The development of PCR technology has greatly accelerated medical research at the genetic and molecular levels. Until recently, the inherent sensitivity of this technique has been limited to isolated preparations of nucleic acids which lack or at best have limited morphological information. With the obvious exception of cell lines, traditional PCR or reverse transcription-PCR (RT-PCR) cannot identify the cellular source of the amplified product. In contrast, in situ hybridization (ISH) by definition, defines the anatomical location of a gene and/or it’s product. However, this technique lacks the sensitivity of PCR and cannot routinely detect less than 10 to 20 copies per cell. Consequently, the localization of rare transcripts, latent viral infections, foreign or altered genes cannot be identified by this technique. In situ PCR or in situ RT-PCR is a combination of the two techniques, exploiting the sensitivity of PCR and the anatomical definition provided by ISH. Since it’s initial description considerable advances have been made in the application of in situ PCR, improvements in protocols, and the development of hardware dedicated to in situ PCR using conventional microscope slides. Our understanding of the importance of viral latency or viral burden in regards to HIV, HPV, and KSHV infections has benefited from this technique, enabling detection of single viral copies in cells or tissue otherwise thought to be normal. Clearly, this technique will be useful tool in pathobiology especially carcinogenesis, gene therapy and manipulations, the study of rare gene transcripts, and forensics.

SummaryNative DNA (dsDNA) induces the aggregation of isolated human platelets. Using isotopically labeled dsDNA (125I-dsDNA) and Scatchard analysis, a single class of platelet receptor was detected with a KD = 190 pM and numbering ~275/platelet. This receptor was discriminatory in that heat denatured dsDNA, poly A, poly C, poly C · I and poly C · poly I failed to substantially inhibit either the platelet binding of, or platelet aggregation induced by, dsDNA; by themselves, these polynucleotides were ineffective as platelet agonists. However, poly G, poly I and poly G · I effectively and competitively inhibited platelet binding of the radioligand, independently activated the platelet and when used at a sub-activating concentration decreased the extent of dsDNA stimulated platelet aggregation. These data depict a receptor on human platelets for dsDNA and perhaps certain additional polynucleotides and relate receptor-ligand interactions to a physiologic platelet function.

Background: Since its initial description in December 2019 in Wuhan, China, coronavirus disease 2019 (COVID-19) has rapidly progressed into a worldwide pandemic, which has affected millions of lives. Unlike the disease in adults, the vast majority of children with COVID-19 have mild symptoms and are largely spared from severe respiratory disease. However, thereare children who have significant respiratory disease, and some may develop a hyperinflammatory response similar to thatseen in adults with COVID-19 and in children with Kawasaki disease (KD), which has been termed multisystem inflammatory syndrome in children (MIS-C).Objective: The purpose of this report was to examine the current evidence that supports the etiopathogenesis of COVID-19 in children and the relationship of COVID-19 with KD and MIS-C as a basis for a better understanding of the clinical course, diagnosis, and management of these clinically perplexing conditions.Results: The pathogenesis of COVID-19 is carried out in two distinct but overlapping phases of COVID-19: the first triggered by the severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) itself and the second by the host immune response. Children with KD have fewer of the previously described COVID-19–associated KD features with less prominent acute respiratory distress syndrome and shock than children with MIS-C.Conclusion: COVID-19 in adults usually includes severe respiratory symptoms and pathology, with a high mortality. Ithas become apparent that children are infected as easily as adults but are more often asymptomatic and have milder diseasebecause of their immature immune systems. Although children are largely spared from severe respiratory disease, they canpresent with a SARS-CoV-2–associated MIS-C similar to KD.

The point of departure for this paper is the publication of two Early Helladic sealing fragments from the coastal settlement of Asine on the north-east Peloponnese in Greece. After an initial description and discussion they are set in the context of sealing custom established on the Greek mainland around 2500 BCE. In the first part of the paper focus is on the apparent qualitative differences between the available seals and the contemporary seal impressions, as well as between different sealing assemblages on northeastern Peloponnese. This geographical emphasis is carried into the second part of the paper which is a review and contextualisation of the representational art of the Aegean Early Bronze Age in general, and northeastern Peloponnese in particular. Seal motifs and figurines are the main media for Early Helladic representational art preserved until today, yet in many ways very dissimilar. These opposites are explored in order to begin to build a better understanding of Peloponnesian representational art, the choices of motifs, and their roles in the lives of the Early Helladic people.

