Varieties of plants including Cymbopogon citratus are traditionally used in controlling hyperglycemia by either stimulating insulin secretion, inhibition α- Glucosidase or α-amylase activity. Cymbopogon citratus leaves were shade dried, grinded into fine powder and then extracted by cold maceration using ethanol. Fractionation was done by VLC using dichloromethane, ethyl acetate and ethanol. OGTT was performed for both crude extract and fractions. Diabetes was induced in mice by intraperitoneal injection of freshly prepared alloxan monohydrate (170 mg/kilogram body weight). The mice were treated with ethyl acetate fraction once daily at 400 mg/kilogram body weight dose for the period of 20 days.FBG and weight were then recorded in days 1, 5, 10, 15 and 20 after six hours of fasting. Safety of crude water extract and ethyl acetate fractions were evaluated in mice by using Lorke’s method, followed by 5 days observation for their mortality and behavioral changes. Comparisons of results among groups were analyzed using One-way ANOVA. The difference between the means of the two population groups (each against negative control) was considered significant at p< 0.05. Results were expressed as mean ± SD. Both crude and ethyl acetate fractions from C. citratus showed significant hypoglycemic activity. Moreover, higher hypoglycemic activity was shown by ethyl acetate fraction (p = 0.004). No mortality was observed at 5000 mg/kilogram body weight dose but sleeping and tremor were observed at a 1000 -5000 mg/kilogram body weight dose. Good hypoglycemic and safety results from ethyl acetate fraction highly suggest that Cymbopogon citratus extracts are effective against insulin-dependent hyperglycemia, which may be contributed by the action of the present of alkaloids, saponins, antraquinone, phenol and tannins. Isolation and testing of the active ingredients from the C. citratus extract are thus warranted for use in developing pharmaceutical anti-hyperglycemic drugs from this herbal plant.

Objective: The prevalence of obesity increases each year globally. Multifactorial etiology of obesity requires therapy management including changing of diet and medicines. Some of obesity drugs have shown ineffectiveness and safety. The previous study showed that water extract of tamarind could reduce body weight (bw). This study aimed to test the activity fraction of water extract tamarind as antiobesity using high carbohydrate diet.Method: The preventive research of antiobesity had done by given water fraction and ethyl acetate fraction of water extract tamarind following with induced high carbohydrate diet during 6th weeks in male Wistar rats. The parameters had observed including consumption of food, body weight, weight of feces, volume of urine, total cholesterols, triglycerides, blood glucose, index of organs, and accumulation of body fat.Result: The ethyl acetate fraction at doses 4.5 mg/kg bw has shown significantly effect to decrease of total cholesterols level and decrease of triglycerides level at weeks 6 (p<0.05). All the tests of fraction have shown activity inhibition of increased body weight, decrease of appetite, total cholesterols, triglycerides, and blood glucose. Meanwhile, mechanism action of antiobesity as increase defecation, urination, and decrease index of organs and accumulation of body fat have not shown by all these test fractions.Conclusion: The ethyl acetate fraction at doses of 4.5 mg/kg bw can inhibit raising of body weight, decrease of total cholesterols, and triglycerides level greater than the other test groups, where increasing of these levels of blood biochemistry was closely related to the pathology of obesity.

BACKGROUND: Drug resistance to malaria is still a problem in various regions, and there have even been developments in resistance to the ACTs (artemisinin-based combination therapies) as standard antimalarial drugs included to artemisinin’s partner drugs. Ethyl acetate fraction of G. mangostana L rind, containing xanthones as an antioxidant, has antimalarial activity in vitro which has a synergistic effect with artemisinin. That’s why the activities of this fraction are needed to be studied in vivo. AIM: To explore the antimalarial and antioxidant activity of ethyl acetate fraction of Garcinia mangostana L rind in mice. METHODS: This was a complete randomised design true experimental study. Six groups of mice: a healthy mice group and 5 groups of Plasmodium berghei inoculated mice treated with various doses of the sample for 3 days compared to artemisinin. Parasitemia and total antioxidant status were examined and analysed using ANOVA, and probit analysis were done.RESULTS: The parasitemia level in all of the treatment groups were lower than the positive control group without treatment (p < 0.01) and the parasitemia level was the lowest in artemisinin group which was not significantly different from the 100 mg/kg body weight dose group (p > 0.05). The parasitemia level in 20 and 4 mg/kg body weight dose groups were higher than the artemisinin group (p < 0.01). Parasite growth inhibition rate from the highest to the lowest consecutively was: artemisinin, 100 mg/kg body weight, 20 mg/kg body weight, 4 mg/kg body weight, and positive control group (p < 0.05) and ED50 was 3.396 mg/kg body weight. Total antioxidant status was the highest in 20 mg/ kg body weight dose and higher than the negative control group (p < 0.05) while the lowest total antioxidant status was in the positive control group. CONCLUSION: Ethyl acetate fraction of G. mangostana L rind potentially showed antimalarial and antioxidant activity in vivo. Further study is needed to explore the detail of its mechanism of action and its quantitative phytochemical analysis to find the leading compound in it.

