Somatic cell count (SCC), mammary pathogens prevalence, total and specific bacterial counts, antimicrobial residues, macroscopic sediment, water addition, aflatoxins and other contaminants constitute the basis for milk payment-schemes, monitoring and improvement of flock hygiene and health management, and development of analytical surveillance programs in the dairy small ruminants. The present work reviews factors influencing the variation of these variables, including milk analytical methods, storage and preservation, along with management implications during the last two decades. Following farmer and cooperative educational programs, progressive reductions have been reported for total bacterial count and antimicrobial residue occurrence in bulk tank milk. These results were consistent, however, with high values for SCC and specific bacterial populations. Thus, mastitis control programs should be intensified to increase hygiene in milk and economic returns for producers and processors. In addition, the implementation of programs to reduce specific bacterial counts (i.e., psychrotrophs, coliforms, Clostridium spp. spores) and mammary pathogen prevalence (i.e., Staph. aureus, Mycoplasma spp.), as well as the use of combined screening methods for an increased rate of antimicrobial detection, are currently required strategies which are positively valuated by milk processors, industry and consumers. Other contaminants may also be present, but cost-effective screening and analytical systems have not yet been implemented. This review aims to be helpful for troubleshooting milk quality and safety, developing future premium payment systems and industry quality-standards, optimizing management, on-farm risk traceability systems and consumer acceptance.

Viable counts were made of heterotrophic bacteria in seawater samples taken from 0, 25, 50, 75, and 100 m at one station off Sydney over a period of 2 years. Populations, which fell largely within the range of 10-1000/ml, tended to be higher in spring and early summer than at other times. Some evidence was obtained of a negative correlation between bacterial population and chlorinity. There was no evidence of a correlation between bacterial populations and several other variables which were examined. A (presumed) species of Chromobacterium occurred intermittently at the lower depths with some suggestion of a correlation with season and total bacterial count.

SummaryA method for detection of proteolysis in milk was evaluated. Amino acids and peptides soluble in trichloroacetic acid were estimated by the Lowry–Folin procedure with expression of results in terms of colour yield equivalent to that of tyrosine (the ‘tyrosine value’ (TV)). Some variables and alternatives in the method of estimation were investigated.TVs for milk freshly drawn from individual cows were extremely varied (0·31–0·92 mg/ml). TVs for samples of refrigerated milk from daily supply farms were in the range 0·40–0·58 mg/ml; no relationships between TV and bacterial counts were evident with these samples. With bulk refrigerated raw milk supplied to Brisbane for liquid consumption from country depots and local farms equipped with bulk vats, significant positive relationships were found between TV and total bacterial count (TBC) for supplies from some sources, but in no instance was a significant relationship found between TV and psychrotroph or proteolytic psychrotroph count. Significant positive relationships were found between TV of bulk milk supplies from some sources and atmospheric temperature, and between TBC of the supplies and atmospheric temperature. The significance of these various relationships for bulk milks is discussed.Natural variation in TV imposed limitations on reliability of the method to provide an index of proteolysis; in general the results indicated that its application was restricted to bulk milk cold-stored for at least 3 days.

Liquid poultry manure was applied to three forest sites at 35, 70, and 225 metric tonnes (1 tonne = 1000 kg) per hectare (1 ha = 10 000 m2) for 1 to 3 years. Two of the sites were white pine plantations, one dry, one moist, and the third site was mixed hardwoods. Over a period of 35 months the total bacterial population in litter and in soil, in both control and manure plots, was studied as well as the ability of the population to produce extracellular proteases, cellulases, lipases, pectinases, and amylases. Total bacterial counts in litter tended to be higher in the autumn and lower in the spring. Litter always contained about 10-fold more bacteria than soil and the number of bacteria in soil fluctuated little with season or treatment. In white pine litter, bacteria able to excrete protease were more numerous than other enzyme-producing bacteria. More bacteria able to excrete lipases were found on moist than on dry sites, especially on manured plots. At the end of the 35-month test period, all bacterial counts in manured plots had returned nearly to the original level of the control. Statistical analysis revealed significant differences in total bacterial count and counts of specific enzyme producers between manured and control plots. Little difference was seen among rates of manure application. Adding large amounts of poultry manure to these forest plots did not greatly change the bacterial ecology of the soil or litter.

