Phytochemical Investigation and Spectrophotometric Studies of Piperine and Thymoquinone Compounds in Methanolic Extracts of Nigella sativa and Piper nigram spices

This study was carried out on some spice samples including Nigella sativa and Piper nigram. Phytochemical screening was applied to the methanolic extraction of both the studied plants. The results showed that methanolic extract of Nigella sativa and Piper nigram revealed the presence of various chemical constituents, such as Alkaloide, Carbohydrates, Glycosides, Tannins, Flavones, Saponins. Piperine is characterizing compound present in fruits of piper nigram used as a bioavailability enhancer. Some spectrophotometric methods as (U.V) and I.R were applied to identify the major chemical compounds of the studied extracts. Polyphenolic enriched extracts from two species of Cyperus, Cyperus glomeratus and Cyperus thunbergii, possess mammalian arginase inhibitory capacities, with the percentage inhibition ranging from 80% to 95% at 100 µg/mL and 40% to 64% at 10 µg/mL. Phytochemical investigation of these species led to the isolation and identification of two new natural stilbene oligomers named thunbergin A-B (1–2), together with three other stilbenes, trans-resveratrol (3), trans-scirpusin A (4), trans-cyperusphenol A (6), and two flavonoids, aureusidin (5) and luteolin (7), which were isolated for the first time from C.thunbergii and C. glomeratus. Structures were established on the basis of the spectroscopic data from MS and NMR experiments. The arginase inhibitory activity of compounds 1–7 was evaluated through an in vitro arginase inhibitory assay using purified liver bovine arginase. As a result, five compounds (1, 4–7) showed significant inhibition of arginase, with IC50 values between 17.6 and 60.6 µM, in the range of those of the natural arginase inhibitor piceatannol (12.6 µM). In addition, methanolic extract from Cyperus thunbergii exhibited an endothelium and NO-dependent vasorelaxant effect on thoracic aortic rings from rats and improved endothelial dysfunction in an adjuvant-induced arthritis rat model.

Ayurvedic medicines are being used increasingly to fight or prevent common diseases. Udhara vayuhara churna is a polyherbal formulation containing Piper longum (Piperceae) Zingiber officinalis (Zingiberaceae), Elettaria cardamom (Zingiberaceae), Plumbago zeylanica (Plumbaginaceae), Carum carvi (Umbelliferae), Acorus calamus (Araceae) and Embilica officinalis (Euphorbiaceae). Methanolic and aqueous extracts of Udhara vayuhara churna were screened for their antimicrobial activity against Bacillus subtilis, Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Aspergillus niger and Candida albicans. The results of antimicrobial activity of aqueous and methanolic extracts of the Udhara vayuhara churna indicated that methanolic extract inhibited the growth of one or more test pathogens than aqueous extract. Udhara vayuhara churna formulation extracts showed a broad spectrum of antimicrobial activity. Phytochemical investigation revealed the presence of tannins, saponins, alkaloids, glycosides, flavonoids and essential oils. This may be due to the multifunctional effect of all the seven plant ingredients of Udhara Vayuhara churn.

Alhagi maurorum (A. maurorum) is one of the medicinally important plants belonging to the family leguminasae, commonly known as camel thorn. This research was amid to identify the chemical compounds in the aerial part of A. maurorum using GC-mass analysis. Three solvents with different polarities were used for the extraction of chemical constituents (water, methanol and petroleum ether). The results of GC-MS analysis led to identification of various compounds. In total, thirty-nine compounds from petroleum ether extract, thirty-two compounds in methanolic extract and seventeen compounds in aqueous extract were identified. Majority of the identified compounds have been reported to possess many biological activities. Among them, we reported 10 new anticancer compounds (Vitamin E; Hexadecanoic acid; Stigmast-5-en-3-ol; Phytol,2-hexadecen-1-ol,3,7,11,15-tetramethyl; Squalene; Hexadecanoic acid; 2-hydroxy-1-(hydroxymethyl) ethyl ester; Oxime,methoxy-phenyl,methyl N-hydroxyben-zenecarboximidoate; Ergost-5-en-3-ol; 9,12- Octadecad-ienoic acid and Farnesol) from A. maurorum using three solvent, while the best effective solvent was petroluem ether. Therefore, we report that A. maurorum has great potential to be developed into anticancer drugs.

