Abstract: Cigarette smoke was one of free radicals source for the body. Free radical contain in cigarette smoke were hydroxide radicals (•OH). Hydroxide radicals would cause erythrocytes damage. Antioxidants were substances that can prevent free radicals. Now, natural antioxidants source has developed, one of that is tempe, because of it contained antioxidants substances such as carotene, Vitamin E, isoflavon, and factor II antioxidants. This resource was for knowing the influence of giving tempe flour toward the amount and morphology of white male rat (Rattus norvegicus) erythrocytes wistar strain was affected by cigarette smoke. A pure experiment which was used the post test only-control group design method. Sample of the resource was white male rat wistar strain aged of 3-4 month as much 30 rats by the weight around 180-220 gram. The samples divided into 3 groups that were control group (K1), treatment group 1 (K2), and treatment group 2 (K3), each group consists of 10 rats. Control group was not given a special treatment, it only got a standard feed, treatment group 1 got special treatment and it was affected by cigarette smoke, furthermore, treatment group 2 got standard feed, it was affected by cigarette smoke and tempe. Analysis of data used one way anova experiment. The test showed the significance P-value = 0,036 that was <α (0,05). The result showed that there was the influence of giving tempe flour toward the amount and morphology of white male rat (Rattus norvegicus) erythrocytes wistar strain was affected by cigarette smoke. Keywords: cigarette smoke, free radicals, erythrocytes, antioxidants, tempe

Abstract: Cigarette smoke contains three main components, carbon monoxide, nicotine, and tar which can cause disturbances in spermatogenesis. Smoke electronic cigarette contains three main components, nicotine, propylene gycol, and glyceryn which can cause disturbances in spermatogenesis. The purpose of this experiment is to compare the quality of spermatozoa Wistar rats by exposure to cigarette smoke and electronic cigarette smoke. This study is an experimental study using an approach of post test only control group design. Subject of the study were 9 rats Wistar male (Rattus norvegicus) were divided into 3 groups: group P0 is not given exposure to cigarette smoke and smoke the electronic cigarette, the group P1 is given exposure to smoke cigarettes 2 rods per day, group P2 is given exposure to smoke an electronic cigarette for two rods lit cigarette. Treatment was done for 50 days. The results showed the concentration of spermatozoa is difference between the treatment groups, but statistical analysis groups P0 and P1 showed a non-significant (p = 0.229), P0 and P2 group (p = 0.559), P1 and P2 group (p = 0.879). Motility P0 with P2 group (p = 0.008) and group P1 and P2 group (p = 0.026) showed significant differences which means there is a treatment effect on motility. While groups of P0 with P1 group (p = 0.209) did not show significant differences which means there is no treatment effect on motility. Morphology Control with treatment group 1 (p = 0.098) while the control group with treatment 2 (p = 0.004) it shows the morphology of the group P0 to P1 has no effect while the P0 to P2 there is an influence on the treatment.Keywords: sperm quality, cigarette smoke, electronic cigarette smoke Abstrak: Asap rokok mengandung tiga komponen utama, yaitu karbonmonoksida, nikotin, dan tar yang dapat menyebabkan gangguan pada spermatogenesis. Asap rokok elektronik mengandung tiga komponen utama, yaitu nikotin, propylene gycol, dan glyceryn yang dapat menyebabkan gangguan pada spermatogenesis. Tujuan penelitin ini adalah untuk melihat perbandingan kualitas spermatozoa tikus wistar yang diberi paparan asap rokok dengan asap rokok elektronik. Penelitian ini merupakan penelitian eksperimental dengan menggunakan pendekatan post test only control group design. Subjek penelitian sebanyak 9 ekor tikus wistar jantan (Rattus norvegicus) yang dibagi menjadi 3 kelompok: kelompok P0 tidak diberikan paparan asap rokok dan asap rokok elektronik, kelompok P1 diberikan paparan asap rokok kretek 2 batang, kelompok P2 diberikan paparan asap rokok elektronik selama 2 batang rokok kretek dinyalakan. Perlakuan dilakukan selama 50 hari. Hasil penelitian menunjukkan perbedaan konsentrasi antar kelompok perlakuan namun analisa statistik kelompok P0 dan P1 menunjukkan hasil yang tidak bermakna (p=0,229), kelompok P0 dan P2 (p=0,559), dan kelompok P1 dan P2 (p=0,879). Motilitas kelompok P0 dengan P2 (p=0,008) dan kelompok P1 dengan kelompok P2 (p=0,026) menunjukkan perbedaan bermakna yang berarti terdapat pengaruh perlakuan terhadap motilitas. Sedangkan kelompok P0 dengan kelompok P1 (p=0,209) tidak menunjukkan perbedaan bermakna yang berarti tidak terdapat pengaruh perlakuan terhadap motilitas. Morfologi kelompok Kontrol dengan Perlakuan 1 (p=0,098) sedangkan kelompok Kontrol dengan Perlakuan 2 (p=0,004) hal ini menunjukkan morfologi kelompok P0 dengan P1 tidak berpengaruh sedangkan kelompok P0 dengan P2 terdapat pengaruh terhadap perlakuan.Kata kunci: asap rokok, asap rokok elektronik, kualitas spermatozoa