ABSTRACT Oral malodor develops mostly from the metabolic activities of indigenous bacterial populations within the oral cavity, but whether healthy or oral malodor-related patterns of the global bacterial composition exist remains unclear. In this study, the bacterial compositions in the saliva of 240 subjects complaining of oral malodor were divided into groups based on terminal-restriction fragment length polymorphism (T-RFLP) profiles using hierarchical cluster analysis, and the patterns of the microbial community composition of those exhibiting higher and lower malodor were explored. Four types of bacterial community compositions were detected (clusters I, II, III, and IV). Two parameters for measuring oral malodor intensity (the concentration of volatile sulfur compounds in mouth air and the organoleptic score) were noticeably lower in cluster I than in the other clusters. Using multivariate analysis, the differences in the levels of oral malodor were significant after adjustment for potential confounding factors such as total bacterial count, mean periodontal pocket depth, and tongue coating score (P < 0.001). Among the four clusters with different proportions of indigenous members, the T-RFLP profiles of cluster I were implicated as the bacterial populations with higher proportions of Streptococcus, Granulicatella, Rothia, and Treponema species than those of the other clusters. These results clearly correlate the global composition of indigenous bacterial populations with the severity of oral malodor.

<p>Total viable counts of bacteria and bacterial diversity of the different soil samples from three different localities in Sharq EL-Neel region: Soba, AL-Aelafoon and Um Dawan Ban sub-regions were carried out. Soil physical and chemical characteristics (pH, EC,SP,solublecations: Na, K, Ca, Mg and anion P, organic carbon, total nitrogen and soil texture)in each studied sub-regions were measured. Qualitative analysis of microorganisms isolated from the studied soil samples reveal a total of thirteendifferent species of bacteria, of which two are unidentified. The ten species are classified under Bacillus genus. In Sharq EL-Neel regionsoil samples, total bacterial counts ranged from 9.5 × 10<sup>4 </sup>cfu g<sup>-1</sup> to 1×10<sup>3</sup> with a mean of 4×10<sup>3 </sup>cfu g<sup>-1</sup>. The quantitative data on microbial population recorded in the present study was analysed using two diversity indices. High Shannon-Weiner diversity Index value for bacteria was obtained in AL-Aelfoon sub-region (1.79361), whereas high Simpson's index value was obtained in Um Dawan Ban sub-region (2.80).ActinomycesActinomyces spp. and Streptomyces spp. Where the most abundant microorganisms identified in the three sub-regions. Total bacterial count in Soba soil was positively correlated with pH (r= 0.0194) and sand (r= 0.3205); the total bacterial count in AL-Aelafoon soilwas positively correlated with EC (r= 0.1062), clay (r= 0.3816), silt (r= 0.1936), SP (r= 0.9302), K (r= 0.6252), Ca (r= 0.0015) and Mg (r= 0.1556), whereas the total bacterial count in Um Dawan Ban soil was positively correlated with clay (r= 0.2614), silt (r= 0.0216), SP (r= 0.565), K (r= 0.9645), P (r= 0.0197), Ca (r= 0.7377), Mg (r= 0.0267), N (r= 0.5215) and O.C (r= 0.3214). There were obvious differences in correlation coefficients among the selected criteria (46 % from the total number of correlation coefficients were positively correlated between bacterial counts and soil physico-chemical properties whereas 54% from the total number were positively correlated between plant type and bacterial counts).</p>

Abstract In this study, the effect of storage temperature (2 or 4°C) on the composition of milk and microbiological load was investigated over 96 h. Milk samples were collected from farm bulk milk tanks after one complete milking and stored at 2 or 4°C over 96 h. Total bacterial count (TBC), psychrotrophic bacterial count (PBC) and proteolytic bacterial count (PROT) were affected by storage time and temperature and varied significantly between farms (P < 0.05). The levels of TBC, PBC and PROT bacterial count increased from 4.37 to 6.15 log cfu/mL, 4.34 to 6.44 log cfu/mL and 3.72 to 4.81 log cfu/mL, respectively, when the milk was stored for 96 h at 2°C. The milk samples stored at 4°C had higher increases in these bacterial counts after 72 h in comparison to milk samples stored at 2°C. The casein fraction content was lower in milk samples stored at 4°C, which could be due to high levels of PROT bacteria or enzyme activity in these samples. Milk stored for 96 h at 2°C has less impact on composition or processability parameters compared to milk stored at 4°C.

