ABSTRAKTujuan dari penelitian ini adalah untuk mengetahui diameter daya hambat air rebusan daun sirih terhadap bakteri penyebab mastitis. Bakteri yang digunakan dalam penelitian ini adalah Staphylococcus aureus dan Escherichia coli. daun sirih yang digunakan adalah daun sirih hijau yang banyak tumbuh di Pulau Timor. Rancangan penelitian yang digunakan adalah faktorial dengan 3 waktu perebusan (30, 45 dan 60 menit) dan 3 konsentrasi (12.5%, 25% dan 50%) dengan 3 pengulangan. Hasil penelitian menunjukkan bahwa air rebusan daun sirih hijau belum mampu menghambat pertumbuhan bakteri Escherichia coli. Air rebusan daun sirih konsentrasi 50% dengan waktu perebusan 45 menit menunjukkan hasil penghambatan paling besar. Air rebusan daun sirih mampu menghambat bakteri Staphylococcus aureus namun belum mampu menghambat bakteri Escherichia coli.Kata kunci: Escherichia coli, rebusan air daun sirih, Staphylococcus aureus, uji antibakteriABSTRACTThe aim of this study is to determine the inhibition of betel leaf decoction water against bacteria that caused mastitis. The bacteria used in this study are Staphylococcus aureus and Escherichia coli. The betel leaves used are non-fragrant green betel leaves which are widely grown on Timor Island. The design used was a factorial pattern with 3 boiling time (30, 45 and 60 minutes) and 3 concentrations of boiled water (12.5%, 25% and 50%) with 3 replications. The results showed that the betel leaf decoction water was not able to inhibit the growth of Escherichia coli. Betel leaf decoction water concentration of 50% with a boiling time of 45 minutes showed the greatest inhibitory effect. Betel leaf decoction water has been proven to be able to inhibit the Staphylococcus aureus but has not been able to inhibit Escherichia coli.Keywords: Escherichia coli, decoction of betel leaf water, Staphylococcus aureus, antibacterial test

