Serum levels of inflammatory biomarkers, glycaemic control indices and leptin receptors expression in adult male Wistar rats exposed to oyrethroids

BACKGROUND: Due to widespread use of pesticides, an individual’s lifetime exposure to pesticide mixtures is unavoidable, resulting in potentiation of toxic effects such as changing metabolic signaling temporarily or permanently due to the formation of free radicals among exposed individuals. OBJECTIVE: This study then looked at the expression of leptin and insulin receptors in Wistar rats exposed to pyrethroid and its effects on adipokines and glycemic indices. METHODS: 84 Wistar rats were randomly selected and divided into two groups. 51 rats were exposed to 1.2 % w/v pyrethroid insecticides, while the remaining 33 rats were grouped as non-exposed. The groups were divided into three different groups, each with 7, 21, and 41 days. The animals were anesthetically sacrificed and samples of blood were collected after days of exposure. The levels of total antioxidant status (TAS), malondialdehyde (MDA), glutathione (GSH), hydrogen peroxide (H2O2), nitric oxide (NO), glucose and lipids were determined. Blood activities of superoxide dismutase (SOD), glutathione peroxidase (GPX), and catalase (CAT) were also determined. ELISA was used to assess leptin, adiponectin, insulin, CRP, IL-6, and TNFα. Glycaemic indices were calculated appropriately. Leptin and Insulin expression was determined using the reverse transcriptase-polymerase chain reaction (RT-PCR) method. Data obtained were statistically compared, and P≤0.05 was considered significant. RESULTS: The mean TChol, TAG, and HDL-C were significantly different (p <  0.05) among the exposed groups. The serum mean levels of SOD, GPx, CAT, GSH, TAS, QUICKI, TNFα, IL6, and CRP were significantly reduced (p <  0.05), while mean levels of MDA, H2O2, NO, insulin, HOMA-IR, FIRI, leptin, and adiponectin were significantly elevated (p <  0.05) in exposed groups. The relative expression of insulin and leptin genes in exposed and non-exposed groups was also shown by column. CONCLUSION: It is self-evident that pyrethroid chemicals exposure may disrupt carbohydrate and lipid metabolism by raising the risk of obesity among the exposed groups.

Abstract. Physical exercise increases free radicals production; antioxidant supplementation may improve the muscle fiber’s ability to scavenge ROS and protect muscles against exercise-induced oxidative damage. This study was designed to examine the effects of all-trans resveratrol supplementation as an antioxidant to mediate anti-oxidation and lipid per-oxidation responses to exercise in male Wistar rats. Sixty-four male Wistar rats were randomly divided into four equal number (n = 16) including training + supplement (TS), training (T), supplement (S) and control (C) group. The rats in TS and S groups received a dose of 10 mg/kg resveratrol per day via gavage. The training groups ran on a rodent treadmill 5 times per week at the speed of 10 m/min for 10 min; the speed gradually increased to 30 m/min for 60 minutes at the end of 12th week. The acute phase of exercise protocol included a speed of 25 m/min set to an inclination of 10° to the exhaustion point. Superoxide dismutase (SOD), glutathione peroxidase (GPx), catalase (CAT) activity, non-enzymatic antioxidants bilirubin, uric acid, lipid peroxidation levels (MDA) and the total antioxidant capacity (TAC) were measured after the exercise termination. The data were analyzed by using one-way ANOVA. The result showed that endurance training caused a significant increase in MDA level [4.5 ± 0.75 (C group) vs. 5.9 ± 0.41 nmol/l (T group)] whereas it decreased the total antioxidant capacity [8.5 ± 1.35 (C group) vs. 7.1 ± 0.55 mmol/l (T group)] (p = 0.001). In addition, GPx and CAT decreased but not significantly (p > 0.05). The training and t-resveratrol supplementation had no significant effect on the acute response of all variables except MDA [4.3 ± 1.4 (C group) vs. 4.0 ± 0.90 nmol/l (TS group)] (p = 0.001) and TAC [8.5 ± 0.90 (C group) vs. 6.6 ± 0.80 mmol/l (TS group)] (p = 0.004). It was concluded that resveratrol supplementation may prevent exercise-induced oxidative stress by preventing lipid peroxidation.

