The Protective Effects of Probiotics on High Fat Diet-Induced Oxidative Damage Using a Comet Assay in Rats

The objective of this study was to investigate the antioxidant capability and preventive effect of probiotics on high fat diet induced oxidative damage. Thirty male rats were randomly divided into three groups.The control group consumed a normal standard diet (5% fat, w:w), the high fat diet group received a high fat diet (20% fat, w:w), and probiotics group received a high fat diet supplemented with 2% probiotics. After 6 weeks, antioxidant capability of liver was determined and DNA fragmentation of liver cell was determined using a comet assay. The results showed that high fat diet could induce oxidative stress, shown as significant increases in lipid peroxidation and nitric oxide free radical, significant decrease in activities of superoxide dismutase and glutathione peroxidase, the percentage of DNA fragmentation was markedly increased, compared with control group. These alterations were significantly reversed in probiotics supplemented group and had no significant difference in antioxidant capability, lipid peroxidation and DNA damage compared with control group. Present observation indicated the probiotics had antioxidant property, which could scavenge free radical and enhance antioxidant defense system, and had the positive protective function on oxidative damage.

Hypertriglyceridemia is one of the risk factors for cardiovascular disease and acute pancreatitis (Pejic and Lee, 2006; Brahm and Hegele, 2013; Afari et al., 2015). One way to treat it by using the apple vinegar (Malus sylvestris Mill) of manalagi varieties which contained high antioxidants and acetic acid as well as more durable than the fresh fruit. The study aimed to know the effect of the Apple vinegar of manalagi varieties (Malus sylvestris Mill) to blood triglyceride levels of white male rats (Rattus norvegicus) wistar strain with high-fat diet and induced by alloxan. The study used the post test only control group design, with 24 rats samples which were divided into three groups. The negative control group (K0) only received standard diet for 24 days, the positive control group (K1) received a high-fat diet for 7 days and induced by alloxan with doses of 90 mg/kgBW of rats, and treatment group (K2) was given a high-fat diet for 7 days, induced by alloxan and received the apple vinegar (Malus sylvestris Mill) of manalagi varieties with a dose of 0,27 ml/100 g BW of rats for 14 days. The results by using one way ANOVA showed the significance value of the data p = 0,974 (p>0,05) so that there is no significant difference of the mean of blood triglycerides levels between K0, K1 and K2. This study concluded that apple vinegar (Malus sylvestris Mill) of manalagi varieties can not significantly lower the blood triglyceride levels of white male rats (Rattus norvegicus) wistar strain with high-fat diet and induced by alloxan. This study aims to analyze the effect of combination of motor vehicle particular matter exposure and high-fat diet in kidney histopathology, creatinine levels, and MDA levels in Wistar rats. This study used a posttest-only control group design. Eighteen healthy male Wistar rats were divided into three groups. The intervention groups received motor vehicle fume exposure for 100 s with normal diet (X1) or high-fat diet (X2), and the control group received no exposure (C). Data analysis was processed with a SPSS 25.0 computer program by using the one-way ANOVA test followed by post hoc LSD. The degree of kidney histopathological damage showed significant differences between the X1 and X2 groups when compared with the control group (p < 0.05). The results of the creatinine level examination found a significant difference between the X2 and C groups (p < 0.05) and the treatment groups X1 and X2 (p < 0.05). The results of kidney MDA level examination showed a significant difference between the treatment groups (X1 and X2) and the control group (p < 0.05). The combination of particular matter of motor vehicle fumes exposure and high-fat diet could induce kidney damage through histopathological change and increased creatinine levels and kidney MDA levels in Wistar rats.

High-fat diet (HFD) consumption has been linked to dyslipidemia, low-grade inflammation and oxidative stress. This study investigated the effects of a mixed formulation with Limosilactobacillusfermentum 139, L. fermentum 263 and L. fermentum 296 on cardiometabolic parameters, fecal short-chain fatty acid (SCFA) contents and biomarkers of inflammation and oxidative stress in colon and heart tissues of male rats fed an HFD. Male Wistar rats were grouped into control diet (CTL, n = 6), HFD (n = 6) and HFD with L. fermentum formulation (HFD-Lf, n = 6) groups. The L.fermentum formulation (1 × 109 CFU/mL of each strain) was administered twice a day for 4 weeks. After a 4-week follow-up, biochemical parameters, fecal SCFA, cytokines and oxidative stress variables were evaluated. HFD consumption caused hyperlipidemia, hyperglycemia, low-grade inflammation, reduced fecal acetate and propionate contents and increased biomarkers of oxidative stress in colon and heart tissues when compared to the CTL group. Rats receiving the L. fermentum formulation had reduced hyperlipidemia and hyperglycemia, but similar SCFA contents in comparison with the HFD group (p < 0.05). Rats receiving the L. fermentum formulation had increased antioxidant capacity throughout the colon and heart tissues when compared with the control group. Administration of a mixed L. fermentum formulation prevented hyperlipidemia, inflammation and oxidative stress in colon and heart tissues induced by HFD consumption.

