SummaryEthylene oxide is currently recommended for sterilization of antibiotic impregnated beads; however this method carries health risks to personnel and is becoming less available.To perform a pilot study of the effect of radiation for sterilization of polymethylmethacrylate (PMMA) and plaster of Paris (POP) beads impregnated with amikacin, enrofloxacin, and ceftiofur.Radiation would effectively sterilize the beads without affecting the efficacy of the antibiotic.Beads of PMMA and POP were prepared in a clean but nonsterile manner with one of the three antibiotics (amikacin, enrofloxacin, ceftiofur) or no antibiotic. Beads were then exposed to radiation for a total dose of 0 kiloGray (kGy), 10 kGy and 25 kGy. Beads were incubated on Mueller-Hinton agar plates seeded with Escherichia coli, Staphylococcus aureus or Pseudomonas aeruginosa for 24 hours or cultured in brain-heart infusion broth for 48 hours. Zones of inhibition were measured on the agar plates and statistics were performed on the diameters of the zones of inhibition using an analysis of variance.There were no differences in the diameters of inhibition for all levels of radiation for all PMMA beads. The same was true with POP beads with the exception of enrofloxacin which had a significantly decreased zone of inhibition with increased levels of radiation, though the clinical significance of this finding was not assessed. Only beads without antibiotics and not exposed to radiation had bacterial growth.Radiation may be an effective method of sterilization for antibiotic impregnated beads.

Staphylococcus aureussmall-colony variants (SCVs) emerge frequently during chronic infections and are often associated with worse disease outcomes. There are no standardized methods for SCV antibiotic susceptibility testing (AST) due to poor growth and reversion to normal-colony (NC) phenotypes on standard media. We sought to identify reproducible methods for AST ofS. aureusSCVs and to determine whether SCV susceptibilities can be predicted on the basis of treatment history, SCV biochemical type (auxotrophy), or the susceptibilities of isogenic NC coisolates. We tested the growth and stability of SCV isolates on 11 agar media, selecting for AST 2 media that yielded optimal SCV growth and the lowest rates of reversion to NC phenotypes. We then performed disk diffusion AST on 86S. aureusSCVs and 28 isogenic NCs and Etest for a subset of 26 SCVs and 24 isogenic NCs. Growth and reversion were optimal on brain heart infusion agar and Mueller-Hinton agar supplemented with compounds for which most clinical SCVs are auxotrophic: hemin, menadione, and thymidine. SCVs were typically nonsusceptible to either trimethoprim-sulfamethoxazole or aminoglycosides, in accordance with the auxotrophy type. In contrast, SCVs were variably nonsusceptible to fluoroquinolones, macrolides, lincosamides, fusidic acid, and rifampin;mecA-positive SCVs were invariably resistant to cefoxitin. All isolates (both SCVs and NCs) were susceptible to quinupristin-dalfopristin, vancomycin, minocycline, linezolid, chloramphenicol, and tigecycline. Analysis of SCV auxotrophy type, isogenic NC antibiograms, and antibiotic treatment history had limited utility in predicting SCV susceptibilities. With clinical correlation, this AST method and these results may prove useful in directing treatment for SCV infections.

Neste trabalho foi avaliada a ação dos óleos essenciais de Thymus vulgaris (tomilho), Syzygium aromaticum (cravo-da-índia) e Rosmarinus officinalis (alecrim) e dos conservantes benzoato de sódio e sorbato de potássio como agentes antimicrobianos. As cepas de Staphylococcus aureus (ATCC 25923) e Escherichia coli (ATCC 25922) foram utilizadas no teste de susceptibilidade antimicrobiana usando-se a técnica de microdiluição em microplaca de 96 poços para avaliação da Concentração Inibitória Mínima (CIM) e, posteriormente, subcultivo em Mueller Hinton Agar para avaliaçãoda Concentração Bactericida Mínima (CBM). As concentrações dos óleos e conservantes sintéticos testados variaram de 15,6 a 1000μg/mL. As microdiluições utilizando inóculos bacterianos nas concentrações de 104 UFC/mL foram incubadas a 37ºC/24h. As CIM para os óleos essenciais de cravo, tomilho e alecrim foram de 550, 650 e >1000μg/mL para E. coli e 550, 800 e 1000μg/mL para S. aureus, respectivamente. No entanto, a ação bactericida dos óleos essenciais do cravo-da-índia e do tomilho foi encontrada apenas em relação a E. coli, na concentração de 550 e 850μg/mL, respectivamente. Para os dois conservantes sintéticos testados, a CIM foi >1000μg/mL, portanto não apresentaram atividade antibacteriana contra os microrganismos testados. Trabalhos futuros deverão ser realizados para verifi car a efi ciência dos antimicrobianos naturais anteriormente citados e para avaliar a possibilidade de serem utilizados na indústria de alimentos.

