Myostatin, or growth and differentiation factor-8(GDF-8) is a member of the transforming growth factor (TGF)-ß superfamily. The objective of this study was to investigate the association between genetic polymorphism of the myostatin gene with the body-weight of Iraqi local ducks by DNA sequence analysis. The results of DNA sequence showed nitrogen base change (G?A) at 129 base pair sites of the exon1 of MSTN gene. Therefore, two alleles, G and A and two patterns of genotypes, GG, AG were observed. The observed numbers of GG and AG genotypes were 25 and 6, respectively. The G allele was of the highest frequency (0.9) while A allele showed the lowest frequency (0.1). The GG genotype showed the highest frequency (80.65%) and AG genotype showed the lowest frequency (19.35%). Additionally, the percentages of observed and expected heterozygosity were 0.19 and 0.17 respectively. Moreover, the percentages of observed and expected homozygosity were 0.80 and 0.82, respectively. The average heterozygosity was 0.17. The results revealed no significant (P?0.05) effect of the MSTN gene on body weight.

Polymorphism of myostatin gene and its association with body weight traits in a hybrid of GAMA chicken (Gallus gallus domesticus Linn. 1758). Biodiversitas 20: 3207-3212. An experiment was conducted to detect SNP of the myostatin gene and its association with the body weight of hybrid chicken crossbreed from F1 Kamper and BC1 Broiler. Four F1 Kamper hens were crossbred with BC1 Broiler rooster. Day old chick (DOC) hatched were maintained for 49 days with body weight measurement every seven days. The blood samples from 49 days old chicken were taken for DNA isolation by Chelex 5% method and then amplification of the myostatin gene. PCR products were sequenced, and sequence alignment was performed using Clustal Omega to obtain SNP. The SNP obtained was analyzed by the Pearson correlation test with the body weight of forty nine-days-old chickens. The body weight of the hybrid chicken is higher than of Pelung chicken but lower than the Broiler. There are 7 SNPs in myostatin gene exons included 2 Adenine insertions, 1 Guanine deletion, and four substitutions (C2244G, G2283A, T4842G, G7378T) that yield nine haplotypes. Six haplotypes had different protein sequences with Myostatin protein, while three haplotypes were identical to Myostatin protein. The correlation analysis showed that there was a strong positive correlation (r = 0.736) between normal Myostatin protein and mutant to chicken body weight at 49-days-old. Adenine insertion to nucleotide 2099-2100 of myostatin gene had a very strong positive correlation (r = 0.800) to 49-days-old chicken body weight, although T4842G substitution had a strong negative relationship (r = -0.773) to 49-days-old chicken body weight. Adenine insertion to nucleotide 2099-2100 of myostatin gene could be a genetic marker of heavier body weight of the hybrid chicken.

Background and Aim: Myostatin (MSTN), a member of the transforming growth factor-β family, is a negative regulator of muscle mass. This study aimed to detect the genetic variation of the 1160 bp fragment of exon 1 and part of intron 1 of the MSTN gene in several cattle populations raised in Indonesia. Materials and Methods: Polymerase chain reaction products of the MSTN gene amplified from 92 animals representing 10 cattle populations (Peranakan Ongole [PO], Belgian Blue x PO cross, Rambon, PO x Bali cross, Jabres, Galekan, Sragen, Donggala, Madura, and Bali) were sequenced, compared, and aligned with bovine MSTN of Bos taurus (GenBank Acc. No. AF320998.1) and Bos indicus (GenBank Acc. No. AY794986.1). Results: Four nucleotide substitutions (nt 1045 and 1066 in intron 1; nt 262 and 418 in exon 1) and two indels (nt 807 and 869 in intron 1) were synonymous mutations. Among these substitutions, only the nt 262G>C and nt 418A>G loci were polymorphic in all populations, except Bali cattle. The frequencies of the nt 262C (0.82) and nt 418A (0.65) alleles were highest. For the nt 262G>C locus, the CC genotype had the highest frequency (0.66) followed by GC (0.30) and CC (0.03). For the nt 418A>G locus, the AG genotype had the highest frequency (0.52) followed by AA (0.39) and GG (0.09). Conclusion: The results, showing genetic variations in exon 1 and intron 1 of the MSTN gene, might be helpful for future association studies.

