The aim of the current study was to do the analysis of the total protein and its fraction in the blood samples of horses, which are involved in recreational horseback riding in the Pomeranian region (Pomeranian Voivodship, northern Poland). Thirteen healthy adult horses from the Pomeranian region in Poland (Strzelinko village, N54°30´48.0´´ E16°57´44.9´´), aged 9.5±2.4 years, including 5 Hucul ponies, 2 Thoroughbred horses, 2 Anglo-Arabian horses, and 4 horses of unknown breed, were used in the current study.Training started at 10:00 AM, lasted 1 hour, and consisted of a ride of cross country by the walking (5 min), the trotting (15 min), the walking (10 min), the trotting (10 min), the walking (5 min), the galloping (5 min), and the walking (10 min). Blood samples were taken from the jugular veins of the animals in the morning time, 90 minutes after feeding, while the horses were in the stables (between 8:30 and 10 AM), and immediately after the exercise session (between 11:00 AM and 2:00 PM). To obtain serum, the blood was collected in plain tubes without anticoagulants. Blood was stored in tubes with K3-EDTA and held on ice until centrifugation at 3,000g for 15 minutes. The plasma was removed.The total protein and its fractions were measured at +23°C by the biuret method with the use of commercially available reagents and a compact semi-automated analyzer RX Monza (Randox Laboratories LTD., UK) according to the procedures described by the manufacturer. The biuret method is the most widely used colorimetric method for the determination of the total protein concentration in serum because of its simplicity, precision, and accuracy. The absorbance of each sample was measured in duplicate.Results are expressed as mean ± S.E.M. All variables were tested for normal distribution using the Kholmogorov-Smirnov test (p>0.05). To find significant differences (significance level, p<0.05) between at the rest and after exercise, the Wilconson signed-rank test was applied to the data. All statistical analyses were performed using STATISTICA 8.0 software (StatSoft, Krakow, Poland). The total protein level in the blood of horses exhibited a non-significant increase (by 7.1%, p>0.05) immediately after exercise as compared to the resting period. Also, the albumin and globulin levels in the blood of horses were non-significantly increased by 5.9% (р>0.05) and 8.1% (р>0.05) after the training sessions. There were no significant differences in serum albumin/globulin ratio between the resting period and after exercise (0.997±0.09 vs. 0.977±0.08). The results of our current study showed that exercise has a statistically non-significant effect on the total proteins and their fractions in equine serum. The fractions and the A/G ratio were within the range of values obtained in horses in other studies. Thus, it was found that total protein and its fractions were increased in horses after training, and this increase was insignificant. This increase has a direct correlation with exercise. In this paper, it is shown that training can change the physiology and affect the biochemistry of hematobiochemical blood parameters in horses subjected to physical exertion.

Abstract We evaluated a rate colorimetric method (Beckman) for measuring total protein in cerebrospinal fluid. The automated instrument we used was Beckman's ASTRA TM. A 100-microL sample of spinal fluid is introduced into the biuret reagent in the reaction cell and the increase in absorbance at 545 nm is monitored for 20.5 s. Solid-state circuits determine the rate of alkaline biuret-protein chelate formation, which is directly proportional to the total protein concentration in the sample. The linear range of measurement is 120 to 7500 mg/L. Day-to-day precision (CV) over the range of 150 to 1200 mg/L ranged from 15.2 to 2.3%. The method was unaffected by radical alteration of the albumin/globulin ratio, but there is a positive interference in the presence of hemoglobin, a suppression in the presence of bilirubin, and no effect by xanthochromia. The method is precise, accurate, rapid, and convenient. The method was compared with the trichloroacetic acid method as performed on the Du Pont aca III, giving a correlation coefficient (r2) of 0.9693. The method is precise, accurate, rapid, and convenient.

In this study, some biochemical parameters of hair goat kids which had consumed excessive amounts of oak leaves and acorns in the autumn season and displayed toxicity symptoms during clinical inspections were evaluated. Blood samples were collected from the jugular veins of the goat kids. Glucose, AST, ALT, ALP, GGT, Na, K, Cl, cholesterol, and triglyceride levels of poisoned animals were found to be within the reported reference intervals. Creatinine, total protein, albumin, Ca, Mg and Fe levels, on the other hand, were found to be below the reference intervals. As a result, it was concluded that goats reflect the clinical findings of oak poisoning described in the literature, which courses with lowered amounts of mineral (Fe, Mg, Ca), TP, albumin and creatinine, yet without a significant change in other biochemical parameters. It can also be concluded that excessive consumption of oak by goats was capable of leading to toxicity.

