Success in competition is ensured by long-term preparation, but direct pre-competition training (DPT) is of fundamental importance, with the help of which athletes must bring themselves to a state of high or the highest working capacity. Modern views on the essence and construction of preliminary training stage most intensively began to be formed since the 1960s. The reason for this was the appearance of such fundamental projects, such as "Periodization of sports training" by L.P. Matveva, "The current system of sports training" by N.G. Ozolina, "The coach of athletics trainer" ed. L.S. Khomenkovoi. The success in competitions is provided by long-term training, but fundamental importance has a direct pre-competition training (DPT), by which athletes should lead themselves to a state of high or the highest efficiency. If the goal of competition is only of training or control nature and athlete is not eager to achieve a high result, then DPT is not necessary before the starts. Such a combination of the process of training and participation in competitions may occur in the first competitions at the beginning of the competitive period. However, when there is a participating in high-level competitions or even more in the culminating competitions, in such an event, DPT is essential. Mistakes in the building of the phase of the DPT are often the reason not only for an unsuccessful speech in the main competition, but it is also the reason for a slowed down growth of achievements and even deterioration of them. All training of an athlete can be considered successful if he shows the best result in the crucial competition with a significant progress in his accomplishments.

Cryptography is a study discussing mathematic technics that related to information security aspects such as secrecy, data integrity, and authenticity. Cryptography is also a method to secure data or information in the form of a password that makes it hard to understand the meaning. This research discusses how to modify a scheme while looking for random points using different pickup points and income points. While looking for the random point in the modified scheme research using the rice planting and field plow techniques, it has three testing processes which were runtest, monobit, and blockbit tests. This research used square transpositions 64-bit with the size 8x8 and it got a high result with p-value 7.32395E-10, p-value monobit 1.00000000 and p-value blockbit 1 that showed non-random result with the smallest p-value was 0.011662392 with p-value monobit 1.00000000 and the p-value blockbit 0.99990476 that made thisresearch got a random result.

During the training process before the competition, the athlete often shows a high result similar to, and sometimes exceeding, his previously achieved record. However, in the conditions of the following competitive activity, especially in tough sports competition, there is a decrease in effectiveness. This circumstance is often explained by a poor shape of a sportsman. While a more correct objective indicator of the decrease in effectiveness during the competition, as many experts claim, is the psycho-pedagogical mobilisation unpreparedness of an athlete. The aim of this research was to mobilise the strong-willed qualities of highly qualified swimmers, which allow a sportsman to demonstrate his or her best or equal to previously achieved sporting results during important competitions, involving psychological and pedagogical methods. During the implementation of this goal, methods helping to form person’s strongwilled qualities were used, including psychological and pedagogical methods. The results were obtained. They positively characterize applied psychological and pedagogical methods, which effectiveness was proved by the higher level of record achievements shown by the overwhelming majority of participants in the pedagogical experiment during the competitive activities implementation, as well as in the moral and volitional state rise when overcoming the competitive distance. It has been shown that the use of psychological and pedagogical activities together with autotraining contributes to the formation of an internal mood, the state of assemblage and eagerness to achieve one’s best result.

At present the Precast building are more focus for the building industrialization and main objective of this study to find the most appropriate types of beam-column connection, the purpose of this study is to investigate and high result of precast beam column joint with two types of dry mechanical connection & two types of wet connection were analysis and compared with monolithic connection to know the performance of the joints. the primary test variable were the type of connection cleat angle, dowel bar, fibre reinforced concrete, steel plate, anchor rod and the pre tension bolt are used in this joints the advantage of the precast concrete structure components are assemble in which are fabricated off-site and then connect on-site in term of less resource consumption, more rapid construction, better quality control and less field wet operation.

