Total protein describes the liver's ability to synthesize proteins and metabolize substances in the blood. Bovine serum can be used as a control material because the total protein analyte in bovine blood serum is almost identical to the analyte in human serum. The researchers intend to determine the homogeneity and stability of bovine serum with the addition of 7,5% ethylene glycol preservative after being stored for 12 weeks at -20°C as an alternative to control serum for total protein levels. This research used survey analytics using a one-group pretest-posttest research design.  Data analysis used ISO 13528 2005 calculations. From the examination of the total protein level in the serum, the value of Xr – Yr = 0.23817, which is where the value meets the test criteria according to |  Xr - Yr |  <0.36, that is 0.23817 <0.91585. The total protein content of bovine serum was homogeneous and stable with the addition of 7.5% ethylene glycol stored at -20oC for 12 weeks.

Abstract Commercial NR latex has a higher total protein content than guayule (GR) latex. Some NR proteins are allergens bound to the rubber particle surface. Washing NR latex with a non-ionic surfactant displaced these particle-bound proteins and reduced allergens by more than 95%. The cost of such deproteination was reduced vulcanization efficiency, as determined by crosslink density. The extent of vulcanization correlated well with both total protein and allergen levels. Compared with films prepared from untreated NR latex, films from both surfactant-treated NR latex and GR latex had lower states of cure. Where particle-bound proteins were still present, as they are in GR latex, crosslink development could be completed by heat aging. In contrast, crosslink development in the film from surfactant-treated NR was complete after dipping and drying. The resulting films yielded high levels of extractable protein allergens.

The protein concentration and volume of the haemolymph may change with no apparent relation to one another in normal, ovariectomized, allatectomized, and median-neurosecretorycell-cauterized (mNSC-cauterized) females. Therefore, protein levels in the haemolymph are more meaningfully expressed in terms of the total protein content. In normal females, fluctuations in the haemolymph volume tend to parallel changes in the protein concentration during the first and subsequent gonotrophic periods. However, significantly less protein accumulates during the latter periods. The suggestion that these fluctuations partly reflect changes in the vitellogenic requirements of the oocytes is supported by the finding that both the volume and protein concentration increase significantly after ovariectomy.Allatectomy or mNSC cautery prevents the normal accumulation of protein in the haemolymph. In allatectomized females, the slight increase in protein concentration is accompanied by a decline in haemolymph volume. Cautery of the mNSC, provided it is performed within 3 h of emergence, results in a low protein concentration but has no effect on the haemolymph volume. The observations are discussed in terms of the corpora allata and mNSC control of haemolymph protein synthesis.

Background: Gradual alteration of serum copper and some plasma protein levels may occur with advancement of pregnancy, which is associated with increased maternal and infant morbidity and mortality. Objective: To observe serum copper and plasma protein levels in normal pregnant women of different trimesters in order to find out their nutritional status. Methods: This cross sectional study was carried out in the Department of Physiology, Sir Salimullah Medical College (SSMC), Dhaka, between 1st January 2010 and December 2010. Ninety normal pregnant women of different trimesters with age 20-30 years were included in the study group. They were selected from Out Patient Department of Obstetrics and Gynaecology, SSMC. Age matched 30 non-pregnant women were taken as control. Serum copper level was measured by Spectrophotometric method, serum total protein and albumin levels were estimated by standard method. Statistical analysis was done by one way ANOVA, Bonferroni and Pearson’s correlation coefficient test as applicable. Results: Serum Cu levels were significantly higher in all trimesters of pregnant women compared to control. Again, this value was significantly higher in 3rd trimester than that of in 1st and 2nd trimester and also in 2nd trimester than that of in 1st trimester. In addition, mean serum total protein level was significantly lower in 3rd trimester than control but no statistically significant difference was observed among different trimesters. Again, mean serum albumin level was significantly lower in 2nd and 3rd trimester than 1st trimester and control. In addition, serum Cu concentration showed significant positive correlation with different trimesters of gestation. Conclusion: This study reveals that hypercupremia along with hypoproteinemia occur in pregnant women from 1st to 3rd trimester of gestation. This gradual alteration of micro and macronutrients become more profound with advancement of pregnancy. Elderly people are particularly vulnerable to oral disease. Decreased function of the body can affect the aging process of the organic, and anorganic content in the elderly. The aim of this study was to determine the content of organic elements (total protein level), volume and pH of saliva in the elderly. This descriptive observational study with crosssectional design was performed in Tresna Werdha Gau Mabaji Social Institution, Gowa (n=30)with a simple random sampling technique using 30% of the total population and in Werdha Theodora Makassar nursing home with saturated sampling technique (n = 13). Anorganic contents were measured using spectophotometric and organic content of the total protein level of saliva was measured using an autoanalyzer tool kyltec in BPTP Laboratory Maros. Statistical analysis was performed with t-test using SPSS version 16. There were no significant differences between the anorganic (Ca2+,Mg2+, and phosphate) and organic elements content of saliva (total protein level), the volume of saliva and salivary pH in the elderly by sex and age groups. Yet, there are significant differences in the volume of saliva of elderly who consume drugs for systemic diseases. It is concluded that gender and age do not affect the anorganic (Ca,and phosphate)and organic element content of saliva (total protein content), the volume of saliva, and saliva pH in the elderly. Drugs consumed for systemic diseases does not affect the anorganic and organic element content of saliva and the pH of saliva in the elderly.

