Hypertriglyceridemia is one of the risk factors for cardiovascular disease and acute pancreatitis (Pejic and Lee, 2006; Brahm and Hegele, 2013; Afari et al., 2015). One way to treat it by using the apple vinegar (Malus sylvestris Mill) of manalagi varieties which contained high antioxidants and acetic acid as well as more durable than the fresh fruit. The study aimed to know the effect of the Apple vinegar of manalagi varieties (Malus sylvestris Mill) to blood triglyceride levels of white male rats (Rattus norvegicus) wistar strain with high-fat diet and induced by alloxan. The study used the post test only control group design, with 24 rats samples which were divided into three groups. The negative control group (K0) only received standard diet for 24 days, the positive control group (K1) received a high-fat diet for 7 days and induced by alloxan with doses of 90 mg/kgBW of rats, and treatment group (K2) was given a high-fat diet for 7 days, induced by alloxan and received the apple vinegar (Malus sylvestris Mill) of manalagi varieties with a dose of 0,27 ml/100 g BW of rats for 14 days. The results by using one way ANOVA showed the significance value of the data p = 0,974 (p>0,05) so that there is no significant difference of the mean of blood triglycerides levels between K0, K1 and K2. This study concluded that apple vinegar (Malus sylvestris Mill) of manalagi varieties can not significantly lower the blood triglyceride levels of white male rats (Rattus norvegicus) wistar strain with high-fat diet and induced by alloxan. Further study about apple vinegar of manalagi varieties is required to define its effects.  Keywords—: High-Fat Diet, Alloxan, Triglyceride, Apple Vinegar (Malus sylvestris Mill) of Manalagi Varieties

The objective of this study was to investigate the antioxidant capability and preventive effect of probiotics on high fat diet induced oxidative damage. Thirty male rats were randomly divided into three groups. The control group consumed a normal standard diet (5% fat, w:w), the high fat diet group received a high fat diet (20% fat, w:w), and probiotics group received a high fat diet supplemented with 2% probiotics. After 6 weeks, antioxidant capability of liver was determined and DNA fragmentation of liver cell was determined using a comet assay. The results showed that high fat diet could induce oxidative stress, shown as significant increases in lipid peroxidation and nitric oxide free radical, significant decrease in activities of superoxide dismutase and glutathione peroxidase, the percentage of DNA fragmentation was markedly increased, compared with control group. These alterations were significantly reversed in probiotics supplemented group and had no significant difference in antioxidant capability, lipid peroxidation and DNA damage compared with control group. Present observation indicated the probiotics had antioxidant property, which could scavenge free radical and enhance antioxidant defense system, and had the positive protective function on oxidative damage.

Objective: To compare the number of apoptosis, necrosis and viability of germinal epithelial cells in the contralateral testicle (CT) and plasma testosterone (PT) levels in white male rats which administered Nifedipine after unilateral testicular torsion (TT) compared with control group. Material & Methods: This research was an experimental study using male white rats (Rattus Norvegicus, Wistar strain) aged 10-12 weeks and body weight 150-200 gram. A total of 30 rats were then randomly divided into 5 groups (n=6) which were negative control group (KN), positive control group (KP1 and KP2) and Nifedipine-administered group (N1 and N2). Each group performed unilateral left side torsion of testicular of 1080o anticlockwise except the KN group. There was 4-hour ischemic duration in the KP1 and N1 groups while 10 hours in the KP2 and N2 groups. Administration of Nifedipine 30 minutes before detorsion by intraperitoneal injection dosed 100 μg/kgBW. All groups performed right orchidectomy and plasma blood sampling. Measurement of apoptosis, necrosis and viability of germinal epithelial cells in the CT using flowcytometry. Measurement of PT levels using Enzyme-Linked Immunosorbent Assay (ELISA). Results: The number of apoptosis, necrosis and viability of contralateral testicular germinal epithelial cells and PT levels in the KN group compared with KP1 and KP2 groups were significantly different (p<0.05). There was no significant discrepancy in apoptosis (p>0.05) in KP1 group compared with N1 group, as well as in KP2 group compared with N2 groups. The number of necrosis, viability of germinal epithelial cells in the CT and PT level in KP1 group in compared with N1 group, as well as in KP2 group in compared with N2 group were significantly different (p<0.05). Conclusion: Nifedipine administration prior to testicular detorsion can maintain cell viability and decrease the amount of necrotic germinal epithelial cells in the CT and prevent the decrease in PT levels after unilateral TT.