This study investigate the inhibiting effect of fengycin on respiration and nutrient utilization of Penicillium expansum. The respiratory inhibition rate of the P. expansum was determined by the test of dissolved oxygen fengycin, The effect of fengycin treatment on the activity of P. expansum mitochondrial complex enzyme was detected by mitochondrial enzyme activity assay. The ability of fengycin treatment to P. expansum the utilization of total sugar and total protein was determined by DNS colorimetric method and biuret method. After fengycin treatments, the TCA pathway of respiratory metabolism in P. expansum was inhibited. Besides, fengycin could block the gene expression in P. expansum by binding P. expansum mitochondrial complex enzyme II and III related genes. Therefore, the activity of mitochondrial enzymes was affected. With the increasement of fengycin concentration, the absorption and utilization capacity of P. expansum to total sugar and total protein decreased significantly. Fengycin could inhibit the respiratory metabolism and reduce the biochemical metabolism level in P. expansum and finally caused the growth inhibition.

Fucoidan is a sulfate polysaccharide extracted from brown algae. Fucoidan has various pharmacological properties, such as anti-tumor, anti-mutagenic, anti-inflammatory, antiviral, antioxidant, anti-fibrogenic activity and anti-complementary activities Objective: This study was aimed to explore the effect of fucoidan on biochemical parameters (ALT, AST, GGT, total protein, albumin, globulin) and protein fractions in hepatotoxicity induced by CCl4 in rats. Materials and Methods: The rats used in the study were randomly divided into four groups of 8 rats each: Control group, fucoidan group, fucoidan+CCl4 group and CCl4 group. After 24 hours from the process of an eight-day experiment, blood samples were taken. The analysis of ALT, AST, GGT activities and total protein, albumin, globulin levels were done by an autoanalyser and serum protein fractions (albumin, á1- globulin, á2-globulin â- globulin, ã- globulin and A/G ratio) were electrophoretically determined. Results: In the group of fucoidan+CCl4, it was determined that the levels of AST (p>0.001), GGT (p>0.001), total protein (p>0.01), globulin (p>0.01), â-globulin % (p>0.01), ã-globulin % (p>0.001) statistically decreased compared to CCl4 group. Conclusion: It can be said that fucoidan has the property of hepatoprotectant by looking to some biochemical parameters and changes in protein fractions that examined in hepatotoxicity induced by CCl4.

The aim of the current study was the analysis of the oxidative stress biomarkers, as well as the osmotic-induced resistance of erythrocytes in mares and stallions of ponies involved in recreational horseback riding in Pomeranian regions. Ten healthy adult Hucul ponies (5 stallions and 5 mares), 5-11 years old, from Pomeranian regions in Poland (Ustka city, Pomeranian Voivodship, Poland) were used in our study. All horses participated in recreational horseback riding and were subjected to the resembling type of management. The training was continued 1 hour and included a ride of cross country by walking (10 min), trotting (15 min), walking (10 min), galloping (15 min), and walking (10 min). Blood was drawn from jugular veins of the animals in the morning, 90 minutes after feeding, and immediately after the exercise session. Marker of lipid peroxidation (2-Thiobarbituric acid reactive substances, TBARS), aldehydic and ketonic derivatives in the erythrocytes, total antioxidant capacity (TAC) level in the erythrocytes of both mares and stallions exhibited non-significant changes immediately after exercise as compared to the resting period. Both at the rest and after the training session, the levels of TBARS, aldehydic and ketonic derivatives in the erythrocyte suspensions of mares were non-significantly higher compared to stallions. In a like manner, differences of aldehydic and ketonic derivatives of oxidatively modified proteins between mares and stallions after training sessions were noted. Both at the rest and after the training session, the TAC level in the erythrocyte suspensions of stallions was non-significantly higher compared to mares. Among both mares and stallions, a non-significantly decreased erythrocyte hemolysis was observed after training sessions compared to the rest period. A comparison of erythrocyte hemolysis in mares and stallions at the rest period showed increased values of hemolysis in the stallions. After the training session, decreased hemolysis was observed in the stallions compared to mares. Efforts should be directed toward a thorough characterization of antioxidant defenses, as well as the correlation links between oxidative stress biomarkers and antioxidant defenses including age- and gender-related differences in the training programs of horses involved in the recreational horseback ridings.