The aim of present study was to investigate myostatin gene polymorphism and its association with weaning weight and body weight in Batur sheep, 30 heads of Batur all lambs were fed by concentrated feed. Body weight measured monthly after weaning to the six months of age. DNA Extraction used 200 ul of whole blood samples. To amplify exon 3 region of MSTN gene a specific primer designed using the Primer3 software. The 25 μl volume contained 25 ng of genomic DNA, 12.5 μl 2x Reaction mix of each primer. The cycling protocol was 5 minutes at 95°C as initial denaturation, 35 cycles of denaturing at 94°C for 45 seconds, annealing at 73.9 for 45 seconds, extending at 72°C for 40 s, with final extension at 72°C for 10 minutes. Eleven polymorphic sites were observed in the in 3rd exon region transversions at c.*121 G instead of A, del-T at c.*129, one individual at c.*139 and one individual at c.*158 positions however, one individual sequence disrupted reading frame in whole MSTN sequenced, also weaning weight and 6-month weight of Batur lambs were 21.13±5.70 and 31.64±7.06 to the homozygous whilst 19.99±4.33 and 30.50±5.44 for heterozygote respectively but there is no significe differences seen above all the studied parameters. Homozygous AA lambs had fairly low weaning weight and body weight compared to the heterozygous AB lambs but might have a higher carcass weight, further investigation is needed into the interaction of MSTN with other genes involved in muscle growth.

The aim of the present study is to analyze the effects of 5-aminoisoquinoline (5-AIQ), a poly(ADP-ribose) polymerase-1 (PARP1) inhibitor, over renal dysfunction and fibrosis during recovery phase of cisplatin (CisPt)-induced acute kidney injury (AKI) in rats. Male Wistar rats were distributed in three groups (n=8 each group): control, CisPt, and CisPt + 5-AIQ. Control and CisPt groups received a subcutaneous injection of either saline or 7 mg/kg CisPt, respectively. CisPt + 5-AIQ group received two intraperitoneal injections of 10 mg/kg 5-AIQ 2 h before and 24 h after CisPt treatment. Thirteen days after the treatment, rats were housed in metabolic cages and 24-h urine collection was made. At day 14, CisPt-treated rats showed increased diuresis, N-acetyl-β-d-glucosaminidase (NAG) excretion, glucosuria and sodium fractional excretion (NaFE), and decreased creatinine clearance (CrCl). 5-AIQ significantly increased CrCl and decreased NAG excretion, glucosuria, and NaFE. In plasma, CisPt increased sodium, urea, and creatinine concentrations, while 5-AIQ treatment decreased these variables to the levels of control group. 5-AIQ completely prevented the body weight loss evoked by CisPt treatment. CisPt also induced an increased renal expression of PAR polymer, α-smooth muscle actin (α-SMA), transforming growth factor-β1 (TGF-β1), and collagen-IV. These variables were decreased in CisPt + 5-AIQ group. Tubular lesions and renal fibrosis were also decreased by 5-AIQ treatment. We conclude that inhibition of PARP1 with 5-AIQ can attenuate long-term nephrotoxic effects associated with the CisPt treatment, preventing renal dysfunction and body weight decrease and ameliorating tubular lesions and collagen deposition.

This study aimed to identify the variation of 5’UTR Myostatin (MSTN) gene and its association to body weight in Boer, Kacang, and Boerka goats. DNA samples were obtained from 149 heads of the goats from the Indonesian Goat Research Center, Sungei Putih, North Sumatera. Polymorphism identification was conducted by direct sequencing and PCR-RFLP with DraI as the restriction enzyme for indel g.1256/TTTTA. Analysis of variance for body weight was carried out using the General Linear Model (GLM). The 5’UTR MSTN gene|DraI was monomorphic in Kacang but polymorphic in Boer and Boerka. Genotype frequencies for Boer 0.40(AA), 0.43(AB), 0.17(BB); Kacang 1.00(AA); Boerka 0.53(AA), 0.37(AB), 0.10(BB). The allele frequencies for Boer 0.62(A), 0.38(B); Kacang 1.00(A); Boerka 0.72(A), 0.28(B), respectively. AB was the most frequent genotype among Boer, but AA was the most frequent in Kacang and Boerka. Indel g.1256/TTTTA has a significant effect (P<0.05) only on birth weight (BW) of Boer, but no significant effect on other bodyweight parameters both in Boer and Boerka.AA genotype has the highest BW (P<0.05) than AB, but it’s not significantly different from BB. Indel g.1256/TTTTA could be used as a genetic marker for the birth weight of Boer but not in Boerka goats.

The aim of the study was to identify polymorphisms within nuclear DNA genes and determine their association with the performance traits of farmed raccoon dogs. The study involved 354 animals and their breeding documentation data. Myostatin (MSTN), insulin-like growth factor 1 (IGF1), and growth hormone (GHR) genes were amplified, sequenced, and subjected to bioinformatics and statistical analysis. Estimation of variance components was performed with the residual maximum likelihood (REML) method, and best linear unbiased prediction (BLUP) of the breeding values was carried out. The predictors of the traits of direct additive, maternal additive, and random-specific maternal environmental effects were analysed. Within the nucleotide sequences of the analysed genes, one silent single nucleotide polymorphism (SNP) was identified in exon 1 of the MSTN gene. The analysis of the variance of the fixed-specific maternal environmental effect revealed statistically significant differences in the body weight among raccoon dogs with alternative polymorphisms. The mean effects of the estimators of polymorphism addition and dominance measured by MSTN gene regression had mostly a significant impact on the level of the traits estimated in animals. There was statistically significant association of the polymorphism in the MSTN gene with the body weight in raccoon dogs, which validates selection thereof as a candidate gene for this economically most important performance trait of raccoon dogs.