<p><span id="docs-internal-guid-2cebd816-7fff-d997-4dfd-796d046d0d3b"><span>Daun pucuk idat merupakan tanaman lokal Bangka Belitung yang memiliki aktivitas antibakteri terhadap </span><span>Staphylococcus aureus </span><span>dan</span><span> Escherichia coli.</span><span>Sebagai upaya pemanfaatan potensi lokal maka dibuat suatu produk sediaan gel </span><span>hand sanitizer</span><span> yang praktis dan ekonomis. </span><span>Ekstrak daun pucuk idat diperoleh dengan metode maserasi menggunakan pelarut aseton. Ekstrak yang dihasilkan diformulasikan dalam berbagai konsentrasi ekstrak daun pucuk idat (40%, 60%, 80%, dan 100%) dengan penambahan </span><span>carboxymethyl cellulose sodium</span><span> (</span><span>CMC-Na) 0,25 g, gliserin 1 mL, propilenglikol 0,5 mL serta akuades        10 mL. Setiap formulasi gel </span><span>handsanitizer</span><span> dilakukan pengujian stabilitas fisik meliputi uji organoleptis, uji homogenitas, uji daya sebar, uji pH, uji iritasi kulit serta uji aktivitas antibakteri.</span><span> </span><span> Hasil uji organoleptik menunjukkan bahwa semua konsentrasi </span><span>hand sanitizer </span><span>berbentuk gel, berwarna kuning muda, berbau dan homogen. Uji pH dan daya sebar gel</span><span> hand sanitizer</span><span> telah memenuhi ketentuan SNI No. 06-2588. Pengujian iritasi kulit tidak menimbulkan kemerahan, gatal-gatal dan tidak mengakibatkan kulit menjadi kasar pada punggung tangan sukarelawan. Berdasarkan uji sifat fisik gel</span><span> hand sanitizer </span><span>yang dihasilkan memiliki sifat fisik yang serupa dengan gel </span><span>hand sanitizer </span><span>komersial. Hasil uji antibakteri hand sanitizer ekstrak daun pucuk idat terhadap </span><span>E. coli</span><span> dengan konsentrasi ekstrak daun pucuk idat 40%, 60%, 80%, dan 100% diperoleh diameter zona hambat berturut-turut sebesar 11,5 mm, 17,4 mm, 19,3 mm dan 19,3 mm, sedangkan pada bakteri </span><span>S. aureus</span><span> diperoleh diameter zona hambat berturut-turut sebesar 17,1 mm, 24,2 mm, 27,4 mm, dan  18 mm. Hasil uji antibakteri menunjukkan bahwa sediaan gel </span><span>hand sanitizer </span><span>ekstrak aseton daun pucuk idat memiliki aktivitas antibakteri terhadap bakteri </span><span>E. coli </span><span>dan </span><span>S. aureus</span><span>.</span></span></p><p><span><span><span id="docs-internal-guid-50d6ff2c-7fff-2e66-45f1-73cf49e6527a"><strong>Hand Sanitizer of Leaves Extract of Cratoxylum glaucum as Antibacterial of Staphylococcus aureus and Escherichia coli. </strong><span>Cratoxylum glaucum</span><span> leaves is local plants of Bangka Belitung which have antibacterial activity against </span><span>S. aureus</span><span> and </span><span>E. coli</span><span>. As an effort to explore local potential, a practical and economical gel hand sanitizer product was prepared. </span><span>Cratoxylum glaucum</span><span> leaves extract was obtained by maceration method using acetone. The extract was formulated in various concentrations of </span><span>Cratoxylum glaucum</span><span> leaf extract (40%, 60%, 80%, and 100%) with the addition of 0.25 g carboxymethyl cellulose sodium (CMC-Na), 1 mL glycerin, 0.5 mL propylene glycol and 10 mL distilled water. Each concentration of the hand sanitizer gel was tested for physical stability including organoleptic, homogenity, spreadability, pH, skin irritation and antibacterial activity test. The organoleptic test results showed that all concentrations of hand sanitizers formed gel with light yellow color, smelly odor and homogeneous form. The analysis of pH and spreadability of hand sanitizer gel has fulfilled SNI No. 06-2588. The skin irritation testing does not cause redness, itching, and rough skin on the back of volunteer hands. Based on the physical properties test, the resulting hand sanitizer gel has physical properties similar to commercial hand sanitizer gel. The results of the antibacterial test of hand sanitizer against </span><span>E. coli</span><span> with </span><span>Cratoxylum glaucum</span><span> leaf extract concentrations of 40%, 60%, 80%, and 100% obtained the diameter inhibitory zones of 11.5 mm, 17.4 mm, 19.3 mm, and 19.3 mm, respectively. Meanwhile, the results of the antibacterial test of hand sanitizer against </span><span>S. aureus</span><span> bacteria with concentration of 40%, 60%, 80%, and 100% shows that the inhibition zone diameters were 17.1 mm, 24.2 mm, 27.4 mm, and 18 mm, respectively. The antibacterial test results shows that the gel hand sanitizer prepared has antibacterial activity against </span><span>E. coli</span><span> and </span><span>S. aureus</span><span> bacteria.</span></span></span></span></p>

