Kaposi’s sarcoma-associated herpesvirus (KSHV) induces life-long infections and has evolved many ways to exert extensive control over its host’s transcriptional and post-transcriptional machinery to gain better access to resources and dampened immune sensing. The hallmark of this takeover is how KSHV reshapes RNA fate both to control expression of its own gene but also that of its host. From the nucleus to the cytoplasm, control of RNA expression, localization, and decay is a process that is carefully tuned by a multitude of factors and that can adapt or react to rapid changes in the environment. Intriguingly, it appears that KSHV has found ways to co-opt each of these pathways for its own benefit. Here we provide a comprehensive review of recent work in this area and in particular recent advances on the post-transcriptional modifications front. Overall, this review highlights the myriad of ways KSHV uses to control RNA fate and gathers novel insights gained from the past decade of research at the interface of RNA biology and the field of KSHV research.

C-N bond formation is a particularly important step in the generation of many biologically relevant heterocyclic molecules. Several methods have been reported for this purpose over the past few decades. Well-known named reactions like Ullmann-Goldberg coupling, Buchwald-Hartwig coupling and Chan-Lam coupling are associated with the C-N bond formation reactions. Several reviews covering this topic have already been published. However, no comprehensive review covering the synthesis of drugs/ lead compounds using the C-N bond formation reactions was reported. In this review, we cover many modern methods of the C-N bond formation reactions, with special emphasis on metal-free and green chemistry methods. We also report specific strategies adopted for the synthesis of drugs, which involve the C-N bond formation reactions. Examples include anti-cancer, antidepressant, anti-inflammatory, anti-atherosclerotic, anti-histaminic, antibiotics, antibacterial, anti-rheumatic, antiepileptic and anti-diabetic agents. Many recently developed lead compounds generated using the C-N bond formation reactions are also covered in this review. Examples include MAP kinase inhibitors, TRKs inhibitors, Polo-like Kinase inhibitors and MPS1 inhibitors.

Patrinia scabiosaefolia Fisch. Trev. and Patrinia villosa (Thunb.) Juss, are two species of Patrinia recorded in the Chinese Pharmacopoeia with the same Chinese name “Baijiangcao” and similar therapeutic effect in traditional Chinese medicine. The present article is the first comprehensive review on the chemical composition and pharmacological activities of these herbs. In this review, data on chemical constituents and pharmacological profile of the two herbs are provided. This review discusses all the classes of the 223 compounds (phenylpropanoids, flavonoids, terpenes, saponins and volatile components, etc.) detected in the two herbs providing information on the current state of knowledge of the phytochemicals present in them. In the past three years, our research group has isolated and identified about more than 100 ingredients from the two herbs. Therefore, we published a systematic review of our research papers and studies on the two herbs were carried out using resources such as classic books about Chinese herbal medicine and scientific databases including Pubmed, Web of Science, SciFinder, CNKI. etc. The present review discusses the most thoroughly studied pharmacological activities (antioxidant, anti-inflammatory, immunomodulatory, antimicrobial, antitumor and antiviral activities) of the two herbs. This comprehensive review will be informative for scientists searching for new properties of these herbs and will be important and significant for the discovery of bioactive compounds from the two herbs and in complete utilization of Patrinia scabiosaefolia Fisch. ex Trev. and Patrinia villosa (Thunb.) Juss.

The rapid changes in science and technology witnessed in recent decades have significantly contributed to the arousal of the awareness by decision-makers and the public as a whole of the need to strengthen the connection between outreach activities of universities and research institutes and the activities of educational institutions, with a central role played by schools. While the relevance of the problem is nowadays unquestioned, no unique and fully satisfactory solution has been identified. In the present paper we would like to contribute to the discussion on the subject by reporting on an ongoing project aimed to teach Particle Physics in primary schools. We will start from the past and currently planned activities in this project in order to establish a broader framework to describe the conditions for the fruitful interplay between researchers and teachers. We will also emphasize some aspects related to the dissemination of outreach materials by research institutions, in order to promote the access and distribution of scientific information in a way suited to the different age of the target students.

In the past centuries, small towns in Podlaskie Voivodeship were dominated by wooden buildings. Nowadays, there are not many of them left. However, they can still be found in the centers of towns, including some market squares. These are often inconspicuous objects, mainly wooden houses. This paper discusses the issue of the significance of wooden buildings, especially houses, in maintaining and restoring historic values of market spaces in small towns of Podlaskie Voivodeship in the examples of Bielsk Podlaski and Kleszczele. The research determined the moment of rapid changes, during which most marketplace buildings lost their historic form. The existing condition was analyzed in terms of its historical values. Conclusions were formulated on the scope and type of restoration works for individual market squares. Finally, general conclusions from the research on the market squares of Bielsk Podlaski and Kleszczele were presented.

The coronavirus disease 2019 (COVID-19) outbreak emerged in December 2019 in China and rapidly spread worldwide. Inflammatory bowel disease (IBD) patients are likely to be more susceptible to viral infections, and this is significantly influenced by the type of therapy they receive. Thus, issues specifically concerning the medical treatment of IBD patients were shortly addressed at the beginning of the pandemic. However, recently available data on the occurrence and outcome of SARS-CoV-2 infection in IBD patients does not address the concerns raised at the beginning of the pandemic. Growing evidence and the rapid changes happening over the past few weeks have helped elucidate the current situation, contribute to our understanding of the disease, and many previously raised questions could now be answered. We hereby summarise available evidence regarding viral infections and IBD, focusing on SARS-CoV infections, and we provide practical recommendations related to patient management during the COVID-19 pandemic era.

