This study aims to obtain the bacteria of Pseudomonas from the land of TPA Air Sebakul Bengkulu City as a plastic degrading agent and measure the ability of bacteria to degrade Low Density Polyethylene (LDPE) and Oxium plastic. The research was conducted from February to August 2019, at the Laboratory of Biology and Chemistry Learning, Faculty of Teacher Training and Education, University of Bengkulu. Air Sebakul landfill samples were taken at the coordinate point 3o49 '27.8 "S 102o20 '48.4" E. Isolation of plastic degrading bacteria using selective media King's B Agar added with 2% Polyethylen Glycol (PEG) to test the ability to develop bacterial isolates in plastic-based media. The steps of this research are bacterial isolation, bacterial purification and bacterial selection, macroscopic and microscopic identification of bacteria (Gram staining) and plastic biodegradation test with Mineral Salt Agar (MSM) media. Determination of the% weight loss of LDPE and oxium plastics in the biodegradation process was carried out for 30 days with time variations of 10, 20 and 30 days. The results of bacterial isolation based on morphological characteristics and gram staining test of P-1 bacterial isolate have similarities with Pseudomonas aeruginosa. So that the P-1 bacterial isolate is thought to be a Pseudomonas aeruginosa bacterium. The bidoegradation of LDPE and oxium plastics with isolates of P-1 bacteria for 10, 20 and 30 days respectively was able to degrade oxium plastics by 2.43, 5.17 and 9.86% while LDPE plastics by 1.13, 2 and 1 , 17%.

MICP (Microbially Induced Calcite Precipitation) is a new biological method in soil stabilization. This cheap and eco-friendly technique improves strength parameters of the ground such as shear strength and decreases the permeability of gravelly and sandy soil. There are variety of microorganisms that can be used in calcite precipitation. The most popular method is precipitation of calcium carbonate by bacteria. The main purpose of the article is to present the results from Gram staining of bacteria isolated from construction sites, which is the first step of their identification. Gram’s method allows to find out which morphological groups of bacteria are adapted to conditions present in soil from construction sites and therefore are potentially able to produce calcite. The article describes the methodology of isolation, staining and determination of morphological types of bacteria.

The quality of food that is good in bacteriological, chemical and physical must always be maintained in order to avoid diseases or health problems. Healthy and safe food is an important factor to improve the standard of public health. Seblak is a ready-to-eat Indonesian food which until now has never been carried out research about what kinds of bacteria in it. Seblak is a food made from raw crackers which is then deliberately soaked using hot water to have a chewy texture. This study aims to determine what types of bacteria are contained in seblak, through the process of isolation and identification in the macroscopic, microscopic and biochemical way. This study used 5 samples, which was repeated 5 times for each sample. Bacteria were isolated from the sample using spread plate techniques and observed by growing colonies on the plate. Each different colonies was observed microscopically through gram staining and endospore staining. To strengthen the data, biochemical tests were also carried out, biochemical tests that have been done in this study were the oxidase test, catalase test, and indole test. The results of the study showed that the bacteria that were found from the samples are in the genus of Bacillus sp. because they show the morphological characteristics of the colonies that form concentric circles, meanwhile, microscopic observations show morphological characteristics of cells in the form of gram-negative bacilli and have the endospores.

<p>Antang Furniture Center in Makassar is one of the wood industries and is still traditionally. The wood processing produces waste containing cellulose that is difficult to degrade. This study aims to isolate and observe the characteristics of cellulose degrading bacteria. This research was conducted at the Microbiology Laboratory, Faculty of Mathematics and Natural Sciences, Hasanuddin University, Makassar from March 2019 to November 2019. The method used was an experimental laboratory consisting of <em>Carboxyl Methyl Cellulose</em> (CMC) media for bacterial selection and characterization including microscopic morphology with Gram staining and morphology macroscopic with colony structure. The results showed that cellulose degrading bacteria were characterized by the formation of clear zones formed on CMC media in petri dishes. Based on macroscopic and microscopic morphological characteristics, eleven types of cellulose degrading bacteria isolates were obtained. The isolates obtained can be a source of bacterial isolates for further application in wood waste or other wastes that contain of cellulose.</p>