AbstractAluminium (Al) is a neurotoxic metal that contributes to the progression of several neurodegenerative diseases. The aim of the present study was to evaluate the protective effect of dietary eugenol supplementation against aluminium (Al)- induced cerebral damage in rats. Male Wistar rats were divided into four groups: normal controls, rats fed a diet containing 6,000 μg g-1eugenol, rats intoxicated daily with aluminium chloride (84 mg kg-1body weight) p. o. and fed either a basal diet or a eugenol-containing diet. Daily oral administration of Al for four consecutive weeks to rats significantly reduced brain total antioxidant status (TAS) (11.42±0.31 μmol g-1tissue, p<0.001) with a subsequent significant enhancement of lipid peroxidation (MDA) (32.55±1.68 nmol g-1tissue, p<0.002). In addition, Al enhanced brain acetylcholinesterase activity (AChE) (46.22±4.90 U mg-1protein, p<0.001), tumour necrosis factor alpha (TNF-α) (118.72±11.32 pg mg-1protein, p<0.001), and caspase 3 (Casp-3) (8.77±1.26 ng mg-1protein, p<0.001) levels, and in contrast significantly suppressed brain-derived neurotrophic factor (BDNF) (82.74±14.53 pg mg-1protein, p<0.002) and serotonin (5-HT) (1.54±0.12 ng mg-1tissue, p<0.01) levels. Furthermore, decreased glial fibrillary acidic protein (GFAP) immunostaining was noticed in the striatum of Al-intoxicated rats, compared with untreated controls. On the other hand, co-administration of dietary eugenol with Al intoxication restored brain BDNF (108.76±2.64 pg mg-1protein) and 5-HT (2.13±0.27 ng mg-1tissue) to normal levels, enhanced brain TAS (13.43±0.24 μmol g-1tissue, p<0.05), with a concomitant significant reduction in TNF-α (69.98±4.74 pg mg-1protein) and Casp-3 (3.80±0.37 ng mg-1protein) levels (p<0.001), as well as AChE activity (24.50±3.25 U mg-1protein, p<0.001), and increased striatal GFAP immunoreactivity, compared with Al-treated rats. Histological findings of brain tissues verified biochemical data. In conclusion, eugenol holds potential as a neuroprotective agent through its hydrophobic, antioxidant, and anti-apoptotic properties, as well as its neurotrophic ability against Al-induced brain toxicity in rats.

In the present study, we examined gross changes in the mass of whole adrenal glands and that of the adrenal cortex and medulla in mature male Wistar rats subjected to three different stress types: acute, chronic, and combined, i.e., chronic followed by acute stress. These parameters were correlated with adrenal activity as judged from serum levels of corticosterone and catecholamine, respectively, as well as with serum levels of ACTH and glucose. Under all three conditions, we observed bilaterally asymmetric and stress-type-independent hypertrophy of whole adrenals, as well as adrenal cortices and medullas. Under acute and combined stress, adrenal hypertrophy was followed by increase of adrenal hormones in the blood serum. However, under chronic stress, both cortical and medullar activities as judged from low or unaltered levels of the respective hormones and glucose were compromised and disconnected from the input signal of ACTH. Since all of the studied adrenal activities could be restored by subsequent acute stress, it is concluded that chronic isolation can be viewed as partly maladaptive stress with characteristics resembling stress resistance rather than the stress exhaustion stage of the general adaptation syndrome.

