The term cicatricial alopecia results from irreversible damage to epithelial stem cells located in the bulge region of the hair follicle with subsequent scarring. Based on the mechanism involved in follicular destruction, cicatricial alopecia is divided into primary and secondary forms. Primary cicatricial alopecia are divided into four groups according to their prominent inflammatory infiltrate: with lymphocytic, neutrophilic, mixed or nonspecific cell inflammation pattern. The review presents the main clinical, histological and dermatoscopic signs of various types of primary cicatricial alopecia.

We studied CD34+ stromal cells/telocytes (CD34+SCs/TCs) in pathologic skin, after briefly examining them in normal conditions. We confirm previous studies by other authors in the normal dermis regarding CD34+SC/TC characteristics and distribution around vessels, nerves and cutaneous annexes, highlighting their practical absence in the papillary dermis and presence in the bulge region of perifollicular groups of very small CD34+ stromal cells. In non-tumoral skin pathology, we studied examples of the principal histologic patterns in which CD34+SCs/TCs have (1) a fundamental pathophysiological role, including (a) fibrosing/sclerosing diseases, such as systemic sclerosis, with loss of CD34+SCs/TCs and presence of stromal cells co-expressing CD34 and αSMA, and (b) metabolic degenerative processes, including basophilic degeneration of collagen, with stromal cells/telocytes in close association with degenerative fibrils, and cutaneous myxoid cysts with spindle-shaped, stellate and bulky vacuolated CD34+ stromal cells, and (2) a secondary reactive role, encompassing dermatitis—e.g., interface (erythema multiforme), acantholytic (pemphigus, Hailey–Hailey disease), lichenoid (lichen planus), subepidermal vesicular (bullous pemphigoid), psoriasiform (psoriasis), granulomatous (granuloma annulare)—vasculitis (leukocytoclastic and lymphocytic vasculitis), folliculitis, perifolliculitis and inflammation of the sweat and sebaceous glands (perifolliculitis and rosacea) and infectious dermatitis (verruca vulgaris). In skin tumor and tumor-like conditions, we studied examples of those in which CD34+ stromal cells are (1) the neoplastic component (dermatofibrosarcoma protuberans, sclerotic fibroma and solitary fibrous tumor), (2) a neoplastic component with varying presentation (fibroepithelial polyp and superficial myxofibrosarcoma) and (3) a reactive component in other tumor/tumor-like cell lines, such as those deriving from vessel periendothelial cells (myopericytoma), epithelial cells (trichoepithelioma, nevus sebaceous of Jadassohn and seborrheic keratosis), Merkel cells (Merkel cell carcinoma), melanocytes (dermal melanocytic nevi) and Schwann cells (neurofibroma and granular cell tumor).

Biological macromolecules often exhibit correlation in fluctuations involving distinct domains. This study decodes their functional implications in RNA-protein recognition and target-specific binding. The target search of a peptide along RNA in viral TAR-Tat complex is closely monitored using atomistic simulations, steered molecular dynamics simulations, free energy calculations, and a machine-learning-based clustering technique. An anti-correlated domain fluctuation is identified between the tetraloop and the bulge region in the apo form of TAR RNA that sets a hierarchy in the domain-specific fluctuations at each binding event and that directs succeeding binding footsteps. Thus, at each binding footstep, the dynamic partner selects an RNA location for binding where it senses higher fluctuation, which is conventionally reduced upon binding. This event stimulates an alternate domain- fluctuation which then dictates sequential binding footstep/s and thus, the search progresses. Our cross-correlation maps show that the fluctuations relay from one domain to another specific domain till the anti-correlation between that inter-domain fluctuations sustains. Artificial attenuation of that hierarchical domain fluctuation inhibits specific RNA binding. The binding is completed with the arrival of a few long-lived water molecules that mediate slightly distant RNA-protein sites and finally stabilizes the overall complex. The study underscores the functional importance of naturally designed fluctuating RNA motifs (bulge, tetraloop) and their interplay in dictating the directionality of the search in a highly dynamic environment.

