Detection Limits of β-Lactam Antibiotics in Ewe Milk by Penzym Enzymatic Test

The Penzym is an enzymatic test widely used for the detection of β-lactam antibiotic residuals in milk. It is a specific method with good sensitivity to this group of antibiotics and enables results to be obtained within a short time. In the present work, the detection limits of 10 β-lactam antibiotics were determined in ewe milk, given the lack of previous studies of the Penzym test in ovine milk. For each antibiotic, eight concentrations were tested on 20 ewe milk samples proceeding from individual ewes (160 analyses per drug). The limits of the Penzym test were determined by means of logistic regression models, as follows: 5 μg/kg amoxicillin, 4 μg/kg ampicillin, 33 μg/kg cloxacillin, 3 μg/kg penicillin “G,” 43 μg/kg ce-phadroxil, 10 μg/kg cephalosporin “C,” 16 μg/kg cephalexin, 900 μg/kg cephoperazone, 120 μg/kg Ceftiofur, and 77 μg/kg cephuroxime. The percentages of positive results for those antibiotics at the maximum residue limit (MRL) concentration established by the European Union (EU) were: 100% (penicillin “G”), 93.3% (ampicillin), 93.3% (cloxacillin), 56.7% (Ceftiofur), and 56.7% (amoxicillin).

Atotal of 75 different clinical samples were collected from different hospitals in Baghdad Biochemical and morphological characterization tests showed that forty isolates were identified as Staphylococcus aureus Antibiotic susceptibility tests of all isolates towards ten antibiotics were carried out and results showed that many isolates (97.5 %) were resistant to ?-lactam antibiotic , 70 % were resistant to Tetracyclinee , 62.5% were resistant to co-trimoxazole , 60 % were resistant to ciprofloxacin , 55% were resistant both of chloramphenicol and erythromycin , 52.5% were resistant to gentamicin , 35% were resistant to rifampicin , 10% were resistant to vancomycin . According to the above results the S.aureus I1 which is isolated from patients with osteomyelitis showed resistant to all ten antibiotics therefore was used in the followed experiments. The minimum inhibitory concentration (MIC) of S.aureus I1 vancomycin, cefotaxim , penicillin G, amoxicillin , ciprofloxacin , co-trimoxazole ,gentamicin, rifampicin was checked.The results showed that isolates had MIC between (390-12500) ?g/ml. The combination of different antibiotics with vancomycin showed synergistic effect based on the Fractional inhibitory concentration index (FIC).

AbstractHerpes zoster (HZ), also known as shingles, occurs due to reactivation of varicella zoster. It is manifested by unilateral, painful, vesicular rashes following a dermatome. Homoeopathy is well known for its tremendous results in chronic cases. This article presents the evidence-based significant result of single homoeopathic remedy in an acute case of HZ within a short time period without any post-herpetic neuralgia which usually follows the HZ for months. Three to four doses of Hypericum 30 CH provided significant relief within a day and complete recovery within 10 days. Modified Naranjo score is 9, indicating definite causal attribution. This case proves that by selecting appropriate similimum, promising and positive results are achieved by homoeopathy, be it acute or chronic cases in a safe and gentle way. No such existing homoeopathic literature provides any characteristic information particularly related to HZ in Hypericum. So, future research and clinical trials could be beneficial for better validation of Hypericum as a therapeutic for HZ.

Performance of the Delvo-X-Press β-lactam antibiotic assay was examined using bulk-tank milk samples and milk samples from individual cows. Bulk-tank milk samples fortified with bovine lactoferrin at a concentration of 1 mg/ml or more consistently tested positive. False-positive results were also obtained from bulk-tank milk samples fortified with bovine plasma at concentrations of 20 and 40%. The assay yielded positive results for milk with antibiotic concentrations as low as 2 ppb. Individual milk samples were collected from 144 healthy lactating cows and from 34 cows with chronic Staphylococcus aureus mastitis. Specificity estimates for samples from healthy and mastitic cows were 0.88 (95% confidence interval [CI], 0.82, 0.93) and 0.94 (95% CI, 0.86, 1.00), respectively. Individual milk samples were collected from three cows with experimentally induced mastitis for 21 consecutive days. False-positive results occurred as late as 12 days postchallenge. A moderate but significant (P < 0.01) positive linear correlation (r = 0.61) was observed between test result and somatic cell count (SCC) values in milk samples with SCCs of >106/ml.

Abstract A novel, simple, and rapid single-drop microextraction (SDME) procedure combined with GC has been developed, validated, and applied for the determination of multiclass pesticide residues in honey samples. The SDME was optimized using a Plackett-Burman screening design considering all parameters that may influence an SDME procedure and a consequent central composite design to control the parameters that were found to significantly influence the pesticide determination. The developed analytical method required minimal volumes of organic solvents and exhibited good analytical characteristics with enrichment factors ranging from 3 for -endosulfan to 10 for lindane, procymidone, and captan and method quantification limits ranging from 0.03 g/kg for phosalone to 10.6 g/kg for diazinon. The relative recoveries obtained ranged from 70.8 for captan to 120 for fenarimol, and the precision (RSD) ranged from 3 to 15. The proposed SDME procedure followed by GC with an electron capture detector for quantification and GC/MS for identification was applied with success to the analysis of 17 honey samples. Monitoring results indicated a low level of honey contamination by diazinon, chlorpyrifosethyl, procymidone, bromopropylate, and endosulfan (-, -, and endosulfan sulfate) residues that were far below the maximum residue limit values specified by the European Union for endosulfan (10 g/kg) and bromopropylate (100 g/kg) in honey samples.

