Purpose: To evaluate the toxicity of the dried seaweed, Sargassum plagiophyllum, extract (SPE) pretreatment in constipated mice.Methods: The dried seaweed powder was mixed with distilled water and extracted by autoclave at 121°C. Antioxidant activity of the extract was determined by 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay. Human normal colon cells were pretreated with SPE at 0 - 100 μg/mL for 24 h before challenging them with 100 μM hydrogen peroxide (H2O2). Intracellular reactive oxygen species (ROS) were quantified using 2',7'- dichlorodihydrofluorescein diacetate (H2DCFDA). Male ICR mice were pretreated for 14 consecutive days with SPE at 100, 500 and 1,000 mg/kg or lactulose at 500 mg/kg. Body weight and food intake were recorded daily. Constipation was induced with loperamide on days 12, 13 and 14 and fecal pellets evacuated over a 4-hr period. The ileum, liver, kidney, and spleen were collected for histopathological examination.Results: The IC50 for the radical scavenging capacity of SPE was 343.90 ± 4.21 μg/mL compared to 14.14 ± 0.71 μg/mL for ascorbic acid. Pretreatment with SPE was significantly reduced ROS production in human normal colon cells. Oral administration of all doses of SPE and lactulose for 14 consecutive days had no effect on food intake or body weight when compared to the normal control group. Defecation was significantly more frequent in mice pretreated with SPE at 100 mg/kg than in the constipation control group. Histopathological examination of the ileum, liver, kidney and spleen of pretreated constipated mice revealed no toxic effect from either SPE or lactulose. On the other hand, the loss of mucus-producing cells in the ileum of constipated mice was significantly lower in mice pretreated with SPE.Conclusions: These findings support the safety of SPE supplementation and may broaden itsapplication in clinical fields as an alternative drug or supplement for constipation management.

Objective: Evaluation of the anti-obesity effect of aqueous extract of Mucuna pruriens seeds on rats.Methods: Male Sprague-Dawley (SD) rats were subjected to high-fat diet (HFD) for 12 wk. L-DOPA (12.5 mg/kg, p. o.) as standard drug and aqueous extract of Mucuna pruriens (AEMP) seeds (200 mg/kg, p. o. and 400 mg/kg, p. o.) as test drugs were administered in last 4 wk along with HFD. Body weight, food intake, body mass index (BMI), serum total cholesterol (TC), triglyceride (TG) and high-density lipoprotein (HDL) levels were measured at the end of fourth, eighth and twelfth wk, while white adipose tissue (WAT) mass and brain dopamine levels were measured at the end of the twelfth wk.Results: AEMP (200 mg/kg, p. o.) and (400 mg/kg, p. o.) treated groups showed a significant decrease in food intake and weight gain without altering BMI. Moreover, TG levels were lower in treated groups as compared to the HFD group, but no significant changes were observed in TC and HDL levels. L-DOPA-treated group showed a significant decrease in body weight, food intake, BMI and WAT. Both AEMP and L-DOPA-treated groups showed an increase in brain dopamine levels as compared to disease control group (p<0.05).Conclusion: L-DOPA and AEMP showed anti-obesity activity by reducing body weight gains, food intake and WAT weights; modulating TG with increased brain dopamine level which correlates to the inhibitory action of dopamine on reward mechanism.

Rats were divided into two groups such that mean weight and hemoglobin and hematocrit levels were not significantly different. One group (controls) was then fed a normal chow ad libitum. The other group was fed 6 g daily (30% of normal intake) for 2 weeks. The hemoglobin levels of rats fed the restricted diet rose 1.4–3.5 g/100 ml and hematocrit level rose 2–6%. At the end of 2 weeks total red cell mass (Cr51) was 5.5–6.0 ml in the underfed groups compared with 6.8 ml in the control group. Body weight fell proportionally more than did red cell mass, elevating the calculated red cell mass per unit body weight. Serum osmolality and K+ were not significantly different from control values, and there was a slightly higher serum Na+ and Cl– in the restricted diet group. It is concluded that restriction of food intake produced a relative polycythemia. At the end of 2 weeks of restriction an isosmotic reduction in plasma volume was present.

