Excessive salt consumption is one of the hypertension and kidney disease factors, while telmisartan is one of antihypertensive drugs used in the therapy. Telmisartan not only blocks angiotensin receptor which leads to the decrease of blood pressure, but also activates peroxisome proliferator activated receptor gamma (PPAR-γ) and inhibits transforming growth expression factor of beta-1 (TGFβ-1). Whether telmisartan decreases the kidney collagen volume fraction of excessive NaCl-induced Wistar rats are studied in this experiment. Twenty five male Wistars 2.5-3 months of age and 100-150 g BW rats were used in this research. They were grouped into 5, each consists of 5 rats. Group I (G I) as first negative control did not receive NaCl and telmisartan. G II as second negative control received NaCl but not telmisartan. G III, IV and V received NaCl and telmisartan 3, 6 and 12 mg/kg BW. The treatments were given every day within 8 weeks. At the day of 56 all rats were sacrificed by mean of neck dislocation and operated to take the kidney. The collagen was stained by picrosirius red staining. Data were expressed as mean ± standard deviation. They were analyzed by parametric test (analysis of variance-ANOVA and paired samples t-test) or nonparametric test (Kruskal-Wallis). A value of p0.05 was considered statistically significant. The results showed that intraglomerular and extraglomerular collagen volume fraction were lower in telmisartan-treated Wistar rats group than negative control group (0.05p0.05). In conclusion, intraglomerular and extraglomerular collagen volume fraction were lower in 8% sodium chloride-induced and telmisartan-treated male Wistar rats than the items of negative control group.

Abstract: Monosodium glutamate (MSG) is one of the ingredients used as flavour additive. However, excessive consumption of MSG could damage the liver. This study was aimed to obtain the impact of MSG exposure on histopathological findings of the liver of Wistar rats administered with tomato juice. This was an experimental study with the post-test only control group design. Subjects were Wistar rats divided into three groups, as follows: the negative control group, treatment group I, and treatment group II. The negative control group was given AD2 pellets; the treatment group I was given AD2 pellets and MSG, while the treated group II was given MSG and tomato juice. This study was conducted for 14 days. The histopathological examination of the treatment group I showed fatty degeneration of hepatocytes and infiltration of inflammatory cells meanwhile of the treatment group II, there was less fatty degeneration of hepatocytes compared to the treatment group I and no inflammatory cells. Moreover, there was no fatty degeneration and inflammatory cells in the negative control group. Conclusion: Wistar rats treated with MSG showed fatty degeneration of hepatocytes and infiltration of inflammatory cells which could be reduced by administration of tomato juice along with MSG.Keywords: tomato juice, MSG Abstrak: Monosodium glutamat (MSG) merupakan salah satu bahan makanan yang sering digunakan sebagai penyedap rasa namun konsumsi MSG berlebihan dapat merusak hati. Penelitian ini bertujuan untuk mengetahui pengaruh pajanan MSG terhadap gambaran histologik hati tikus Wistar dengan dan tanpa disertai pemberian jus tomat. Jenis penelitian ialah eksperimental dengan post-test only control group design. Tikus dibagi menjadi 3 kelompok, yaitu kelompok kontrol negatif, kelompok perlakuan I, dan kelompok perlakuan II. Kelompok kontrol negatif hanya diberikan pelet AD2; kelompok perlakuan 1 diberikan pelet AD2 dan MSG; kelompok perlakuan 2 diberikan MSG dan jus tomat. Penelitian ini dilakukan selama 14 hari. Hasil pengamatan gambaran histologik hati tikus Wistar pada kelompok perlakuan I mendapatkan adanya degenerasi lemak pada hepatosit dan sel-sel radang PMN. Pada kelompok perlakuan 2 masih ditemukan adanya degenerasi lemak pad hepatosit tetapi dengan jumlah yang lebih kurang daripada kelompok perlakuan I, serta tidak ditemukan sel-sel radang PMN. Pada kelompok kontrol negatif tidak ditemukan adanya degenerasi lemak pada hepatosit dan sel radang. Simpulan: Pada tikus wistar dengan pemberian MSG terdapat degenerasi lemak pada hepatosit dan adanya sel-sel radang. Pemberian jus tomat bersamaan dengan pemberian MSG berefek menurunkan terjadinya degenerasi lemak pada hepatosit dan tidak disertai sel-sel radang.Kata kunci: jus tomat, MSG

