AbstractThe apparent and true amino acid (AA) digestibility from soya-bean meal (SBM), vetch meal (VM), field pea meal (FPM) and bitter vetch meal (BVM) were determined in the lower ileum of growing chickens force-fed on semisynthetic diets (120 g crude protein and 13·1 MJ metabolizable energy per kg dry matter) based on each meal as the sole source of protein. The average apparent and true digestibility values were 0·82, 0·73, 0·76 and 0·60 and 0·90, 0·91, 0·87 and 0·74 for diets SBM, VM, FPM and BVM, respectively. Marked differences in AA digestibility among diets were found. The apparent digestibility of methionine, the most limiting essential AA in poultry diets, was significantly higher in diet SBM than in diets FPM and BVM (P < 0·05). It is concluded that the seeds of vetch and field pea may be suitable for inclusion in poultry diets as partial substitutes for soya bean. The use of bitter vetch is not recommended.

AbstractAn experiment was carried out in growing chickens to study the effect of supplementation of a semi-synthetic diet containing soya-bean meal as the sole protein source with DL-methionine, to improve its biological value, on the excretion of endogenous protein and amino acids measured in lower ileum and total tract using traditional methods. Thirty-two White Rock male broilers (10 days old) were randomly divided into eight groups each of four birds, of similar body weight (mean live weight: 142·8 (s.e. 0·68) g), and individually housed in metabolism cages. Following a paired-feeding design based on metabolic body weight (kgM0·75), each group of birds was given, for an experimental period of 20 days, each of four levels of protein (60, 120, 180 or 240 g/kg; 5 days each) in two groups of isoenergetic (14·5 kJ metabolizable energy per g dry matter) and semi-synthetic diets based on soya-bean meal, either not supplemented or supplemented with 2 g/kg DL-methionine (diets S and SM, respectively). After 3 days of each treatment excreta were collected for 48 h, frozen and stored at –20ºC. At the end of the fourth treatment three chickens of each group were killed and their lower ileal contents collected. The remaining chick of each treatment was fasted for 24 h and given a protein-free diet for 8 days and excreta were collected for the last 4 days. Then (day 39 of age), chickens were killed and lower ileum contents removed and stored at –20ºC. Samples of excreta and lower ileum contents were subjected to nitrogen (N) analysis by Kjeldahl procedure and amino acid (AA) analysis by high-performance liquid chromatography. Supplementation with DL-methionine of the soya-bean meal-based diets halved total tract endogenous AA losses. Regression analysis produced a higher estimation of ileal and faecal endogenous AA excretion than feeding a protein-free diet. Endogenous AA excretion determined in the lower ileum was higher than in excreta no matter which estimation procedure was utilized. In conclusion, supplementation of dietary protein with the first limiting AA to improve its protein quality, causes an important drop in endogenous AA losses, that may have an important effect on the N economy and energy requirements in poultry. The use of regression analysis on excreta data where graded amounts of protein are given to growing chickens, seems a suitable method for determining endogenous AA losses provided that good quality proteins are used.

AbstractForty-eight 4-week-old White Rock growing chickens (mean live weight 500 (s.e. 9·3) g) were given, by crop intubation, four isoencrgetic (13·1 M] metabolizable energy (ME) per kg dry matter (DM)) and isonitrogenous (120 g crude protein (CP) per kg DM) semisynthetic diets based on vetch (V) or bitter vetch (B) seeds, untreated or antoclaved at 120°C for 30 ruin (diets V, AV, B and AB, respectively). Autoclaving of the seeds caused some changes in amino acid (AA) composition but did not reduce the availability of A As, especially of lysine.The average apparent AA digestibility values derived from ileal content were 0·75, 0·85, 0·59 and 0·85 for diets V, AV, B and AB, respectively, corresponding to true absorption values of 0·90, 1·00, 0·74 and 0·97, respectively. Although the apparent digestibility values of several AAs were relatively low (methionine, isoleucine, tyrosine and cystine), most were higher than 0·70. Heat treatment resulted in a significant increase (P < 0·05) in the AA apparent digestibility of both seeds. It is concluded that heat treatment is useful for improvement of vetch and bitter vetch seeds for inclusion in poultry diets.

AbstractAn experiment was carried out to study the effect of changes in either the quality or the quantity of dietary protein intake on the free essential amino acid profiles in plasma, muscle or liver of growing chickens. Following a randomized paired-feeding design based on metabolic body weight (kg M0·75), White Rock male broilers were allocated to one of three isoenergetic (14·5 kJ metabolizable energy per g dry matter (DM)) semisynthetic diets containing different levels of protein (60, 120, 180 or 240 g/kg DM). All diets were based on soya-bean meal, as the sole source of protein, either unsupplemented (diets S) or supplemented with 20 g/kg L-lysine (diets SL) or 2 g/kg DL-methionine (diets SM). Samples of blood, biceps muscle and liver were taken and amino acid analysis was determined by high performance liquid chromatography. Plasma was only adequate to detect the effect of the supplementation with methionine to balance the dietary amino acid (AA) profile. Plasma concentrations of free methionine significantly increased as a result of the supplementation with methionine (P < 0·001) and remained unaffected by the amount of protein ingested. When lysine was added to diets S to induce an AA imbalance, a significant increase in muscle concentrations of free valine, isoleucine and phenylalanine (P <0·05) and a significant decrease in liver concentrations of free arginine and phenylalanine were observed (P<0·05). The supplementation with DL-methionine gave rise to a significant fall in the concentrations of free histidine, glycine and threonine (P < 0·05) and increased those of isoleucine and methionine in the skeletal muscle (P < 0·05), while in liver it significantly lowered the concentrations of free arginine, valine, leucine, phenylalanine and methionine (P < 0·001 to P < 0·05) and raised that of free glycine (P < 0·001). The ratios lysine: phenylalanine, lysine: valine and methionine: glycine in muscle together with that of methionine: glycine in liver seem to be appropriate indexes of the adequacy of the dietary AA profile to requirements.