ABSTRACT The aim of the present study was to evaluate variations in proteinogram occurring during the estrus cycle of animals infected or not by caprine arthritis encephalitis. Forty blood samples were collected from female goats in different phases of the estrus cycle (estrus, proestrus, metaestrus and diestrus). Samples were classified as positive (n = 5) and negative (n = 5), according to the results of the survey of antibodies against caprine arthritis encephalitis as performed by immunodiffusion in agar gel. Samples were also used in the determination of albumin, total protein and alpha, beta and gammaglobulin by means of electrophoresis and biuret method, respectively. Electrophoresis showed that estrus, proestrus, metaestrus and diestrus positive animals presented total protein mean equal to 7.96 ± 0.69, 7.67 ± 1.14, 7.77 ± 0.36, 7.48 ± 0.83; mean albumin equal to 3.22 ± 0.41, 3.11 ± 0.68, 3.30 ± 0.67, 3.28 ± 0.57; mean alphaglobulin equal to 0.70 ± 0.09, 0.70 ± 0.12, 0.69 ± 0.12, 0.61 ± 0.11; mean betaglobulin 1 equal to 0.85 ± 0.04, 0.87 ± 0.04, 0.90 ± 0.12, 0.83 ± 0.11; mean betaglobulin 2 equal to 0.77 ± 0.18, 0.68 ± 0.17, 0.60 ± 0.15, 0.54 ± 0.09; and mean gammaglobulin equal to 2.43 ± 0.47, 2.31 ± 0.54, 2.28 ± 0.49, 2.21 ± 0.54. Negative animals presented mean total protein equal 7.76 ± 1.31, 8.03 ± 1.10, 7.50 ± 0.51, 6.49 ± 1.14; mean albumin equal to 3.06 ± 0.46, 3.19 ± 0.46, 3.03 ± 0.62, 2.93 ± 0.96; mean alphaglobulin equal to 0.69 ± 0.19, 0.75 ± 0.18, 0.70 ± 0.24, 0.58 ± 0.20; mean betaglobulin 1 equal to 0.79 ± 0.03, 0.79 ± 0.23, 0.80 ± 0.15, 0.66 ± 0.16; mean betaglobulin 2 equal to 0.67 ± 0.20, 0.95 ± 0.37, 0.81 ± 0.40, 0.73 ± 0.41; and mean gammaglobulin equal to 2.35 ± 1.42; 2.42 ± 1.30, 2.23 ± 1.03, 1.92 ± 0.70, respectively. Variance analysis did not show any statistically significant differences. Animals infected by caprine arthritis encephalitis (CAE) did not present any changes in proteinogram, regardless of the phase of the estrus cycle they were in.

Objectives. Preeclampsia is a pregnancy specific syndrome. Studies have shown that preeclampsia has multiorgan dysfunction effects. This study evaluated biomarkers of renal and liver function among preeclamptic Nigerian women. Study Design. This was a cross-sectional study conducted among 49 preeclamptic women and 50 normotensive healthy pregnant women. Method. The baseline data comprising age, gestational age, and blood pressure were obtained. Venous blood and spot urine samples were collected from each participant. Plasma obtained from blood samples taken into lithium heparinized vacutainer bottles was assayed for electrolytes, urea, creatinine, total protein, albumin, and uric acid, while sera samples from blood samples taken into serum separation tube- (SST-) gel vacutainer were assayed for aspartate transaminase and alanine transaminase using ion selective electrode technique and Cobas autoanalyzer. Spot urine samples were assayed for protein and creatinine using Pyrogallol’s reagent and Jaffe’s methods, respectively. Microalbuminuria (protein/creatinine ratio) was generated from spot urine protein and creatinine data. Result. The plasma sodium, total protein, and albumin in preeclamptic group were significantly decreased (p<0.05) when compared with control. There was statistically significant increase (p<0.05) in microalbuminuria, plasma potassium, urea, creatinine, uric acid levels, serum AST, and ALT activities in preeclamptic group. A positive association (p<0.05) between alanine aminotransferase and biomarkers of renal function was observed. Conclusion. Preeclampsia has deleterious effects on renal and liver function as shown by alteration of these parameters.

Serum profile is generally considered as comprehensive data which directly reflected animal health and their potential resistance to environmental, nutritional and pathological stress. The present study aimed to provide physiological reference values for selected biochemical parameters in Noi chickens, a famous Vietnamese native breed, at 56 days old. The collected blood samples of 355 Noi birds (164 males and 191 females) were used to evaluate biochemical serum profile. Glucose, total protein, albumin, globulin, albumin/globulin ratio, triglycerides, cholesterol, creatinine, and uric acid in the serum were measured. The variation in these values arising from different sexes was also investigated. The results indicated that there were no differences (P>0.05) in the assessed indices between male and female Noi chickens. In overall for the whole group, these values were obtained as 250.86 mg/dL, 7.34 g/dL, 3.40 g/dL, 3.94 g/dL, 1.75, 148.54 mg/dL, 190.86 mg/dL, 0.44 mg/dL, and 1.44 mg/dL, respectively. The relationship among the biochemical parameters showed relatively low coefficients (r=0.79 in maximum). The females obtained higher correlation coefficients between levels of total protein and glucose (r=0.42) as well as between total protein and globulin (r=0.79), compared to the males. It was concluded that the male and female chickens performed similar values of serum indices. The contribution of this study might expand the knowledge on the biochemical profile and improved breeding strategies of Vietnamese indigenous Noi chickens.