The success of hematopoietic cell transplantation (HCT) is limited by a high rate of disease relapse. Post-HCT chimerism analysis is typically used to evaluate donor cell engraftment and success of therapy in allogeneic HCT. The detection of microchimerism, especially in the early period after allogeneic HCT, is an important high-risk factor for disease relapse (Ahci et al, 2017). However, common approaches based on semi-quantitative fluorescent PCR of short tandem repeats (STRs) suffer from low sensitivity and high result variability and often fail to detect microchimerism and predict patient outcomes (Schuurhuis et al, 2018). Here, we describe development and characterization of AlloHemeTM, a novel sensitive and precise chimerism surveillance solution with high accuracy for detection and quantification of low levels of patient cells which ultimately can lead to clinical disease recurrence. The method is compatible with chimerism assessment in whole blood, bone marrow, or specific cell subtypes. CD3, CD15, CD33 and CD34 markers are used to separate T cells, myeloid progenitors and granulocytes from blood and hematopoietic stem cells from bone marrow, respectively. A minimum of 8ng genomic DNA is used to amplify hundreds of single nucleotide polymorphisms (SNPs) using target specific primers in a single multiplex reaction. Because there is no need to select genetic variants that differ for specific donor-patient pair, the method is universally applicable to any allogeneic HCT. Relative contribution of SNP alleles is measured by Next Generation Sequencing (NGS) and % chimerism of post-HCT samples is calculated using an analysis pipeline incorporating open source and custom bioinformatics tools. To evaluate reproducibility of cell subtype separation, blood from 3 healthy volunteers was subjected to cell separation for CD3, CD15 and CD33 cells at four different timepoints after draw (24, 48, 72 and 96 hours). Flow cytometry analysis using non-competing antibodies targeting same cell populations demonstrated that the cell subtype isolation step consistently yields sub-populations with an average purity of 96% across all cell subtypes and timepoints evaluated (93-99% range). To evaluate assay sensitivity, accuracy and precision, genomic DNA samples (gDNA, Coriell Institute) were used to prepare 2 panels, A and B, each containing different mixtures of gDNAs to simulate post-HCT samples with varying contribution of patient's cells. Panel A contained mixtures of gDNA samples from unrelated individuals ranging from 0.01-50% and panel B from related (parent-child) ranging from 0.01-75%. Each mixture was tested in 6 (Panel A) or 5 (Panel B) independent replicates. The results show that the assay is accurate from 0.03% through 50% for panel A and 0.1% through 75% for panel B with minimal proportional and systematic bias (Figure 1). The assay precision is high, with a mean coefficient of variation (CV) of 2.6% for panel A and 6.1% for panel B across the entire linear range. For sample mixtures with chimerism levels greater than 1%, mean CV was 0.76% for panel A and 1.62% for panel B. AlloHeme results show that the test can detect microchimerism down to 0.03%. High result reproducibility is achieved through the use of a proprietary NGS process and automated data analysis and workflow, generating results in the CLIA lab with a turnaround time of 3 days. The test was developed with rigorous QC steps to ensure high purity and accuracy for each cell population analyzed, increasing confidence and sensitivity of results. Given previously published data on clinical value of microchimerism, AlloHeme has the potential for early relapse detection post-HCT. Future studies will evaluate the utility of AlloHeme in clinical setting with the goal to improve patient outcomes and survival rates. In summary, we have developed a sensitive, accurate and precise method for the universal detection and quantification of chimerism and potential early disease recurrence detection for allogeneic HCT patient surveillance. Because the method provides high sensitivity and consistent results for microchimerism detection, it may be particularly suited to help identify recurrence of primary disease in hematologic cancer patients with or without known cancer-associated genetic determinants. The purpose of this study was to find out the implementation of the 2013 Curriculum in Bekasi Elementary Schools. The method used in this study is the survey method. The sample in this study was a physical education teacher who taught at the Bekasi Elementary School amount of 30 people. The instrument used by the questionnaire of teaching indicators which include: (a) preparation factors, (b) application factors, and (c) evaluation factors. Data analysis technique were descriptive statistics by using the SPSS Version 23 program. The result showed that the 2013 curriculum implementation of physical education teachers is viewed from 3 factors which are as follows: (1) Preparation shows the very high result as many as 32.6%, while as many as 14.8% answered very low, 7.3% answered low, 16.3% answered moderate, and 29.1% answered high, (2) Application shows the high result as many as 35.1%, while as many as 16.4% answered very low, 14.1% answered low, 12.6% answered moderate, and 21.9% answered very high, (3) Evaluation shows the high result as many as 41.9%, while 7.6% answered very low, 7.6% answered low, 11% answered moderate, and 31.9% answered very high. So, the implementation of the 2013 curriculum in Physical Education subjects in Bekasi is already good in the preparation factor and needs improvement in application and the evaluation.