A field survey involving 180 adult white Fulani cattle was carried out in the Guinea Savannah and Derived Savannah vegetational zones of South Western Nigeria to determine the effect of natural trypanosomal infection on plasma protein levels. Enzyme-linked immunosorbent examination were used as diagnostic indices. The results showed a prevalence of 36.7% mixed infection in the animals sampled. It was also observed that the albumin level was lowered, globulin level increased and the total protein level was slightly on the increase. Trypanosomal infection in this study therefore affects the concentration of total and differential plasma protein levels in indigenous cattle.

SummaryCultures of human hepatoblastoma (HepG2) cells were treated with vitamin K1 or warfarin and prothrombin antigen and mRNA levels were determined. With 3 and 6 h of 10 µg vitamin K1 treatment secreted prothrombin antigen levels, relative to total secreted protein levels, were increased 1.5-fold and 2.1-fold, respectively, over ethanol-treated control levels as determined by an enzyme-linked immunosorbent assay. Dose-response analysis with 3 h of 25 µg/ml vitamin K1 treatment demonstrated a maximal increase of 2.0-fold in secreted prothrombin antigen levels, relative to total secreted protein levels, over ethanol-treated control levels. Pulse-chase analysis with 35S-methionine and immunoprecipitation of 35S-labelled prothrombin demonstrated that, with vitamin K1 treatment (25 µg/ml, 3 h), the rate of prothrombin secretion increased approximately 2-fold and the total amount (intra- and extracellular) of prothrombin synthesized increased approximately 50% over ethanol-treated control levels. Warfarin treatment (1, 5, or 10 µg/ml, 24 h) resulted in decreases in secreted prothrombin antigen levels, relative to total protein levels to approximately 85%, 87% or 81% of ethanol-treated control levels. Analysis of total RNA isolated from these cultures by Northern and solution hybridization techniques demonstrated that prothrombin mRNA was approximately 2.1 kb and that neither vitamin K1 nor warfarin treatment affected the quantity of prothrombin mRNA (ranging from 240–350 prothrombin mRNA molecules per cell). These results demonstrate that vitamin K1 and warfarin, in addition to effects on γ-carboxylation, affect prothrombin synthesis post-transcriptionally, perhaps influencing translation, post-translational processing and/or secretion mechanisms.

Lipidomics provides an overview of lipid profiles in biological systems. Although blood is commonly used for lipid profiling, cerebrospinal fluid (CSF) is more suitable for exploring lipid homeostasis in brain diseases. However, whether an individual’s background affects the CSF lipid profile remains unclear, and the association between CSF and plasma lipid profiles in heathy individuals has not yet been defined. Herein, lipidomics approaches were employed to analyze CSF and plasma samples obtained from 114 healthy Japanese subjects. Results showed that the global lipid profiles differed significantly between CSF and plasma, with only 13 of 114 lipids found to be significantly correlated between the two matrices. Additionally, the CSF total protein content was the primary factor associated with CSF lipids. In the CSF, the levels of major lipids, namely, phosphatidylcholines, sphingomyelins, and cholesterolesters, correlated with CSF total protein levels. These findings indicate that CSF lipidomics can be applied to explore changes in lipid homeostasis in patients with brain diseases.

In our previously described Hep G2/24-hour/total protein assay, protein levels were measured by using the Lowry method. This assay was the best acute in vitro assay for the prediction of human toxicity within the Multicentre Evaluation of In Vitro Cytotoxicity (MEIC) study. In order to increase the MEIC data-base with a wider range of chemicals, we were interested in introducing the more practical 3-(4-carboxybenzoyl)-quinoline-2-carboxaldehyde (CBQCA) method for the quantification of the total protein content. Therefore, we investigated whether the same good results for the prediction of acute human toxicity would be obtained with the CBQCA method. The cells were treated for 24 hours, then cytotoxicity was determined by measuring the total protein content with CBQCA. The results were quantified by using the PI50c: the concentration (in mM) of test compound required to reduce the total protein content measured with the CBQCA-method by 50% as compared to the control cells. The results were compared with the PI50, the corresponding value when the Lowry method was used. A relatively low correlation was observed between PI50 and PI50c, reflecting the large and unexpected, differences when using the two protein assays. However, when comparing the log PI50c with the human toxicity, a correlation coefficient of r2 = 0.761 ( n = 44) was obtained for exactly the same series of MEIC chemicals. This value is clearly higher than that for the Lowry method ( r2 = 0.695). Compared to the Lowry method originally used, the Hep G2/24-hour/CBQCA total protein assay has the additional important advantage that it can be very easily adapted for large-scale analyses with robotic systems, including the on-line calculation of the results.
