Studies on the frozen storage of human blood products have benefited veterinary transfusion medicine in recent years, but the long-term cryopreservation of canine red blood cells (RBCs) has not yet been thoroughly investigated. Further, no studies are available with respect to the frozen storage of leukocyte-depleted canine red blood cells (LD-RBCs). The objective of the current study was to investigate time-dependent effects of long-term frozen storage on leukocyte-depleted canine RBCs. Twelve healthy adult dogs meeting the criteria for blood transfusion were used in the study. Whole blood samples (450 ± 45 ml) collected from each dog were centrifuged for 5 min at 22 °C and 4200 × g in a cryogenic microcentrifuge and concentrated RBC (pRBC) suspensions were obtained. Leukocyte depletion was achieved by filtration (2.6 log<sub>10</sub>). Then, the filtrated samples were prewashed three times in 0.9% NaCl solution and were allocated into three subgroups to be evaluated at three different time points (Day 0, Month 4 and Month 6). The samples for cryopreservation were subjected to glycerolisation and then stored at –80 °C for 4- and 6-month periods. At the end of this period pRBC units were thawed by manual agitation in a water bath maintained at 36–38 °C, centrifuged and then washed in a consecutive series of 12%, 1.6% and 0.9% of NaCl + 0.2 dextrose solutions. 2,3-Diphosphoglycerate (2,3-DPG), adenosine triphosphate (ATP), supernatant haemoglobin (SupHb), sodium (Na<sup>+</sup>) and potassium (K<sup>+</sup>) levels, residual glycerol concentrations and haemograms of thawed and deglycerolised pRBC samples were evaluated together with those of Day 0. Sterility tests were performed on all samples for bacterial contamination. No statistically significant differences were noted except for Hct and SupHb levels. No bacterial contamination was noted in any of the samples on the basis of sterility tests. It was found that the described glycerolisation procedure could be a method of choice in the cryopreservation of leukocyte-depleted pRBCs (LD-pRBCs) since no negative effect was observed on the quality of the products and long-term frozen storage did not cause RBC destruction.

The biobank is a structure established with the goal of long-term responsible storage of biological samples and the associated data for their further use in scientific and clinical research. The objectives of biobanking are the creation of unified recommendations on: the planning of premises and the selection of equipment for storage; development of management methods and staff training; standardization of methods for the collection, shipping, processing and storage of biomaterial of various origins, as well as methods for quality control and validation of the applied methods; creation and use of databases of information accompanying biospecimens. The lack of common standards for conducting the preanalytical phase has been the cause of low accuracy and poor reproducibility of research results. To date, a large number of guidelines and best practices have been published that provide an answer to a wide range of problems in organizing the biobanking process. The article provides an overview of the most famous biobanking guidelines that can be used to solve various research problems. Biobanking in Russia is actively developing. Since 1996 there is a work on the legislative regulation of biobanking activities, as a result of which a number of regulatory documents have been issued. An important stage in the development of biobanking in Russia was the establishment of the “National Association of Biobanks and Biobanking Specialists” (NASBio) in 2018, which included representatives of medical and research institutions, commercial firms, and qualified specialists in the field of biobanking. One of the key tasks of NASBio is the adaptation and implementation of the best biobanking practices in Russian research institutes and centers. The use of modern guidelines and best practices on biobanking will lead to an increase in the quality of research and publications.