Weight loss resulting from decreased caloric intake raises levels of the orexigenic hormone, ghrelin. Because ingested nutrients suppress ghrelin, increased ghrelin levels in hypophagic weight loss may result from decreased inhibitory input by ingested food, rather than from lost weight. We assessed whether ghrelin levels increase in response to exercise-induced weight loss without decreased caloric intake. We randomized 173 sedentary, overweight, postmenopausal women to an aerobic exercise intervention or stretching control group. At baseline, 3 months, and 12 months, we measured body weight and composition, food intake, cardiopulmonary fitness (maximal oxygen consumption), leptin, insulin, and ghrelin. Complete data were available for 168 women (97%) at 12 months. Exercisers lost 1.4 ± 0.4 kg (P &lt; 0.05 compared with baseline; P = 0.01 compared with stretchers) and manifested a significant, progressive increase in ghrelin levels, whereas neither measure changed among stretchers. Ghrelin increased 18% in exercisers who lost more than 3 kg (P &lt; 0.001). There was no change in caloric intake in either group and no effect on ghrelin of exercise per se independent of its impact on body weight. In summary, ghrelin levels increase with weight loss achieved without reduced food intake, consistent with a role for ghrelin in the adaptive response constraining weight loss and, thus, in long-term body weight regulation.

AbstractThe present study aimed to investigate the protective effect of β-carotene on the oxidative stress injury of human normal colon cell line CCD-18Co triggered by tert-Butyl hydroperoxide (tBHP). XTT examination was used to determine cell viability after β-carotene supplementation and to determine the optimal concentration of antioxidant in spectroscopic studies. Cell biochemistry for CCD-18Co control group, after tBHP adding and for cells in β-carotene - tBHP model was studied by using label-free Raman microspectroscopy. Results for stress treated CCD-18Co human colon normal cells and human colon cancer cells Caco-2 based on vibration features were also compared. Pretreatment with β-carotene alleviated damages in CCD-18Co human normal colon cells induced by tBHP and showed the preventative effect on cells apoptosis. Treatment with β-carotene altered the level of ROS investigated based on intensities of Raman peaks typical for lipids, proteins and nucleic acids. Presented study confirmed the antioxidant, protective role of β-carotene against ROS by using spectroscopic label-free Raman techniques.

Despite its nutritional content and huge application in the food, pharmaceutical and cosmetic industries, the use of anthocyanin remains restricted due to the difficulty in its extraction process. In this study, we examined the antioxidant effects of anthocyanin (one of the numerous flavonoids known in modern enthno-medicinal practice) on some serum free radicals in wistar rats. Twenty (20) adult wistar rats of between 100 – 250 g were procured and housed in the animal house of the Ambrose Alli University, Ekpoma, Edo State. After two (2) weeks of acclimatization, the animals were then grouped into five groups of four (4) rats each (n=4). Control (Group I) rats were fed normal rat chow and water ad libitum, whereas, group II rats received 100 mg/kg body weight (bw.) of anthocyanin extract. Groups III, IV and V rats also received 200 mg/kg bw., 300 mg/kg bw., and 400 mg/kg bw of anthocyanin extract respectively for a period of four weeks, using the oro-gastric cannula post prandial. For each week, body weights were obtained, and serum assayed for changes in the levels of anti-oxidant enzyme activities [Catalase (CAT), Malondialdehyde (MDA), Glutathione Peroxidase (GPx) and superoxide Dismutase (SOD)] using appropriate biochemical procedures. Obtained results were subjected to statistical comparisons using the one way analysis of variance (ANOVA). Tukey post hoc tests were also performed (where necessary) to ascertain the source(s) of mean differences between groups. We found a statistically significant decrease (at p < 0.05) by week in serum CAT, MDA, GPx and SOD levels with the administration of test substance. This also proved to be dose-dependent as increasing dose cause an increase in the enzyme activities over the period of administration. Body weight changes were also duration and dose dependent. Anthocyanin therefore showed potency in improving serum antioxidant enzyme activities. We recommend similar studies on other body tissues than blood.

1. Male chickens, 1 week old, were fed alternately for 6 or 12 h periods with diets complementary in their amino acid composition. The isonitrogenous, isoenergetic diets were based on fish meal or sunflower meal as the sole source of protein, or fish meal–sunflower meal (40:60, w/w); each diet contained 120 g crude protein/kg. Other groups of chickens were given these diets ad lib.Net protein utilization (NPU) values obtained after a 1-week experimental period indicated that sequential feeding for both 6 and 12 h resulted in significantly improved responses compared with chickens given the same diets ad lib. These results suggested that the two protein sources, although provided at separate times, could complement each other.2. In certain treatments there was a 6 or 12 h fasting period between feeding periods to reduce the effect of nutrients from the previous period.3. For alternate feeding and fasting periods of 6 h with the combined diet, food intake and gain in body-weight were both significantly reduced compared with the corresponding values for the combined diet fed ad lib. The NPU value was virtually the same for both treatments. For 12 h periods of alternate feeding and fasting the gain in body-weight was only just significantly lower than that for the control group (combined diet fed ad lib.), but the NPU value was highly significantly better than that for the control group.4. For the treatments which included 6 and 12 h fasting periods between the two singleprotein diets, the food intake and gain in body-weight were significantly lower compared with the treatment groups given alternate feeding and fasting periods of either 6 or 12 h with the combined diet. The NPU values for the two groups given 12 h alternate feeding and fasting periods differed significantly, whereas the NPU values for the two groups given 6 h feeding and fasting periods were the same.