Antibiotic residue in chicken is a human health concern due to its harmful effects on consumer health. This study aims at screening the antibiotic residues from 80 chicken samples collected from farms located in different regions of Lebanon. An optimized multi-class method for identification and quantification of 30 antibiotics from four different chemical classes (sulfonamides, tetracyclines, quinolones, and beta-lactams) has been developed by using liquid chromatography–mass spectrometry. The evaluation of antibiotics residues in 80 chicken muscles samples has shown that 77.5% of samples were at least contaminated with antibiotics residues, out of which 53.75% were exposed to co-occurrence of multidrug residues. The screening of the four antibiotics families has shown that ciprofloxacin (quinolones) represents the highest occurrence percentage (32.5%), followed by amoxicillin (β-lactams) (22.5%) and then tetracyclines (17.5%). Means of sarafloxacin, amoxicillin, and penicillin G residues levels were above the Maximum Residue Limit (MRL) recommended limit according to the European Union EC. This study revealed that chicken samples collected from Lebanese farms contain antibiotic residues. Guidelines for prudent use of antimicrobials agents for chicken should be adopted to reduce the prevalence of resistant Salmonella in chicken.

Objectives: To explore whether the influence of a partner’s socioeconomic status (SES) on health has an additive or a combined effect with the ego’s SES. Methods: With data on 4533 middle-aged (30–59) different-sex couples from the 2012 Spanish sample of the European Union Statistics on Income and Living Conditions (EU-SILC) survey, we apply separate sex-specific logistic regression models to calculate predicted probabilities of having less than good self-perceived health according to individual and partner’s characteristics separately and combined. Results: Both approaches led to similar results: Having a partner with better SES reduces the probabilities of not having good health. However, the combined approach is more precise in disentangling SES effects. For instance, having a higher educated partner only benefits health among Spanish low-educated men, while men’s health is worse if they have a working spouse. Conversely, women’s health is positively influenced if at least one couple member is economically active. Conclusions: There are significant health differences between individuals according to their own and their partner’s SES in an apparently advantageous population group (i.e., individuals living with a partner). The combinative approach permits obtaining more precise couple-specific SES profiles.

Abstract A collaborative study was performed on a rapid Bacillus stearothermophilus agar diffusion ampule method to detect low levels of penicillin G in 7 types of fluid milk products. A multitest technique for processing a large number of samples simultaneously was also studied. Slight modifications were made in the original method to establish more uniformity and to eliminate doubtful responses by specifying a confirmation procedure. Twenty samples spiked with penicillin G (0.000 to 0.008 IU/mL) and tetracycline hydrochloride were frozen and sent to 20 laboratories in the ampule test, and 16 laboratories in the multitest. Each analyst was asked to do a screening run and a confirmation run. Results were reported by color reaction and also as positive or negative for β-lactam inhibitors. The concentrations (penicillin G) where percent positive results equal 100 or not significantly less than 100 (α = 0.05) ranged from 0.005 to 0.007 IU/mi in the ampule test and from 0.004 to 0.007 IU/mL in the multitest. Both techniques have been adopted official first action.

After a short introduction of the LIS landscape in Austria, the development of LIS education for academic librarians will be discussed. Until 2004 library education in Austria consisted mainly of vocational training to which only university graduates and eventually high-school graduates were admitted. In the nineteen-nineties, with Austria joining the European Union and the creation of universities of applied sciences, LIS education became possible at the university level. For various reasons, the two established LIS programmes were discontinued after a relatively short time period. Major changes in university legislation concerning the legal autonomy of Austrian universities meant, however, that a post-graduate inter-university library and information studies programme could be launched in 2004. With the fourth revision of the curriculum in 2019, the programme has become even more aligned with international practice in terms of content, didactic methods and flexibility. The increased dynamism in library education has also had an impact on the creation of non-academic training and continuing education programmes.

In early 2017 high particulate matter (PM) levels were observed across mid-Europe, including Austria. Here we characterize PM pollution in the city of Graz during January to March 2017, a period with substantial exceedances (34 days) of the European Union (EU) PM10 short time limit value. This study evaluates whether the observed exceedances can be attributed to the accumulation of pollutants emitted by local sources or to a larger scale pollution episode including transport. The analyses are based on the ratios of PM10 concentrations determined at an urban and background site, and the analyses of chemical composition of PM2.5 samples (i.e., water soluble ions, organic and elemental carbon, anhydro-sugars, humic-like substances, aluminum, and polycyclic aromatic hydrocarbons). Source apportionment was realized using a macro-tracer model. Overall, the combination of different approaches (PM10 ratios, chemical composition, and macro-tracer derived source apportionment) enabled a conclusive identification of time periods characterized by the accumulation of emissions from local sources or regional pollution episodes.

AbstractAntibiotic-resistant and biofilm-associated infections brought about by methicillin-resistant Staphylococcus aureus (MRSA) strains is a pressing issue both inside as well as outside nosocomial environments worldwide. Here, we show that a combination of two bacteriocins with distinct structural and functional characteristics, garvicin KS, and micrococcin P1, showed a synergetic antibacterial activity against biofilms produced in vitro by S. aureus, including several MRSA strains. In addition, this bacteriocin-based antimicrobial combination showed the ability to restore the sensitivity of the highly resilient MRSA strain ATCC 33591 to the β-lactam antibiotic penicillin G. By using a combination of bacterial cell metabolic assays, confocal and scanning electron microscopy, we show that the combination between garvicin KS, micrococcin P1, and penicillin G potently inhibit cell viability within S. aureus biofilms by causing severe cell damage. Together these data indicate that bacteriocins can be valuable therapeutic tools in the fight against biofilm-associated MRSA infections.
