In Colombia, the cultivation of deciduous fruit trees such as pear is expanding for socio-economic reasons and is becoming more and more important for the local population. Since organized cultivation is slowly replacing sustenance cultivation, scientific information on the present agro-environment is needed to proceed in this change in an organic and environmentally friendly way. In particular, this study is an accurate description of the mycobiota present in the bulk soil of two different high altitude pear orchards in the Colombian Andes. The metabarcoding of soil samples allowed an in-depth analysis of the whole fungal community. The fungal assemblage was generally dominated by Ascomycota and secondly by Mortierellomycota. As observed in other studies in Colombia, the genus Mortierella was found to be especially abundant. The soil of the different pear orchards appeared to host quite different fungal communities according to the soil physico-chemical properties. The common mycobiota contained 35 fungal species, including several species of Mortierella, Humicola, Solicoccozyma and Exophiala. Moreover, most of the identified fungal species (79%) were recorded for the first time in Colombian soils, thus adding important information on soil biodiversity regarding both Colombia and pear orchards.

Three compost samples were prepared in this study from Siam weed (Chromolaena odorata) and cowdung. Sample A was prepared from Cow dung and siam weed at ratio 100g: 100g, Sample B was prepared from 200g chopped siam weed and sample C contained 200g cowdung. These three sam-ples were composted in plastic drums perforated for aeration and each sample were replicated three times. The content in the drums were regularly turned and monitored at 1, 10, 30 and 60 days for mi-crobiological and physicochemical properties. The microbiological and physicochemical analyses of the compost were carried out using standard procedures. Bacterial, Coliform and Fungal count in-creased from day 1 to the 30th day and thereafter decreased from 30th day to the 60th day in all the composting samples. The bacteria species isolated and identified were Pseudomonas fragilis, Pseu-domonas nitrificans, Proteus mirabilis, E. coli, Streptococcus faecium, Micrococcus luteus, Clostridium perfringes, Bacillus cereus, Proteus morganii, Micrococcus acidophilus. Fungal species were Aspergil-lus flavus, Aspergillus fumigatus, Fusarium oxysporium, Penicillum chrysogenum, Aspergillus niger, Mucor sp. and Saccharomyces cerevisiae. The pH of the composted samples ranges between 5.8 to 6.9. The nitrogen, phosphorus and potassium content increased with days of composting but the heavy metals decreased with days of composting. The sulfatase, phosphatase, dehydrogenase, amyl-ase and cellulose enzymes in the three samples increased from day 1 to the 60th day. Sulfatase en-zyme which was the highest ranged from 25 to 76.5% in the three sample, phosphatase (14 to 60.5%), dehydrogenase (20.5 to 55.0%), cellulose (16.5 to 49%) and amylase which was the least enzyme recorded ranged from 5.0 to 38%.

The chemical properties in the rhizosphere of fruit tree species were investigated during different season at CCS HAU, Hisar. Results revels that pH value, OC, N, P, K, Zn contents increased significantly with all the fruit orchards over control (uncultivated land). However, maximum pH, EC, OC, CN ratio, available N, P, K, Zn and Fe (8.30, 0.33 dS/m, 0.54 %, 11.63, 126.26 kg/ha, 32.24 kg/ha, 284.93 kg/ha, 2.10 ppm and 3.34 ppm) was found in aonla, sweet orange, jamun, jamun, guava, sweet orange, guava, ber and sweet orange, respectively. Highest EC, CEC, OC and available K were found in winter season but pH, EC, available N, Zn, Mn and B contents were recorded maximum in summer season.CN ratio and available P were maximum in rainy season. Inter row spaces soil had highest pH, CEC, CN ratio and Zn contents while maximum OC, Available N, P, K, Fe, Mn and B contents was observed under the canopy of fruit trees.

