<p style="text-align: justify;">In a laboratory, an erythrocyte is done by using hemocytometer and microscope. The task is to measure and assess the size and shape of the erythrocyte. But this procedure is time consuming, complex and tedious. As a solution to this problem, to provide an automated, cost-effective and efficient alternative to detection and counting of erythrocyte, hematology analyzers are used. However, false results related either to erythrocyte or other parameters from complete blood count may be observed in several instances. The objective study was to compare the accuracy of erythrocyte count results of automatic hematological analysis by the manual method. The study is an analytic survey with a cross-sectional design. Erythrocyte counting is done by using three types of control blood, namely high, normal and low, with 9 repetitions. In high control blood, the mean erythrocytes count by the manual method was 7.08 million/μl with a bias value of 1.4%, while that by the automated method was 7.03 million/μl with a bias value of 0.7%. The mean erythrocyte count in normal control blood by the manual method was 4.50 million/μl with a bias value of 0.9%, while that by the automated method was 4.4 million/μl with a bias value of 2.4%. And in low control blood, the mean erythrocyte count by the manual method was 1.72 million/μl with 4.4% bias value, while that the automated method was 1.67 million/μl with 1.2% bias value. Statistical analysis showed no significant difference in accuracy between the erythrocyte count of manual methods with automated methods.</p>

Abstrak Penyebab dismenore belum semuanya diketahui, ada dugaan peningkatan proses peroksida lipid yang akan mengaktivasi mediator inflamasi pada endometrium yang menimbulkan rasa nyeri haid (dismenore). Tujuan penelitian ini adalah menentukan perbedaan kadar malondialdehide dan tromboksan B 2  pada dismenore dan tanpa dismenore. Studi observasional ini menggunakan desain potong lintang komparatif. Subjek penelitian terdiri dari dua kelompok yaitu 23 remaja dismenore dan 23 remaja tanpa dismenore dengan waktu penelitian dari Juni sampai Juli 2014. Analisis sampel dilakukan di Laboratorium Biomedik dan Biokimia Universitas Andalas Padang. Pemeriksaan kadartromboksan B 2  menggunakan metode ELISA dan kadar malondialdehide menggunakan metode Asam Thiobarbiturat (TBA). Hasil penelitian diperoleh bahwa rerata kadar malondialdehid pada remaja dengan dismenore yaitu 2,60±0,63 µmol/ml, rerata remaja tanpa dismenore 1,98±0,12 µmol/ml dengan probabilitas p<0,05 (0,000), sedangkan reratakadar Tromboksan B 2  pada remaja dengan dismenore 20,043±9,56 ng/ml, rerata remaja tanpa dismenore 19,222±10,79 ng/ml, dengan probabilitas p>0,05 (0,786). Kesimpulan penelitian ini adalah terdapat perbedaan yang signifikan rerata kadar malondialdehid pada remaja dengan dismenore dan tanpa dismenore dan tidak terdapatperbedaan signifikan pada kadar tromboksan B 2 pada remaja dengan dismenore dan tanpa dismenore.Kata kunci: remaja, dismenore, malondialdehide, tromboksan B2 Abstract The precise cause of dysmenorrhea is still unclear, there may be increased lipid peroxidation process will activate the inflammatory mediators at endometrium that cause menstrual cramps (dysmenorrhea). The objective of this study was to determine the difference of malondialdehyde levels and thromboxane B 2  levels in dysmenorrhea and without dysmenorrhea. It was an observational study with comparative cross-sectional design. The subjects consisted of two groups, they are 23 adolescent with dysmenorrhea and 23 adolescents without dysmenorrhea, done in Juny -July 2014. Sample analysis was conducted in Laboratory of Biochemistry and Biomedical Laboratory of Andalas University Padang. The examination of Thromboxane B 2  levels used ELISA and the examination of malondialdehyde levels used a Thiobarbituric acid method. The results showed the mean of malondialdehyde levels in adolescents withdysmenorrhea was 2.60±0.63 µmol/ml, the mean level in adolescent without dysmenorrhea was 1.98±0.12 µmol/ml with probability p<0.05 (0.000), while the mean levels of thromboxane B 2  in adolescents with dysmenorrhea was 20.043±9.56 ng/ml, the mean level in adolescent without dysmenorrhea was 19.222±10.79 ng/ml, with probabilityp>0.05 (0.786). It can be concluded that there is a significant difference in the mean of malondialdehyde levels between adolescents with dysmenorrhea and without dysmenorrhea and there is no significant differences in thromboxane B 2 level between adolescents with dysmenorrhea and without dysmenorrheaKeywords: adolescent, dysmenorrhea, malondialdehyde, thromboxane B2

