Synthesis of Selenium Nanoparticles in the Presence of Oyster Polysaccharides and the Antioxidant Activity

A simple method for synthesis of nanoselenium using oyster polysaccharides as soft template was investigated. The uniform stable selenium polysaccharides were obtained under the condition of reaction temperature 40°C, reaction time 3 h-5 h, the content of oyster polysaccharides 400mg/L and the content of sodium selenite 1mM. The size distribution and morphology of the product were confirmed by scanning electronic microscopy (SEM) and tansmission electronic microscope (TEM). The antioxidant activity of nanoselenium polysaccharides were studied in vitro. The results showed that nanoselenium polysaccharide is an effective hydroxyl radical and DPPH radical scavenger.

This study investigated the in-vitro antioxidant activity of ethanol leaf extract of Justicia carnea and its effect on antioxidant status of alloxan-induced diabetic albino rats. The in-vitro antioxidant activity was assayed by determining the total phenol, flavonoids, ascorbic acid, β-carotene and lycopene contents and by using 2,2 diphenyl-1-picrylhydrazyl (DPPH) radical, reducing antioxidant power and inhibition of lipid peroxidation antioxidant systems. Oxidative stress was produced in rats by single intraperitoneal injection of 150 mg/kg alloxan and serum concentration of malonaldehyde (MDA), superoxide dismutase (SOD) and catalase (CAT) were determined. Five experimental groups of rats (n=6) were used for the study. Two groups of diabetic rats received oral daily doses of 100 and 200 mg/kg Justicia carnea leaf extract respectively while gilbenclamide (5 mg/ml); a standard diabetic drug was also given to a specific group for 14 days. From the result, the leaf extract contained a higher concentration of flavonoids followed byphenols, ascorbic acid, lycopene and β-carotene. The extract displayed more potent reducing power ability with EC50 of 40 µg/ml compared to BHA (EC50 of 400µg/ml). The percentage DPPH radical scavenging activity of the extract was also higher with EC50 of 200µg/ml and increased with increase in concentration while BHA had EC50of 320µg/ml. The inhibition of lipid peroxidation also increased with increase in concentration with EC50 of 58µg/ml and comparable with BHA (EC50=60µg/ml). The effect of the plant extract on antioxidant enzyme activities was concentration-dependent. Administration of 100mg/kg of the plant extract resulted in a significant decrease (p<0.05) in serum MDA concentration, while 200 mg/kg of the extract caused a significant (p˂0.05) increase in superoxide dismutase (SOD) and catalase activities with a non-significant increase (p>0.05) in the serum level of MDA when compared with the diabetic untreated group. These findings suggest that ethanol leaf extract of Justicia carnea have antioxidant properties and could handle diabetes-induced oxidative stress.

The processes of formation different types of calli, as well as the morphogenesis pathways in morphogenic calli, were studied by scanning electron microscopy (SEM) during anther culture in vitro in hybrid line Fotos of spring soft wheat. The microspore haploid origin of calli has been proven. The morphological status of the obtained calli was determined. It was shown that morphogenic callus consists of small densely packed meristematic cells covered with extracellular substance. This type of calli was obtained using a variant of the Potato II induction culture medium, added by 1.0 mg/l synthetic auxin 2,4-D. Nonmorphogenic callus consists of large, elongated, loosely located cells with a smooth surface. This type of calli was obtained using a variant of the Potato II culture medium, added by 2.0 mg/l 2,4-D. It was found that the introduction of various IAA concentrations into the Blaydes nutrient medium for regeneration in morphogenic calli implements the following pathways of morphogenesis in vitro: embryoidogenesis (without IAA addition), gemmorhizogenesis (0.5 mg/l), and rhizogenesis (1.5 mg/l). Revealed degenerative changes in cells of nonmorphogenic calli. The fundamental possibility of regulating of the morphogenesis pathways of in vitro of morphogenic calli in the direction necessary for research in biotechnological research has been confirmed.

The research aimed to provide scientific basis for developing Proanthocyanidins from peanut skin. In this study, the antioxidant activity of Proanthocyanidins form peanut skin was studied at different system (O2-•, •OH and DPPH•) in vitro, with Vc. For O2-•, •OH and DPPH•, the IC50 of Proanthocyanidins form peanut skin was 0.143mg/mL, 0.097mg/mL and 0.274mg/mL; the IC50 of Vc was 0.163mg/mL, 0.097mg/mL and 0.306mg/mL. The result showed that proanthocyanidins from peanut skin had stronger antioxidant activity than Vc, and that Proanthocyanidins would be a effective natural free radical scavenger.