ABSTRACT Background: Recent studies showed that Interleukin-8 (IL-8), activated cytokine immune response which plays an important role in the development of acute and chronic gastritis. Harmless anti-inflammatory therapeutic alternatives have been proposed, for example, the consumption of Sechium Edule Jacq. Swartz (chayote). Antioxidant (flavonoid) and cell regeneration (alkaloid) agents were found in chayote. This study aimed to determine the effect of chayote Sechium Edule Jacq. Swartz extracts on the level of IL-8 in Wistar rats with aspirin- induced gastritis. Subjects and Method: This was a randomized controlled trial (RCT) conducted at the laboratory of Mathematics and Natural Science, Universitas Sumatra Utara from January to February 2020. A total of 35 male Wistar rats was selected for this study and randomly allocated into 7 groups: (1) Negative control; (2) Positive control; (3) 100 mg/ kg BW chayote ethanol extract ; (4) 200 mg/kg BW chayote ethanol extract; (5) 100 mg/ kg BW chayote ethyl acetate fraction; (6) 200 mg/kg BW chayote ethyl acetate fraction; and (7) 20 mg omeprazole. The rats in positive control and treatment groups were induced with aspirin (200mg/ kg BW). The negative control group received no intervention. The dependent variable was level of IL-8 measured by ELISA. The independent variables were treatment status. The data were analyzed by One Way Anova and post hoc test. Results: The mean differences of IL-8 level were not statistically significant between study groups (p= 0.327). Mean of IL-8 level was higher in positive control group (Mean= 160.80; SD= 6.90) than in negative control group (Mean= 141.20; SD= 10.98). The lowest IL-8 level was in 100mg/ kg BW chayote ethanol extract group (Mean= 149.94; SD= 40.4), followed by 200mg/ kg BW (Mean= 152.4; SD= 30.73) and 100mg/ kg BW (Mean= 164.60; SD= 25.04) chayote ethyl acetate fraction groups, 20 mg omeprazole group (Mean= 170.60; SD= 21.58), and 200 mg/ kg BW chayote ethanol extract group (Mean= 176.80; SD= 10.98). Conclusion: The low dose (100mg/ kg BW) chayote ethanol extract has the most potential antiinflammation effect on in vitro gastritis with the lowest IL-8 level of all doses of chayote ethanol extract, chayote ethyl acetate fraction, and omeprazole.   The niacin in energy drinks has metabolic product that cause oxidative stress and liver damage, while the liver damage can be prevented by hepatoprotective agents. Scoparone in Artemisia vulgaris L. can act as a hepatoprotector by its antioxidant effect. This study aimed to investigate the effectivity of Artemisia vulgaris L. extract as a hepatoprotector in wistar hepatocytes induced by niacin. This study used 25 male rats which were divided into 5 groups: normal, the negative control, the positive control, the treatment group I, and II. Treatment was conducted for 28 days. The samples were terminated and the hepatocyte were prepared for histological examination. Histological appearance was catagorized as mild, moderate, and severe damage with or without inflamatory cells activity. The data analysis by Kruskal Wallis showed significant difference (p<0,001). Further analysis by Mann Whitney revealed significantly difference (p<0,05) between normal group and all groups, negative control group and positive control group, and positive control group and treatment group I, but not significantly difference between negative control group and treatment group I, negative control group and treatment group II, positive control group and treatment group II, and between treatment groups. The study concluded that the effectivity of Artemisia vulgaris L. extract has not been proven as a hepatoprotector but further study is needed to draw a definite conclusion.   Keywords: energy drink, niacin, Artemisia vulgaris L., hepatoprotector

Background: Terminalia macroptera Guill. &Perr. (Combretaceae) is a flowering plant with several ethno-medicinal claims. However, the dearth of information on its analgesic property has necessitated this study.Objectives: to evaluate the anti-nociceptive potential of ethanol extract of Terminalia macroptera stem bark (TMSB) in mice.Materials and Methods: Male and female mice of weight range 22 – 25g were randomly allotted into seven groups (n= 5) and treated as follows: Group I received 0.5 mL distilled water orally (negative control), Groups II-V were orally administered ethanol extract of T. macroptera stem bark (TMSB) at 50, 100, 200, and 400 mg/kg respectively while groups VI-VII received piroxicam 10 mg/kg and pentazocine 2 mg/kg intraperitoneally respectively as standards. The same treatment pattern was adopted for both pain models: tail immersion and acetic acid-induced writhing assays. Data were expressed as mean ± standard error of mean (SEM) using two-way analysis of variance (ANOVA) followed by Tukey’s and Bonferroni's multiple comparisons tests with p < 0.05 taken as significance.Results: The ethanolic extract of Terminalia macroptera stem bark showed significant dose-dependent anti-nociceptive activity at 100 and 400 mg/kg (2.95±0.41 and 2.9±0.31 respectively) 60 min post-treatment compared to the negative control group in the tail immersion test. Significant inhibition of nociception (0.20±0.20) was obtained at 400 mg/kg compared to the negative control group in the acetic acid-induced writhing test.Conclusions: The ethanol extract of Terminalia macroptera stem bark exhibited dose-dependent anti-nociceptive potential in both tail immersion and acetic acid-induced writhing assays in mice.