Abstract. Large round bales are commonly fed to livestock; however, it has been well documented that outdoor storage can negatively impact dry matter (DM) and forage quality. To reduce storage losses and improve feeding value, new wrap types and alfalfa cultivars have been developed; however, these options have not been extensively investigated. Therefore, the objectives of this research were to evaluate the time required to bind large round bales, determine changes in DM and forage quality, and examine the economics of reduced-lignin and conventional alfalfa hay bound in twine, net wrap, and B-Wrap® while in outdoor storage. Hay was baled into 24 large round bales; 12 bales each of reduced-lignin and conventional alfalfa. Within each cultivar, four replicates were bound with each wrap type. Using stopwatches, the time to bind each bale was recorded. At the time of harvest, and every 90±3 days for 365 days, individual bales were weighed and cored to determine changes in DM and forage quality. Significance was set at P = 0.05. Time to bind a bale was longest for twine, intermediate for B-Wrap®, and shortest for net wrap (P &lt; 0.01). Alfalfa cultivar had minimal effects on the parameters measured; therefore, the interaction between wrap type and storage length was reported. After 365 days in outdoor storage, DM losses were 7% for twine bales, 5% for net wrap bales, while B-Wrap® bales maintained DM. Changes in forage quality were observed at =180 days of storage where nonstructural carbohydrates were decreased and insoluble fiber components were concentrated. B-Wrap® bales had a higher value compared to net wrap and twines bales at 180 and 270 days in storage. These results suggest that B-Wrap® was better able to shed precipitation which preserved forage quality and bale value compared to large round bales bound in net wrap and twine in long term, outdoor storage. When improving software packages such as MOUSE and SAMBA for designing sewers and storage basins, and simulating overflows and flooding the quality of the input becomes important. The essential input to these modelling tools are the historical rain series. This paper presents the procedures for collection and quality control of rain data from a network of tipping bucket rain gauges in Denmark carried out by the Danish Meteorological Institute. During rain, the RIMCO gauge counts the number of tips (each of 0.2 mm of precipitation) every minute. The quality control of the rain data includes an automatic and a manual bit marking, where the automatic control basically is pointing out minutes with extreme intensities. In the manual control, the maximum intensities as well as the daily totals of precipitation are inspected, using weather charts, intensity plots and precipitation sums of nearby Hellmann gauges. Shortcomings and improvements of the quality control are discussed. Although, it is possible to improve the efficiency of the quality control, long term corrections will always be necessary.

Button mushrooms have a very short shelf life after harvesting and are sensitive to mechanical damage and browning. This can be a severe problem in enlarging the market and the long-distance exportation of this product. In this respect, edible coatings could be an alternative treatment to extend the shelf life of button mushrooms, maintaining their quality during long-term storage. The aim of this study was to investigate the impact of gum, agar, sodium alginate, egg white protein, and lecithin on the postharvest weight loss, color, browning, respiration rate, ethylene production, and storage life of button mushrooms. The results showed that the above-mentioned edible coatings are a promising way to extend the life and maintain the quality of button mushrooms. Significant differences (p < 0.05) were observed between the control and edible coating-treated samples in all parameters. Sodium alginate and gum were more effective in preventing weight loss, coloring, and browning than other edible coatings. On the other hand, the respiration rate and ethylene production were more suppressed by the agar and lecithin coatings compared to the others. In conclusion, it can be recommended that the above-mentioned edible coatings could be used as novel coatings in commercial treatments for maintaining the quality of button mushrooms during a long-term storage period.

Phytolith-occluded carbon (PhytOC), a promising long-term biogeochemical carbon sequestration mode, plays a crucial role in the global carbon cycle and the regulation of atmospheric CO2. Previous studies mostly focused on the estimation of the content and storage of PhytOC, while it remains unclear about how the management practices affect the PhytOC content and whether it varies with stand age. Moso bamboo (Phyllostachys heterocycla var. pubescens) has a great potential in carbon sequestration and is rich in PhytOC. Here, we selected four management treatments, including control (CK), compound fertilization (CF), silicon (Si) fertilization (SiF) (monosilicic acid can form phytoliths through silicification), and cut to investigate the variation of phytoliths and PhytOC contents in soil, leaves, and litters, and their storage in Moso bamboo forests. In soil, the SiF fertilizer treatment significantly (P &lt; 0.05) increased phytolith content, PhytOC content, and storage compared to CK, while there were no significant differences between the treatments of CF and cut. In leaf, compared with CK, phytolith content of the second-degree leaves under SiF and the first-degree leaves under cut treatment significantly increased, and the three treatments significantly increased PhytOC storage for leaves with three age classes. In litter, the phytolith and PhytOC contents under the three treatments were not significantly different from that under the CK treatment. The PhytOC storage increased by 19.33% under SiF treatment, but significantly decreased by 40.63% under the CF treatment. For the entire Moso bamboo forest ecosystems, PhytOC storage of all the three management treatments increased compared with CK, with the largest increase by 102% under the SiF treatment. The effects of management practices on the accumulation of PhytOC varied with age. Our study implied that Si fertilization has a greater potential to significantly promote the capacity of sequestration of carbon in Moso bamboo forests.