Abstract The field of genetic epidemiology is relatively young and brings together genetics, epidemiology, and biostatistics to identify and implement the best study designs and statistical analyses for identifying genes controlling risk for complex and heterogeneous diseases (i.e., those where genes and environmental risk factors both contribute to etiology). The field has moved quickly over the past 40 years partly because the technology of genotyping and sequencing has forced it to adapt while adhering to the fundamental principles of genetics. In the last two decades, the available tools for genetic epidemiology have expanded from a genetic focus (considering 1 gene at a time) to a genomic focus (considering the entire genome), and now they must further expand to integrate information from other “-omics” (e.g., epigenomics, transcriptomics as measured by RNA expression) at both the individual and the population levels. Additionally, we can now also evaluate gene and environment interactions across populations to better understand exposure and the heterogeneity in disease risk. The future challenges facing genetic epidemiology are considerable both in scale and techniques, but the importance of the field will not diminish because by design it ties scientific goals with public health applications.

Charophytes in the genus Lamprothamnium exhibit a large amount of diversity, particularly in the examples from Australia, although little of that variation has been recognised at species level in the past. The Australian members of the genus are revised here on the basis of extensive new collections, examination of specimens in herbaria and comprehensive review of the literature and available type material. The existing species Lamprothamnium macropogon (A.Braun) Ophel, L. inflatum (Fil. & G.O.Allen ex Fil.) A.García & Karol and L. heraldii A.García & Casanova are retained, eight new species are described (L. australicum Casanova, L. beilbyae Casanova, L. capitatum Casanova, L. compactum Casanova, L. coorongense Casanova, L. diminutum Casanova, L. macroanthum Casanova and L. stipitatum Casanova) and two taxa variously treated at infraspecific rank in Lychnothamnus are transferred to Lamprothamnium at species rank (L. cockajemmyense Casanova, L. tasmanicum (A.Braun) Casanova). Neither L. papulosum (Wallr.) J.Groves nor L. succinctum (A.Braun) R.D.Wood are confirmed for Australia after examination of the type material of these species. Species are distinguished by the arrangement of the gametangia, morphology of the fertile whorls and characteristics of the oospores. Four of these species are dioecious and nine are monoecious, which supports published conjectures concerning the biogeography of charophyte species (Proctor (1980): J. Phycol. 16, 218–233, doi:10.1111/j.1529-8817.1980.tb03023.x).

This paper explores Dworkin's ‘law as a chain novel’ analogy and considers the recent work of Dworkin and MacCormick through close scrutiny of two recent judgments of Lord Hoffmann, in Barlow Clowes v Eurotrust International [2005] UKPC 37 and Barker v Corus [2006] UKHL 20. The aim is to examine Dworkin's theory in the context of recent English private law decisions and determine whether Lord Hoffmann's approach to interpretation is consistent with that of Dworkin (as his Lordship has contended in the past). It is argued that Lord Hoffmann's treatment of recent decisions on which he himself sat raises significant questions regarding fidelity, coherence and the institutional structure of the House of Lords.

Abstract Why do modern states recruit legionnaires—foreigners who are neither citizens nor subjects of the country whose military they serve? Rather than exclusively enlist citizens for soldiers, for the past two centuries states have mobilized legionnaires to help wage offensives, project power abroad, and suppress dissent. A supply-and-demand argument explains why states recruit these troops, framing the choice to mobilize legionnaires as a function of political factors that constrain the government's leeway to recruit domestically and its perceptions about the territorial threats it faces externally. A multimethod approach evaluates these claims, first by examining an original dataset of legionnaire recruitment from 1815 to 2020, then by employing congruence tests across World War II participants, and finally by conducting a detailed review of a hard test case for the argument—Nazi Germany. The prevalence of states’ recruitment of legionnaires calls for a reevaluation of existing narratives about the development of modern militaries and provides new insights into how states balance among the competing imperatives of identity, norms, and security. Legionnaire recruitment also underscores the need to recalibrate existing methods of calculating net assessments and preparing for strategic surprise. Far from being bound to a state's citizenry or borders, the theory and evidence show how governments use legionnaires to buttress their military power and to engineer rapid changes in the quality and quantity of the soldiers that they field.

Accelerating the development and approval of novel therapeutics has emerged as a key public health priority given the mortality, morbidity, and economic costs associated with infections caused by drug-resistant bacteria. However, there is limited empirical evidence to guide policymaking, such as the factors that may disadvantage antibiotics compared to other classes of drugs. In this Article, we empirically examine characteristics of the key clinical trials underpinning FDA's approval of antibiotics and other drugs over the past decade. Despite perceptions that antibiotic trials are larger and more difficult to conduct, we find that antibiotic trials are no larger than those conducted for drugs approved in other disease areas with high unmet medical needs, suggesting that policymakers may need to target other levers to meaningfully stimulate innovation. We discuss the risks and benefits of harnessing new and existing regulatory pathways to speed the approval of new drugs, particularly those intended to treat patients with serious and life-threatening infections, and we evaluate ways that proposals for new regulatory pathways could be improved to better prioritize and expedite the approval of therapies with the greatest potential for patient health benefits.
