótica. CHARACTERISTICS OF PROBIOTIC CULTURES FOR THE USE IN FERMENTED MEAT PRODUCTS - SENSIBILITY TO CURING SALTS AND ANTIBIOTIC USE FOR THE SELECTIVE ENUMERATION Abstract The objective of this study was to evaluate the resistance of probiotic species of Lactobacillus in the presence of curing salts and to test their sensibility in the presence of antibiotics for the development of a selective culture media. The probiotic cultures, Lactobacillus casei, Lactobacillus paracasei spp paracasei and Lactobacillus casei spp rhamnosus, were plated in MRS agar with concentration of 0 to 3% of sodium chloride and 0 to 200 ppm of sodium nitrite. The inhibitory effect of 3% sodium chloride and 200 ppm sodium nitrite in simultaneous use was evaluated for the probiotic strains. The sensibility of the starter culture, Pediococcus pentosaceus and the probiotic cultures in the presence of antibiotic was carried out using 20 different antibiotic discs by the disc-diffusion technique. The growth of the probiotic cultures wasn’t affected at different concentrations of sodium chloride, and even to concentrations of 200 ppm of sodium nitrate. Resistance was verified by simultaneous use of sodium chloride and nitrate. Among the tested antibiotics, tetracycline showed a satisfactory inhibition effect for the probiotic strains, since the growth of starter Pediococcus pentosaceus was not affected.