SummaryIntroduction: The liver is involved in the metabolism of xenobiotics and their metabolites and it is vulnerable to oxidative damage. Hyperglycaemia is highly implicated in the progression of diabetes mellitus, and adversely affects the liver. Though, conventional hypoglycaemic drugs may be effective in reducing blood glucose, they do not appear to be effective in attenuating the progression of diabetes and its complications.Objective: This study evaluated the ameliorative effects of Anchomanes difformis on hyperglycaemia and hepatic injuries in type 2 diabetes.Methods: Type 2 diabetes was induced in male Wistar rats with a single intraperitoneal injection of streptozotocin (40 mg/kg BW) after two weeks of fructose (10%) administration. Aqueous extract of A. difformis (200 and 400 mg/kg BW) and glibenclamide (5 mg/kg BW) were administered orally for six weeks. Blood glucose concentrations were measured. Serum levels of liver dysfunction markers (ALT, AST, and ALP), total cholesterol, triglycerides, HDL- and LDL-cholesterol were investigated. Total protein, albumin, and globulin were also assessed. Antioxidant parameters: ORAC, GSH, GSSG, SOD, CAT and FRAP were evaluated in the liver while ORAC, FRAP and lipid peroxidation were determined in the serum. Histological examination of the liver tissue was carried out.Results: Treatment with aqueous extract of A. difformis significantly (p<0.05) reduced blood glucose and reversed steatosis in the diabetic-treated rats. The antioxidant status of diabetic-treated rats was significantly (p<0.05) improved. Serum levels of liver dysfunction markers were significantly (p<0.05) reduced in diabetic-treated rats.Conclusion: The findings in this study revealed that 400 mg/kgBW Anchomanes difformis was more effective than 200 mg/kg BW in ameliorating diabetes-induced hepatopathy, however, both doses of Anchomanes difformis demonstrated more antidiabetic ability than glibenclamide. Anchomanes difformis may be a novel and potential therapeutic agent in the management of diabetes and resulted hepatic injuries.

Background: Oxidative stress may play an important role in the pathogenesis of pulmonary tuberculosis (PTB). To our knowledge there is paucity of data on the status of oxidative stress biomarkers among PTB patients in Gombe, North-eastern Nigeria. Our study was designed to evaluate the oxidative stress biomarkers in pulmonary tuberculosis patients in Gombe, North-eastern Nigeria. Aims and Objectives: To determine the serum levels of oxidative stress biomarkers among patients with pulmonary tuberculosis in Gombe metropolis, North-eastern Nigeria and to assess the correlation between the oxidative stress biomarkers in pulmonary tuberculosis patients. Materials and Methods: A cross sectional comparative study was conducted in a tertiary health care facility with 40 pulmonary tuberculosis (PTB) patients on anti-TB drugs treatment (ATT), 40 newly diagnosed PTB patients not yet on anti-TB drugs treatment (ATT-naïve) and 40 age- and sex-marched apparently healthy subjects (controls). Serum total antioxidant status (TAS), total oxidant status (TOS), malondialdehyde (MDA), nitric oxide (NO) and oxidative stress index (OSI) determined using standard techniques. Data was analysed using INSTAT® (Graph Pad Software Inc., La Jolla, CA, USA). Results: Serum levels of TOS, OSI, MDA and NO were significantly (p ˂ 0.001) increased in PTB patients (ATT and ATT-naïve) when compared with healthy individuals. Serum TAS and body mass index (BMI) were significantly (p ˂ 0.001) decreased in PTB patients when compared with healthy individuals. Serum TOS significantly correlated with serum OSI, MDA and NO in ATT-naïve PTB patients. Conclusion: This study observed an increased oxidative stress biomarkers and decreased total antioxidant status in newly diagnosed pulmonary tuberculosis patients and those on treatment. Our findings suggest that antioxidants supplementation and improved nutrition in the management of pulmonary tuberculosis patients may go a long way in preventing the oxidative onslaught and further complications in PTB patients.

