Increase in antimicrobial resistance coupled with successful treatment of various diseases with herbal medications has triggered the upsurge in research geared towards harnessing the medicinal potentials of various plants. The aim of this study was to evaluate the phytochemical composition of Bryophyllum pinnatum, Ocimum gratissimum, Jatropha curcas and Ficus exasperata and their combined antibacterial activity on Escherichia coli, Salmonella typhi, Staphylococcus aureus, and Klebsiella pneumonia. Plant leaves were air–dried, extracted with ethyl-acetate and water with subsequent qualitative analyses for phytochemical compounds. Bacterial pathogens were obtained from University of Benin Teaching Hospital (UBTH), confirmed, and their zones of inhibition studied at 50, 40, 30, 20 and 10 mg/ml of extract combinations. The presence of phenols, tannins and saponins were confirmed in all extracts while alkaloids were present in all ethyl acetate extracts and terpenoids in all aqueous extracts. Different zones of inhibition were measured with the different extract combinations with E. coli having its highest zone of inhibition with the combination of Bryophyllum pinnatum and Ocimum gratissimum in 30 mg/ml (27.25 ± 1.70 mm). However, the highest zone of inhibition observed in the study was with S. aureus in 10 mg/ml concentration of ethyl acetate plant extracts of Ocimum gratissimum and Ficus exasperata (31.75 ± 3.07 mm). The Gram positive cell wall of S. aureus is less complex and therefore, more susceptible to bio-agents. Antibacterial activities of these extracts are attributed to the presence of secondary metabolites that make them good bio-agents for production of antibacterial drugs. Human pathogenic microorganisms have developed resistance in response to indiscriminative use of commercial drugs. Plants produce many secondary metabolites with microbiocidal activity hence their use in traditional medicine. Herbalists in Kenya use medicinal plants including Solanum incanum in treating microbial infections. Though S. incanum has been used to treat different diseases in humans and animals, there is little information on antimicrobial activities of its extracts against Escherichia coli and Staphylococcus aureus. In this study, phytochemical analysis and antibacterial activity of solanum incanum leaves, roots and seeds extracts were determined. Ethanolic and aqueous extracts of leaf, root and seed of concentrations 25, 50, 75 and 100, and amoxicillin 25 mg/ml (control) with three replications were used for antibacterial analysis by the agar-well diffusion method. The results were subjected to analysis of variance at P < 0.05. Phytochemical screening revealed the presence of alkaloids, flavonoids, glycosides, saponins, steroids and tannins. Solanum incanum exhibited significant antibacterial effect against the two test bacteria. Ethanol extracts were more active than extracts against the bacteria. Ethanol extracts at 100% inhibited growth of Staphylococcus aureus more than the Escherichia coli. The zones of inhibition for Staphylococcus aureus were 35.0±0.6 mm, 30.94±0.3 mm and 30.14±0.64mm for seed, root and leaves respectively. On the other hand, the zones of inhibition for Escherichia coliat 100% ethanol were 27.20±0.06, 23.14±0.12 and 21.0±0.4 seed, root and leaves respectively. The results validate the use of these plants in ethnomedicine and potential of this plant in treating infections caused by the two bacteria.

Phytochemical analysis, antimicrobial and antioxidant properties of the leaf extract and fractions of Sabicea brevipes were studied. The leaves were defatted and the marc extracted with methanol. The extract was further purified by solvent-solvent partitioning using n-hexane, ethyl acetate and n-butanol to obtain the three solvent fractions. They were screened for phenolics, flavonoids, tannins, saponins, terpenoids, glycosides, and steroids. Total phenolics, flavonoids and tannins were determined quantitatively. The antimicrobial test was screened in vitro by agar diffusion method. Analysis of variance (ANOVA) was used to test for significant difference at p ≤ 0.05 in all study groups. The methanol extract exhibited the most significant amount of phenolics (110.78 ± 1.06 mg GAE/g) while ethyl acetate fraction had the least total phenolics content (50.55 ± 2.91 mg GAE/g). The same trend was observed for the total flavonoids content whereas the methanol extract measured (418.40 ± 14.03 mg QE/g) while ethyl acetate fraction had 192.40 ± 3.06 mg QE/g. Total tannins contents were: methanol extract (102.22 ± 7.58 mg GAE/g) and ethyl acetate (27.33 ± 0.77 mg GAE/g). The antioxidant results showed that the methanol extract had the highest DPPH free radical scavenging ability (93.69%) with half maximal inhibitory concentration (IC50) of 0.601± 0.02 and also highest ferric ion reducing power (50.381 ± 1.56 µmol Fe2+/g). Also, the methanol extract showed high total antioxidant capacity (96.79 ± 0.31 mg AAE) and IC50 of 0.798± 0.01. The antimicrobial results revealed that the methanol extract showed better activity against Escherichia coli, Bacillus subtilis, Pseudomonas aeruginosa, Staphylococcus aureus, and Candida albicans than the solvent fractions at concentrations of 200, 100, 50, 25, 12.5, mg/mL having various inhibition zone diameters (IZDs). The methanol extract and fractions of S. brevipes compared favourably in terms of zone of inhibition and minimum inhibition concentration (MIC) with the standard drug disc (Gentamycin and Ketoconazole) against the tested microorganisms. The MIC of the extract and solvent fractions ranged from 6.31 mg/mL to 50.12 mg/mL. The continual use of the extract of Sabicea brevipes in preventing oxidative stress and in the treatment of common infection is justified by these results.