Eggs were coated with propolis in order to determine its effect on egg quality and total bacterial count. Effect of storage time prior to propolis coating was also studied. Eggs were obtained from a local farm, coated with 0, 3, 6 or 9% of propolis and stored at 4 ºC with a humidity of 75% for 30 days. A total of 18 eggs were used for egg quality traits and 6 eggs were chosen for microbial activity for each group. Bacteria were recovered with sterile cotton swabs from egg surfaces. Colonies with different morphology and color were detected. 11 bacterial species were determined following the characterization of 14 isolates. Elevated levels of bacterial counts were detected for daily eggs, which were significantly higher than those stored for 30 days. The quality traits of the daily eggs have also been found higher compared with the other groups. It was observed that coating the eggs with varying concentrations of propolis lead to significant differences in the total bacterial counts of the eggshells. Moreover, the egg quality traits which decreased with the increase in storage time have not been further affected with propolis coating.

The study of microbial populations of biohydrometallurgic processes is generally more focused on the solutions involved than on the minerals. This is mainly due to complexity and costs associated with taking mineral samples during the leaching cycle. The present study compares the microorganisms in the solutions and the minerals in 1 m test columns and in samples taken from two strips of an industrial heap from the Escondida Copper Mine in Chile. The column test was carried out in six 1 m columns that were operated with similar parameters than the industrial heap. The industrial heap is divided into strips, and samples were collected from Strip 1 (after the end of the operation) and from Strip 9 (during the irrigation period). To analyze the bacterial population Real-Time PCR and CARD-FISH techniques were used. From the results of the column leaching tests a slightly greater cellular density was observed for the mineral than the solution, when the total bacterial count was considered. In both types of samples the predominant microorganisms were Acidithiobacillus thiooxidans, Acidithiobacillus ferrooxidans D2 and Leptospirillum ferriphilum. In the samples from the industrial heap A. ferrooxidans predominated at the start of operation and A.thiooxidans at the end of the leach cycle.

Summary Eleven different brands (93 packages) of frozen and unfrozen (breaded and unbreaded) pork and veal cutlets were purchased during 12 months from 4 large volume stores. Initial bacterial counts ranged from 15,000 to 25 million per g. Samples taken from 62 packages of frozen pork and veal cutlets and stored at 2 to 3 C for 3 to 4 days showed bacterial populations varying from 400,000 to more than 6 billion per g. The average of the median counts of all brands was 64 million. In samples from the 62 packages stored 7 days at 2 to 3 C, the average of the median counts from all brands was 1.8 billion per g, or a 28-fold increase in the bacterial count during 3 to 4 extra days of storage. Obvious spoilage was much greater in the 7 day samples. Obvious spoilage (odor, sliminess) was greater in 31 stored samples of unfrozen pork cutlets compared with previously frozen pork and veal stored similarly, i.e., at 2 to 5 C.

Indirect immunofluorescence was used to quantify the seasonal variation of four halophilic aerobic Antarctic bacteria in Antarctic saline lakes from July 1990 to January 1991. Antibodies were raised against type strains from the Australian Collection of Antarctic Microorganisms. During summer, all four serogroups were identified in the aerobic waters of lakes with total dissolved salts above 61%. Maximal abundances of Halomonas meridiana, H. subglaciescola and Flavobacterium gondwanense serogroups were observed at discrete depths within the water column in the two most hypersaline lakes at about midsummer, coincident with the time of maximum sunlight and the commencement of the summer thaw. At this time the Halomonas spp. serogroups comprised up to 40% of the total bacteria and the F. gondwanense serogroup up to 10% of the total bacteria. The F. salegens serogroup was in low numbers (>2% of total bacteria) in some aerobic waters. Up to 2% of the total bacterial populations in the lakes were autofluorescent or stained non-specifically. Dissolved organic carbon values were measured throughout the sampling period and correlated well with total bacterial numbers but not with changes in species composition. Change in species abundance, as indicated by immunofluorescence, was not reflected in the total bacterial count, indicating compositional change of the total bacterial population.