National data shows the prevalence of coronary heart disease is 3,6%. The prevalence of dyslipidemia is around 35,9%. Dyslipidemia has a causal relationship with cardiovascular disease. Based on previous research, 70% ethanolic extract of Ridge Gourd has been shown to have antihyperlipidemic activity. This study aimed to study the activity of 70% ethanolic extract fraction of Ridge Gourd in decreasing the levels of total cholesterol, low-density lipoprotein (LDL-C), and triglycerides in hyperlipidemic hamsters. A total of 32 hamster were divided into 8 groups. Negative control, normal control, positive control (using atorvastatin dose 1,24 mg/kg body weight), positive control (fenofibrate dose 12,35 mg/kg body weight), ethyl acetate fraction, n-hexane fraction, water fraction (fraction doses 36,75 mg/kg body weight), and ethanol extract group (extract dose of 240 mg/kg body weight). The results showed that fraction group (n-hexane, ethyl acetate, water) and ethanol extract group experienced a decrease in total cholesterol, LDL-C, and triglycerides levels. Statistical analysis with one-way ANOVA showed an effect on the treatment given (p = 0,005). Tukey's test showed that ethyl acetate fraction group had an effect comparable to positive control with a decrease in total cholesterol,  LDL-C, and triglycerides level by 56,03%, 52,14%, and 59,51% consecutively. Ethyl acetate fraction (doses 36,75 mg/kg body weight) was the most effective fraction in reducing total cholesterol, LDL-C, and triglycerides.

Objective: Noni fruit (Morinda citrifolia L) and cinnamon bark (Cinnamomum burmannii) are plants that have the hypoglycemic effect on their ethyl acetate fraction in the single dose. This study was to determine the hypoglycemic activity of the combination of ethyl acetate fraction (FEM) and cinnamon bark (FEC) in glucose-induced mice, as well as the dose of the combination of FEM and FEC which could have a hypoglycemic effect. Methods: The methods were including extraction using liquid-liquid extraction, identification by TLC, and assay of hypoglycemic activity in mice induced by oral glucose for a moment in the eight groups that were normal control, negative control, positive control, dose 1-5. The activity tests were performed at 30th, 60th, 90th and 120th of minutes. Results: The research showed that the yield of FEM and FEC were 5.25% and 8.05%. Identification of compounds by TLC showed that the FEM and FEC were 4 and 3 spots. The results of a hypoglycemic test showed that the combination of FEM and FEC on glucose-induced mice showed a decreased effect of mice blood glucose levels better than a single dose of these fractions. Conclusion: The hypoglycemic activity test at a combined dose of FEM 175 mg/kg BW and FEC 150 mg/Kg BW have provided hypoglycemic activity better compared to other dose groups with a percentage decrease in average relative blood glucose at the 30th, 60th, 90th, 120th minute were 29.57%, 44.94%, 43.40% and 40.55%, respectively. Thus, the combination of the FEM and FEC in glucose-induced mice gave a hypoglycemic effect in mice.

ABSTRACT Background: Recent studies showed that Interleukin-8 (IL-8), activated cytokine immune response which plays an important role in the development of acute and chronic gastritis. Harmless anti-inflammatory therapeutic alternatives have been proposed, for example, the consumption of Sechium Edule Jacq. Swartz (chayote). Antioxidant (flavonoid) and cell regeneration (alkaloid) agents were found in chayote. This study aimed to determine the effect of chayote Sechium Edule Jacq. Swartz extracts on the level of IL-8 in Wistar rats with aspirin- induced gastritis. Subjects and Method: This was a randomized controlled trial (RCT) conducted at the laboratory of Mathematics and Natural Science, Universitas Sumatra Utara from January to February 2020. A total of 35 male Wistar rats was selected for this study and randomly allocated into 7 groups: (1) Negative control; (2) Positive control; (3) 100 mg/ kg BW chayote ethanol extract ; (4) 200 mg/kg BW chayote ethanol extract; (5) 100 mg/ kg BW chayote ethyl acetate fraction; (6) 200 mg/kg BW chayote ethyl acetate fraction; and (7) 20 mg omeprazole. The rats in positive control and treatment groups were induced with aspirin (200mg/ kg BW). The negative control group received no intervention. The dependent variable was level of IL-8 measured by ELISA. The independent variables were treatment status. The data were analyzed by One Way Anova and post hoc test. Results: The mean differences of IL-8 level were not statistically significant between study groups (p= 0.327). Mean of IL-8 level was higher in positive control group (Mean= 160.80; SD= 6.90) than in negative control group (Mean= 141.20; SD= 10.98). The lowest IL-8 level was in 100mg/ kg BW chayote ethanol extract group (Mean= 149.94; SD= 40.4), followed by 200mg/ kg BW (Mean= 152.4; SD= 30.73) and 100mg/ kg BW (Mean= 164.60; SD= 25.04) chayote ethyl acetate fraction groups, 20 mg omeprazole group (Mean= 170.60; SD= 21.58), and 200 mg/ kg BW chayote ethanol extract group (Mean= 176.80; SD= 10.98). Conclusion: The low dose (100mg/ kg BW) chayote ethanol extract has the most potential antiinflammation effect on in vitro gastritis with the lowest IL-8 level of all doses of chayote ethanol extract, chayote ethyl acetate fraction, and omeprazole. Amebiasis caused by <em>Entamoeba histolytica</em> is a major public health problem in tropical and subtropical countries. Treatment failure with specific chemotherapy has been reported suggesting the possibility of drug resistance. This study investigated the anti-amoebic effects of four plant extracts on cecal amebiasis in rats. The cecal amebiasis was induced by the injection of 3.0×105 troph/mL of <em>E. histolytica</em> parasite directly into the rat’s caecum. A total of 137 rats were used for these studies; five rats in each group for both positive and negative control, 15 rats in each group to test the four plant extracts and metronidazole. The infected rats were treated for cecal amebiasis using each of the four plant extracts at graded doses of 100 mg/kg, 200 mg/kg and 400 mg/kg and with metronidzole at a dose of 62.5 mg/kg,100 mg/kg and 125 mg/kg for five consecutive days. The efficacy of the four plant extracts were evaluated based on Neal’s, 1951 method. The plant extracts of Garlic, <em>Guava</em>, Pawpaw and Pumpkin at 400 mg/kg and 200 mg/kg body-weight gave a cure rate of 80%, 100%, 60%, 40% and 40%, 80%, 40%, 0%, respectively. The mean parasite count in the cecal contents of the treated rats at a dose 400 mg/kg were 18.5±1.6, 0.0±0.0, 33.3±1.8 and 49.5±4.0, respectively. The difference was statistically significant (P&lt;0.05). This study has revealed that Guava at a high dosing level (400 mg/kg body weight) is as good as the standard drug in reducing the both parasite load (probably with limited side effect).