Abstract Introduction The aim of obturation in the root canal is to completely seal the canal space to eliminate all the portals of entry and exit between root canal and periodontal space. Various techniques have been developed to achieve a hermetic seal. Materials and Methods As many as 150 extracted human maxillary central incisors were taken for the study. Biomechanical preparation was done up to F5 protaper file. According to different obturation techniques, samples were divided into six groups, keeping 30 samples in experimental and 15 samples in control groups. Group I–Lateral Condensation, Group II–Thermafil, Group III–Beefill, Group IV–GuttaFlow, Group V–Positive Control group, Group VI–Negative Control group. After obturation, the samples were immersed in 2% Rhodamine-B dye for 24 hours. Each sample was longitudinally sectioned to examine under confocal laser scanning microscope. Statistical Analysis The results were evaluated with ANOVA and posthoc Tukey honest significant difference (HSD) comparison test. Results The mean values of dye penetration of different groups were Group I (Lateral Condensation) 1.51 ± 0.451, Group II (Thermafil) 0.918 ± 0.399, Group III (Beefill) 1.30 ± 0.559. Group IV (GuttaFlow) 0.655 ± 0.396, Group V (Positive Control group) 1.96 ±0.046, Group VI (Negative Control group) 0 ± 0. The lowest mean value of apical microleakage was found in GuttaFlow amongst all experimental groups. Conclusion It can be concluded that the GuttaFlow obturating material exhibited better apical sealing ability with canal walls.

Abstract At any moment, the continuous usage of medications can accompanied by DNA damage and the accumulation of such damages can cause serious consequences. Antidepressants are long-term used drugs and the incidence of their genotoxic impacts cannot be excluded. Therefore, this work was designed to investigate the possible genotoxic effects of the commonly used antidepressants (fluoxetine and amitriptyline) in adult male rats. Detection of DNA damage in individual cells was assessed by comet and micronucleus assays in three different cell populations i.e. liver, testis and bone marrow tissues of 24 swiss albino adult male rats. The animals were randomly allocated into three groups of 8 rats each: Group I - rats orally-administered distilled water via gavage tube for four weeks as a negative control. Group II - rats orally-treated with fluoxetine hydrochloride solution (7.2mg/kg/day) via gavage tube for four weeks. Group III - rats orally-treated with amitriptyline hydrochloride solution (27mg/kg/day) via gavage tube for four weeks. The results showed that both drugs (Group II and Group III) induced the same extent of DNA damage, as evidenced by a significantly higher DNA fragmentation in liver and testis tissues with increased frequencies of micronuclei formation in bone marrow tissues as compared with the negative control (Group I). These findings indicates that both Fluoxetine and Amitriptyline have genotoxic potentials and can induce the same extent of cytogenetic damage in rats. Special precautions and medical supervision should be taken in consideration with their uses.