Previously, we have found that the addition of L-ascorbic acid to chitosan enhanced the reduction in body weight gain in guinea pigs fed a high-fat diet. We hypothesized that the addition of L-ascorbic acid to chitosan would accelerate the reduction of body weight in humans, similar to the animal model. Overweight subjects administered chitosan with or without L-ascorbic acid for 8 weeks, were assigned to three groups: Control group (N = 26, placebo, vehicle only), Chito group (N = 27, 3 g/day chitosan), and Chito-vita group (N = 27, 3 g/day chitosan plus 2 g/day L-ascorbic acid). The body weights and body mass index (BMI) of the Chito and Chito-vita groups decreased significantly (p < 0.05) compared to the Control group. The BMI of the Chito-vita group decreased significantly compared to the Chito group (Chito: -1.0 kg/m2 vs. Chito-vita: -1.6 kg/m2, p < 0.05). The results showed that the chitosan enhanced reduction of body weight and BMI was accentuated by the addition of L-ascorbic acid. The fat mass, percentage body fat, body circumference, and skinfold thickness in the Chito and Chito-vita groups decreased more than the Control group; however, these parameters were not significantly different between the three groups. Chitosan combined with L-ascorbic acid may be useful for controlling body weight.

Introduction: Prevalence of obesity is 30 % in the Czech Republic and is expected to increase further in the future. This disease complicates surgical procedures but also the postoperative period. The aim of our paper is to present the surgical technique called hand-assisted laparoscopic nephrectomy (HALS), used in surgical management of kidney cancer in morbid obese patients with BMI >40 kg/m2. Methods: The basic cohort of seven patients with BMI >40 undergoing HALS nephrectomy was retrospectively evaluated. Demographic data were analyzed (age, gender, body weight, height, BMI and comorbidities). The perioperative course (surgery time, blood loss, ICU time, hospital stay and early complications), tumor characteristics (histology, TNM classification, tumor size, removed kidney size) and postoperative follow-up were evaluated. Results: The patient age was 38−67 years; the cohort included 2 females and 5 males, the body weight was 117−155 kg and the BMI was 40.3−501 kg/m2. Surgery time was 73−98 minutes, blood loss was 20−450 ml, and hospital stay was 5−7 days; incisional hernia occurred in one patient. Kidney cancer was confirmed in all cases, 48–110 mm in diameter, and the largest removed specimen size was 210×140×130 mm. One patient died just 9 months after the surgery because of metastatic disease; the tumor-free period in the other patients currently varies between 1 and 5 years. Conclusion: HALS nephrectomy seems to be a suitable and safe surgical technique in complicated patients like these morbid obese patients. HALS nephrectomy provides acceptable surgical and oncological results.

The article presents the data on the study of the influence of mycotoxins combination (T-2 toxin at the concentration of 0.095 mg/kg and aflatoxin B1 in the concentration of 0.019 mg/kg) on the body of quails and the results of pharmacocorrection of toxicosis with a complex consisting of beet pulp and lecithin. Structural changes in the intestines of quais at fodder mycotoxicosis are described. The use of antitoxic feed additives in poultry led to a weakening of the action of xenobiotics, which was confirmed by an increase in the safety of poultry and increase in body weight of quails, a decrease in the clinical manifestations of intoxication, as well as in positive changes in the structure of the intestine of the poultry during histological examination.

This study was conducted to find out hepatoprotective activity of hesperidin (HES) 100mg/kg body weight (b.w.) against ciprofloxacin (CPX) 100 mg/kg induced hepatotoxicity in local breed rabbits .CPX is a broad spectrum antibiotic used for treatment of many bacterial infections. Twenty four male rabbits were divided into four groups ,group1: control, (1 ml/kg Saline orally) group 2: CPX (100 mg/kg orally) for (14) consecutive days , group 3: HES (100 mg//kg) orally for (14) consecutive days group 4: CPX (100 mg/kg orally) plus HES (100 mg//kg orally ) for (14) consecutive days. All the rabbits were killed on the (15) day of the experiment, and then the blood, and livers samples were taken. CPX induced hepatotoxicity was proved by a significant (p less than 0.01) reduction in the body weight ,and a significant (p less than 0.01) increased serum aspartate transaminase (AST), alanine transaminase (ALT) , Malonaldehyde enzyme (MAD) and histopathological changes. Protective hepatic toxicity effect and oxidative damage caused by CPX significantly (p less than 0.01) increasing in body weight and significantly (p less than 0.01) decreasing AST , ALT, MAD and improving tissue morphology in HES (100 mg//kg) . These results assure that HES (100 mg//kg) antioxidant effects can protect CPX-induced hepatotoxicity in rabbits.