The essential oil from the roots of Bupleurum longiradiatum, obtained by hydrodistillation was analyzed by gas chromatography/mass spectrometry (GC/MS) and evaluated for antimicrobial activity and antioxidant activity. Fifty-one compounds were identified, representing 99.3% of the total oil. The major constituents were thymol (7.0%), butylidene phthalide (6.8%), 5-indolol (5.6%), heptanal (5.3%), 4-hydroxy-2-methylacetophenone (5.3%), 4,5-diethyl-octane (5.3%), bormeol (5.1%) and hexanoic acid (5.1%). The oil was tested against 4 bacteria at different concentrations using disc diffusion and 96-well dilution methods. The inhibition zones and minimum inhibitory concentration values for bacterial strains were in the range of 7.0–18.0 mm and 250 −500 μg/mL, respectively. The in vitro antioxidant activity was assessed by DPPH radical scavenging and inhibition of lipid peroxidation methods. The oil showed a potent free radical scavenging activity, as evidenced by the low IC50 value for DPPH radical (566.2μg/mL) and inhibition of lipid peroxidation (induced by FeSO4, H2O2 and CCl4) with IC50 values of 275.2 μg/mL, 296.9 μg/mL and 118.7 μg/mL, respectively.

Objective: The present study was carried out to evaluate the in vitro preliminary phytochemical analysis and antioxidant activity of methanolic leaf extracts of Corchorus olitorius L. The in vitro antioxidant activity was evaluated by DPPH radical scavenging activity method.Methods: Fresh jute leaves (immature, mature and senescence) were collected, air dried and the crude powder was prepared for phytochemical and antioxidant analysis. The powder was mixed with 70% methanol and the supernatant was separated. The antioxidant activity of this methanolic extract was measured on the basis of the scavenging activity of the stable 1, 1-diphenyl 2-picrylhydrazyl (DPPH) free radical with slight modifications. Phytochemical analysis was performed according to standard laboratory protocol.Results: The results indicated the presence of different phytochemicals viz. glycosides, steroids, cholesterol, alkaloids, phenols, flavonoids, riboflavin, saponins and terpenoids. The sample also showed antioxidant activity by inhibiting DPPH radical. The significant antioxidant activity of methanolic leaf extract of might be due to the presence of saponins, phenols, flavonoids and alkaloids found in the preliminary phytochemical analysis.Conclusion: Present study reveals that the jute leaf possesses different phytonutrients and exhibited DPPH radical scavenging activity, and therefore, may be used for therapeutic purposes.

Dihydromyricetin-zinc(II) complex was synthesized. The scavenging activity of DPPH radical, hydroxyl radical, superoxide anion radical and alkane radical by dihydromyricetin-zinc(II) complex were determine, moreover compared with Vc. The results showed that dihydromyricetin-zinc(II) complex possessed strong scavenging activities on DPPH•, hydroxyl radical, superoxide anion radical and alkane radical with correspounding maximum scavenging rate 74%, 70% , 65% and 50% at the concentration from 0.1 mmol/L to 0.5 mmol/L, respectively, which were higher than Vc. These results suggested that dihydromyricetin-zinc(II) complex would be an effective free radical scavenger.

Background: Tetracarpidium conophorum (African walnut) is an African plant with ethnobotanical uses. Objectives: The purpose of this study was to evaluate the phytochemical screening and in vitro antioxidant activities of methanol extract and fractions (F) [n-hexane (HEX-F), dichloromethane (DCM-F) and, ethyl acetate (EA-F)] of T. conophorum seeds. Methods: Phytochemical screening and in vitro antioxidant activity study were carried out using DPPH, ABTs radical scavenging assays, nitric oxide inhibitory and reducing potential assays. Results: Methanol extract and its fractions contain phenols, flavonoids, saponins, tannins, terpenoids, and alkaloids. The concentrations of total phenols and flavonoids content were significantly higher in EA-F and crude methanol extract compared to other fractions. Crude methanol and EA-F contain higher concentrations of tannin while hexane fraction had the lowest tannin content but relatively higher proanthocyanidin content compared to other fractions. The antioxidant activity study showed that both methanol crude extract and fractions of T. conophorum seeds have significant activities for DPPH radical scavenging, reducing power, ferric reducing antioxidant potential, nitric oxide inhibitory activities, ABTS and hydroxyl radical scavenging for. DPPH radical scavenging activities of EA-F showed the lowest IC50 of 33.11 µg/mL, followed by Hex-F, DCM-F and crude methanol extract with IC50 of 33.43, 42.09 and 45.44 µg/mL, respectively, when compared to ascorbic acid with IC50 of 17.08 µg/mL. Conclusion: The study showed that T. conophorum seed is a rich source of secondary metabolites, which may be responsible for its antioxidant activities.

This study was designed to assess the protective effects of Lycium europaeum methanol extract (LEM) on liver and kidney injuries induced by cisplatin. The phytochemical composition, the antioxidant activity, and hepatorenal injury biomarkers were investigated. Results revealed that LEM exhibited a significant antioxidant activity in vitro on DPPH radical and H2O2 scavenging assays. In the animal studies, treatment with LEM significantly reduced the effects of cisplatin intoxication on serum liver biomarkers and serum renal biomarkers. Meanwhile, LEM diminishes significantly the effect of cisplatin on the level of lipid peroxidation in liver and kidney tissues. The activities of the antioxidant enzymes (reduced glutathione, glutathione peroxidase, superoxide dismutase, and catalase) were increased in groups pretreated with LEM and quercetin. Additionally, the normal histological structures of the liver and kidney were restored after treatment with LEM. This work clearly demonstrated that L. europaeum may be useful as a drug with hepato-nephroprotective potentials.