Background: Sleep disorders are among the most common psychiatric and medical conditions. Objective: The present study aimed at investigating the hypnotic activity of the hydro-alcoholic extract of Capparis spinosa (HAE) in mice. Method: Three doses of HAE (30, 60 and 120 mg/kg) and three fractions of it, namely n-hexane fraction (NHF), water fraction (WF), and ethyl acetate fraction (EAF), were given in comparison with diazepam (3mg/kg body weight i.p.) as positive control and saline as negative control. After 30 min, pentobarbital (30 mg/kg body weight i.p.) was administrated. In addition, LD50 of HAE was examined and the cytotoxicity of HAE was assessed in l929 cells using the MTT assay. Moreover, for motor-coordination ability 30 mins after administration of HAE rota-rod test was performed. Results: The results exhibited that the HAE and all the fractions significantly augmented pentobarbital induced sleeping time which was comparable to that of induced by diazepam. The LD50 value was 2.4 g/kg. The extract did not induce any cytotoxic effects in L929 fibroblast cells. HAE did not affect the animals’ performance on the rota-rod test. Conclusion: Our finding suggests that hydro-alcoholic extract of C. spinosa possesses hypnotic potential that may require further scientific investigations.

The ethanol-water (7 : 3, v/v) extract of Cotoneaster multiflorus sarcocarp was sequentially fractionated by liquid-liquid partition using n-hexane, diethyl ether, methylene dichloride, and ethyl acetate. The contents of total polyphenols, total flavones, and oligomeric proanthocyanidins in the five parts (including the ethanol-water extract) were determined. In addition, 2,2-diphenyl-1-picrylhydrazyl free radical-scavenging, 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt radical cation decolorization, reducing power, ferric reducing antioxidant power, and lipid peroxidation inhibition assays were conducted to test the antioxidant activities of Sample 1 (the ethanol-water fraction) and Sample 2 (the ethyl acetate fraction) in vitro. In the above five assays, Sample 2 showed greater antioxidant capacities than Sample 1. Furthermore, Sample 2 was better able to protect low-density lipoproteins from oxidation in a dose-dependent manner. The test results show that C. multiflorus sarcocarp, especially the ethyl acetate-soluble fraction, may be a potential source of natural antioxidants.

Ethyl acetate fraction of Kalanchoe pinnata (Lmk) Pers leaves (EF-KP) is a potential material to be developed for lupus treatment. The aim of this research was to measure the effects of the EF-KP on lupus arthritis mice.  The research was performed by testing of CD123+IFN-α+ dendritic cells and CD68+IL-6+ macrophages as the main biomarkers by using flow cytometry methods, and then the outcomes were directly observed in the joint’s tissue structure. The results of the research were analyzed by using statistics. The EF-KP reduces the relative percentages of CD123+IFN-α+ dendritic cells significantly (p<0.05) with the percentage of 32.95±8.25% (negative control); 23.28±9.31%* (EF-KP); 22.98±10.39%* (positive control). It also reduces the relative percentages of CD68+IL-6+ macrophages but not significantly (p>0.05). Finally, the outcome to the grade of joint damage was scored by using Pritzker method. The treated groups have one grade lower, and the joint spaces were narrower than the untreated group. The results show the ability of the active compounds in EF-KP, which are comparable to 0.042 mg/Kg quercetin, to inhibit the progress of lupus arthritis pathogenesis in mice. It reveals the effectiveness of the EF-KP in human with lupus arthritis. However, the further clinical research is necessary.