Abstract: High blood cholesterol is a major risk factor for atherosclerosis. Gedi plants contain polyphenolic compounds that can lowering blood cholesterol levels. This study aimed to find out the effect of gedi (Abelmoschus manihot L) leaves extract on histopatologic features of aorta of those wistar rats, which had been administered an atherogenic diet. This was an experimental study on animal using 20 Wistar rats as samples. The rats were divided into 4 different groups, 5 rats for each group. Negative control group was fed only pelet, positive control group was fed pork fat as much as 2 ml/day, group C was fed on pork fat and gedi leaf extract, and group D was fed on pork fat then continued with gedi leaf extract. The results indicated that administration of 30 mg gedi leaf extract showed a histopathologic feature which is decreasing of foam cells. Conclusion: Microscopically, aorta of those rats fed on pork lard diet showed a number of foam cells in tunica intima and media whereas the rats that were fed on pork lard along with gedi leaf extract, generally there were no foam cells, but there aresome parts which contain few foam cells. On the group of Wistar rats which were fed on pork lard, continued with gedi leaf extract showed less foam cells compared to the positive control group.Keywords: Gedi leaf extract, pork lard, foam cell.Abstrak: Kadar kolesterol darah yang tinggi merupakan faktor risiko utama terjadinya aterosklerosis. Tumbuhan gedi mengandung senyawa polifenol yang dapat menurunkan kadar kolesterol darah. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak daun gedi terhadap gambaran histopatologi aorta tikus wistar dengan diet aterogenik. Hewan uji yang digunakan berupa 20 ekor tikus Wistar yang dibagi ke dalam 4 kelompok, masing-masing kelompok terdiri dari 5 ekor tikus. Kelompok kontrol negatif hanya diberikan pelet biasa, kelompok kontrol positif diberikan diet lemak babi 2ml/hari, kelompok C diberikan diet lemak babi bersamaan dengan ekstrak daun gedi, dan kelompok D diberikan lemak babi kemudian dilanjutkan dengan pemberian ekstrak daun gedi. Hasil penelitian memperlihatkan bahwa pemberian ekstrak daun gedi 30 mg mampu memberikan gambaran histopatologi aorta berupa berkurangnya jumlah sel busa. Simpulan: Secara mikroskopik, aorta tikus wistar yang diberi lemak babi menunjukkan adanya penumpukan sel-sel busa pada tunika intima dan media, sedangkan pada aorta tikus wistar yang diberi lemak babi bersamaan dengan ekstrak daun gedi maupun yang dilanjutkan dengan ekstrak daun gedi ditemukan berkurangnya jumlah sel-sel busa.Kata Kunci : Ekstrak daun gedi, Lemak babi, Sel busa.

Trigona honey (TH) is well known for its therapeutic characteristics. To date, the study of Trigona honey as a prophylactic or immune booster prior to the bacterial infection of the invivo model is not well covered. This study aims to investigate anti-inflammatory and immune activities in Wistar rats infected with respiratory infection following with Trigona honey. 25 Wistar rats were assigned to possitive groups, negative control group, positive control group was fed TH (5 g / kg body weight) orally, the untreated group was infected with Staphylococcus aureus to induce respiratory infection, the treated group has been infected with S. aureus followed by treatment with TH at a dose of 1.5 ×108 CFU / mL and the preventive group ingested TH one week before S. aureus infection. Blood was obtained for biochemical analysis. Lung tissues have been collected for molecular examination. The results showed a significant decrease in serum levels of ALT, AST, urea and creatinine in the preventive and treated groups, serum IgG increased significantly (P<0.05) in the preventive and treated groups, IFN-y increased in the preventive group while decreased in the treated group, and IL-8 increased in the treated group while decreased in the preventive group. The mRNA expression of AGP is up-regulated in the positive control, preventive and treated groups. The α2-MG, TNF-α , and mRNA expressions showed lower regulation after administration of TH in preventive and treated groups. The results show the ability of TH to counteract immune and inflammatory changes in serum levels and gene expressions.

