We aimed to analyze the chemical compositions in Arabica coffee bean extracts, assess the relevant antioxidant and iron-chelating activities in coffee extracts and instant coffee, and evaluate the toxicity in roasted coffee. Coffee beans were extracted using boiling, drip-filtered and espresso brewing methods. Certain phenolics were investigated including trigonelline, caffeic acid and their derivatives, gallic acid, epicatechin, chlorogenic acid (CGA) and their derivatives, p-coumaroylquinic acid, p-coumaroyl glucoside, the rutin and syringic acid that exist in green and roasted coffee extracts, along with dimethoxycinnamic acid, caffeoylarbutin and cymaroside that may be present in green coffee bean extracts. Different phytochemicals were also detected in all of the coffee extracts. Roasted coffee extracts and instant coffees exhibited free-radical scavenging properties in a dose-dependent manner, for which drip coffee was observed to be the most effective (p < 0.05). All coffee extracts, instant coffee varieties and CGA could effectively bind ferric ion in a concentration-dependent manner resulting in an iron-bound complex. Roasted coffee extracts were neither toxic to normal mononuclear cells nor breast cancer cells. The findings indicate that phenolics, particularly CGA, could effectively contribute to the iron-chelating and free-radical scavenging properties observed in coffee brews. Thus, coffee may possess high pharmacological value and could be utilized as a health beverage.

An indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) for ochratoxin A (OTA) detection in green, roasted and instant coffees was developed using anti-OTA monoclonal antibody. Immunological reagents prepared were OTA-BSA (4.76 mg/mL), anti-OTA.7 MAb (2x10³-fold dilution) and HRP-anti IgG (10³-fold dilution). The detection limit was 3.73 ng OTA/g and correlation coefficients (r) between this immunoassay and high performance liquid chromatography were 0.98 for green coffee, 0.98 for roasted and 0.86 for instant. OTA levels detected by ic-ELISA were higher than by HPLC, with ELISA/HPLC ratio of 0.66 - 1.46 (green coffee), 0.96 - 1.11 (roasted) and 0.93 - 1.82 (instant). ELISA recoveries for OTA added to coffee (5 - 70 ng/g) were 81.53 % for green coffee, 46.73 % for roasted and 64.35 % for instant, while recoveries by HPLC were 80.54 %, 45.91 % and 55.15 %, respectively. Matrices interferences were minimized by samples dilution before carrying out the ELISA assay. The results indicate that MAb-based ic-ELISA could be a simple, sensitive and specific screening tool for OTA detection, contributing to quality and safety of coffee products.

Due to concerns about meeting the strict legislation currently in force, anaerobic treatment is being investigated for the treatment of a variety of waste streams. Instant coffee wastes are one type of industrial effluent that appears to be amenable to anaerobic digestion, and this paper presents some results on the biodegradation of this effluent. The method used was a batch bioassay technique known as the Biochemical Methane Potential assay (BMP), which gave 84% degradation of a composite coffee waste sample. Various fractions of the coffee waste revealed mixed degradation results mostly attributed to structural differences, with the liquid fractions being almost 60% degradable, and the solids filter cake fraction, containing most of the lignocellulosic material being only 9% degradable. Volatile fatty acid (VFA) analysis revealed high concentrations of formate being formed and subsequently degraded in the bioconversion process, and a possible role for formate production was postulated. Variations in structure did not appear to affect the route by which VFAs were produced. Bioconversion of 5-6 major classes of pure organic compounds thought to be potentially recalcitrant in coffee effluent was investigated to determine possible mechanisms of degradation, and the extent to which structural variation affected degradability. The phenolics and chlorogenic acids gave the highest degradation of 70% and 60% respectively, and the cyclic volatiles the least degradation at 40%.

Abstract This study developed and validated a method for measuring concentrations of ochratoxin A (OTA) in coffee beverages, not coffee beans. The new method involved extraction using immunoaffinity columns and ultra-performance LC (UPLC)-MS/MS using isotope-dilution techniques. The combination of a fused-core column and UPLC significantly shortened chromatographic time to 3 min compared to reported UPLC methods. The method was sensitive, with an LOD and LOQ of 0.52 and 1.73 pg/mL, respectively. Quantitative intraday (n = 4) and interday (n = 4) biases and RSD were both below 15%. The OTA levels in 40 samples of freshly brewed coffee from chain stores, 24 samples of canned ready-to-drink coffee, and 6 beverages made from instant coffee granules ranged from 1.60 to 93.2 pg/mL (90% positive), 6.00 to 131 pg/mL (100% positive), and 21.8 to 59.0 pg/mL (100% positive), respectively. Based on published tolerable daily intake, men and women in Taiwan should consume no more than 6.3 and 5.1 fifteen gram packages of instant coffee per day, respectively. Specific suggestions were not made for brewed coffee and canned coffee because of their large variation in OTA concentrations. This study should be more relevant to actual human exposure than those studying OTA in green, roasted, and ground coffee beans alone.

This study is aimed to improve the antioxidant property of instant coffee by using microencapsulation technique and spray drying. Concentrated coffee extract was mixed with Konjac glucomannan hydrolysate (KGMH) and Maltodextrin (MD). The mixture of coating material and coffee extract was then spray dried at 160 - 180 °C inlet air temperature and at 85-90 °C outlet air temperature. KGMH can preserve retention of phenolic compounds, DPPH scavenging activity and antioxidant activity of FRAP (p&lt;0.05 of instrant coffee better than other treatment. Coffee consumption is increasing because many types of coffee are circulating, such as instant coffee among the public. The instant coffee studied in this study is the Iwan Fals edition of instant coffee. This study aimed to analyse the lifestyle and effectiveness of instant coffee advertising on instant coffee consumption among peri-urban farmers in Bogor Regency. This study used a cross-sectional study design located in Cibungbulang District, Bogor Regency. The location was chosen purposive because it is a peri-urban area that has more than 20% of agricultural land. A total of 100 respondents in this study were selected using a purposive sampling method with the criteria of a farmer and in the last 12 months consuming and viewing Iwan Fals edition coffee advertisements. The results of this study found that the maker's lifestyle is the most (32%) owned by the respondents. The effectiveness of instant coffee advertising in this study is included in the effective category (3.54). The results showed a relationship between gender, believing lifestyle, trying hard lifestyle with instant coffee consumption. The influence test results found an influence of male gender and lifestyle beliefs on instant coffee consumption. In general, respondents are classified as effective in consuming coffee because of their lifestyle and increase energy.

The higher demands of fossil energy usage are currently imbalance to its reserves. This issue will give a potency of fossil fuel scarcity that contributes on rising of its cost and disturbing economic stabilization in the future. It is desirable to find another resource which is sustainable and renewable to reduce the dependency on this resource. This study aimed to utilize the oil from instant coffee waste as biodiesel feedstocks. The coffee oil was extracted from instant-coffee waste by Soxhlet extractor and characterized its physical and chemical properties. The solvent extraction through the solid bed of coffee ground performed by five cycles interval using n-hexane as a solvent. Separation processing oil from its hydrocarbon solvent was in a rotary vacuum evaporator and the oil extract collected for further purposes. There was about 17.6% (on a dry weight basis) of coffee oil gained on 20 cycles of extraction. Analysis regarding its physicochemical properties reported that the crude oil has 0.89 g/mL of density, 43.82 mm2/s of kinematic viscosity, 44.47 mg KOH/g of acid value, and 176.40 mg KOH/g of saponification value. The fatty acids composition of the oil provided by GC-MS analyzer showed that unsaturated fatty acids contained, observed as trans-fatty acids rather than cis-fatty acids.