The effect of pregnancy and lactation on sodium appetite, water, and food intake of young adult BALB/c mice was studied. Voluntary sodium intake increased fourfold during the last 3 days of gestation. Water and food intake and body weight also increased significantly. During the last 14 days of lactation, sodium intake was increased significantly three- to fivefold, relative to base-line period and a control group. Large significant increase of water and food intake occurred also during lactation. After weaning, intake returned to base line. Calculation of sodium sequestered in young in utero and provided to pups during lactation showed increased voluntary intake in great excess of metabolic need, suggestive of hormonal determination of sodium appetite during reproduction as in wild rabbits.

Cisplatin (cis-diamminedichloroplatinum (II) or CDDP), used in the treatment of many solid-tissue cancers, has its chief side-effect in nephrotoxicity. Hence, this study sought to investigate and compare the protective effect of gallic acid (GA) and tannic acid (TA) against cisplatin induced nephrotoxicity in rats. The rats were given a prophylactic treatment of GA and TA orally at a dose of 20 and 40 mg/kg body weight for 7 consecutive days before the administration of a single intraperitoneal (i.p.) injection of cisplatin (CP) at 7.5 mg/kg bwt. The protective effects of both GA and TA on CP induced nephrotoxicity were investigated by assaying renal function, oxidative stress biomarkers, and histopathological examination of kidney architecture. A single dose of cisplatin (7.5 mg/kg bwt) injected i.p. caused a significant increase in some biomarkers of renal function (creatinine, uric acid, and urea levels), with a marked elevation in malondialdehyde (MDA) content accompanied by a significant (P<0.05) decrease in reduced glutathione (GSH) content (103.27%) of kidney tissue as compared to control group. Furthermore, a significant (P<0.05) reduction in kidney antioxidant enzymes (SOD, catalase, GPx, and GST) activity was observed. However, pretreatment with oral administration of tannic acid and gallic acid at a dose of 20 and 40 mg/kg body weight, respectively, for 7 days prior to cisplatin administration reduced histological renal damage and suppressed the generation of ROS, lipid peroxidation, and oxidative stress in kidney tissues. These results indicate that both gallic and tannic acids could serve as a preventive strategy against cisplatin induced nephrotoxicity.

The purpose of this study was to examine the effect of short-term (7 days) undernutrition on Type I (soleus) and Type II (plantaris, gastrocnemius) muscles in rats. Male Sprague-Dawley rats ( N = 20) were randomly assigned to one of two groups: a control group ( n = 10) in which animals were allowed to have water and pellets ad libitum and an undernourished group ( n = 10) in which animals were allowed to have 37% of the total food intake of the control group and water ad libitum. Body weight and food intake were measured daily. After 7 days, rats were anesthetized and the soleus, plantaris, and gastrocnemius muscles and liver were dissected. Body weight, liver weight, muscle weight, Types I and II fiber cross-sectional area, and myofibrillar protein content were determined. After 7 days of undernutrition, the undernourished group showed significant decreases ( p < .05) compared to the control group in body weight, liver weight, muscle weight of soleus, plantaris, and gastrocnemius muscles, and cross-sectional areas of Types I and II fiber of the plantaris and gastrocnemius muscles.

The anti-inflammatory and analgesic activities of the ethyl acetate (EAFSM) and n-butanol (NBFSM) fractions of the alcoholic extract of S. mangifera bark were evaluated using carrageenan-induced rat paw edema and by the tail-flick method in rats. The radical scavenging activity of the ethanolic extract, aqueous extract and fractions was determined with the DPPH radical scavenging capacity assay. Two fractions of the alcoholic extract, EAFSM and NBFSM, at doses of 75, 150, 300 mg/kg b.w. administered orally, showed a significant reduction in paw volume when compared with the respective control group challenged by carrageenan. Different doses of extract fractions also showed a significant prolongation of the tail-flick latency of the rat (P<0.01). Different concentrations of alcoholic, aqueous extracts and fractions of alcoholic extract showed significant free radical scavenging capacity against DPPH generated free radicals.