Background: Pre-eclampsia is a serious disorder of pregnancy with unknown ethological factors that may occur at any stage of second or third trimester of pregnancy. The objectives of the present study were to assess changes in complete blood counts including platelets, liver enzymes and serum uric acid in pre-eclamptic cases compared to second-half normal pregnant and non-pregnant Sudanese women and their correlations to other biomarkers.Methods: This was a cross-sectional, case-control study performed from December 2008 to December 2010; in Omdurman Maternity Hospital, in concomitance with other studies in pre-eclampsia. The sample size included three groups, 72 up pre-eclamptic cases in their recent pregnancies, 96 normal pregnant in their second half of pregnancy and 63 non- pregnant (control) women; a total of 231 subjects. Questionnaire Interviews and clinical examination were done for all participants. Laboratory investigations were done including complete blood picture, liver enzymes and uric acid.  Results: The mean Hb concentration of the pre-eclamptic (11.3g/dl±1.7) was statistically significantly lower than that of the non-pregnant (12.1g/dl±0.2) (P=0.01) but not from that of the normal pregnant (11.4g/dl±0.1) (P=0.882) .There was no statistical significant difference in the mean WBC count between the pre-eclamptic (7.4x103/mm3±0.3) and non-pregnant (7.3x103/mm3±0.3) (P=0.797) and between the pre-eclamptic and normal pregnant (7.7x103/mm3±0.2) (P=0.270). There was a considerable statistical significant decrease in the mean platelets count of the pre-eclamptic (236.4/mm3±8.3) compared to the non-pregnant group (322.0/mm3±10.4) (P=0.0001) s well as to the normal pregnant (275.0/mm3±8.9) (P = 0.003). In the pre-eclamptic cases, serum ALT correlated significantly with TWCC (r=0.26, P=0.03) and serum AST (r=0.65, P=0.000). In the pre-eclamptic cases, serum AST correlated significantly with Hb (r=0.26, P=0.03), serum ALT and serum uric acid (r=0.36, P=0.01).Conclusions: There was a considerable statistical significant decrease in mean platelets count of the pre-eclamptic compared to the non-pregnant group and to the normal pregnant may be explained by hemodilution; whereas further decrease was due to pre-eclampsia. ALT and AST are strong prognostic indicators of pre-eclampsia.

Background: Acne vulgaris is a disease of the pilosebaceous unit that often occurs in adolescents and young adults. The pathogenesis of acne vulgaris involves several factors, one of which is the acidity (pH) of the skin.  Objective: This study aimed to determine the difference in the degree of acidity of facial skin, chest and back of patients with acne vulgaris.  Methods: This was a comparative analytic study with cross sectional design. The sample collection technique was consecutive sampling. Data were analyzed using ANOVA-test.  Results: The majority of patients with acne vulgaris who came to Murni Teguh Memorial Hospital Medan and the private clinic of dr. Rudyn Reymond Panjaitan, M.Ked (KK), Sp.KK was a 17-25 year old. The mean acidity of facial skin with acne vulgaris was 5.66, the mean acidity of chest skin was 5,84 and the mean acidity of back skin was 6.09. There was a significant difference in the mean value of acidity in facial, chest and back skin of patients with acne vulgaris (p = 0,000). Conclusion: There is a significant difference in average degree of acidity on the facial, chest and back skin in patients with acne vulgaris.

An automated ankle-brachial index device could lead to potential time savings and more accuracy in ankle-brachial index-determination after vascular surgery. This prospective cross-sectional study compared postprocedural ankle-brachial indices measured by a manual method with ankle-brachial indices of an automated plethysmographic method. Forty-two patients were included. No significant difference in time performing a measurement was observed (1.1 min, 95% CI: −0.2 to +2.4; P = 0.095). Mean ankle-brachial index with the automated method was 0.105 higher (95% CI: 0.017 to 0.193; P = 0.020) than with the manual method, with limits of agreement of −0.376 and +0.587. Total variance amounted to 0.0759 and the correlation between both methods was 0.60. Reliability expressed as maximum absolute difference (95% level) between duplicate ankle-brachial index-measurements under identical conditions was 0.350 (manual) and 0.152 (automated), although not significant ( p = 0.053). Finally, the automated method had 34% points higher failure rate than the manual method. In conclusion based on this study, the automated ankle-brachial index-method seems not to be clinically applicable for measuring ankle-brachial index postoperatively in patients with vascular disease.

