Phytochemical screening, total phenolic and flavonoid methanolic extract of pomegranate bark (Punica granatum L): Evaluation of the inhibitory effect in acidic medium 1 M HCl

Introduction: Punica granatum is a plant used in traditional and alternative medicine for the management of several diseases. Objective: The objective of the present work is to compare the phytochemical characteristics and the antioxidant properties of extracts of leaves powders of Punica granatum L. collected for standardization. Methodology: We worked on seven samples of powders of leaves of Punica granatum L collected in seven different regions of Burkina Faso. Two types of extracts, aqueous and hydroethanolic were prepared with each sample. We performed a phytochemical screening by thin layer chromatography (TLC), then determined the content of the various extracts in total phenolic and flavonoids as well as a study of the antioxidant activity of the aqueous and hydro-ethanolic extracts of the plant. Results: Fourteen extracts of Punica granatum are obtained and these contain secondary metabolites such as tannins, sterols, triterpenes, saponosides and flavonoids. The anti-free radical activities at the DPPH* were more important in the samples from the towns of Dedougou, Banfora and Fada. The anti-free radicals at ABTS of extracts from the towns of Manga, Banfora, Fada and Kaya were found to be the most active. The FRAP test shows better activity of samples from the cities, Manga, Dedougou and Banfora. Conclusion: At the end of this work, the towns of Dedougou and Banfora may be the sites to be favored as harvesting sites because their samples were the richest in phenolic compounds and had the best antioxidant activities compared to the tests carried out.

Objectives: The objectives of this study are to analyze phytoconstituents in Rhododendron arboreum leaves (qualitatively and quantitatively) and to estimate the immunomodulatory effects of extracts using different in vitro methods for intracellular and phagocytic killing potency of neutrophils, which are subsequent, involved in the process of phagocytosis by neutrophils.Methods: R. arboreum leaves extracts were separately extracted in 100% methanol, 50% methanol, and aqueous by a hot percolation technique using Soxhlet apparatus and concentrated with a vacuum rotary evaporator apparatus. The percentage yield of the extracts wares calculated. The extract was used to analyze phytoconstituents qualitatively, and total phenolic and flavonoid content were estimated. In another study, already purified methanolic extract of R. arboreum leaves, namely cytoplasmic male sterility (CMS-3) (kaempferol), RAM fr2, and transcranial magnetic stimulation (TMS-10) (ursolic acid) was used to evaluate immunomodulatory activities by nitroblue tetrazolium test, phagocytosis of killed Candida albicans.Results: Phytochemical screening of the extracts showed the presence of major classes of phytochemicals, i.e., phytosterols, flavonoids, phenols, alkaloids, carbohydrate, glycoside, sterols and steroids, terpenoids, and tannin while saponin was absent in all extract. The percent yield of leaves extract was 12.97%. Total phenolic content (TPC) expressed as gallic acid equivalents (GAE) ranged 1327.5±0.009 (in 100% methanolic extract), 1309.5±0.011 (in 50% methanolic), and 1468.5±0.006 mg GAE g−1 dry weight (in aqueous extract). Total flavonoid content expressed as rutin equivalents (RE) ranged 219±0.025 (in 100% methanolic extract), 184±0.02 (in 50% methanolic), and 262±0.58 mg (in aqueous extract) mg RE g−1 dry weight. R. arboreum purified extract CMS-3 (kaempferol), RAM fr2, and TMS-10 (ursolic acid) stimulated the neutrophils to phagocytic activity to the extent of 67.67±2.08%, 40.3±1.5%, and 79.67±0.57%, respectively. The above-said extracts stimulated the phagocytosis of killed C. albicans. The mean particle numbers for CMS- 3, RAM fr2, and TMS-10 were found to be 6.7, 6.3, and 7, respectively, at concentration 1 mg/ml.Conclusion: The phytochemical screening of all the extracts showed the presence of various phytochemicals that are biologically important. The total phenolic and flavonoid content of the plant are comparable to other medicinal plants. Even though the enormous progress on the phytochemistry and pharmacology of R. arboreum have been made, there still require more conclusive studies on the safety, efficacy, and in vivo toxicity of extracts and pure compounds to gain a better understanding.