Therapeutic properties of the medicinal plant are due to the presence of phytochemical constituents. The phytochemical constituents of Plumeria alba flower were investigated by phytochemical screening assays and gas chromatography-mass spectrometry (GC-MS). The phytochemical screening of hexane, dichloromethane, ethyl acetate, butanol, and aqueous extracts of P. alba flower showed it contains a various concentration of saponins, flavonoids, tannins, steroids, volatile oil and phenolic compounds. Several major chemical constituents that were identified is squalene, bis(2-ethylhexyl) phthalate, methyl (methyl 4-O-methyl-α-d-mannopyranoside) uronate and tricyclo [7.2.0.0(2,6)] undecan-5-ol, 2,6,10,10-tetramethyl- (isomer 2) by using GC-MS technique.

Aims: To determine chemical constituents of the Leaf extracts of Centella asiatica using the LC-MS and GC-MS and their antimicrobial activities. Study Design: Structural determination of compounds from the leaf extracts was done using GC-MS and LC-MS analysis. The antimicrobial properties of the extracts were done using disc diffusion method. Place and Duration of Study:  Pure and Applied Chemistry Department, Masinde Muliro University of Science and Technology, Kenya: Between 2016-2019. Methodology: Plant materials of C. asiatica were sequentially extracted separately based on the polarity viz., hexane, ethyl acetate and methanol. Determination of chemical constituents was done using LC-MS and GC-MS analysis and phytochemical screening. The extracts were assayed against Staphylococcus aureus, Klebsiella pneumonia, Escherichia coli and Candida albicans. Standard antimicrobials viz. ampicillin and Nystatin were used as the control. Disc diffusion method was used and zones of inhibition, after respective incubation periods, were used to quantify antibacterial and antifungal activity. Results: Phytochemical screening of the hexane, ethyl acetate and methanolic extracts showed that terpenoids, flavonoids, saponins, alkaloids, steroids, amino acids and proteins, reducing sugars and carbohydrates were present. LC-MS and GC-MS analyses of the methanolic extracts identified 22 and 33 compounds, respectively, by use of the national institute of standards and technology (NIST) library. The extracts showed appreciable activity against common microbes tested. Conclusion: This study forms the basis for the biological characterization and significance of the compounds identified in the leaf and stem extracts of C. asiatica. These compounds are known to possess antibacterial and antifungal activities that could be established as potential candidates for future drug development. However, these extracts, need to be subjected to further chromatographic procedures to isolate the identified compounds and their bioactivities determined.

The present study was aimed to investigate the antimicrobial potential of leaf extracts of Euphorbia paralias and Melilotus sulcatus against four bacterial species Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli and Klebsiella sp. and two fungal species Asperigillus niger and Aspergillus flavus. The agar well diffusion assay was used to evaluate the antimicrobial activity. The effect of these extracts was most effective against the bacterial species compared to the fungal species at a used concentration (100 mg/ml). Methanolic extracts of selected plants displayed good antimicrobial activity against all tested microorganisms species, while, no activity for aqueous extracts against tested fungal species. Methanolic extracts were the most effective plant extracts against all tested bacterial species, with MIC and MBC reached 6.2 and 12.5 mg/ml, except Klebsiella sp. which was less sensitive to M. sulcatus methanolic extract and its MIC and MBC reached 12.5 and 25 mg/ml, respectively. These plant extracts which proved to be potentially effective can be used as bioactive agents to control microorganisms caused for diseases and they can be used naturally in the human and veterinary healthcare systems.