BACKGROUND: Purslane (Portulaca oleracea L.) is a wild plant that grows in Indonesia and is considered a weed, but it has the highest antioxidant activity in the leaves and stems. Cigarette smoke is one of the exogenous free radicals that can enter blood circulation, interfering with all cells and tissues in the body, including reproductive organs. AIM: The research aims to examine the antioxidant effect of purslane leaf extract on oxidative stress levels (ovarian malondialdehyde [MDA] levels) and reproductive hormone levels (follicle-stimulating hormone [FSH] and 17ß-estradiol) in female white rats exposed to cigarette smoke. METHODS: This study is an experimental study with randomized post only control group design, using 12-week-old female Wistar strain Rattus norvegicus weighing about 200–250 g divided into four treatment groups (n = 6). Negative control without exposure to cigarette smoke and purslane leaf extract (K), cigarette group with cigarette smoke 1 stick/day administration without purslane leaf extract (P1), purslane group with purslane leaf extract 200 mg/Kg BW without exposure to cigarette smoke (P2), and group the group with cigarette smoke 1 stick/day accompanied by purslane leaf extract 200 mg/Kg BW administration (P3). Ovarian MDA measurement measures MDA levels as an indicator of oxidative stress in the ovaries determined by the spectrophotometry method. The enzyme-linked immunosorbent assay method measured the measurement of FSH and 17ß-estradiol levels in blood taken intracardially through the right ventricle of the heart. RESULTS: The results showed that purslane leaf extract at a 200 mg/Kg BW dose reduced ovarian MDA levels of female white rats exposed to cigarette smoke significantly with p < 0.05. CONCLUSION: However, there has been no significant change in FSH and 17ß-estradiol.

Cigarettes are the biggest health problem around the world. Nicotine in cigarettes has been proved insulin receptor resistance and can reduce insulin secretion in pancreatic β cells, which causes an increase in blood glucose levels. One of the efforts to reduce blood glucose levels is to consume vitamin D3. Vitamin D3 is a prohormone that plays a role in metabolic functions in cells and repair functions of cells. Purpose: to know the effect of vitamin D3 toward glucose blood level of male Rattus norvegicus Wistar exposed by cigarette smoke. Methods: This research conducted with pure experimental pre- and post-test with control group design. The samples used were 24 male rats. The sample divided into four groups, including K1, K2, K3, and P1. K1, which is not exposed by cigarette smoke and not given of vitamin D3. K2 is a group exposed by cigarette smoke but not given vitamin D3. K3 is a group that is not exposed by cigarette smoke but given vitamin D3 a dose of 0.2 µgr /head. P1 is a group exposed by cigarette smoke and given a vitamin D3 dose of 0.2 µgr /head. Results: Paired T-test showed increased blood glucose levels that are meaningful (p<0,05) In groups K2 (p=0,018) and P1 (p=0,035). One-way Anova showed there were differences in blood glucose levels that are meaningful between groups (p=0,001). Statistical analysis Post Hoc LSD showed significant differences in group K1 with K2 (p=0,004), group K1 with K3 (p=0,038), group K1 with P1  (p=0,038), group K2 with K3 (p=0,000),  group K2 with P1  (p=0,004). Conclusion: Vitamin D3 can reduce fasting blood glucose levels of Rattus norvegicus exposed by cigarette smoke.

Background: Cigarette smoke contains various carcinogenic substances such as polycyclic aromatic hydrocarbons and nitrosamines. These chemicals not only have the potential to damage DNA, but can also induce genetic mutations and activate genes that function during apoptosis. Thus, if the gene is dysregulated, it will cause cells to survive, proliferate and subsequently lead to the development of cancerous ones. Histologically, the carcinogenic process affecting the oral cavity starts with hyperplasia and dysplasia, followed by severe dysplasia then leading to invasive cancer and metastatic processes in other bodies. Purpose: This study aims to reveal the correlation between exposure to cigarette smoke and the degree of epithelial dysplasia evident in research subjects. Methods: This study used 27 samples of Rattus norvegicus tongue, divided into three groups, namely; a control group, a treatment group subjected to four weeks’ exposure to cigarette smoke, and a treatment group subjected to exposure lasting eight weeks. Each rat was placed in an individual chamber and exposed to smoke from 20 cigarettes introduced by a pump via a pipe for 7.5 minutes. The degree of epithelial dysplasia in each case was subsequently observed microscopically using HE staining technique. Results: Mild epithelial dysplasia increased by 0.82%, during the fourth week of exposure to cigarette smoke and by 2.99% during the eighth week. Similarly, moderate epithelial dysplasia rose by 5.29% during the fourth week of exposure and 5.99% during the eighth week. Severe epithelial dysplasia also increased by 2.2% during the fourth week of exposure and by 2.66% during the eighth week. Conclusion: The longer the exposure to cigarette smoke, the higher the degree of ensuing dysplasia.

This study aims to analyze the administration of mangosteen peel ethanol extract at different doses can reduce levels of malondialdehyde (MDA) and leukotriene B4 (LTB4) serum in male white rats (Rattus norvegicus) Wistar strains exposed to electric cigarette smoke. Electric cigarette induces mitochondrial ROS production and triggers oxidative stress. Antioxidants are needed by the body if there are many free radicals, for example mangosteen peel. This laboratory experimental study uses a randomized post test only control group design. Thirty male white rats were divided into 5 groups randomly (KK, KR, P1, P2, P3). KK given 0.5% Na-CMC, KR given smoke and 0.5% Na-CMC, P1, P2, P3 given smoke and mangosteen peel ethanol extract 100, 200, 300 mg / kg weight. Data were tested for normality and homogeneity tests. Data distribution was found not normal. Data were tested by Kruskal-Wallis Test and Mann-Whitney. Kruskal-Wallis Test results showed significant differences in MDA and LTB4 levels in each experimental group (KS, KR, P1, P2 and P3), p <0.05. Mann-Whitney test results showed a significant difference in the MDA levels of the KR against P3 and in the LTB4 level of the KR against P2 (p <0.05). The conclusion of the study was the administration of mangosteen peel ethanol extract can reduce MDA and LTB4 levels serum in male white rats (Rattus norvegicus) Wistar strains exposed to electric cigarette smoke.