Abstract Background: Coronavirus Disease 2019 (COVID-19) is an infectious disease caused by Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2). In COVID-19 there is a process of thrombosis and coagulopathy, which are systemic inflammation and endothelial disorders resulting in hypercoagulability. D-dimer is a laboratory test that can determine coagulation activation (hypercoagulability) and fibrinolysis. D-dimer can be used as a guide of anticoagulant and thrombolytic therapy and prognostic parameter in COVID-19 patients, where its value runs parallel to the severity of the disease and is associated with increased mortality. Materials and methods: The whole sampling was taken from 59 medical records of inpatients confirmed with COVID-19 through the RT-PCR examination at Immanuel Hospital, Bandung City for the period of October 1 st - December 31 st, 2020. The value of d-dimer was taken from whole blood, tested with sandwich ELISA method with cut-off value (normal value) &lt; 0.5μg/ml. COVID-19 patient classification was based on the COVID-19 guideline from Indonesian Ministry of Health. The research method was observational analytical with cross-sectional design. Statistical test used was Kruskal-Wallis test with Mann-Whitney advanced test (α = 0.05). Results: The mean d-dimer value in patients with mild, moderate, and severe COVID-19 was 0.3034 µg/mL; 0.5138 µg/mL; and 1.1751 µg/mL. The results of Kruskal-Wallis test showed a very significant difference in mean d-dimer values in mild, moderate, and severe COVID-19 patients. Mann-Whitney test showed that there was a very significant difference in the mean d-dimer value between mild and severe COVID-19 patients, also between moderate and severe COVID-19 patients with a value of p = 0.000 (p &lt;0.01). But there was no difference in the mean d-dimer value between mild and moderate COVID-19 patients, p = 0.454 (p&gt; 0.05). Conclusion: There is a very significant difference in d-dimer values between mild, moderate and severe COVID-19 patients. The d-dimer value increases with the severity of COVID-19. The aim of this study was to see the difference in the mean ratio of sodium levels between pregnancy with severe preeclampsia and eclampsia. The method in this research is an analytical study using a cross sectional design. The research was conducted in the delivery room Dr. M. Djamil Padang from 15 January 2016 to 31 December 2017. There were 60 patients as research subjects, the sample was divided into 2 groups, namely severe preeclampsia (PEB) and eclampsia. Anamnesis and physical examination were then carried out to obtain data and clinical diagnosis. The data were recorded in a research form that had been provided, then the blood electrolytes were examined for sodium and potassium. Statistical analysis to assess meaning using the T-test. The results showed that in the PEB and Eclampsia groups, it was found that multiparity parity had the highest respondents. This is in accordance with the literature where the incidence of preeclampsia is more often found at gestational age near term. The conclusion of this study there was no significant difference in the mean sodium ratio between preeclampsia, PEB and eclampsia. The use of anticoagulants is one of the important pre-analytic factors in hematological tests. Both dipotassium (K2) andtripotassium (K3) Ethylene Diamine Tetraacetic Acid (EDTA) are widely used anticoagulants. International CouncilStandardization of Hematology (ICSH) and several researchers recommend the use of K2 EDTA due to its less hyperosmolareffect on blood cells compared to K3 EDTA. This study aimed to compare the results of Complete Blood Count (CBC) andErythrocyte Sedimentation Rate (ESR) using anticoagulant K2 EDTA and K3 EDTA. This study was an analytic observationalstudy with a cross-sectional design conducted from April to December 2018. The subject of the study were 103 healthyadults selected by consecutive sampling. Blood samples were collected in both anticoagulant tubes with a volume of 3 mLeach. Samples were tested twice, in the first 0 hours and the next 6 hours using Sysmex XN 1000 and Alifax Roller 20 LC.Kolmogorov-Smirnov test, paired T-test and Wilcoxon rank test were used for statistical analysis. The agreement testbetween both anticoagulants was carried out using the Bland Altman plot for parameters with a significant difference. Therewas a significant difference between both anticoagulants for the parameters of hemoglobin, hematocrit MCV, MCHC, RDW,PDW, MPV, PLC-R, and erythrocyte sedimentation rate in both the first and second tests. The agreement test using the BlandAltman plot showed that the difference in these parameters was within the Limit of Agreement (LOA) range of 95%. Thisstudy showed that there were differences in some parameters of complete blood count and erythrocyte sedimentation ratebetween the two anticoagulants (K2 K3 EDTA), but these differences were within the LOA range.