ABSTRACT Preservation of the quality of coffee seeds is hindered by their intermediate behavior in storage. However, long-term storage at sub zero temperatures may be achieved by adjusting the water content of the seeds. The aim of this study was to evaluate the tolerance of coffee seeds to freezing, in relation to physiological and enzymatic modifications. Coffee seeds were dried in two manners, rapid and slow, to water contents of interest, 0.67, 0.43, 0.25, 0.18, 0.11, and 0.05 g H2O g-¹ dw (dry basis). After drying, the seeds were stored at a temperature of -20 ºC and of 86 ºC for 24 hours and for 12 months, and then compared to seeds in cold storage at 10 ºC. The seeds were evaluated through calculation of percentage of normal seedlings, percentage of seedlings with expanded cotyledonary leaves, dry matter of roots and of hypocotyls, and viability of embryos in the tetrazolium test. Expression of the enzymes superoxide dismutase, catalase, and peroxidase were evaluated by means of electrophoretic analysis. Only seeds dried more slowly to 0.18 g H2O g-1 dw present relative tolerance to storing at -20 °C for 12 months. Coffee seeds do not tolerate storage at a temperature of -86 ºC for 12 months. Water contents below 0.11g H2O g-¹ dw and above 0.43 g H2O g-¹ dw hurt the physiological quality of coffee seeds, regardless of the type of drying, temperature, and storage period. Coffee seed embryos are more tolerant to desiccation and to freezing compared to whole seeds, especially when the seeds are dried to 0.05 g H2O g-¹ dw. The catalase enzyme can be used as a biochemical marker to study tolerance to freezing in coffee seeds.

The objective of this study was to assess the sperm membrane integrity in extended boar semen during storage time using specific spectrum laboratory methods. Boar semen was diluted with the long-term liquid commercial extenders Androhep (A), Androstar (AS), Androstar plus (AS<SUP>+</SUP>), LD and M III and was stored up to 96 h. The sperm membrane integrity was evaluated by motility, viable spermatozoa, short hypoosmotic swelling test (sHOST) and by the activity of the enzyme aspartate aminotransferase (AST). Negative changes in the quality of sperm membrane in relation to storage time were observed after 48 h for sHOST, after 72 h for viable spermatozoa and after 72 h for motility. The percentage of viable spermatozoa was decreased by 0.27% each hour. A statistically significant difference between extenders A and LD was observed in sHOST after 72 h and 96 h storage (<I>P</I> &lt; 0.05). The AST activity did not show any statistically significant differences in extenders and in storage time. In overall assessment Androhep was the best of the tested extenders, followed by AS, AS<SUP>+</SUP>, M III and LD in terms of motility, viable spermatozoa and sHOST. The correlations among laboratory methods were highly significant (<I>P</I> &lt; 0.001). In conclusion, the results documented that the sperm membrane integrity was statistically significantly affected by extenders and storage time (<I>P</I> &lt; 0.001). Boar semen quality was the best in extender A. sHOST is a very sensitive and relatively simple method for the assessment of sperm membrane integrity in diluted semen.

A Fog Computing architecture consists of edge nodes that generate and possibly pre-process (sensor) data, fog nodes that do some processing quickly and do any actuations that may be needed, and cloud nodes that may perform further detailed analysis for long-term and archival purposes. Processing of a batch of input data is distributed into sub-computations which are executed at the different nodes of the architecture. In many applications, the computations are expected to preserve the order in which the batches arrive at the sources. In this paper, we discuss mechanisms for performing the computations at a node in correct order, by storing some batches temporarily and/or dropping some batches. The former option causes a delay in processing and the latter option affects Quality of Service (QoS). We bring out the trade-offs between processing delay and storage capabilities of the nodes, and also between QoS and the storage capabilities.

Revolution 4.0 is the first milestone in the birth of globalization in the world economy. The current movement of business progress is accompanied by developments in the field of information and communication technology. Social media is a social interaction between individuals in sharing and exchanging information. Social media can encompass various ideas, opinions, ideas and content in virtual communities and be able to present and translate new ways of communicating with technology that is completely different from traditional media. Advertising with social media even offers long-term value. This research was conducted to determine the effect of social media advertising (attention, understanding, acceptance, and storage) on purchasing decisions at the University of Bali International. The sample in this study were 96 students at the University of Bali International where the results of data collection were processed using multiple regression analysis techniques. The results of the study prove that attention, understanding, acceptance, and storage of social media advertisements have a positive and significant effect on consumer purchasing decisions at the University of Bali International. This shows that purchasing decisions can increase not only influenced by conventional advertising but the existence of Instagram social media ads can increase consumer purchasing decisions perceptions of students can also improve the quality of higher education itself.