Laboratory waste produced unique characteristics, contrast to waste produced by industrial activities. Material waste that comes from the laboratory has greater variety of waste types; although the amount of material discarded is not many. The research objective was to obtain bacterial isolates that were able to survive in laboratory waste as potential waste-degrading bacteria. Research method is observartional laboratory with isolate reaction testing that was detected by the ability to degrade starch, cellulose, proteins and non-organic compounds. The sampling method was purposive sampling. The stages in this study were divided into two; first, the manufacture of pure cultures from the inoculants previously diluted, then microscopic observations. The second, identification and biochemical test according to Bergey's Manual of Bacteriology Determination. Bacteria were rejuvenated on medium nutrient so that the isolates were obtained twenty four hours old. Then an examination was carried out include Gram staining. Enzymatic test of amylase, protease, and cellulose, and biochemical test to identify microbes that degrade chemical compounds includes; oxidase test, motility, nitrate, lysine, ornithine, H2O, Glucose, Mannitol, xylose, ONPG, Indole, urease, V-P, citrate, TDA. The results of the study were found Pseudomonas stutzeri, Proteus mirabilis, and Pseudomonas aeruginosa. Isolates that have an amylolytic index are C1, C2 and O7 namely Pseudomonas stutzeri, Proteus mirabilis, and Pseudomonas aeruginosa. The resulting index was C1 = 0.45, C2 = 0.65 and O7 = 0.87. Limbah bahan yang berasal dari laboratorium memiliki jenis sampah yang lebih banyak, meskipun jumlah bahan yang dibuang tidak banyak. Tujuan penelitian adalah untuk memperoleh isolat bakteri yang mampu bertahan hidup di dalam limbah laboratorium sebagai bakteri pengurai limbah potensial. Metode penelitian adalah observasional laboratorium dengan melakukan tes reaksi isolat untuk mengetahui kemampuan degradasi pati, selulosa, protein dan senyawa non-organik. Teknik pengambilan sampel adalah purposive sampling. Tahapan dalam penelitian ini dibagi menjadi dua yaitu pertama, pembuatan kultur murni dari inokulan yang sebelumnya diencerkan, kemudian pengamatan mikroskopis. Kedua, identifikasi dan uji biokimia sesuai dengan Manual Penentuan Bakteriologi Bergey. Bakteri diremajakan pada nutrisi sedang sehingga isolat diperoleh dalam dua puluh empat jam. Uji pemeriksaan adalah pewarnaan Gram. Selanjutnya uji enzimatik amilase, protease, dan selulosa, dan uji biokimia untuk mengidentifikasi mikroba yang mendegradasi senyawa kimia meliputi; uji oksidase, motilitas, nitrat, lisin, ornithine, H2O, Glukosa, Mannitol, xilosa, ONPG, Indole, urease, V-P, sitrat, TDA. Hasil penelitian ditemukan Pseudomonas stutzeri, Proteus mirabilis, dan Pseudomonas aeruginosa. Isolat yang memiliki indeks amilolitik adalah C1, C2 dan O7 yaitu Pseudomonas stutzeri, Proteus mirabilis, dan Pseudomonas aeruginosa. There is an information how to identify hydrocarbon degrading bacteria for bioremediation of marine oil spill. We have Bioremediation treatment for degradation of oil spill on Pari island and need two kind of experiment there are tanks experiment (sampling 0 to 90 days) and semi enclosed system (sampling 0 to 150 days). Biostimulation with nutrients (N and P) was done to analyze biodegradation of hydrocarbon compounds. Experiment design using fertilizer Super IB and Linstar will stimulate bacteria can degrade oil, n-alkane, and alkane as poly aromatic hydrocarbon. The bacteria communities were monitored and analyzed by Denaturing Gradient Gel Electrophoresis (DGGE) and Clone Library; oil chemistry was analyzed by Gas Chromatography Mass Spectrometry (GCMS). DNA (deoxyribonucleic acid) was extracted from colonies of bacteria and sequence determination of the 16S rDNA was amplified by primers U515f and U1492r. Strains had been sequence and had similarity about 90-99% to their closest taxa by homology Blast search and few of them suspected as new species. The results showed that fertilizers gave a significant effect on alkane, PAH and oil degradation in tanks experiment but not in the field test. Dominant of the specific bacteria on this experiment were Alcanivorax, Marinobacter and Prosthecochloris. Keywords: Bioremediation, Biostimulation, DGGE, PAH, Pari Island

AbstractThis study deals with the determination of new natural fibers extracted from the Corypha taliera fruit (CTF) and its characteristics were reported for the potential alternative of harmful synthetic fiber. The physical, chemical, mechanical, thermal, and morphological characteristics were investigated for CTF fibers. X-ray diffraction and chemical composition characterization ensured a higher amount of cellulose (55.1 wt%) content and crystallinity (62.5%) in the CTF fiber. The FTIR analysis ensured the different functional groups of cellulose, hemicellulose, and lignin present in the fiber. The Scherrer’s equation was used to determine crystallite size 1.45 nm. The mean diameter, specific density, and linear density of the CTF fiber were found (average) 131 μm, 0.86 g/cc, and 43 Tex, respectively. The maximum tensile strength was obtained 53.55 MPa for GL 20 mm and Young’s modulus 572.21 MPa for GL 30 mm. The required energy at break was recorded during the tensile strength experiment from the tensile strength tester and the average values for GL 20 mm and GL 30 mm are 0.05381 J and 0.08968 J, respectively. The thermal analysis ensured the thermal sustainability of CTF fiber up to 230 °C. Entirely the aforementioned outcomes ensured that the new CTF fiber is the expected reinforcement to the fiber-reinforced composite materials.

