Extraction and Screening of ß-Sitosterol Compounds from Chloroform Extracts Barks of Kelor Plant (Moringa oleifera Limn) as a Candidate of Cancer Treatment Compounds

Treatment for cancer disease by using traditional plant has been used since the ancient era. many plants have the potential to treat cancer, one of them is β-sitosterol. for over a decade, β-sitosterol has many functions in the human body such as treatment for the cancer patient or cholesterol treatment. for this reason, the aims of these studies were to get the best solvent system to the gain β-sitosterol compound from the bark of kelor. Extraction section showed the best solvent system is hexane: ethyl acetate by increasing polarity index and from the pharmacognostic test, hexane extract has a positive result for alkaloid and steroid but not for flavonoid test. The result of the partition of extract hexane founded that there were 4 (A-D) fractions but only C fraction has crystal yellow. Base on the IR fragment, fraction C of the bark of kelor, (Moringa oleifera Limn) has similarity with the β-sitosterol compound.

Nowadays, knowledge of ancient botanical medicinal practices and application of modern phytochemical techniques have provided the excellent tools for the purification and structural elucidation of various phyto-compounds, which, in turn, has given insights into their mode of action on the human body. This study has been designed to investigate for the first time the antioxidant effects of the ethyl acetate, methanolic, and water extracts of Kitaibelia balansae. Different chemical methods were performed and the observed abilities depend on the solvent used. The best antioxidant ability was noted in water extract, followed by methanol and ethyl acetate extracts. The highest level of phenolic was also detected in water extract. The present findings suggest that K. balansae can be considered as a potential source of bioactive compounds for novel phytopharmaceuticals development

The aim of this study was to investigate the antibacterial effect and identify the phytochemical constituents of Moringa oleifera leaves and seeds extract on Staphylococcus aureus (S. aureus) clinical isolates using agar well diffusion method. The samples were collected from the premises of Usmanu Danfodiyo University, Sokoto. The Seeds and Leaves were collected fresh. They were extracted using methanol and ethyl acetate. Various concentrations from 100mg/ml to 500mg/ml were prepared. The test bacteria used is Staphylococcus aureus obtained from Microbiology laboratory of the Usmanu Danfodiyo University. The bacteria were re-identified using biochemical tests. The bacterial inoculums were standardized to McFarland scale 0.5. Zone of inhibition were read after 24 hours of incubation at 370C.The results of the antibacterial study revealed that the methanolic leaves extracts at 500 mg/ml had effect on S. aureus with zone of inhibition of 20mm. The methanolic seed extract have effect on S. aureus with zone of inhibition of 19.5mm. The MIC for the leave and seed extracts for Staphylococcus aureus was 250mg/ml. The MBC was 500mg/ml. The results of the phytochemical analysis revealed the presence of flavonoid, tannins, saponins, cardiac glycosides, alkaloids, volatile oil, saponin glycosides, and glycosides but anthraquinone and steroids were absent in the extracts. The zones of inhibition showed that both the methanolic and ethyl acetate extracts at 500mg/ml were active to all the tested bacteria. ANOVA and Duncan Multiple Mean Range test was used to analyze the data. Based on Duncan’s grouping, there is significant difference between the solvents and the concentrations used.

The natural naphthopyranones paepalantine (1), paepalantine-9O-β-ᴅ-glucopyranoside (2) and paepalantine-9-O-β-ᴅ-allopyranosyl-(1→6)-O-β-ᴅ-glucopyranoside (3) were separated in a preparative scale from the ethanolic extract of the capitula of Paepalanthus bromelioides by high-speed counter-current chromatography (HSCCC). The solvent system used was composed of water-ethanol-ethyl acetate-hexane (10:4 : 10:4, v/v/v/v). This technique led to the separation of the three different naphthopyranone glycosides in pure form in approximately 7 hours. Paepalantine showed a good antioxidant activity when assayed by the DPPH radical spectrophotometric assay.

Background: This study examined the in vitro antibacterial property of extracts of Moringa oleifera and the effect of different concentrations of the ethyl-acetate extract on cutaneous wound using an ointment delivery base. The aim of the study was to screen the extracts with best antibacterial property and evaluate effectiveness of different concentrations of the best extract when delivery is modified to ensure prolonged contact and reduced frequency of administration using an ointment base delivery vehicle. Materials and Methods: Dried and pulverized leaves of Moringa oleifera were screened for secondary metabolites. Successive gradient extraction was carried out using n-hexane, ethyl acetate and methanol. Leaf extracts were screened against clinical wound isolates of Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Klebsiella pneumoniae and Proteus mirabilis using agar cup diffusion technique. The ethyl acetate extract was investigated for its healing efficiency on excision wound model in rats. The study made use of wistar rats (150-180 kg), randomized into Gentamicin, M. oleifera ointment (5% and 3.25%), and ointment base treatment groups. Treatments were applied topically on days 0, 7 and 14. The percentage wound closure rate was measured and histopathology of the healed wounds carried out. Results: In vitro antimicrobial screening showed that ethyl-acetate extract was effective against the test isolates. Topical application of ointment with 3.25% of the plant extract resulted in faster wound closure rate, rapid epithelization, resolution of granulation tissue, and remodeling at histology. Conclusion: This may be due to less interference by components of M. oleifera which appear to retard wound healing at higher concentrations. Therefore, M. oleifera 3.25% ointment preparation is recommended topically for wound healing.

In-situ transesterification method without catalysts to produce biodiesel (fatty acid ethyl esters, FAEE) from rice bran using subcritical water ethanol mixture has been investigated. This method was found to be efficient since the rice bran oil (RBO) extraction and reaction of RBO into FAEE occur simultaneously. In this process other chemical (ethyl levulinate, EL) was also formed along with FAEE. EL can be used to improve the biodiesel quality by improving the low temperature properties of biodiesel. In this study effect of co-solvent types (without co-solvent, ethyl acetate, chloroform, and n-hexane) and water ethanol ratio (20%, 40%, 50%, 60% and 80%, v/v) on the content and yield of FAEE and EL at subcritical water ethanol mixture (T= 160°C, P= 80 bar, and t= 2 h) were investigated systematically. The content and yield of FAEE and EL obtained was found to be affected by the type of co-solvent. The content of FAEE and EL obtained without co-solvent (ethanol and water polarity index were PI=5.2 and PI=10.2, respectively) and with co-solvent of ethyl acetate (PI= 4.4), chloroform (PI= 4.1) and n-hexane (PI= 0.1) were 55.80% and 3.92%, 68.63% and 1.15%, 65.56% and 2.14%, and 62.00% and 0.93%, respectively. Higher polarity index of co-solvent extracted more RBO, as consequent the yield of FAEE (79.79%) obtained was higher using ethyl acetate as co-solvent. This data also suggested that RBO contains more free fatty acids (FFA= 63.59%) rather than of triglycerides (TG= 24.94%).The content and yield of FAEE and EL decreased with increasing water ethanol ratio. The highest content of FAEE (60.57%) and EL (8.48%) and yield of FAEE (78.03%) and EL (10.92%) were obtained using water ethanol ratio of 20%, v/v.