Physalis minima a medicinally important plant of the family Solanaceae has been screened for its anticancer activity. The results of preliminary phytochemical screening of the leaf extract revealed the presence of flavonoids, alkaloids, tannins, saponins, steroids, cardiac glycosides, reducing sugars and terpenoids. Determination of total phenolic contents revealed that methanolic extract showed 78.3 mg/g of phenolic compounds. Evaluation of total flavonoid content showed 61.3 mg/g of flavonoid. It was found that the % viability of HeLa cell line & Hep2 cell line are 80% & 71.8% respectively. The percentage of growth inhibition of methanolic extract in SRB assay was found to be increase with increasing concentration against both HeLa and Hep2cell lines (68 and 58% respectively). The methanolic extract showed the strongest growth inhibitory effect on both HeLa and Hep2 cell lines (85 and 73% respectively) in MTT assay. Flavonoids have been isolated and purified from the methanolic leaf extract.

In Peru, Caulerpa filiformis is a marine algae listed as an invasive species. For years, its distribution has been considered to be in the north coast (Isla Lobos de Afuera and Piura) until a recent report of its distribution in the central coast (Ancash, Lima, and Ica). The present investigation aims to determine the main groups of secondary metabolites, total phenol content, and antioxidant activity of the methanolic extract of C. filiformis from Sechura Bay (Piura) and Paracas Bay (Ica). The main chemical groups were determined through phytochemical screening, the content of phenols by the Folin–Ciocalteu method, and antioxidant activity by the ABTS method (2,2-azinobis-[3-ethylbenzothiazoline-6-sulfonic acid]) and 2,2′-diphenyl-1-picrylhydrazyl (DPPH). The phytochemical screening of the methanolic extract of C. filiformis from Sechura Bay and Paracas Bay revealed the presence of carbohydrates, polyphenols, tannins, flavonoids, lipids, alkaloids, steroids, and triterpenes for both extracts. The total phenol content of the extract of C. filiformis from Sechura Bay (39.31 ± 0.39 mg of AGE/g extract) was significantly higher (p < 0.05) than that from Paracas Bay (18.78 ± 0.31 mg of AGE/g extract). In the ABTS and DPPH assays, the antioxidant capacity of the Sechura C. filiformis extract (IC50 = 3.49 ± 0.01 and 2.18 ± 0.02 mg/mL) was significantly higher (p < 0.05) than that of the Paracas C. filiformis extract (IC50 = 6.41 ± 0.02 and 2.42 ± 0.04 mg /mL). These findings suggest that the methanolic extract of C. filiformis is a source of secondary metabolites with an antioxidant potential.