Multidrug-resistant tuberculosis (MDR-TB) is a tuberculosis infection that is resistant to the treatment at least two of the most powerful anti-tuberculosis drugs, such as Isoniazid and Rifampisin. Increased cases of MDR-TB in morbidity and mortality become obstacles in the control of tuberculosis (TB), thus requiring supportive treatment of natural ingredients that can contribute in the treatment of TB, such a noni fruit. The main objective of this study was extract of noni fruits to inhibition the growth of strain MDR-TB bacteria, and compered it with the anti-TB drugs. The Morinda c. Linn (Noni) fruits was extraced by ethanol (96%). The extract was filtered through whatman No.1 filter paper, evaporated to dryness on a water bath until the solvent evaporated completely and yield of the crude extract. The experiment were divided into 3 groups, i.e.: negative control: group I; positive control: group II; crude extracts noni fruit: group III: combinations of crude extracts noni fruit and anti-TB drugs (K, AK, and OF). Each group was divided into three groups’ doses of 30 mg/ml, 40 mg/ml and 50 mg/ml. The Anti-tuberculosis activities of extracts noni fruit and K, AK, and OF against TB-MDR bacteria were tested by susceptibility test using proportion method in Lowenstein-Jensen (LJ) media.The anti-tubercular activity of noni fruits was determined by the minimum inhibitory concentration (MIC) of the bacterial growth at various doses 30 mg/ml, 40 mg/ml, and 50 mg/ml. The research design used post-test only contol group, and analyzed using analysis of variance and post hoct test. The extracted of Morinda c.Linn (noni) fruits have antitubercular activity to inhibiton of growth MDR-TB bacteria at various doses (p value=0,000). At a dose 30 mg/mlthe mean rate of the growth colonies of MDR TB-bacteria whit the mean rate 59,00 ± 27,81, and at a dose 40 mg/ml was1,50 ± 2,81. While at a dose of 50 mg/ml the bacterial colonies of MDR-TB did not grow in media. The combination of Morinda c.Linn (noni) fruit with anti-tuberculosis drugs, was the smallest groups to inhibit and eliminate MDR-TB bacteria at a dose 30 mg/ml(0,00 ± 00.00). The experimental results confirmed the extracted of Morinda c.Linn (noni) fruits have antitubercular activity as well as anti-TB drugs, and the combination of the extracted of Morinda c.Linn (noni) fruits and anti-TB drugs was the best groups to inhibiton of growth MDR-TB bacteria.   Keywords: TB-MDR Bacteria, Morinda c. Abstract: Okra (Abelmoschus esculentus) is one of many herbal medicine that have been used worldwide to prevent and treat diabetes mellitus. This research aimed to evaluate the effect of garlic bulbs extract administration on elevated blood glucose levels on Wistar rats (R. norvegicus) induced by alloxan. The subject in this research were male Wistar rats with total of 15 samples divided into 5 groups, consisting of one negative control group, one positive control group who were induce by 130 mg/kgBW of alloxan that cause the Rats in conditions of hyperglycemic, and three groups of Rats were given okra extract with dose of 0.375 ml/100 gBW of rat, 0.75 ml/100 gBW of rat , and 1.5 ml/100 gBW of rat. Blood glucose levels was measured on day zero, day one and day two every thirty minutes on 0, 30, 60, 90, 120, and every six hours on 0, 6, 12, 18, and 24. Data from the measurement showed that 0.375 ml/100 gBW, 0.75 ml/100 gBW , and 1.5 ml/100 gBW of okra extract can’t reduce elevated blood glucose levels on Wistar rats.Keywords: Abelmoschus esculentus, okra, blood glucose levels, alloxan Abstrak: Buah Okra (Abelmoschus esculentus) diyakini masyarakat sebagai salah satu tanaman obat yang mampu mencegah dan mengobati penyakit diabetes. Subjek penelitian berupa tikus Wistar berjumlah 15 ekor yang dibagi dalam 5 kelompok, terdiri atas satu kelompok kontrol negatif, satu kelompok kontrol positif yang diberi aloksan dengan dosis 130 mg/kgBB yang menyebabkan tikus Wistar dalam keadaan hiperglikemik yang kemudian diinsuksi insulin, dan tiga kelompok Tikus hiperglikemik diberi ekstrak buah okra dengan dosis 0,375 ml/100 gBB tikus, 0,75 ml/100 gBB tikus, dan 1,5 ml/100 gBB tikus. Data diperoleh dari pemeriksaan kadar glukosa darah pada semua kelompok tikus Wistar pada hari ke nol, pertama, dan kedua pada menit ke-0, 30, 60, 90, 120 dan jam ke-0, 6, 12, 18, dan 24. Hasil penelitian menunjukkan bahwa pemberian ekstrak buah okra dengan dosis 0,375 ml/100 gBB tikus, 0,75 ml/100 gBB tikus, dan 1,5 ml/100 gBB tikus tidak mempunyai efek menurunkan kadar glukosa darah tikus Wistar.