It has been proposed that there is a correlation between high-fat diet (HFD), oxidative stress and decreased γ-aminobutyric acid (GABA) levels, but this has not been thoroughly demonstrated. In the present study, we determined the effects of strawberry extract intake on the oxidative stress and GABA levels in the frontal cortex (FC) of obese rats. We observed that an HFD increased lipid and protein oxidation, and decreased GABA levels. Moreover, UV-irradiated strawberry extract (UViSE) decreased lipid peroxidation but not protein oxidation, whereas non-irradiated strawberry extract (NSE) reduced protein oxidation but not lipid peroxidation. Interestingly, NSE increased GABA concentration, whereas UViSE was not as effective. In conclusion, our results suggest that an HFD increases oxidative damage in the FC, whereas strawberry extract intake may ameliorate the disturbances associated with HFD-induced oxidative damage.

Abstract Diet-induced obesity (DIO) reduces the ability of centrally administered insulin to reduce feeding behavior and also reduces the transport of insulin from the periphery to the central nervous system (CNS). The current study was designed to determine whether reversal of high-fat DIO restores the anorexic efficacy of central insulin and whether this is accompanied by restoration of the compromised insulin transport. Adult male Long-Evans rats were initially maintained on either a low-fat chow diet (LFD) or a high-fat diet (HFD). After 22 weeks, half of the animals on the HFD were changed to the LFD, whereas the other half continued on the HFD for an additional 8 weeks, such that there were 3 groups: 1) a LFD control group (Con; n = 18), 2) a HFD-fed, DIO group (n = 17), and 3) a HFD to LFD, DIO-reversal group (DIO-rev; n = 18). The DIO reversal resulted in a significant reduction of body weight and epididymal fat weight relative to the DIO group. Acute central insulin administration (8 mU) reduced food intake and caused weight loss in Con and DIO-rev but not DIO rats. Fasting cerebrospinal fluid insulin was higher in DIO than Con animals. However, after a peripheral bolus injection of insulin, cerebrospinal fluid insulin increased in Con and DIO-rev rats but not in the DIO group. These data provide support for previous reports that DIO inhibits both the central effects of insulin and insulin's transport to the CNS. Importantly, DIO-rev restored sensitivity to the effects of central insulin on food intake and insulin transport into the CNS.

Abstract BackgroundOptimization of experimental conditions in streptozotocin induced diabetic model in Sprague Dawley (SD) rats to evaluate the stability of the model.MethodsMale and female SD rats were randomly divided into control group, STZ 45 group (STZ: 45 mg / kg), STZ 65 group (STZ: 65 mg / kg), STZ 85 group (STZ: 85 mg / kg), high fat diet with STZ 45 group (STZ: 45 mg / kg), high fat diet with STZ 65 group (STZ: 65 mg / kg), high fat diet with STZ 85 group (STZ: 85 mg / kg). N = 6 in each group. The changes of body weight and blood glucose were observed dynamically.ResultsThere was no significant difference in blood glucose or body weight between the STZ 45 group and the control group in both male and female rats, whether or not they were on a high-fat diet. However, there were significant differences in blood glucose between the high-dose STZ group and the control group in both male and female rats, regardless of whether the rats were on a high-fat diet or not (P < 0.05 or P < 0.01). Compared with the control group, there were significant differences in blood glucose levels (P < 0.05 or P < 0.01) and higher blood glucose levels in the male rats fed with the normal diet than that in those fed with the high-fat diet.ConclusionsIn this study, male rats fed with ordinary feed and injected STZ dose of 65 mg / kg were the most stable and ideal diabetic rat.

Purpose: This study was aimed to evaluate the antioxidant and anti-inflammatory effects of vitamin D and Simvastatin (SIM) on a high-fat diet (HFD) induced-obese rats. Methods: 40 adult male rats were divided into four groups: control group, HFD, HFD + vitamin D, and HFD + SIM for 14 weeks. Vitamin D or SIM supplementation was done for the last 6 weeks. Vitamin D dosage was 500 IU/kg, while SIM dosage was 10 mg/kg. Interleukin-6 (IL-6) concentration and markers of oxidative stress including malondialdehyde (MDA), superoxide dismutase (SOD), glutathione peroxidase (GPx), and reduced glutathione(GSH) concentrations in serum were determined using ELISA kits and spectrophotometry methods, respectively. Results: Treatment with vitamin D or SIM could significantly reduce IL-6 and MDA and increases SOD, GPx activities, and GSH levels. Oxidative stress can result not only from increased ROS production but also from dysfunctional antioxidant defenses. Conclusion: From the experimental results, it was observed that SIM and vitamin D could attenuate oxidative stress and inflammation markers associated with obesity.