<p>Following substorm auroral onset, the active aurora region usually expands poleward toward the poleward auroral boundary. Such poleward expansion is often associated with a bulge region that expands westward and forms the westward travelling surge (WTS). In this paper we show all-sky imager and Poker Flat Advanced Modular Incoherent Scatter Radar (PFISR) radar observations of two surge events to investigate the relationship between the surge and flow from the polar cap. For both events, we observe auroral streamers, with an adjacent flow channel consisting of decreased density and cool electron temperature plasma flowing equatorward. This flow channel appears to impinge and lead/feed surge formation, and to stay connected to the surge as it moves westward. Also, for both events, streamer observations indicate that, following initial surge development, similar flows led to explosive surge enhancements. The observation that the streamers connected to the auroral polar boundary and that the flow channels consisted of low density, low electron temperature plasma indicates that the impinging plasma came from the polar cap. For both events, the altitude variations of F region plasma within the surges are related with aurora emission and the poleward/equatorward flow, and the surges develop strong auroral streamers that initiate along the poleward auroral boundary when contacted with flow from the polar cap. These results suggest that the polar cap flow channels play a crucial role in auroral surges by feeding low entropy plasma into surge initiation and development, and also playing an important role in the dynamics within a surge.</p>

AbstractIn mammals, organ induction occurs only during embryonic development except for hair follicles (HFs). However, HF-resident epithelial stem cells (HFSCs), which are responsible for repetitive HF regeneration, are not fully characterized. Here, we establish in vitro culture systems that are capable of controlling the ability of HFSCs to regenerate HFs. Based on a method that precisely controlled the number of HFs for regeneration, functional analysis revealed that CD34/CD49f/integrin β5 (Itgβ5)-triple-positive (CD34+/CD49f+/Itgβ5+) cells have multipotency and functional significance for continual hair regeneration. In native HFs, these cells reside in the uppermost area of the bulge region, which is surrounded by tenascin in mice and humans. This study unveils the subpopulation of HFSCs responsible for long-term hair cycling of HFs regenerated from bioengineered HF germ, suggesting the presence of functional heterogeneity among bulge HFSCs and the utility of our culture system to achieve HF regenerative therapy.

AbstractThis study focuses on mechanisms of shelf valley bathymetry affecting the spread of riverine freshwater in the nearshore region. In the context of Changjiang River, a numerical model is used with different no-tide idealized configurations to simulate development of unforced river plumes over a sloping bottom, with and without a shelf valley off the estuary mouth. All simulated freshwater plumes are surface-trapped with continuously growing bulges near the estuary mouth and narrow coastal currents downstream. The simulations indicate that a shelf valley tends to compress the bulge along the direction of the valley long axis and modify the incident angle of the bulge flow impinging toward the coast, which then affects the strength of the coastal current. The bulge compression results from geostrophic adjustment and isobath-following tendency of the depth-averaged flow in the bulge region. Generally, the resulting change in the direction of the bulge impinging flow enhances down-shelf momentum advection and freshwater delivery into the coastal current. Sensitivity simulations with altered river discharges (Q), Coriolis parameter, shelf bottom slope, valley geometry, and ambient stratification show that enhancement of down-shelf freshwater transport in the coastal current, ΔQc, increases with increasing valley depth within the bulge region and decreasing slope Burger number of the ambient shelf. Assuming potential vorticity conservation, a scaling formula of ΔQc?Q is developed, and it agrees well with results of the sensitivity simulations. Mechanisms of valley influences on unforced river plumes revealed here will help future studies of topographic influence on river plumes under more realistic conditions.

Stars with higher levels of aluminium and nitrogen enrichment are often key pieces in the chemical makeup of multiple populations in almost all globular clusters (GCs). There is also compelling observational evidence that some Galactic components could be partially built from dissipated GCs. The identification of such stars among metal-poor field stars may therefore provide insight into the composite nature of the Milky Way (MW) bulge and inner stellar halo, and could also reveal other chemical peculiarities. Here, based on APOGEE spectra, we report the discovery of 29 mildly metal-poor ([Fe/H] ≲ −0.7) stars with stellar atmospheres strongly enriched in aluminium (Al-rich stars: [Al/Fe] ≳ +0.5), well above the typical Galactic levels, located within the solar radius toward the bulge region, which lies in highly eccentric orbits (e ≳ 0.6). We find many similarities for almost all of the chemical species measured in this work with the chemical patterns of GCs, and therefore we propose that they have likely been dynamically ejected into the bulge and inner halo from GCs formed in situ and/or GCs formed in different progenitors of known merger events experienced by the MW, such as the Gaia-Sausage-Enceladus and/or Sequoia.