AbstractIn the present work, a novel continuous flow system (CFS) is developed for the preconcentration and determination of Cr (VI) using Pseudomonas aeruginosa static biomass immobilized onto an effective and low-cost solid support of powdered eggshells. A mini glass column packed with the immobilized biosorbent is incorporated in a CFS for the preconcentration and determination of Cr (VI) from aqueous solutions. The method is based on preconcentration, washing and elution steps followed by colorimetric detection with 1,5-diphenyl carbazide in sulphuric acid. The effects of several variables such as pH, retention time, flow rate, eluent concentration and loaded volume are studied. Under optimal conditions, the CFS method has a linear range between 10 and 100 μg L-1 and a detection limit of 6.25 μg L-1 for the determination of Cr (VI). The sampling frequency is 10 samples per hour with a preconcentration time of 5 mins. Furthermore, after washing with a 0.1 M buffer (pH 3.0), the activity of the biosorbent is regenerated and remained comparable for more than 200 cycles. Scanning electron microscopy reveals a successful immobilization of biomass on eggshells powder and precipitation of Cr (VI) on the bacterial cell surface. The proposed method proves highly sensitive and could be suitable for the determination of Cr (VI) at an ultra-trace level.

The aims of the present study were the determination of antimicrobial and antibiofilm effects of meropenem-loaded mesoporous silica nanoparticles (MSNs) on carbapenem resistant Pseudomonas aeruginosa ( P. aeruginosa) and cytotoxicity properties in vitro. The meropenem-loaded MSNs had shown antibacterial and biofilm inhibitory activities on all isolates at different levels lower than MICs and BICs of meropenem. The viability of HC-04 cells treated with serial concentrations as MICs and BICs of meropenem-loaded MSNs was 92–100%. According to the obtained results, meropenem-loaded MSNs display the significant antibacterial and antibiofilm effects against carbapenem resistant and biofilm forming P. aeruginosa and low cell toxicity in vitro. Then, the prepared system can be an appropriate option for the delivery of carbapenem for further evaluation in vivo assays.

Abstract Study question Is there a relationship between the serum progesterone and estradiol levels and certain morphological characteristics of human endometrium during the mid-luteal phase? Summary answer Serum progesterone is associated with the stromal edema and the abundance and size of basal vacuoles in the endometrium of women during the mid-luteal phase. What is known already Progesterone and estrogen are essential hormones that are necessary to prepare the endometrium for pregnancy. Their serum concentrations during the mid-luteal phase are important criteria for prediction of successful embryo implantation. In addition, a variety of endometrial morphological markers, such as the presence of pinopodes, subnuclear and supranuclear vacuoles, glandular secretion, and stromal edema have been applied for determination of the window of implantation and endometrial receptivity. However, the relationship between these endometrial morphological characteristics and serum levels of progesterone and estradiol is still scarcely studied. Study design, size, duration This is an observational study of 98 women, 25 to 46 years of age (mean 37 years), who had a blood sample and an endometrial biopsy during the mid-lutheal phase (LH + 7) in a natural cycle. The study was conducted between August 2020 and November 2020. Participants/materials, setting, methods Serum progesterone and estradiol were measured by electrochemiluminescence immunoassay (ECLIA) on the Cobas e411 analyser (Roche Diagnostics, Germany). The following endometrial morphological characteristics were assessed using light microscopy: (1) basal vacuoles (mean size and percentage of vacuolated glandular cells) (2) apical vacuoles (mean size and percentage of vacuolated glandular cells), (3) pinopodes (percentage of luminal epithelium covered in pinopodes), (4) glandular intraluminal secretion (6-level scoring system), (5) stromal edema (6-level scoring system). Main results and the role of chance The serum progesterone levels ranged between 0.39 and 145.3 ng/ml, with a median of 24.36 ng/ml. The serum estradiol levels varied between 26.91 and 842.89 pg/ml with a median of 124.75 pg/ml. The percentage of cells with basal vaculoles ranged from 0 to 90%, with a median of 38.57%, apical vacuoles (0–50%, 16.83%), pinopodes (0–80%, 23.87%), glandular intraluminal secretion (0–80%, 28.57%), and stromal edema (1–6, 1.42). To examine the association between the serum progesterone and estradiol and the studied endometrial morphological characteristics, the Spearman’s Rho Correlation coefficient for non-paramentric data was used. No correlation was found between serum estradiol levels and the studied morphological variables (p &gt; 0.05). In contrast, the serum progesterone concentration showed a significant negative correlation with the percentage of glandular epithelial cells with basal vacuoles (R= - 0.28; p = 0.03), the mean size of the basal vacuoles (R= - 0.24; p = 0.5) and a significant positive correlation with the stromal edema (R = 0.34; p &lt; 0.01). Limitations, reasons for caution The study was limited in sample size. Wider implications of the findings: The results of this study revealed that serum progesterone is more strongly associated with the occurrence of certain endometrial morphological characteristics during the mid-luteal phase than serum estradiol. These findings are valuable for development of new methods for accurate determination of the window of implantation. Trial registration number Not applicable
