Kinome Analysis of Cattle Peripheral Lymph Nodes to Elucidate Differential Response to Salmonella spp.

Salmonella spp., contained within the peripheral lymph nodes (PLNs) of cattle, represents a significant source of contamination of ground beef. Herein is the first report where species-specific kinome peptide arrays designed for bovine biology were used to further the understanding of Salmonella spp. within these PLNs. For the purpose of this research, multiple comparisons of sub-iliac lymph nodes were made to include nodes from feedlot cattle that were infected with Salmonella spp. to those that were non-infected; seasonal differences in feedlot cattle harvested in either August or January; cull dairy cows compared to feedlot cattle; and PLNs from cattle experimentally inoculated with Salmonella spp. versus naturally infected animals. The first comparison of Salmonella-positive and -negative PLNs found that considering the kinotypes for these animals, the major distinguishing difference was not the presence or absence of Salmonella spp. in the PLNs but the concentration. Further, the majority of pathways activated were directly related to immune responses including innate immunity, thus Salmonella spp. within the PLNs activates the immune system in that node. Results from the comparison of feedlot cattle and cull dairy cows suggests that a Salmonella spp.-negative animal, regardless of type, has a more consistent kinome profile than that of a Salmonella spp.-positive animal and that the differences between feedlot and cull dairy cattle are only pronounced when the PLNs are Salmonella spp. positive. PLNs collected in the winter showed a much more consistent kinome profile, regardless of Salmonella status, suggesting that in the winter these cattle are similar, and this is not affected by the presence of Salmonella spp., whereas significant variability among kinotypes was observed for PLNs collected in the summer. The most distinct clustering of kinotypes observed in this study was related to how the animal was infected with Salmonella spp. There were significant differences in the phosphorylation state of the immune response peptides between experimentally and naturally infected animals, suggesting that the immune system is activated in a significantly different manner when comparing these routes of infection. Increasing our understanding of Salmonella spp. within cattle, and specifically within the PLNs, will ultimately help design effective pre-harvest intervention strategies as well as appropriate experimentation to validate those technologies.

Animals in flow – Towards the scientific study of intrinsic reward in animals

The concept of flow, a state of complete absorption in an intrinsically rewarding activity, has played a pivotal role in advancing notions of human well-being beyond minimising suffering towards promoting flourishing and thriving. While flow has played a fundamental role in human positive psychology, it has not yet been explored in non-human animals, leaving an enormous void in our understanding of intrinsic motivation in animals. As ethology and related fields keep progressing in uncovering complex cognitive and affective capacities of non-human animals, we propose the time is ripe to translate the concept of flow to animals. We start by embedding flow in the topic of intrinsic motivation and describe its impact on positive human psychology and potentially positive animal welfare. We then disambiguate flow from related concepts discussed in the animal literature. Next, we derive experimental approaches in animals from the canonical characteristics of flow in humans and provide guidelines for both inducing and assessing flow by focusing on two characteristics that do not necessarily depend on self-report, namely resistance to distraction and time distortion. Not all aspects of the human flow experience are (yet) translatable, but those that are may improve quality of life in non-human animals.

Investigating the Reward Cycle of Play in Pigs (Sus scrofa)

Observations of play in animals have been suggested as a promising indicator of positive emotions and thus of positive animal welfare. However, if play can follow the proposed reward cycle concept where animals estimate and value reward differently in different phases of the cycle (anticipation, consummation and post-consummation) is unclear. To investigate if a reward cycle for play exists in growing pigs, we carried out an exploratory study where pigs were tested when they were naïve to a reward cycle test (first occasion) against when they were accustomed to going through the test after having the access to an open play arena with objects. Forty undocked pigs were housed in a weaner stable with two castrated males and two females per pen. Within each litter, we randomly selected and tested one male and one female test pig, each being tested as naïve or accustomed to the testing environment. The first week the pigs (n = 20) were tested four times and regarded as naïve during the first day. After that they were regarded as accustomed, and were tested twice a week for 3 weeks. We observed the behavior of the tested pairs in three subsequent stages: (1) in a holding pen 3 min, (2) in a play arena 15 min, and (3) in their home pen 10 min. When accustomed, pigs showed more locomotor play, social interactions and standing, and a tendency of more orientation toward the play arena and exploring bars facing the play arena (i.e., reward-seeking behavior) in the holding pen than when they were naïve, suggesting an anticipation to enter the play arena. Performing high numbers of object play in all sessions, and for accustomed pigs more exploration and social interaction, but less locomotor play and walking in the play arena may suggest consumption of play and exploration. Finding more lying and sitting in accustomed pigs, but less standing and walking in the home pen is in line with the previous hypothesis of the post-consummatory behaviors. Our study showed mixed results for the existence of a reward cycle for play in pigs and generated questions for future research.