In this research, aloe vera peeling and dicing system is designed and fabricated with high capacity of 5 ton/hr. The system consists of aloe vera fillet, peeling and dicing mechanism. The system is tested for different speed of the roller and leaf thickness. It is concluded that for getting the maximum gel peeling efficiency 98.56%, the system should be carried out at 75 rpm roller speed for 20-22 mm thickness of aloe vera leaves. The dicing efficiency also exhibits high result with mechanism help changing the flexible aloe vera dice size. Moreover, the system also shows the high stability for reaching capacity of 5 ton/hr while running continuously in ten hours.

Biology is a fairly complicated initial subject because it involves knowledge of biodiversity. Google Lens is a unique, mobile software that allows you to recognition species and genus of the plant student looking for. The article devoted to the analysis of the efficiency of the functioning of the Google Lens related to botanical objects. In order to perform the analysis, botanical objects were classified by type of the plant (grass, tree, bush) and by part of the plant (stem, flower, fruit) which is represented on the analyzed photo. It was shown that Google Lens correctly identified plant species in 92.6% cases. This is a quite high result, which allows recommending this program using during the teaching. The greatest accuracy of Google Lens was observed under analyzing trees and plants stems. The worst accuracy was characterized to Google Lens results of fruits and stems of the bushes recognizing. However, the accuracy was still high and Google Lens can help to provide the researches even in those cases. Google Lens wasn’t able to analyze the local endemic Ukrainian flora. It has been shown that the recognition efficiency depends more on the resolution of the photo than on the physical characteristics of the camera through which they are made. In the article shown the possibility of using the Google Lens in the educational process is a simple way to include principles of STEM-education and “New Ukrainian school” in classes.

The latest data stated that the rate of deforestation has reached 2.83 million hectares per year during 1997 to 2000 and was reduced to 1.08 million hectares per year 2000-2005. It included the population ofironwood, population of ironwood is thinning because of the loggers who are logging ironwood as a good effort. Ironwood has been classified as endangered, but in the District of Kintap there are still many loggers of stump ironwood. Respondents need to have knowledge about the population of ironwood plant to measure the extent of respondents understanding related to their habits, especially in terms of ironwood conservation. People’s habit to use the ironwood plants in the district Kintap is the behavior of the villagers who live in the area near the forest who are not concerned about the reduction in the plant population. The purpose of this study are (1) to describe the knowledge and the habitof the loggers and collectors about the scarcity of the ironwood plantpopulation in Kintap district (2) Create public information material that is made by the results of the research know ledge and habits of loggers and collectors of the population ironwood plants in Kintap district as leaflets. The results showed (1) Knowledge of the loggers and the collectors about ironwood plant population, they knewabout the breeding, growth, utilization, distribution and conservation, but not very aware of the rules. Loggers and collectors habit on logging and selling is always done without making purchaselicense. In addition, they did not care about the preservation of ironwood (2) the result of the leafle treadability assessment of loggers, collectors and the public are rated high. Result of the validation by the three validators categorized as very valid.

Internet that has developed into a good distribution tool for digital data, causing a large increase in digital data sharing, especially in the form of digital images, and causing problems that need attention. One of them is about copyright protection. Watermarking is one technique that aims to protect digital image copyright. In this study, watermarking was carried out using the Discrete Wavelet Transform (DWT) and Singular Value Decomposition (SVD) algorithms in digital images with different file extensions. Transformation of 2 levels of DWT and SVD on the host image and SVD transformation on the watermarked image that inserted in the LL2 sub-band of the host image. Watermarking with the proposed method produces good image quality with PSNR values exceeding 40 dB, SSIM reaching 0.99 and NCC reaching 1. This shows a robust and imperceptible watermarking image.