Penelitian ini bertujuan untuk melakukan uji fitokimia terhadap ekstrak etanol Cabai merah (Capsicum annum L.) dan uji aktivitasnya terhadap pertumbuhan bakteri Staphylococcus aureus dan Escherichia coli. Pengujian aktivitas antibakteri ini menggunakan metode difusi agar dengan menggunakan kertas cakram dengan tiga kali pengulangan. Ekstrak etanol cabai merah dibuat dengan konsentrasi 45, 60, 75, dan 90%. Uji aktivitas antibakteri dilakukan dengan metode Kirby Bauer. Media yang digunakan sebagai uji antibakteri pada penelitian ini yaitu Mueller-Hinton Agar (MHA). Hasil skrining fitokimia terhadap ekstrak etanol cabai merah menunjukkan adanya senyawa metabolit sekunder. Senyawa metabolit sekunder tersebut yaitu alkaloid, flavonoid, tanin, saponin, dan Steroid/triterpenoid. Steroid berfungsi sebagai antimikroba. Hasil penelitian ini juga mengungkapkan adanya kandungan senyawa capsaicin yang terkenal sebagai senyawa aktif antimikroba. Ekstrak etanol cabai merah memiliki aktivitas antibakteri terhadap bakteri Staphylococcus aureus dan Escherichia coli zona bening terendah pada konsentrasi 45% dan zona bening tertinggi pada konsentrasi 90%. Dapat disimpulkan bahwa ekstrak cabai merah memiliki aktivitas antibakteri terhadap penghambatan pertumbuhan bakteri.THE ABILITY OF RED CHILI ETHANOL EXTRACT IN INHIBITHING THE GROWTH OF BACTERIAThis study aimed to perform a phytochemical test on the ethanolic extract of red chili (Capsicum annum L.) and test its activity against the growth of Staphylococcus aureus and Escherichia coli bacteria. This antibacterial activity test used the agar diffusion method using disc paper with three repetitions. Red chili ethanol extract was made with concentrations of 45%, 60%, 75% and 90%. The antibacterial activity test was carried out using the Kirby Bauer method. The medium used as an antibacterial test in this study was Mueller-Hinton Agar (MHA). The results of phytochemical screening of red chili ethanol extract showed the presence of secondary metabolites. The secondary metabolites are alkaloids, flavonoids, tannins, saponins, and steroids/triterpenoids. Steroids serve as  antimicrobials. The results of this study also revealed the presence of capsaicin compounds which are known as active antimicrobial compounds. Red chili ethanol extract had antibacterial activity against Staphylococcus aureus and Escherichia coli bacteria in the lowest clear zone at a concentration of 45% and the highest clear zone at a concentration of 90%. It can be concluded that red chili extract has antibacterial activity against bacterial growth inhibition.

A ação antibacteriana in vitro de óleos essenciais de seis plantas foi verificada por meio da Concentração Inibitória Mínima (CIM=%v/v) pela diluição dos óleos em meio de cultura Mueller Hinton Agar, frente a linhagens de Staphylococcus aureus (n=16) e Escherichia coli (n=16) isoladas de casos clínicos humanos, além de 1 amostra padrão ATCC para cada espécie (Sa ATCC 25923 e Ec ATCC 25922), e determinação de curvas de sobrevivência em concentrações equivalentes a CIM90% dos respectivos óleos. O óleo essencial de canela foi o mais eficiente, com valores de CIM90% de 0,047 e 0,09 para S. aureus e E. coli respectivamente, enquanto gengibre (0,09), cravo da índia (0,095) e capim cidreira (0,1) apresentaram eficiências semelhantes para S. aureus. Frente a E. coli, os óleos de gengibre (0,52) e capim cidreira (0,55) foram equivalentes quanto à eficiência. De acordo com as curvas de sobrevivência, foi possível verificar também que os valores de CIM90% obtidos podem ser tanto bactericidas ou bacteriostáticas de acordo com a bactéria testada. Em conclusão, verificou-se que os óleos essenciais testados foram efetivos no controle do desenvolvimento bacteriano, sendo o potencial antimicrobiano diferente em função da espécie bacteriana testada, sendo que a bactéria Gram positiva (S. aureus) mostrou-se mais susceptível aos óleos testados que a Gram negativa (E. coli).