Behavioral diversity as a potential positive indicator of animal welfare in bottlenose dolphins

Accredited zoological facilities are committed to fully understanding the behavioral, mental, and physical needs of each species to continuously improve the welfare of the animals under their professional care and detect when welfare has diminished. In order to accomplish this goal, internally consistent and externally valid indicators of animal welfare are necessary to advance our understanding of the current welfare status of individual animals. Historically, efforts have focused on monitoring visible or observable signs of poor health or problem behavior, but lack of signs or problems does not necessarily demonstrate that an individual animal is thriving. The current study examined fecal hormone metabolite levels and behavior for two species of bottlenose dolphins (Tursiops truncatus and Tursiops aduncus) from 25 different accredited zoological facilities. At the time of the study, all facilities were accredited by the Alliance of Marine Mammal Parks and Aquariums and/or the Association of Zoos and Aquariums. This was part of the multi-institutional study ‘Towards understanding of the welfare of cetaceans in zoos and aquariums” commonly referred to as the Cetacean Welfare Study. Behavioral diversity was calculated using the Shannon Diversity Index on species-appropriate behavioral events. Behavioral diversity was compared to the fecal metabolites of cortisol, aldosterone, and the ratio of cortisol to dehydroepiandrosterone (DHEA) as well as the stereotypic behavior of route tracing. Similar to previous studies on other species, there was a significant inverse relationship between behavioral diversity and both fecal cortisol metabolites and route tracing. Additionally, a significant inverse relationship also exists between behavioral diversity and the ratio of fecal cortisol to DHEA metabolites. Behavioral diversity and fecal aldosterone metabolites were not associated. Additional research is still needed to validate behavioral diversity as an indicator of positive animal welfare for bottlenose dolphins and across species. However, based on current results, facilities could utilize behavioral diversity combined with other measures of welfare to more comprehensively evaluate the welfare of bottlenose dolphins.

Epidemiological situation of bovine tuberculosis in the state of Tocantins, Brazil

The epidemiological situation of bovine tuberculosis was studied from October 2014 to August 2015 in the state of Tocantins, a traditional beef exporter. The state was divided into five regions and the farms were randomly selected in each region. A predetermined number of animals was randomly selected and diagnosed using the Comparative Cervical Tuberculin Test. A total of 11,926 animals from 757 farms were tested. Animals with inconclusive test results were retested with the same diagnostic procedure at least 60 days later. The prevalence of infected herds in the state was 0.16% [0.02-1.15] and the prevalence of infected animals 0.009% [0.001-0.063]. Only one positive animal was detected in the Araguaína region. The state of Tocantins has a very low prevalence of bovine tuberculosis and, therefore, should evaluate the need of structuring a surveillance system to eradicate the disease.

Gastrointestinal Strongyles Egg Excretion in Relation to Age, Gender, and Management of Horses in Italy

Current equine helminth control strategies play a key role in strongyle epidemiology and anthelmintic resistance and have led to the recommendation for new treatment plans, which include diagnostic and efficacy surveillance. Assessing the equine strongyle distribution patterns would thus be useful and this study describes the strongyle prevalence in the equine population in Italy through coprological analysis and coproculture. In addition, individual data on each animal were collected in order to identify risk factors associated with strongyle egg shedding. Of the total number of stables investigated, 86.4% were found to have at least one positive animal and a 39.5% prevalence of strongyle egg shedding with a mean eggs per gram (EPG) of 245. A total of 80% of the total recorded EPG was shed from 12.8% of positive horses, thus confirming the need for new targeted intervention strategies. Significant differences in parasite prevalence were found based on season, sex, geographical distribution, management and rearing system, and breed. Significantly lower EPG values were found in horses that had received anthelmintic treatment, and macrocyclic lactones (MLs) were the most effective. Lastly, although large strongyles are more pathogenic, horses in Italy are mainly burdened by small strongyles, which pose an important animal health risk requiring continuous parasitological monitoring.