Background: Statins can reduce reproductive damage induced by obesity or high-fat diet (HFD), but the specific regulatory mechanisms are largely unknown. Since mTOR/p70s6k sinaling promotes spermatogonia proliferation and spermatogenesis, we hypothesized that this pathway will be involved in the protective effects of statin in HFD-induced reproductive dysfunction. Methods: Male Sprague Dawley rats (3 weeks old) were randomly divided into a control group (standard diet), HFD group, and a fluvastatin group (HFD + fluvastatin at 6mg/kg, once daily by oral gavage). After 8 weeks, body weight was obtain and rats were sacrificed. Weights of the testes, gross morphology, sperm parameters, circulating levels of sex hormones, lipid levels, and tissue mTOR, p-P70s6k were measured. Another set of male rats were treated with rapamycin or vehicle. Flow cytometry was used to detect the spermatogonia marker c-kit and cell cycle. p-P70s6k expression was analyzed by Western blot. Results: HFD not only results in rat obesity but also leads to spermatogenetic damage and fluvastatin was able to partially block the effects of HFD. Fluvastatin also partially reversed the suppression of mTOR and p-p70s6k expresson. Conclusion: Our data suggest that fluvastatin has protective effects on reproductive function in obese male rats most probably through enhanced signaling of mTOR.

Abstract Nowadays, high-fat foods are eaten in most societies, which causes memory impairment and anxiety through the oxidative stress pathway. Sesame oil (SO) has potential antioxidant properties. Therefore, the effects of sesame oil on memory impairment and anxiety caused by a high-fat diet (HFD) in male rats were investigated. Eighty male Wistar rats were divided into eight groups (n = 10): control (standard diet; SD), the HFD, SD + SO (0.5, 1, or 2 ml/kg; once/day, gavage), and HFD + SO (0.5, 1, or 2 ml/kg; once/day, gavage) groups. All diets were given to the animals for three months. Finally, behavioral and oxidative stress parameters were measured. The step-through latency of retention test in SD + SO (0.5 or 1 ml/kg) groups increased more than the control group. Also, the Barnes test on training days revealed that the latency time to find the target hole increased in the HFD group compared with the control group. Moreover, the time spent on the open arms in the SD + SO (0.5 ml/kg) group improved remarkably than the control group. Total oxidant (TOS) level in the HFD + SO (0.5, 1, and 2 ml/kg) groups was lower than the HFD group. The level of total antioxidant capacity (TAC) in the SD + SO (2 ml/kg) group was higher than the SD + SO (0.5 ml/kg) group and the amount of thiol in the HFD group decreased compared with the control group. These findings suggest that the positive effects of SO on memory and anxiety are probably due to its antioxidant properties and the elimination of free radicals.

Alcohol (ethanol) has long been used as liquor. In some areas in Indonesia, the use of ethanol is an insaperable part of family rites and traditional festivities.  However, due to its serious consequences, alcoholism becomes a chronic social problem. Ethanol effects the metabolism in body organs and tissues, including male reproductive organ. The objective of this study was to prove that ethanol comsumption may reduce the weight of testis. This study used Posttest Only Group Design with experimental animals of 30 male rats (Wistar strain Rattus norvegicus) that were subjected in five types of treatment, i.e,(1) feeding with aquades 2ml/day as control group (2) feeding with 10% 1 gr/Kg/day of ethanol, (3) feeding with 10% 3gr/Kg/day of ethanol, (4) feeding with 30% 1 gr/Kg/day of ethanol and (5) feeding with 30% 3 gr/Kg/day of ethanol. Treatment was given daily for 45 days. At the end of the study, on the day 46th, the rats was sacrified for examination. Sample of testis were taken for weight examination.  Data were analysed using Anova and when the difference was found, the analysis was followed with Least significant Difference at the level of significance of 95%. The result of this study showed that ethanol reduced the weight of testis compared to control. The results of Anova  test revealed that higher dosage and concentrations of ethanol resulted in significantly different decrease of the testis’s weight (p<0,05) compared to other dosage and other concetration. In conclusion, ethanol has effect in the reduction of the weight of the testis.Key words: ethanol, male reproduction, Rattus norvegicus, weight of testis.

This study aims to analyze the effect of combination of motor vehicle particular matter exposure and high-fat diet in kidney histopathology, creatinine levels, and MDA levels in Wistar rats. This study used a posttest-only control group design. Eighteen healthy male Wistar rats were divided into three groups. The intervention groups received motor vehicle fume exposure for 100 s with normal diet (X1) or high-fat diet (X2), and the control group received no exposure (C). Data analysis was processed with a SPSS 25.0 computer program by using the one-way ANOVA test followed by post hoc LSD. The degree of kidney histopathological damage showed significant differences between the X1 and X2 groups when compared with the control group (p < 0.05). The results of the creatinine level examination found a significant difference between the X2 and C groups (p < 0.05) and the treatment groups X1 and X2 (p < 0.05). The results of kidney MDA level examination showed a significant difference between the treatment groups (X1 and X2) and the control group (p < 0.05). The combination of particular matter of motor vehicle fumes exposure and high-fat diet could induce kidney damage through histopathological change and increased creatinine levels and kidney MDA levels in Wistar rats.
