Direct Detection of Theileria annulata in Bovine Blood Samples Using Standard and Isothermal DNA Amplification Approaches

Abstract Fifty-one laboratories from 14 countries participated in a survey on the determination of selenium (Se) in 8 bovine blood samples with Se concentrations ranging from 0.2 μmol/L (0.016 μg/mL) to 14 μmol/L (1.1 μg/mL). The methods used (and the percentage of participants using each method) were fiuorometry (61), hydride-generation atomic absorption spectrophotometry (AAS) (23), graphitefurnace AAS (6), gas chromatography (4), neutron activation analysis (4), and X-ray fiuorometry (2). There was little difference in the mean Se results obtained by fiuorometry or hydride-generation AAS (P > 0.05). Mean intralaboratory coefficients of variation (CVs) from known replicates ranged from 4 to 14% for all samples. Interlaboratory CVs were related to blood Se concentration and increased to 55% at Se levels below 0.4 μmol/L (0.032 μg/mL). Laboratories that used quality control (QC) schemes had lower interlaboratory CVs than those that did not, but the advantage began to diminish at blood Se concentration below 0.4 μmol/L (0.032 μg/mL). The high interlaboratory CVs, coupled with the false assurance from the low intralaboratory CVs and the ineffectiveness of the QC schemes at blood Se concentrations below 0.4 μmol/L (0.032 μg/mL), are of concern in diagnosis of marginal Se deficiency in livestock where the concentrations of interest are in the range 0.15-0.5 μmol/L (0.012-0.039 μg/mL).

Abstract:This study aimed to report the prevalence of Babesia canis vogeli in dogs and ticks in the urban and rural areas of Petrolina, Pernambuco. Serum and peripheral blood samples of 404 dogs were tested by indirect immunofluorescence assay (IFA) and by blood smears, respectively. The presence of tick infestation was evaluated, and some specimens were submitted to DNA amplification by polymerase chain reaction (PCR). The presence of antibodies anti-B. canis vogeli was determinate in 57.9% (234/404) of dogs. The direct detection of Babesia spp was obtained in 0.5% (2/404) dogs by visualization of intraerythrocytic forms. Infestation by Rhipicephalus sanguineus sensu lato was observed in 54.5% (220/404) of dogs in both urban and rural areas. DNA of Babesia canis vogeli were obtained by PCR in 6% individual (3/50) and 8.7% of pool of ticks (7/80). The risk factors for the presence of anti-B. canis vogeli antibodies, as determined through the application of logistic regression models (P<0.05), were the following: medium breed size variables (P<0.001); contact with areas of forest (P=0.021); and access on the street (P=0.046). This study describes, for the first time, the confirmation of infection of B. canis vogeli in dogs and ticks in the semiarid region of Pernambuco, Brazil.

Abstract Coupled particlelight scattering (Copalis™) is a homogeneous immunoassay technology that permits simultaneous determination of multiple analytes in serum, plasma, or whole blood. Copalis differentiates monomeric latex microparticles from latex aggregates and cells on the basis of their unique light scatter properties. Copalis readily discriminates small (∼0.1 μm) differences in latex microparticle size. Therefore, multiple simultaneous assays are configured by the use of mixtures of different-size latex microparticles. The Copalis research immunoassay for hepatitis B surface antigen (HBsAg) is configured in a sandwich format where the extent of light scatter histogram broadening due to HBsAg-mediated binding of colloidal gold to latex provides the basis for antigen quantification. Simultaneous Copalis forward- and wide-angle light scatter measurements allow discrimination of latex microparticles from the cell components of whole blood. Consequently, direct detection of HBsAg in unprocessed whole-blood samples by Copalis is feasible.

Abstract A liquid chromatographic (LC) method is described for the determination of the plant estrogens daidzein, formononetin, and coumestrol and the estrogenically active metabolite equol in bovine blood plasma and urine. The blood and urine samples are incubated overnight with and without β3-glucuronidase/sulfatase for analysis of both free and conjugated forms of estrogens. Samples are applied to Extrelut ® columns, extracted with ethyl acetate, and evaporated to dryness. Residues from urine samples are dissolved in methanol, diluted with water, acidified with HCl, and purified by injection through a Sep-Pak® C,8 cartridge. This eluate is used for LC analysis. Residues from blood samples are dissolved in benzene-petroleum ether (1 + 1), extracted with ammonium hydroxide, acidified with glacial acetic acid, and extracted with ethyl acetate. The ethyl acetate extract is evaporated, dissolved in 80% methanol, injected onto a LC reversephase column, and separated in a linear gradient system between 40 and 80% methanol in phosphate buffer. Quantitation is performed by means of UV and fluorescence responses. The method was sensitive enough to determine 0.4 μg/mL of daidzein and formononetin and 0.1 and 13 ng/mL of coumestrol and equol, respectively, in blood, and 130,80, and 7 ®g/mL of daidzein, formononetin, and coumestrol, respectively, and 4 Mg/mL of equol in urine. The applicability of the method was checked by the determination of total and free plant estrogens in blood samples from a dairy cow fed a normal diet.
