Chikitsa is a process which not only aims at the radical removal of the causative factors of the disease, but also at the restoration of the Dhatu Samyata. It is broadly classified into Shodhana and Shamana Chikitsa. Panchakarma considered under Shodhana Chikitsa includes a specially designed five procedures for internal purification of the body. Panchakarma helps in reversal of the pathology by drawing morbid doshas from Shakha to Koshta and eliminating the same through the nearest possible route. In this process, there will be influence of the procedure at both Somatic and Psychological levels. In order to get optimum therapeutic effect of Panchakarma, in order to provide a favorable environment for the procedure to act on the body as well as to ensure the safety of the patient in preventing any complications that may arise either during or after the treatment, a set of rules and regulations pertaining to Ahara, Vihara and Manasika bhavas are to be followed by the client undergoing the Panchakarma. Among these, the Ashtamahadoshakarabhavas also called as Ashtamahavarjyakarabhavas are of paramount importance and creating the awareness in the patient undergoing panchakarma to avoid these eight impediments facilitates the attainment of optimum therapeutic effect without any complications. Henceforth, it is very important to have a thorough knowledge of Ashtamahadoshakarabhavas and its complications as well as its treatment.

Aam is an important concept described by Ayurvedic Acharyas which is responsible for many diseases. According to Ayurvedic point of view, all diseases are originated from aamdosh, vitiation of Agni i.e malfunction of Agni produces Aam. Aam is unripe, undigested food which is caused due to Agnimandya. Agnimandya produces aam and viceversa. We all know that all diseases are caused due to Agnimandya. Therefore as agnimandya and aam are the causative factors of each other, aam is the root cause of all diseases. Hence aam and agnimandya plays an important role in diagnosis and treatment of disease. Nowadays due to lack of exercise, unhygienic and unhealthy diet, incapability to obey the rules of sound body maintenance and increasing pollution results in agnimandya and aam production in the body and decrease in immunity resulting into various diseases. The concept of aam is the most important fundamental principal of Ayurveda in understanding the physio- pathology of the disease.

Mesenchymal stem cell (MSC)-based cell therapy has been demonstrated as a promising strategy in the treatment of inflammatory bowel disease (IBD), which is considered an immune disease. While the exact mechanisms underlying the therapeutic effect of MSCs are still unclear, MSCs display anti-inflammatory and immunomodulatory effects by interacting with various immunoregulatory cells. Our previous studies have shown that MSCs can be preconditioned and deconditioned with enhanced cell survival, differentiation and migration. In this study, we evaluated the effect of preconditioning on the immunoregulatory function of human umbilical cord-derived MSCs (hUCMSCs) and their therapeutic effect on treating IBD. Our results show that intraperitoneal administration of deconditioned hUCMSCs (De-hUCMSCs) reduces the disease activity index (DAI), histological colitis score and destruction of the epithelial barrier, and increases the body weight recovery more intensively than that of un-manipulated hUCMSCs. In addition, De-hUCMSCs but not hUCMSCs elicit anti-apoptotic effects via induction of the ERK pathway during the early stage of IBD development. In vitro co-culture studies indicate that De-hUCMSCs suppress T-cell proliferation and activation more markedly than hUCMSCs. Moreover, De-hUCMSCs block the induction of inflammatory cytokines such as tumor necrosis factor (TNF)α and interleukin (IL)-2, while promoting the secretion of the anti-inflammatory cytokine IL-10 in T-cells. Mechanically, we find that prostaglandin E2 (PGE2) secretion is significantly increased in De-hUCMSCs, the suppression of which dramatically abrogates the inhibitory effect of De-hUCMSCs on T-cell activation, implying that the crosstalk between De-hUCMSCs and T-cells is mediated by PGE2. Together, we have demonstrated that preconditioning enhances the immunosuppressive and therapeutic effects of hUCMSCs on treating IBD via increased secretion of PGE2.