Abstract Context Previous studies have characterized code comments in various programming languages, showing how high quality of code comments is crucial to support program comprehension activities, and to improve the effectiveness of maintenance tasks. However, very few studies have focused on understanding developer practices to write comments. None of them has compared such developer practices to the standard comment guidelines to study the extent to which developers follow the guidelines. Objective Therefore, our goal is to investigate developer commenting practices and compare them to the comment guidelines. Method This paper reports the first empirical study investigating commenting practices in Pharo Smalltalk. First, we analyze class comment evolution over seven Pharo versions. Then, we quantitatively and qualitatively investigate the information types embedded in class comments. Finally, we study the adherence of developer commenting practices to the official class comment template over Pharo versions. Results Our results show that there is a rapid increase in class comments in the initial three Pharo versions, while in subsequent versions developers added comments to both new and old classes, thus maintaining a similar code to comment ratio. We furthermore found three times as many information types in class comments as those suggested by the template. However, the information types suggested by the template tend to be present more often than other types of information. Additionally, we find that a substantial proportion of comments follow the writing style of the template in writing these information types, but they are written and formatted in a non-uniform way. Conclusion The results suggest the need to standardize the commenting guidelines for formatting the text, and to provide headers for the different information types to ensure a consistent style and to identify the information easily. Given the importance of high-quality code comments, we draw numerous implications for developers and researchers to improve the support for comment quality assessment tools.

The basic stages during automatic qualification of agricultural products are: acquisition of initial information of the quality state, mathematical proceeding of this information and decision making for object qualification to defined sets according to the state of quality. One of the steps in the second stage is transformation the patterns of qualified products to a new space of symptoms. In this paper an approach for transformation the initial description of the objects to a new space of spectral symptoms, independently of the size of the initial description of the object is presented. A relation for synthesis of symptoms for recognition has been generated.

A new numerical method is presented in this paper to simulate rotor/stator interaction in turbomachinery by use of a vortex method based on a Lagrangian frame. The algorithm takes the result from steady solution as input, which can give an initial description of the unsteady disturbance flow field. To calculate the unsteady response to these disturbances, the Lagrangian vortex method is used to capture the convective process, and the deterministic vortex scheme to approximate the viscous diffusion process. The application of Baldwin-Lomax turbulence model in wakes is developed, so as to overcome the difficulties such as the much higher calculated viscosity in the outer region than that in the boundary regions, and the difficulty in continuously tracing moving wake centerlines encounted by other numerical methods. The agreement between the computational and experimental results is generally good. The sweeping characteristic of wakes, the influence of unsteadiness on incidence and the decaying features of unsteady velocities, pressure are included in the paper.

Cat-scratch disease has been reported with increasing frequency since its initial description in 1931. It is now recognized as being so prevalent that it probably represents the most common cause of unilateral lymphadenopathy in children, once nonspecific viral diseases and cutaneous infections have been excluded. The cardinal feature of the disease is a subacute regional granulomatous lymphadenitis. Because of its relatively mild natural course, most cases are treated on an outpatient basis. Forty-four children, representing the more seriously ill portion of the disease spectrum, were treated at Texas Children's Hospital from 1972 to 1984 and constitute the data base for this article. Most patients were symptomatic for nearly 1 month prior to admission, typically complaining of low-grade fever and regional lymphadenopathy. The two most common sites of lymph node involvement were the axilla (54%) and the neck (46%). Although there is, at this time, no specifically recommended treatment, 80% of patients received antibiotics and 70% underwent a surgical procedure—usually total nodal excision. Nodal excision was followed by rapid resolution of symptoms. Because cat-scratch disease is benign, surgery should be reserved for severe cases with signs of significant suppuration. Generally, symptomatic relief can be afforded by warm soaks and analgesics and recovery over the course of several weeks is uneventful.

Experience on the job training will help students improve the competence of graduates to align with what is needed. In addition to developing competencies, learning through experience on the job training will stimulate the development of character and also a work ethic of discipline, responsibility, honesty, cooperation, leadership, and others according to what is needed by the industry. The creation of competence, character and work ethic is expected that graduates will be ready to welcome the competition of the ASEAN Economic Community (AEC). This research was conducted based on a literature study of various theoretical studies regarding experiences on the job training. Where these studies are an initial description in conducting research, especially those related to the influence of experiences on the job training. Based on the results of data analysis with several similar research literature, the effect of on the job training experience on graduate work readiness is very significant and it can be concluded that there is a significant effect on the job training experience on graduate work readiness.