<p>Frontal fibrosing alopecia (FFA) is a primary lymphocytic cicatricial alopecia, characterized by recession of frontotemporal hairline with frequent involvement of eyebrows and affecting occasionally other body areas. Entitled an emerging epidemic, due to its rising incidence, FFA etiology remains unclear without proven effective therapies. This study reviews relevant publications on FFA, regarding pathogenesis, clinical findings, histology along with treatment and prognosis. A bibliographic search was conducted in the main international databases, using the term ‘FFA’. Guidelines, observational studies, randomized controlled trials, reviews, systematic reviews and meta-analysis regardless of publication date, presented in English, Portuguese or Spanish, were included in this review. 50 publications were fully analysed. The majority of FFA cases were observed in postmenopausal women, although both men and women from younger ages can be victims. From an etiological point of view, immune-mediated hypothesis is widely spread, as stem cells localized in the bulge region of the follicle are destroyed by an inflammatory infiltrate. However, environmental factors raised interest, since sunscreen use was related to a bigger incidence of FFA. Three clinical patterns have been described. Yet the failure to use standardized assessment methods in addition to the absence of prospective studies evaluating available therapies, do not concede comparable data. Spontaneous stabilization of disease can occur, but regrowth was only seen in scarce cases. Outcomes from randomized controlled trials and prospective studies are missing to draw further understanding of FFA.</p>

ABSTRACT We use data of ∼13 000 stars from the Sloan Digital Sky Survey/Apache Point Observatory Galactic Evolution Experiment survey to study the shape of the bulge metallicity distribution function (MDF) within the region |ℓ| ≤ 11° and |b| ≤ 13°, and spatially constrained to RGC ≤ 3.5 kpc. We apply Gaussian mixture modelling and non-negative matrix factorization decomposition techniques to identify the optimal number and the properties of MDF components. We find that the shape and spatial variations of the MDF (at [Fe/H] ≥ −1 dex) are well represented as a smoothly varying contribution of three overlapping components located at [Fe/H] = +0.32, −0.17, and −0.66 dex. The bimodal MDF found in previous studies is in agreement with our trimodal assessment once the limitations in sample size and individual measurement errors are taken into account. The shape of the MDF and its correlations with kinematics reveal different spatial distributions and kinematical structure for the three components co-existing in the bulge region. We confirm the consensus physical interpretation of metal-rich stars as associated with the secularly evolved disc into a boxy/peanut X-shape bar. On the other hand, metal-intermediate stars could be the product of in-situ formation at high redshift in a gas-rich environment characterized by violent and fast star formation. This interpretation would help us to link a present-day structure with those observed in formation in the centre of high-redshift galaxies. Finally, metal-poor stars may correspond to the metal-rich tail of the population sampled at lower metallicity from the study of RR Lyrae stars. Conversely, they could be associated with the metal-poor tail of the early thick disc.

The pilosebaceous unit is the triad comprising of hair follicle, arrector pilli muscle, and sebaceous gland. Drug delivery to and through the hair follicles has garnered much attention of the researchers and the hair follicles represent an attractive target site via topical applications. They are bordered by capillaries and antigenpresenting cells, connected to the sebaceous glands and the bulge region of the hair follicle anchors the stem cells. The nano lipid carriers have the propensity to penetrate through the skin via transcellular route, intracellular route and follicular route. It has been established that nano lipid carriers have the potential for follicular drug delivery and provide some advantages over conventional pathways, including improved bioavailability, enhanced penetration depth, fast transport into the skin, tissue targeting and form a drug reservoir for prolonged release. This review describes the pilosebaceous unit (PSU) and related diseases and the recent lipid-based nanotechnology approaches for drug delivery to the follicular unit as well as related issues. Different types of nano lipid carriers, including ethosomes, liposomes, nanoparticles, solid lipid nanoparticles (SLNs), and nano lipid carriers (NLCs) have been reported for follicular drug delivery. Targeted drug delivery with nano-lipid carriers has the potential to augment the efficacy of drugs/bioactives to treat diseases of PSU. This review systematically introduces the activities of different formulations and the use of nano lipid carriers in treating PSU related disorders like alopecia, acne, and hirsutism.