It has been established that in propolis, which is a biologically active product of beekeeping contains several dozen compounds, trace elements and vitamins. It contains most of the compounds of flavonoid and terpenoid nature, phytoncides, unsaturated aromatic acids, trace elements: copper, cobalt, potassium, sodium, manganese, zinc, calcium, barium, titanium, nickel, chromium, vanadium, tin, vitamins (A, B, C, E, PP), glucose, fructose, cinnamon alcohol. This composition provides a favorable complex effect of propolis on the body: bactericidal and virusocidal (kills bacteria and viruses), bacteriostatic (slows their growth), regenerative (restoring tissues, heals wounds) and fungicidal (kills fungi). The main therapeutic effect in this biologically active beekeeping product is provided by ferulic, caffeic and benzoic acids, galantine, chrysin, isalpine, pinocebrin. They are combined with trace elements and vitamins, as well as probiotics and prebiotics, which provide the greatest therapeutic effect. This circumstance is important for fungal diseases of poultry and especially for candidiasis. Geese candidiasis has been causing great economic damage to poultry farming all over the world in recent decades. The search for effective prevention measures and therapies remains an unsolved problem to date. The data on the development of reactions basophils, pseudoeosinophils, eosinophils, lymphocytes, monocytes on the background of candidiasis and their enzyme adaptogens: probiotic and propolis for traditional microbiotheria is not effective and does not contribute to the restoration of cytological reactions and meat production of poultry have been presented in the article. At the same time as shown by our studies in farms affected by candidiasis geese timely application of enzyme therapy on a background of immune propolis and propitiatory not only contributes to the full restoration of all biological indicators, but more intense live weight gain of poultry and improve the livability of their livestock. This underlines the importance of taking measures to prevent candidiasis in goose farms, together with good conditions for feeding and housing poultry.

A previous study has shown that laser acupuncture is a useful healing method for the treatment of visceral postmenopausal obesity in combination with a low-calorie diet. We observe and evaluate the therapeutic effect of laser acupuncture in subjects of simple obesity with a non-restrictive diet protocol. Subjects included 73 women and 22 men with simple obesity and body mass indices ≥ 27 kg/m2. Daily energy intake recommendations for obese females and males were 1620.0 and 1894.2 kcal in average, respectively. The gallium aluminum arsenide Handylaser Trion was used to apply 0.25 J of energy to each of the following acupuncture points three times per week for four consecutive weeks: Stomach, Hunger, ST25, ST28, ST40, SP15, and CV9. The subjects' body weights and body mass indices were recorded before treatment, and four weeks after treatment, and the percent reduction in each parameter was calculated. Statistically significant reductions in body weight and body mass index were detected after four weeks of treatment. The mean reduction and mean percent reduction in body weight were 3.17 kg and 3.80% (p < 0.0001), respectively. The corresponding values for the body mass index were 1.22 kg/m2 and 3.78% (p < 0.0001), respectively. We concluded that laser acupuncture was found to exert a therapeutic effect on simple obesity by reducing both body weight and body mass index. Moreover, subjects showed good compliance with this comfortable and non-restrictive diet protocol.

In Ayurveda, all skin diseases have been described under the umbrella of Kushtha. Ekakusta is considered as the most commonest skin disease that can affect an individual. Acharya Charak has described the involvement of Vata-Kapha in Eka Kushtha. In the present study Eka kushtha is compared with psoriasis due to resemblance of signs symptoms and causative factors with it. Psoriasis is a papulosquamous disorder of the skin, characterized by sharply defined erythematosquamous lesions this illness exhibits a prompt response if treated carefully, left it may run a very chronic course extending into larger area of the body. Ayurveda focuses more on curative management than palliative management. The drug used in the treatment of Ayurveda does not have complications like modern treatment. Repeated Sodhana prevent the recurrence of the disease. Similarly various preparations have been advocated for the management of Psoriasis. The drugs used in this study are Guduchyadi Kashaya & Rajrukshadi Pachana Kashaya. Both the Kashayas are said to be effective in skin diseases. As per Yogaratnakara, Guduchyadhi Kwath has been mentioned as one of the drug having curative effect on all types of Kustha. Hence for present study the orally Guduchyadi Kwath is taken for EkaKustha treatment. The drugs in Guduchyadi Kwatha are Kusthaghana, Twachya, Swedal, Tridosh shamaka, as well as with Rasayan properties, which is beneficial to break down the pathogenesis of disease. As per Gadanigraha, as mentioned in Kayachikitsakhanda, Rajvrukshadipachana Kashaya is mentioned for the treatment of Ekakushtha. Ingredients in Rajvrukshadi Pachana Kashaya reduce Kapha dosha and most of them act on the skin. Kwatha dravyas are Rasadushtihara, Kaphapittahara, Deepana, Twachya, and also having antibacterial, antimicrobial, antiparasitic and anti-inflammatory properties, Hence, it is helpful for clearing the skin infections.