Background: To evaluate the complete blood count of Laotian students in Hue city. Materials and method: A cross-sectional study on 400 Lao students who were examinated the complete blood count at Department of Hematology, Hue University of Medicine and Pharmacy from September 2019 to May 2020. Results: 400 Lao students (male/female = 1/1.2), the students who came from Salavan province accounted for the highest percentage (22.25%). The mean of age was 24.78 ± 6.44 (17 - 44). The mean of red blood cells (RBC) was 5.13 ± 0.73 T/L and of hemoglobin in the study group was 129.27 ± 18.58 g/L. The percentage of students with anemia was 13.5% and there were 3 students with severe anemia. The mean of mean corpuscular volume (MCV) and mean corpuscular hemoglobin (MCH) were respectively 81.27 ± 9.32 fL (49.2 - 103.9) and 25.50 ± 3.56 pg (11.8 - 33.2). The mean of white blood cells (WBC) and platelets (PLT) were respectively 7.42 ± 2.29 G/L and 272.33 ± 83.62 G/L. There was a correlation between anemia and age (p < 0.05), mainly in 26 - 40. There was significant difference about RBC and Hb between anemic group and non-anemic group (p < 0.05). Conclusion: It is essential to check the complete blood count of Laotian students to evaluate anemia and screen hemoglobinopathies. AbstrakPreeklampsia-eklampsia sampai saat ini masih merupakan disease of theory. Kelainan yang terjadi pada penyakit ini adalah disfungsi endotel yang dapat disebabkan oleh perubahan kadar profil lipid. Tujuan penelitian ini adalah mengetahui perbedaan rerata kadar profil lipid pada preeklampsia dengan kehamilan normal pada etnik Minangkabau. Penelitian observasional dengan rancangan cross sectional ini dilakukan di RSUP dr. M. Djamil Padang, RS Reksodiwiryo Padang dan UPTD Laboratorium Kesehatan Daerah Sumatera Barat. Subjek terdiri dari 2 kelompok yang diambil secara consecutive sampling dengan jumlah masing-masing 27 orang. Pemeriksaan profil lipid dilakukan dengan metode kalorimetri. Analisis data dilakukan secara uji statistik independent t-test. Hasil penelitian diperoleh rerata kadar total kolesterol pada kelompok preeklampsia dan kehamilan normal adalah 270,19+68,955 mg/dL dan 247,56+44,415 mg/dL dengan nilai p=0,158, rerata kadar trigliserida pada kelompok preeklampsia dan kehamilan normal adalah 296,07±157,993 mg/dL dan 272,89±84,608 mg/dL dengan nilai p=0,504, rerata kadar HDL pada kelompok preeklampsia dan kehamilan normal adalah 51,93±19,882 mg/dL dan 63,33±11,222 mg/dL dengan nilai p=0,012, rerata kadar LDL pada kelompok preeklampsia dan kehamilan normal adalah 159±52,038 mg/dL dan 129,67±38,692 mg/dL dengan nilai p=0,023, rasio perbandingan LDL/HDL pada kelompok preeklampsia dan kehamilan normal adalah 3,86 dan 2,04. Dari hasil penelitian dapat disimpulkan pada kedua kelompok tidak ada perbedaan kadar kolesterol total dan trigliserida, tetapi ada perbedaan kadar HDL dan LDL , rasio perbandingan LDL/HDL lebih tinggi pada preeklampsia daripada kehamilan normal.Kata kunci: preeklampsia, total kolesterol, trigliserida, HDL, LDL, etnik Minangkabau AbstractPreeclampsia-eklampsia still being a disease of theory. The data analyzed statistically by independent t-test. The result of this study showed mean level of total cholesterol in preeclamsia and normal pregnancy were 270.19±68.955 mg/dL and 247.56±44.415 mg/dL with p-value 0.158. The mean levels of triglyceride in preeclampsia and normal pregnancy were 296.07±157.993 and 272.89±44.415 mg/dL with p-value 0.504. The mean level of HDL in preeclampsia and normal pregnancy were 51.93±19.882 mg/dL and 63.33 mg/dL p-value 0.012. The mean level of LDL in two groups were 159±52.038 mg/dL and 129.67±38.692 mg/dL with p-value 0.023 and the mean levels of LDL/HDL ratio were 3.86±3,09 mg/dLin  preeclampsia and 2,08±0,64 in normal pregnancy. It can be concluded that there is no significant difference in total cholesterol and triglyseride between preeclampsia and normal in HDL and LDL and there is significant difference in LDL/HDL ratio between preeclampsia and normal pregnancy at Minangkabau Ethnic.Keywords: preeclampsia, total cholesterol, triglyceride, HDL, LDL, Minangkabau Ethnic

Background: Deoxynivalenol (DON) is one of the most common mycotoxins found in cereal products. Objective: The purpose of this study is to compare the DON contamination level in raw wheat flour and bread. Methods: In this analytical study with cross-sectional design, a total of 44 wheat flour and bread samples (Lavash flour and bread, and Barbari flour and bread) were collected. The DON level was measured using ELISA method. Collected data were analyzed in SPSS software by using ANOVA and t-test considering a significance level of P≥0.05. Findings: The Mean±SD total DON level in the flour and bread samples was 0.03±0.04 and 0.12±0.21 µg/kg. The mean DON level in the Lavash and Barbari flour samples was 0.01±0.02 and 0.01±0.01 µg/kg, and in the Lavash and Barbari bread samples as 0.04±0.03 and 0.16±0.27 µg/kg, respectively. There was no statistically significant difference in the DON levels between flour and bread samples (P≥0.05). Conclusion: The presence of DON in the studied samples was observed, but its contamination level was lower than the permissible limit.