The present study was aimed to investigate the effect of physico-chemical parameters and soil macro-nutrients to know the nutrient uptake status during sowing time (ST) and after the harvesting (AH) of crops of Leh-Ladakh. In this context, total 55 no. of soil samples were collected from the eleven villages. Thereafter, soil texture, pH, electrical conductivity (EC), total dissolved solids (TDS), organic carbon (OC), nitrogen (N), phosphorus (P), and potassium (K) were analyzed as per the standard methods. The results exhibited variation in different studied parameters at ST and AH, are OC (ST- 1.70 ± 0.11; AH-2.31±0.08), N (ST- 171.54±11.40; AH- 212.03±13.18), P (ST- 75.62±8.16; AH- 96.32±11.56), pH (ST- 8.12±0.05; AH- 8.16±0.06), EC (ST- 0.48±0.04; AH- 0.58±17), TDS (ST-309±22.41; AH-189±16.42) and soil texture gradient (Sand: ST-75.16±1.27 & AH-71.75±1.26, Silt: ST- 18.55±1.09 & AH- 20.66±1.02 and clay: ST- 6.33±0.53 & AH- 7.76±0.63). The comparison of physico-chemical parameters, macronutrients, soil texture, and organic carbon at sowing time (ST) and after harvesting (AH) revealed significant difference in some macronutrients, EC, and organic carbon, whereas no changes were observed in soil texture, pH and phosphorus. Hence, this study highlights the need of physico-chemical parameters management during crops sowing for enhancing macronutrients availability to crops in trans-Himalayan high altitude region.

An in-depth analysis of wheat flour (WF) substituted with 0.5–2.5% rosehip powder (Rp) concerning the proximate composition, dough farinographic properties, and bread physico-chemical characteristics was performed. The purpose of this study was to investigate whether the use of Rp as a natural alternative for synthetic ascorbic acid in breadmaking was appropriate. A sample of wheat flour with an ascorbic acid addition of 2 mg/100 g was also used. Rp showed higher ash, carbohydrates, and fibre content, as well as lower moisture and protein content compared to wheat flour, and a vitamin C content of 420 ± 16.09 mg/100 g. A proximate composition analysis revealed a decrease in moisture, protein, and wet gluten, and an increase in ash, carbohydrates, and fibres for the flour mixtures compared with WF. Farinographic properties were positively influenced by the Rp addition and the high fibre content in the flour mixtures. Water absorption increased from 58.20% (WF) to 61.90% (2.5% Rp). Dough stability increased for the 0.5–1.0% Rp addition, then slightly decreased. The physico-chemical properties of bread prepared from flour mixtures showed a significant increase in height: 100.10 ± 0.14 mm (WF)–115.50 ± 0.14 mm (1.5% Rp), specific volume: 142.82 cm3/100 g (WF)–174.46 cm3/100 g (1.5% Rp), moisture: 41.81 ± 0.40% (WF)–43.92 ± 0.15% (2.0% Rp), and porosity: 87.75 ± 1.06% (WF)–89.40 ± 0.57% (2.5% Rp). The results indicated that the Rp used in breadmaking to replace synthetic ascorbic acid could be suitable.

Fracturing under ultrahigh vacuum (UHV, p ≤ 10-9 Torr) produces membrane fracture faces devoid of contamination. Such clean surfaces are a prerequisite foe studies of interactions between condensing molecules is possible and surface forces are unequally distributed, the condensate will accumulate at places with high binding forces; crystallites will arise which may be useful a probes for surface sites with specific physico-chemical properties. Specific “decoration” with crystallites can be achieved nby exposing membrane fracture faces to water vopour. A device was developed which enables the production of pure water vapour and the controlled variation of its partial pressure in an UHV freeze-fracture apparatus (Fig.1a). Under vaccum (≤ 10-3 Torr), small container filled with copper-sulfate-pentahydrate is heated with a heating coil, with the temperature controlled by means of a thermocouple. The water of hydration thereby released enters a storage vessel.

SummaryPhysico-chemical properties of recombinant desulphatohirudin expressed in yeast (CIBA GEIGY code No. CGP 39393) were reinvestigated. As previously reported for natural hirudin, the recombinant molecule exhibited abnormal behaviour by gel filtration with an apparent molecular weight greater than that based on the primary structure. However, molecular weight estimation by SDS gel electrophoresis, FAB-mass spectrometry and Photon Correlation Spectroscopy were in agreement with the theoretical molecular weight, with little suggestion of dimer or aggregate formation. Circular dichroism studies of the recombinant molecule show similar spectra at different pH values but are markedly different from that reported by Konno et al. (13) for a natural hirudin-variant. Our CD studies indicate the presence of about 60% beta sheet and the absence of alpha helix in the secondary structure of recombinant hirudin, in agreement with the conformation determined by NMR studies (17)