Background Rasbora agrotynea (local name : Seluang ) is a fish native to Sumatera, Malaya and Borneo. This fish has a potential as a local wisdom for supplementation of omega 3 and omega 6. In Sumatera, Seluang fish was used as traditional food.   Aim of Study Aim of this study to explore the efficacy of seluang fish oil in body weight, lipid profile, adiponectin and leptin level in Wistar Rats-Induced High Fat Diet   Methods This study was an experimental study , pre-post test with control group design. The sample in this study was  30 male rats, 8 weeks old, weight 150-200 gram. Rats were given high fat diet and seluang fish oil  at dose of 0,1 , 0,2  and 0,4 mL/200 gr BW/day for 2 weeks. Seluang fish oil was extracted by distilation methods. The results of this study were assayed by SPSS 18.   Results Seluang fish oil 0,4 mL/200 gr BW  was more potent to reduce body weight gain, triglyseride level,  leptin and increase adiponectin level than seluang fish oil 0,1 mL/200 gr BW, 0,2 mL/200 gr BW, negative control and positive control.   Conclusion Seluang fish oil had a potention to reduce body weight, triglyceride, leptin and increase adiponectin level.   Keywords: Seluang fish Oil – body weight- triglyserida – leptin - adiponectin

Abstract. The effects of instant cooked rice made from a combination of white rice and pigmented giant embryonic Keunnunjami rice, in comparison with those of instant regular white or brown rice and instant non-pigmented giant embryonic brown rice, on the glucose metabolism and antioxidant defense status in high-fat diet-fed mice were investigated. 56 male C57BL/6N mice were randomly divided into 7 dietary groups: normal control, high fat (23 %, HF), and HF supplemented with normal white (HF + NW) or brown rice (HF + NB), non-pigmented giant embryonic rice (HF + GB), and white rice with 8 % Keunnunjami (HF + KJ8) and 18 % Keunnunjami (HF + KJ18). After 7 weeks, HF mice showed marked increases in blood glucose (156 mg/dL), plasma insulin (12.1 mg/mL), and lipid peroxidation, and a significant decrease in hepatic glycogen (14.2 mg/g) relative to the control group (p < 0.05). However, addition of instant NB, GB, KJ8, andKJ18) rice suppressed this high-fat diet-induced hyperglycemia and oxidative stress through altering glucose-regulating enzymes (glucokinase, glucose-6-phosphatase, and phosphoenolpyruvate carboxykinase) and activation of antioxidant enzymes (superoxide dismutase, glutathione peroxidase, catalase, glutathione reductase, and paraoxonase). Compared with HF mice, HF + KJ8 and HF + KJ18 groups exhibited significantly lower glucose (139–141 mg/dL), insulin (10.6–10.9 mg/mL), and lipid peroxidation and higher glycogen (15.3–16.4 mg/g) (p < 0.05). The hypoglycemic and antioxidant effects of instant KJ8 and KJ18 rice were generally comparable to those of instant NB and GB rice. These findings illustrate that instant rice made from white rice with 8 % Keunnunjami rice may be useful as a functional food with therapeutic potential against hyperglycemia and oxidative damage.

Background: Xuezhikang capsule, which contains cholesterol synthase inhibitors and a large number of natural statins, is put in the clinical application of lipid-lowering and so on. However, the specific use of dose, lipid-lowering effect and the relationship between metabolites are to be further studied. Introduction: Metabonomics is the study of the relationship between the change of quantity and physiological changes from metabolites. At present metabolomics has been widely used in drug development and testing. In this study, we developed a metabolomic method based on gas chromatography-mass spectrometry (GC-MS) to find out hyperlipemia-related substances, and study the lipid-lowering mechanism of Xuezhikang. Method: Fifty SD rats (220 ± 20 g) were given high-fat diet. After four-weeks modeling, they were randomly divided into semi-control group, high fat group, simvastatin intervention group and Xuezhikang intervention group (0.23, 0.69, 1.15 mg/kg, low, medium, high), each dosage in eight rats. The control group (rest eight rats) were given normal diet, and no specific treatment. The rats were sacrificed at the end of the experiment. Result: The biochemical and body weight indexes of the normal control group and the high fat group were significantly different (P <0.05), which indicated that the model of hyperlipidemia was established success. There was significant difference (P <0.05) between Xuezhikang intervention group and high fat control group (P <0.05), and hyperlipemia metabolomics related markers, oxalic acid, butyric acid, mannitol, glucose, glucuronic acid were found. Glucuronic acid and non-binding bilirubin combined with bilirubin, combined with some of the liver harmful substances, play a detoxification effect. Conclusion: The results of metabonomics showed that the high fat group and the control group were significant difference. Mannose, glucose content is relatively stable, lipid metabolism in high-fat group stearic acid, palmitic acid levels decreased, suggesting that high-fat diet disorders rat body lipid metabolism. It is worth mentioning that the experimental evaluation of rats such as biochemical indicators and pathological results are prompted to model success, Xuezhikang intervention effect is more significant, consistent with the expected.