The study was designed to evaluate the phytoconstituents present in the methanolic extract of aerial parts of Abelmoschus moschatus. The qualitative phytochemical screening of different extracts of aerial parts revealed the presence of some bioactive compounds. GC – MS analysis was performed using Shimadzu Gas chromatography-mass spectroscopy (Model Number: QP2010S) instrument. GC-MS detection of phytoconstituents was done by computer evaluation of mass spectra of samples through National Institute Standard and Technology (NIST II) and WILEY 8 library. GC – MS analysis detected the presence of 14 compounds. GC – MS profile of the methanolic extract revealed the presence of megastigmatrienone, phytol, loliolide, farnesyl acetate, methyl linoleates, gamma-sitosterol, cis, cis, cis-7,10,13-Hexadecatrienal, thymine, pyranone, coumarin, 2 – methoxy 4 – vinyl phenol, guanosine, chinasaure and 3- cyclopentyl propionic acid 2 dimethyl aminoethyl ester. The current study suggests that methanolic extracts of aerial parts of Abelmoschus moschatus contain phytoconstituents with antioxidant and cytoprotective activity. The study results will pave a way for the production of therapeutic agents which can be used for the treatment of various diseases.

Inula viscosa (L.) is a medicinal plant that has been used for a long time for its many therapeutic properties, especially to treat pain and inflammation. The aim of this study was to evaluate the anti-inflammatory, analgesic, and antipyretic activities of methanolic extracts and decoctions of Inula viscosa leaves and flowers. The phenol content of the extracts was determined by spectrophotometric methods. The extracts were administered orally (400, 600, and 800 mg/kg) to mice and/or rats in the study of antiinflammatory (carrageenan-induced paw edema test), analgesic (acetic acid-induced writhes test), and antipyretic (Brewer’s yeast-induced pyrexia method) activities. The highest phenolic content was found in leaf methanolic extract. Methanolic extracts and decoctions of leaves and flowers showed significant decrease in mice paw edema after 4 h (P < 0.01; P < 0.001), and an appreciable analgesic effect at the tested doses with a maximum of writhing inhibition observed with leaf methanolic extract at the dose of 800 mg/kg (93.39%; P < 0.001). The rectal temperature of the rats decreased significantly (P < 0.01; P < 0.001) after 4 h in groups treated with leaves and flowers extracts at the doses of 600 and 800 mg/kg. The obtained results confirm the traditional uses of Inula viscosa.

The phytochemical screening of Dipsacus mitis D. Don and Woodfordia fruticosa (L.) Kurz revealed the presence of bioactive secondary metabolites in both of the species but their concentrations were variable. Flavanoids and saponins were found in remarkable amount in both of the samples. The volatile compounds of both plants were extracted by Simultaneous Steam Distillation and Extraction (SDE) apparatus and analyzed by Gas Chromatography/Mass Spectrometery (GC-MS). Total 53 volatile compounds were tentatively identified from roots of Dipsacus mitis D. Don and 78 were identified from flowers of Woodfordia fruticosa (L.) Kurz. Alcohols were dominant in both the plants accounting 45% and 32% respectively. Numerous bioactive volatile compounds were detected in both herbs. The effects of increasing concentrations of the methanolic extracts on the amplitude and frequency of spontaneously contracting uterine tissues were tested. The effect of extracts on the smooth muscle strips from rat uterus showes the dramatic muscular relaxation on spontaneous contractility and was determided most effective at a concentration 6500 µg/ml of methanolic extract of Dipsacus mitis D.DOI: http://dx.doi.org/10.3126/jnpa.v26i1.6627JNPA. <p class="Abstract">The present study is aimed to investigate the phytochemical screening and biological activities of methanolic extract of <em>Cyperus scariosus</em> roots. Dried plant was grounded and extracted with methanol to prepare methanol crud extract. In vitro biological tests were conducted using this methanolic extracts according to the standard procedure. 100% death rate of brine shrimp was perceived at 3 mg/mL of plant extract after 72 hours. The extract showed action against <em>Aspergillus flavius</em> i.e. 90% followed by A. niger (91%) while the highest activity was shown against <em>A. fumegatrus</em> (94%). Important scavenging results were detected during scavenging of free radicals viz; 92.2% against DPPH, 82.2% to ABTS, 75.8% to hydrogen peroxide, 88.1% to β-carotene, 86.1% to hydroxyl radical and 89.4% against phosphomolybdate at 3 mg/mL were obtained. The results obtained in this study point out that extract showed significant biological activities which might be due to the presence of bioactive constituents.</p><p> </p>