Increase in antimicrobial resistance coupled with successful treatment of various diseases with herbal medications has triggered the upsurge in research geared towards harnessing the medicinal potentials of various plants. The aim of this study was to evaluate the phytochemical composition of Bryophyllum pinnatum, Ocimum gratissimum, Jatropha curcas and Ficus exasperata and their combined antibacterial activity on Escherichia coli, Salmonella typhi, Staphylococcus aureus, and Klebsiella pneumonia. Plant leaves were air–dried, extracted with ethyl-acetate and water with subsequent qualitative analyses for phytochemical compounds. Bacterial pathogens were obtained from University of Benin Teaching Hospital (UBTH), confirmed, and their zones of inhibition studied at 50, 40, 30, 20 and 10 mg/ml of extract combinations. The presence of phenols, tannins and saponins were confirmed in all extracts while alkaloids were present in all ethyl acetate extracts and terpenoids in all aqueous extracts. Different zones of inhibition were measured with the different extract combinations with E. coli having its highest zone of inhibition with the combination of Bryophyllum pinnatum and Ocimum gratissimum in 30 mg/ml (27.25 ± 1.70 mm). However, the highest zone of inhibition observed in the study was with S. aureus in 10 mg/ml concentration of ethyl acetate plant extracts of Ocimum gratissimum and Ficus exasperata (31.75 ± 3.07 mm). The Gram positive cell wall of S. aureus is less complex and therefore, more susceptible to bio-agents. Antibacterial activities of these extracts are attributed to the presence of secondary metabolites that make them good bio-agents for production of antibacterial drugs. AbstrakAntibiotik yang digunakan secara tidak tepat dapat meningkatkan jumlah bakteri penyebab gastroenteritis yang resisten terhadap antibiotik. Beberapa kelompok mikroorganisme tanah diketahui memiliki potensi menghasilkan senyawa aktif untuk menghambat dan membunuh bakteri patogen, seperti Actinomycetes. Tujuan dari penelitian ini adalah untuk mengetahui potensi antibiotik Actinomycetes  terhadap bakteri penyebab gastroenteritis yang diisolasi dari sawah tandus di Desa Cempaka Baru, Kabupaten Kapuas Hulu, Kalimantan Barat. Proses penelitian dilakukan dari isolasi Actinomycetes dari sawah tandus, karakterisasi, serta pengujian kemampuan antibiotik terhadap bakteri Escherichia coli ATCC 25922, Salmonella enterica ATCC 14028 dan Staphylococcus aureus ATCC 25923 menggunakan metode sumuran pada media Mueller Hinton Agar. Hasil identifikasi berdasarkan karakteristik morfologi koloni, sel, dan biokimia diperoleh 1 isolat bakteri, yaitu genus Nocardia sp. ATS-4.1 yang mampu menghambat pertumbuhan bakteri-bakteri uji yang dibuktikan adanya rata-rata zona hambat yang terbentuk, yakni 14,51 mm; 16,16 mm; dan 11,10 mm. Hasil uji statistik Friedman diperoleh nilai Asymp. Sig 0,10>0,05, sehingga menunjukkan bahwa pemberian cairan kultur isolat Nocardia ATS-4.1 memberikan pengaruh yang tidak berbeda nyata antara hambatan terhadap bakteri E. coli, S. enterica, dan S. aureus. Hal ini dapat disimpulkan bahwa isolat Nocardia sp. ATS-4.1 berpotensi menghasilkan antibakteri yang setara terhadap bakteri E. coli, S. enterica, dan S. aureus penyebab gastroenteritis.Abstract Using antibiotics incorrectly increases the number of resistant bacteria to gastroenteritis. Soil microorganisms are known to have the potential to produce active compounds to inhibit and kill pathogenic bacteria,  for example, Actinomycetes. The purpose of this study was to determine the potential of antibiotics Actinomycetes isolated from rice fields in Cempaka Baru, Kapuas Hulu Regency, West Kalimantan, against gastroenteritis bacteria. The research order was carried out by isolating Actinomycetes from barren rice fields, and then characterizing and testing the ability of antibiotics against Escherichia coli ATCC 25922, Salmonella enterica ATCC 14028, and Staphylococcus aureus ATCC 25923 using the well method on Mueller Hinton Agar. The identification results based on morphological characteristics of colonies, cells and biochemistry showed that one of the bacteria was from genus Nocardia sp. ATS-4.1 which was able to inhibit the three test bacteria by the average inhibition zone 14.51 mm; 16.16 mm; and 11.10 mm, respectively. Friedman’s statistical test resulted in values asymp. sig 0.10>0.05 which showed that the isolate gave inhibition differences insignificantly among the bacteria. In conclusion, isolate Nocardia sp. ATS-4.1 able to produce equal antibacterial activity against bacteria E. coli, S. enterica and S. aureus caused gastroenteritis.