Staphylococcus aureus dan Escherichia coli merupakan bakteri Gram positif dan Gram negatif yang dapat menimbulkan infeksi atau penyakit dalam tubuh. Penelitian ini bertujuan untuk menguji aktivitas bakteri patogen dengan memakai nanopartikel kitosan sebagai antibakteri yang dibuat dalam empat konsentrasi (0,5%, 1%, 1,5% dan 2%) serta penggunaan kontrol asam asetat 1%, ciprofloxacin dan air steril sebagai pembanding. Metode penelitian yang digunakan yaitu metode gelasi ionik untuk pembuatan nanopartikel kitosan dan difusi agar untuk pengujian antibakteri. Data dianalisis dengan One Way Anova yang dilanjutkan dengan metode BNT (Beda Nyata Terkecil). Hasil penelitian diperoleh penghambatan pertumbuhan bakteri S. aureus dan E. coli tertinggi pada konsentrasi 0,5%, dengan diameter zona hambat hari pertama sampai hari ketiga 12,31 mm, 9,98 mm, dan 20,46 mm pada S. aureus dan 15,88 mm, 18,71 mm, dan 20,43 mm pada E. coli, kategori kuat, dan bersifat bakteriostatik dan penghambatan terendah pada konsentrasi 2% dengan diameter zona hambat pada S. aureus yaitu 5,56 mm, 5,50 mm, dan 5,40 mm, dan pada E. coli yaitu 5,93 mm, 9,64 mm, dan 12,58 mm, kategori sedang, dan bersifat bakteriostatik. Kata kunci: Kitosan, nanopartikel kitosan, aktivitas antibakteri.  Abstract Antibacteria is a substance that can inhibit the growth of bacteria and able to kill bacteria that cause infections. Staphylococcus aureus and Escherichia coli are Gram positive and Gram negative bacteria that able to cause infections or diseases. This study aimed to examine the activity of pathogenic bacteria by using chitosan nanoparticles as antibacterial. The treatments were made in four concentrations (0.5%, 1%, 1.5% and 2%) and, for comparison, there were also acetic acid control, ciprofloxacin and sterile water. The research method used is the ionic gelation method for the manufacture of chitosan nanoparticles and agar diffusion for antibacterial testing. Data were analyzed with One Way Anova followed by LSD (Least Significant Difference) method. The results showed the highest inhibition of growth of S. aureus and E. coli bacteria at a concentration of 0.5%, with a diameter of inhibition zones of the first day to the third day of 12.31 mm, 9.98 mm, and 20.46 mm in S. aureus and 15,88 mm, 18,71 mm, and 20,43 mm in E. coli, the strong category, and are bacteriostatic and the lowest inhibition was at 2% concentration with inhibition zone diameters in S. aureus namely 5.568 mm, 5.50 mm, and 5, 40 mm, and in E. coli, 5.93 mm, 9.63 mm and 12.58 mm, the medium category and bacteriostatic.Key words: Chitosan, nanoparticles chitosan, antibacterial activity.

Taking monascin as the research object, monascin was extracted from red kojic rice by ethanol extraction and extracted with 60%, 70% and 80% ethanol respectively. Finally, it was concluded that when the concentration of ethanol was 70%, the extraction rate of monascin was the highest, reached 75.68%. The bacteriostatic experiments of monascin extract and monascin fermentation showed that it had strong inhibitory effect on Staphylococcus aureus and Bacillus subtilis, weak inhibitory ability on Escherichia coli and Aspergillus niger, and no obvious inhibitory effect on the growth of Saccharomyces cerevisiae.

In effort to combat environmental pollution, improve animal production and avoid drug residue, producers have resorted to the use of phytobiotics to inhibit ammonia producing microbes in the gut. Freshly harvested Azadirachta indica, Carica papaya, Saccharum officinarum, Chromolaena odorata, Eucalyptus camadulensis and Mangifera indica leaves were air dried, blended and extracted using five concentrations of solvent (100% water, 70% water + 30% methanol, 50% water + 50% methanol, 30% water + 70% methanol and 100% methanol). The antibacterial activities of the extracts and antibiotics (doxycycline) as control, were tested according to standard procedures against both gram positive (Bacillus subtilis, Staphylococcus aureus) and gram negative (Escherichia coli, Vibrio cholerae) ammonia producing bacteria. In most cases, it was observed that the inhibitory effect of methanolic extracts of Azadirachta indica, Carica papaya, Saccharum officinarum, Chromolaena odorata, Eucalyptus camadulensis and Mangifera indica leaves were not significantly different from that of synthetic antibiotics, doxycycline, when tested against the test bacteria, Bacillus subtilis, Staphylococcus aureus and Vibrio cholera. However, leave extracts showed significantly better inhibitory effect on Escherichia coli than the antibiotic drug. It can be concluded, therefore, that the leaf extracts of Azadirachta indica, Carica papaya, Saccharum officinarum, Chromolaena odorata, Eucalyptus camadulensis and Mangifera indica can serve as alternative to synthetic drugs in animal production due to their effective actions against microbial organisms.