Using Zoo Welfare Assessments to Identify Common Issues in Developing Country Zoos

Zoo animal welfare is a high priority for many institutions worldwide, with modern zoos now ensuring that animals are housed and cared for to the highest standards. However, in countries where this knowledge is not as available or understood, standards may be lower. The aim of this research was to investigate if there were common zoo welfare concerns across developing country zoos. Wild Welfare is a charity working globally to improve welfare for zoo animals and has an independent welfare audit that is carried out before any intervention occurs. The Wild Welfare Audit, consisting of 110 questions, covering nine topics, was completed at 11 zoos in seven developing countries (Brazil, Egypt, Libya, Indonesia, Thailand, Malaysia and Vietnam) following a Likert scale score (1–3). A principal component analysis was also performed to evaluate the audit questions. The results suggest that common areas of concern were animal behaviour, positive animal mental states and human health and safety. These themes were likely due to the lack knowledge and understanding that may be linked to historical and cultural differences. This research has helped to revise the welfare audit as well as inform future intervention strategies for improving developing country zoo animal welfare.

Moving Beyond the Absence of Pain and Distress: Focusing on Positive Animal Welfare

For many years, researchers, veterinarians, animal ethics committees, and regulators have focused on minimizing pain and distress as a primary goal of refinement when working with animals in science. More recent publications as well as a shift in animal ethics and public opinion have emphasized promotion of positive affective states, culminating in the concept of positive animal welfare. Robust measures are required to know when positive animal welfare states are occurring, and a number of measures are proposed and discussed. Regardless of whether there are newer methods available that focus exclusively on measuring positive affective states, consistent consideration of research animal behavioral programs, refinement, and adopting periodic stand-alone animal welfare assessments for all species involved will help to push the care and practices of research animals towards an increased focus on positive animal welfare.

Behavioral Diversity as a Potential Indicator of Positive Animal Welfare

Modern day zoos and aquariums continuously assess the welfare of their animals and use evidence to make informed management decisions. Historically, many of the indicators of animal welfare used to assess the collection are negative indicators of welfare, such as stereotypic behavior. However, a lack of negative indicators of animal welfare does not demonstrate that an individual animal is thriving. There is a need for validated measures of positive animal welfare and there is a growing body of evidence that supports the use of behavioral diversity as a positive indicator of welfare. This includes an inverse relationship with stereotypic behavior as well as fecal glucocorticoid metabolites and is typically higher in situations thought to promote positive welfare. This review article highlights previous research on behavioral diversity as a potential positive indicator of welfare. Details are provided on how to calculate behavioral diversity and how to use it when evaluating animal welfare. Finally, the review will indicate how behavioral diversity can be used to inform an evidence-based management approach to animal care and welfare.

The continued citation of retracted publications in dentistry

Objective: Publications are retracted for many reasons, but the continued use and citation of retracted publications presents a problem for future research. This study investigated retractions in the dental literature to understand the characteristics of retracted publications, the reasons for their retractions, and the nature and context of their citations after retraction.Methods: In September 2018, the authors identified retracted dentistry publications using the Retraction Watch database. Citations to those publications were retrieved from Scopus and Web of Science. Characteristics of retracted publications and their citations were collected, including study design, reasons for retraction, and nature of citation (positive, negative, or neutral). We used chi-square tests to determine if there were notable differences between retracted publications that were cited following retraction and those that were not, and if there were relationships between the nature of the citation, the study design of the original publication, and its reason for retraction.Results: Of the 136 retracted publications, 84 were cited after retraction. When restricted to English language, 81 retracted publications received citations from 685 publications. Only 5.4% of the citations noted the retracted status of the original publication, while 25.3% of citations were neutral and 69.3% were positive. Animal studies were more likely to be uncited after retraction, while in vitro studies and randomized controlled trials were more likely to be cited. Retracted publications that were cited negatively were more likely to have been retracted due to scientific distortion than those that were cited positively or neutrally. Retracted publications that were cited negatively were also more likely to be observational studies than those cited positively or neutrally.Conclusion: Retracted publications in dentistry are continually cited positively following their retraction, regardless of their study designs or reasons for retraction. This indicates that the continued citation of retracted publications in this field cannot be isolated to certain research methods or misconduct but is, instead, a more widespread issue.