Abstract Background Carbapenems are broad-spectrum antibacterials that have seen increased usage for the Enterobacteriales family in recent years. While carbapenem usage has been associated with increased antibacterial resistance, there is currently a lack of data comparing the risk of reduced susceptibility selection by the two most commonly used carbapenems in the US, ertapenem (ERT) and meropenem (MER). We conducted a novel serial passage experiment with clinical isolates of Enterobacteriales to assess the impact of repeated exposure to ERT or MER on phenotypic susceptibility patterns. Methods Non-duplicate clinical Enterobacteriales isolates were selected randomly for inclusion. Antimicrobial susceptibility testing was performed by CLSI disc diffusion methods. Standardized suspensions of isolates were plated on Mueller-Hinton agar, and ERT (10mcg) and MER (10mcg) discs applied. Zones of inhibition were measured and recorded after 16-18 hours incubation. Growth from the innermost zone of inhibition around each disc was used to prepare subsequent suspensions for serial susceptibility testing. This process would be repeated daily for 10 days. Each subsequent serially-passaged isolated was tested against both ERT and MER. Daily zones of inhibition were measured and interpreted. Baseline & final susceptibilities were determined by automated methods (Vitek 2). Results Seventeen Enterobacteriaceae isolates were selected, including: Klebsiella pneumoniae (n=11), Klebsiella oxytoca (n=2), Escherichia coli (n=1), Morganella morganii (n=1), and Enterobacter cloacae (n=2). Despite a greater degree of reductions in zones of inhibition with repeated ERT exposure (vs MER), the overall 10 day trends were not found to be significant different (P=0.529). Resistance developed to ERT in six isolates compared to one MER-resistant isolate (P = 0.053). E. cloacae was the only species to show a significant change between drugs (P=0.010). Two of three isolates that developed reduced zone changes > 10mm to MER were initially exposed to ERT on an earlier plate. Conclusion This novel experiment identified the development of some nonsignificant reductions in susceptibility with ERT after serial exposure. Results from this pilot study should encourage larger well-designed studies in this area. Disclosures All Authors: No reported disclosures

A resistência microbiana tem se mostrado um problema de proporções mundiais, causando estado de morbidade e mortalidade em diversos pacientes. Em vista disso, tem crescido a busca por métodos alternativos naturais de profilaxia. A investigação clínica sugere que o Extrato de Cranberry está entre as melhores propostas de prevenção natural. O Cranberry (Vaccinium macrocarpon) é um fruto que tem crescido comercialmente pelo sabor e propriedades benéficas à saúde. Dentre as formas comercializadas estão: o suco, o chá e as cápsulas contendo o extrato seco. A ação desta planta está relacionada ao tratamento de doenças do trato urinário, por possuir substâncias que inibem a adesão bacteriana ao epitélio do trato urinário, dificultando sua proliferação e reprodução. Dentre todas as infecções relacionadas à assistência a saúde, a Infecção do Trato Urinário é a mais frequentemente associada a procedimentos invasivos. Se não for tratada, pode resultar em complicações como pielonefrite aguda, bacteremia e pionefrose. Portanto, cranberry pode ser uma nova alternativa para o combate das infecções uroepiteliais, por ser um produto natural de preço acessível, e com formas de comercialização diversificada, ao contrário dos antimicrobianos convencionais, que por sua vez são caros e podem acabar causando resistência nos micro-organismos. Este trabalho teve como objetivo avaliar in vitro a atividade antimicrobiana do extrato de Cranberry, adquirido em farmácia de manipulação, sobre 8 micro-organismos isolados de infecções urinárias. As cepas utilizadas, adquiridas da coleção da FIOCRUZ, foram: Escherichia coli, Pseudomonas aeruginosa, Klebsiella pneumoniae, Proteus mirabilis, Serratia marscecens, Staphylococcus aureus, Enterococcus faecalis e Enterococcus faecium. No estudo, foram utilizados o caldo Mueller Hinton (MH), Extrato de Cranberry e as bactérias patogênicas. O ensaio foi realizado em triplicata, com o uso de um controle de crescimento dos micro-organismos e o experimento para avaliação do crescimento bacteriano na presença do extrato. A turbidez foi medida com o auxílio de um espectrofotômetro, no comprimento de onda de 600 nm, antes e após 24 horas de incubação à 37 ºC. O procedimento forneceu a Densidade Ótica, do qual possibilitou a identificação da inibição microbiana. Para análise estatística foi utilizado o Teste t de Student. O Extrato de Cranberry apresentou atividade antimicrobiana sobre as bactérias Staphylococcus aureus, Klebsiella pneumoniae, Escherichia coli, Serratia marscecens e Enterococcus faecalis (p < 0,05), confirmando seu efeito benéfico em infecções urinárias. No entanto, não teve efeito inibitório significativo sobre Pseudomonas aeruginosa, Proteus mirabilis e Enterococcus faecium (p > 0,05).