Lactobacillus acidophilus exerted antagonistic actions on growth of Staphylococcus aureus, Salmonella typhimurium, enteropathogenic Escherichia coli, and Clostridium perfringens when grown with each in associative cultures. S. aureus and C. perfringens were more sensitive to the inhibition than were S. typhimurium and E. coli. The amount of the antagonism produced varied among strains of L. acidophilus and could not be directly related to amounts of acid produced; hydrogen peroxide produced by the lactobacilli appeared to be partially responsible for the antagonistic interaction. The inhibitory effect was produced also under anaerobic conditions in a pre-reduced medium.

Growth of three strains of Staphylococcus aureus and two strains of Escherichia coli on nutrient agar (NA) supplemented with ethanol and NaCl was investigated. S. aureus did not grow on NA containing ≧10% ethanol (wt/wt) combined with ≧0% NaCl (wt/wt), or 7.5% ethanol combined with 7.5% NaCl. Neither E. coli nor E. coli O157:H7 grew on NA containing ≧7.5% ethanol combined with ≧0% NaCl, 5% ethanol combined with ≧2.5% NaCl, or ≧5% NaCl combined with ≧0% ethanol. It is apparent that NaCl enhanced the inhibitory effect of ethanol on growth of S. aureus and E. coli. When cells were suspended in nutrient broth containing 12.5, 20, or 40% ethanol combined with NaCl, viable cells decreased with an increase of ethanol concentration. Ethanol sensitivity among strains and between genera varied in a limited range. When the cells were exposed to 20% ethanol in combination with 5% NaCl, S. aureus and E. coli lost viability after 30 and 10 min, respectively. When treated with 40% ethanol combined with ≧0% NaCl, all test strains lost viability within 5 min.

Among applications of ceramics in technological context, hydroxyapatite (HAp) stands out in the scientific community due to chemical biocompatibility and molecular similarity with the structures of bone and dental tissues. Such features are in addition to its antimicrobial properties. This work aimed firstly to synthesize hydroxyapatite by two different routes: hydrothermal (HD HAp) and co-precipitation (CP HAp), and secondly to verify the antimicrobial properties of these materials through direct contact tests against Staphylococcus aureus (SA10) and Escherichia coli (EC7) bacteria. These materials were characterized by XRD, Raman, and TEM. Antimicrobial tests showed inhibitory efficacy of 97.0% and 9.5% of CP HAp for SA10 and EC7, respectively. The HD HAp showed inhibitory effect of 95.0% and 0.0% for SA10 and EC7, respectively. The inhibitory effect of the tested materials against Staphylococcus aureus may be related to the HAp hydrophilicity.

The work is focused on the antibacterial effect of four types of blossom honey, one honeydew and two Mānuka honey (MAN100+ and MAN400+) on selected pathogenic microorganisms isolated from cow’s milk (Staphylococcus aureus 51 and S. aureus 428), sheep’s milk (S. aureus 627), and from the Czech Collection of Microorganisms (Streptococcus uberis CCM 4617, Streptococcus agalactiae CCM 6187, Enterococcus faecalis CCM 4224 and Escherichia coli CCM 4787). The concentrations of honey samples were 20% and 30%. The obtained results showed a 100% inhibitory effect of MAN400+ on all tested bacterial strains even at a concentration of 20% and also a comparable inhibitory effect of Mānuka honey with Czech honeydew. The results indicate that honey had an inhibitory effect against the tested bacterial species which may cause mastitis.

As a result of the development of resistance of microorganisms to older antimicrobial agents there is need for a search for new agents, which are effective for the treatment of infections. The crude aqueous and alcoholic extr acts of Garcinia kola fruits were assayed against Staphylococcus aureus, Bacillus cereus, Escherichia coli and Klebsiella pneumoniae. The results revealed that the plant extracts possess inhibitory effect against the microorganisms tested. The minimum inhibitory concentration of the plant extracts ranged between 20mg/ml and 45mg/ml. There was a change in the antibacterial activity of the test extracts on variation of temperature. The results obtained may suggest that the plant extract is thermal stable and could serve as a source of industrial drugs useful in chemotherapy of some microbial infections.