Background: Pomegranate peels are rich in bioactive compounds and could be an alternative natural source such as antioxidants. Aims: The aim of the present study was to assess the abilities of five solvents to extract phenolic antioxidants from pomegranate peels. Methods and Material: Pomegranate peels powder was subjected to extraction and the extraction yield was compared. The total phenolic, flavonoid, condensed and hydrolysable tannins contents were analyzed. The antioxidant activity of the extracts was evaluated by two methods (DPPH* and ABTS•+ scavenging capacities) and results were then compared. Results: Results showed that the mixture methanol/water (50:50) allowed higher extraction yield (37.33±5.3%) than the others solvents (P<0.05). Further, the total phenolic, flavonoid and condensed tannins contents were the highest in mixture water/methanol (50:50) extract. Phenolic antioxidants showed a distinct reducing capacity and a high DPPH* inhibition values were recorded for all extracts with no significant differences (P>0.05) between ethanol and mixture water/methanol (50:50) extracts. All extracts exhibited high inhibition against ABTS•+ but with a considerable variation. Phenolic content and antioxidant activities were well positively correlated with each other. Conclusions: Our findings revealed that the choice of the extracting solvent affects considerably the extraction of phenolic antioxidants from pomegranate peels. Basil (Ocimum spp.) is a traditional herbal medicine abundant in antioxidants such as phenolic compounds. As part of a diet, this herb is proved to have some roles in decreasing the risk of cancer, and in the treatment of inflammation and neurodegenerative diseases. This study aims to explore the total phenolic and flavonoid content of two new basil hybrids growing in Romania, namely “Aromat de Buzau” (AB) and “Macedon” (MB). The antioxidant capacity of those two species was also analyzed by DPPH and cyclic voltammetry. Six different flavonoids, such as catechin (+), rutin, hyperoside, naringin, naringenin, and genistein, were separated, identified, and quantified by HPLC–DAD chromatography, for the first time, from romanian basil hybrids. The main flavonoid of the extracts was found to be naringin which is present in the highest amount (26.18 mg/kg) in “Aromat de Buzau” (O. basilicum) methanolic extract. These results suggest that dietary intake of these new hybrids can be a source of antioxidant compounds.

Belimbing wuluh merupakan tanaman jenis buah dan obat tradisional. Tanaman belimbing wuluh sudah sering dimanfaatkan masyarakat salah satunya untuk mengobati penyakit seperti batuk dan radang rektum. Tujuan penelitian ini adalah menguji senyawa fitokimia, menganalisis kandungan total fenolik dan flavonoid, serta aktivitas antioksidan dan antiinflamasi secara in vitro pada ekstrak etanol daun belimbing wuluh. Metode yang dilakukan pada penelitian ini adalah ekstraksi daun belimbing wuluh, skrining fitokimia, perhitungan total fenol dan flavonoid, uji aktivitas antioksidan, dan uji aktivitas antiinflamasi. Senyawa fitokimia yang terkandung pada ekstrak etanol daun belimbing wuluh adalah saponin, tanin, steroid, flavonoid, dan alkaloid. Kandungan total fenolik dan flavonoid ekstrak etanol daun belimbing wuluh secara berturut-turut sebesar 39,03 dan 97,28 µg QE/mg. Ekstrak etanol daun belimbing wuluh memiliki aktivitas antioksidan yang tergolong sangat kuat, sementara aktivitas antiinflamasinya terutama ditunjukkan pada konsentrasi ekstrak 200 µg/ ml, memiliki nilai persen inhibisi hemolisis yang paling tinggi. Kesimpulannya, ekstrak etanol daun belimbing wuluh dapat menjadi salah satu sumber antioksidan dan antiinflamasi alami.Ethanol Extracts of Averrhoa Bilimbi Leaf Demonstrated Antioxidative and Anti-inflammatory ActivityAbstractAverrhoa bilimbi, a fruit-bearing plant, has been traditionally used for medicinal purposes such as treatment of cough and rectal inflammation. In this current work, ethanol extract of Averrhoa bilimbi leaf was analyzed, with regard to phytochemical composition, i.e. total phenolic, and flavonoid, antioxidant activity, and in vitro antiinflammation activity. Extraction of carambola leaves, phytochemical screening, total phenolic and flavonoid contens, assay of antioxidant activity, and assay of anti-inflammatory activity were done in this research. As a result, the leaf extract positively contained some phytochemical compounds, i.e. saponin, tannin, steroid, flavonoid, and alkaloid. Furthermore, total phenolic and flavonoid of the leaf extract was found at 39.03 and 97.28 µg QE/mg extract, respectively. Additionally, antioxidant activity of the leaf extract was classified as very strong, while its anti-inflammatory feature at extract concentration of 200 µg/ml exhibited the highest inhibition of hemolysis. In summary, the ability of Averrhoa bilimbi leaf ethanol extracts to act as antioxidative and anti-inflammatory agents was determined and this may open the use for natural antioxidant and anti-inflammatory agents.