The ethanol extracts of root, bark and leaf of Bridelia ferruginea was investigated for antibacterial activity against clinical isolate of Staphylococcus aureus and Escherichia coli. The extracts had significant antibacterial activity in vitro at concentration of 25 mg/ml, 50 mg/ml, 100 mg/ml and 200 mg/ml and in vivo at dose of 50 mg/kg and 100 mg/kg. The root extract in vitro had the highest zone of inhibition, followed by the bark extract for both Staphylococcus aureus and Escherichia coli. The concentration of 200 mg/ml had the highest zone of inhibition in vitro. The minimum inhibitory concentration (MIC) showed a decreasing inhibitory effect of the plant extracts for both Staphylococcus aureus and Escherichia coli as the concentration decreases with root having 3.125 mg/ml, bark having 6.25 mg/ml and leaf having 25 mg/ml for Staphylococcus aureus and Escherichia coli. Likewise, the minimum bactericidal concentration (MBC) showed decreasing bactericide effects with decrease concentration with root having 12.5 mg/ml, bark having 12.5 mg/ml and leaf having 25 mg/ml for Escherichia coli while root had 6.25mg/ml, bark had 12.5mg/ml and leaf had 25mg/ml for Staphylococcus aureus. The in vivo investigation showed that the root and bark extract exhibited antibacterial activity on both Staphylococcus aureus and Escherichia coli at doses of 100mg/kg and 50mg/kg; the root extract had higher activity than the bark and root/bark combined. The dose of 100 mg/kg had the highest colonies reduction for Staphylococcus aureus and Escherichia coli in vivo. Preliminary phytochemical screening of root, bark and leaves of Bridelia ferruginea revealed the presence of tannins, flavonoids, carbohydrates, cardiac glycoside (root, bark and leaves), saponins (root and bark). The presence of tannins, saponins, flavonoid, cardiac glycoside and carbohydrate in the bark and root extracts of the plant indicates that the bark and root extracts were pharmacological importance

The aim of this study was to investigate the antibacterial effect and identify the phytochemical constituents of Moringa oleifera leaves and seeds extract on Staphylococcus aureus (S. aureus) clinical isolates using agar well diffusion method. The samples were collected from the premises of Usmanu Danfodiyo University, Sokoto. The Seeds and Leaves were collected fresh. They were extracted using methanol and ethyl acetate. Various concentrations from 100mg/ml to 500mg/ml were prepared. The test bacteria used is Staphylococcus aureus obtained from Microbiology laboratory of the Usmanu Danfodiyo University. The bacteria were re-identified using biochemical tests. The bacterial inoculums were standardized to McFarland scale 0.5. Zone of inhibition were read after 24 hours of incubation at 370C.The results of the antibacterial study revealed that the methanolic leaves extracts at 500 mg/ml had effect on S. aureus with zone of inhibition of 20mm. The methanolic seed extract have effect on S. aureus with zone of inhibition of 19.5mm. The MIC for the leave and seed extracts for Staphylococcus aureus was 250mg/ml. The MBC was 500mg/ml. The results of the phytochemical analysis revealed the presence of flavonoid, tannins, saponins, cardiac glycosides, alkaloids, volatile oil, saponin glycosides, and glycosides but anthraquinone and steroids were absent in the extracts. The zones of inhibition showed that both the methanolic and ethyl acetate extracts at 500mg/ml were active to all the tested bacteria. ANOVA and Duncan Multiple Mean Range test was used to analyze the data. Based on Duncan’s grouping, there is significant difference between the solvents and the concentrations used.