Sukra is the essence of all Dhatus and responsible for the both Prakruta and Vikruta function on the Body in Physiological and Pathological conditions respectively. Androgens are the male gonadal hormones and maintain the male reproductive functions. Acharya Sushruta clears that Sukradhara kala is present in all living organisms and all over the body as like Circulating Androgens in the blood. The dysfunction of Sukra Dhatu will affect all over the body by means of Many Causative factors (Nidanas) or Secondary affection as a result of many systemic disorders. In Bruhatrayi and Laghutrayee explained Ksheena sukra lakshana and Sukrakshaya lakshana to understand Sukra dushti. Here sukra denotes Semen plus androgen. Hyper and hypo androgenic action cause many diseases and itself will get affect by the influence of many systemic disorders so one should meticulously to be evaluated in most of the diseases even other than infertility cases for Sukrakshaya and androgen deficiency.

Abstract In-situ vaccination is a local intervention in which immune enhancing agents are injected locally into one site of tumor, triggering a T cell immune response locally that then travels to attack cancer throughout the body. We have employed a preclinical strategy whereby the same syngeneic tumor is implanted at two separate sites in the body. One tumor is then injected with the test agents and the resulting systemic immune response, if any, is detected by the regression of the distant, untreated tumor. In this test for abscopal therapeutic effects, the combination of unmethylated CG-enriched oligodeoxynucleotide (CpG) - a TLR9 ligand - and agonist anti-OX40 antibody has provided impressive results. This combination lead to durable disease control and long-term treatment-free survival in multiple mouse models of cancer. CpG induced myeloid cells to secrete cytokines, which subsequently induced OX40 expression on T cells. Thus, we hypothesized that administration sequence and timing may affect the anti-tumor responses of in-situ vaccination. In order to screen for the best sequence and timing we implanted A20 lymphoma tumors bilaterally in opposite sides of the abdomen of Balb/C mice. After tumors were established, one tumor was injected at the selected sequence and timing with the test agents and the resulting immune response was monitored by the measuring growth of the distant, untreated tumor. As opposed to our usual schedule of three injections, even a single injection of CpG (50µg) and anti-OX40 (8µg) resulted in a fully protective systemic immune response. In addition, the cured animals were protected from re-challenge with the same A20 tumor but not unrelated tumors. Decreasing the dose even further to 10µg CpG and 1µg anti-OX40 partially preserved the therapeutic response with a long-term survival of 60%. Concurrent administration of CpG and anti-OX40 resulted in eradication of both local and distant disease. Sequential administration of CpG followed by anti-OX40 preserved the therapeutic efficacy. However, the opposite order of anti-OX40 followed by CpG significantly attenuated the therapeutic effect. While CpG followed by a 24- or 48-hour-delayed anti-OX40 treatment preserved the therapeutic efficacy, a 72h delay in anti-OX40 administration resulted in reduced therapeutic effect. These data demonstrate the importance of the administration sequence for fully protective anti-tumor immune responses. Our data suggest that the anti-OX40 antibody should be administered at the same time as CpG or with only a slight delay but not in the reverse order. Low-dose radiotherapy (2×2 Gy) is an effective treatment for patients with indolent non-Hodgkin's lymphoma. This treatment results in high response rates at the treated site. Since immune infiltrating cells in the tumor microenvironment are essential for in situ vaccination of CpG and anti-OX40 we aimed to assess the effect of adding radiation in our pre-clinical models of lymphoma. We found that the addition of 2x2 Gy radiation did not interfere with the induction of a protective immune response by of CpG and anti-OX40. Given the effectiveness of low dose radiotherapy for local control and its lack of interference with the immune related abscopal response in the pre-clinical model, we are including radiation in our current clinical trials. In addition, we have incorporated our findings in the preclinical model regarding dosing and scheduling of CpG and anti-OX40 antibody to the design of our current in situ vaccination lymphoma clinical trial. Abstract In this paper, Mimics is using to reconstruct the 3D model of hemangioma from 2D cerebral angiography images. The process of 3D model reconstruction is formulated. The hemangioma model is extracted and the parameters of the hemangioma are measured, which provided a model basis for the structural design of the spring coil. The shape and structure of the coil are very important for the therapeutic effect. During the treatment, the coil is implanted into the hemangioma from outside the body through a catheter.