Chronic exposure to stress factors contributes to the development of depression by generating excess of reactive oxygen species which leads to oxidative stress and inflammatory processes. The aim of the study was to assess the potential protective properties of α-tocopherol supplementation on the rats exposed to chronic variable stress (CVS). Male Wistar rats (50-55 days old, weighing 200-250 g) were divided into three groups (n=10): control, stressed, and stressed and receiving (+)-α-tocopherol solution in a dose of 100 mg/kg/day. Rats in the stressed groups were exposed to CVS for 40 days. Markers of redox disorders (glutathione reduced and oxidized levels, GSH/GSSG ratio, glutathione peroxidase, glutathione reductase activities, total antioxidant status, and lipid peroxidation) and inflammatory response (IL-1β, IL6, and TNF-α) were determined in the blood. Additionally, molecular biomarkers of depression (expression of Fkbp5 and Tph2) were studied in hippocampus. The biochemical analysis was inconclusive about the presence of oxidative stress in the blood of rats exposed to CVS. However, changes in all parameters suggest presence of redox equilibrium disorders. Similarly, activation of inflammatory processes was observed as a result of CVS. Molecular effects of environmental stress in hippocampus were also observed. Generally, α-tocopherol ameliorated redox equilibrium disorders, tempered inflammatory response, and protected from changes in determined molecular markers of depression.

1. The effects of dietary polychlorinated biphenyls (PCB) and excess tyrosine on serum and liver lipids, urinary ascorbic acid and catecholamines were compared in male Wistar rats.2. Serum levels of cholesterol, urinary ascorbic acid, norepinephrine, epinephrine, dopamine and histamine were significantly increased in rats given either PCB or excess tyrosine.3. The hypercholesterolaemia induced by PCB or excess tyrosine was blocked by the adrenergic α-blocker, phenoxybenzamine.4. The present results suggest causal interrelations between the hypercholesterolaemia induced by dietary PCB or excess tyrosine and the secretion of catecholamines.

Aim: The aim of this study was to assess the antioxidant properties of ethanolic and aqueous extracts of Punica granatum (Pomegranate) seed in testosterone induced benign prostate hyperplastic albino Wistar rats. Study design: This study is an interventional study. Place and Duration of Study: The experimental aspect of this study was conducted at the animal house, Department of Pharmacology, University of Port Harcourt between April and September, 2019. Methodology: Seventy (70) adult albino male wistar rats were used for this study. They were divided into 12 groups of 5 rats each and fed with commercial rat diet and clean drinking water. Aqueous and ethanolic extracts of Punica granatum seed were prepared using the maceration method. Benign Prostate Hyperplasia was induced in rats after they submitted to bilateral orchiectomy by daily injections of testosterone propionate (TP) (4 mg/kg b.wt.sc). Rats were treated with 500 or 1500 mg/kg b.wt. of aqueous or ethanoI extracts of Punica granatum seed, dutasteride or in combination. Administration of extracts was done by gavage. Plasma total oxidant status (TOS), total antioxidant status (TAS), superoxide dismutase (SOD) activity, were analyzed using sandwich ELISA Kits by Shanghai Korain Biotech Co., Ltd, China, while oxidative stress indices (OSI) were calculated. Statistical analysis was done using SPSS version 22.0 of Windows Stat Pac and p values less than 0.05 were considered statistically significant. Results: The results showed that the mean TOS, TAS, SOD and OSI for the rats in the normal control group were 1.66±0.2U/ml, 2.71±0.25U/ml, 41.8±2.9pg/ml and 0.62±0.10 respectively. After BPH induction (group 2), the values were 3.25±0.5U/ml,1.17±0.14U/ml, 23.38±2.09 pg/ml and 2.81±0.60pg/ml respectively. There were significant decreases for TOS and OSI, and significant increases for TAS and SOD when the rats where treated with lower and higher doses of both aqueous and ethanolic extracts of Punica granatum and Dutasteride. Conclusion: In conclusion, both doses of Punica granatum seed for ethanolic and aqueous extracts individually and in combination and with dutasteride markedly reduced total oxidant status, oxidative stress indices and improved the activities of antioxidant parameters like superoxide dismutase and total antioxidant status.