1. In a 4 x 4 Latin square experiment four cows were given, twice daily, diets consisting of (g/kg dry matter (DM)) 500 barley, 400 grass silage and 100 soya-bean meal. The diets were given at either 1.15 (L) or 2.3 (H) times maintenance energy requirements and the soya-bean meal was either untreated (U) or formaldehyde (HCH0)-treated (T).2. The passage of digesta to the duodenum was estimated using chromic oxide as a flow marker;35S was used to estimate the amount of microbial protein entering the small intestine. A microbial fraction was prepared by differential centrifugation from duodenal digesta. Samples of bacteria and of protozoa from rumen digesta were also prepared.3. The total amino acid contents of feedingstuffs, duodenal digesta, duodenal microbial material, rumen bacteria and rumen protozoa were determined by ion-exchange chromatography. The D-alanine and D-glutamic acid contents of the samples were determined by gas–liquid chromatography.4. The quantity of each amino acid entering the small intestine was significantly (P < 0,001) increased by increasing DM intake and tended to be increased by formaldehyde-treatment of the soya-bean meal. There were net losses of all amino acids across the forestomachs except for lysine, methione, o-alanine and D-glutamic acid for which there were net gains.5. There were significant (P < 0.05) differences in amino acid composition between rumen bacteria and duodenal microbial material; differences in amino acid composition between rumen bacteria and rumen protozoa were also observed.6. D-Alanine and D-glutamic acid were present in the silage but not in the barley or either of the soya-bean meals. All samples of microbes and digesta contained D-alanine and D-glutamic acid.7. The use of D-ahine and D-glUtamiC acid as markers for microbial nitrogen entering the small intestine was assessed. Estimates of the quantities of microbial N entering the small intestine based on the D-alanine or D-glutamic acid contents of rumen bacteria or duodenal microbes were significantly higher than those determined using 35S as a marker.

1. Three growth trials were done using male broiler chicks. In the first two trials, groundnut meal was used, with and without supplementary methionine and lysine. In the third trial, soya-bean meal was used with and without supplementary methionine. Protein levels ranged in the first trial from 120 to 420 g/kg diet and in the third trial from 120 to 300 g/kg diet. Thus the assumed minimal amino acid requirements of the chick were supplied by high levels of low-quality dietary protein.2. Diets based on cereals and groundnut meal did not support maximum live-weight gain or maximum efficiency of food utilization at any level of dietary protein. When the principal deficiencies of lysine and methionine were corrected, this protein mixture was capable of supporting the same growth rate as a control diet of cereals and herring meal.3. Diets based on maize and soya-bean meal did not support quite the same growth rate as similar diets supplemented with methionine, even though the protein level in the unsupplemented diets was sufficient to meet the assumed methionine requirements.4. These results are interpreted as examples of amino acid imbalance in diets composed of familiar feeding-stuffs. It is concluded that one cannot assume that the poor quality of a protein source can always be offset by increasing the concentration of dietary protein.

1. Young male rats were used in five experiments to study the utilization for growth of methionine sulphoxide, and the relationship between the sulphoxide content in the diet and the level of microbiologically determined methionine activity in blood or blood plasma. In one nitrogen-balance experiment methionine and methionine sulphoxide were compared as supplements to a casein diet and a fish-meal diet.2. Methionine sulphoxide was poorly utilized for growth when tested as the sole sulphur amino acid in an amino acid diet. Substitution of one-third of the sulphoxide with cystine improved utilization so that it approached that of methionine.3. Methionine alone and in combination with methionine sulphoxide were added to a soya-bean-meal diet. The sulphoxide showed no adverse effect on growth.4. Fish meal in which methionine had been oxidized to methionine sulphoxide was tested alone and in combinations with unoxidized fish meal. Only when the oxidized meal was given alone was there an appreciable effect on growth. The fish meals used were low in cystine.5. Whereas both methionine and methionine sulphoxide improved the N balance when a casein diet was given, there was no effect when a fish-meal diet was given.6. There was a linear relationship between methionine sulphoxide content in the amino acid diets and the methionine activity in the blood plasma. Methionine sulphoxide added to a soya-bean-meal diet or present in oxidized fish meal gave a curvilinear relationship, and the observed activities were lower than with the amino acid diets. Methionine activity in blood could not be used as an indicator of moderate amounts of methionine sulphoxide in protein-containing diets.

Abstract Two experiments were conducted to test the hypothesis that both the degree of heating and the time that heat is applied will affect the concentration of DE and ME, and the apparent ileal digestibility (AID) and the standardized ileal digestibility (SID) of amino acids (AA) in 00-rapeseed meal (00-RSM) fed to growing pigs. The nine treatments were prepared using a conventional 00-RSM that was either not autoclaved or autoclaved at 110 °C for 15 or 30 min or at 150 °C for 3, 6, 9, 12, 15, or 18 min. In experiment 1, 20 growing barrows with an average initial BW of 21.2 ± 1.2 kg were randomly allotted to the 10 diets in a replicated 10 × 4 Youden square with 10 diets and four periods in each square. A corn-based basal diet and nine diets containing corn and each source of 00-RSM were formulated. Urine and fecal samples were collected for 5 d after 7 d of adaptation. In experiment 2, nine diets contained one of the nine sources of 00-RSM as the sole source of AA, and an N-free diet that was used to measure basal endogenous losses of AA and CP was formulated. Twenty growing barrows with an initial BW of 69.8 ± 5.7 kg had a T-cannula installed in the distal ileum and were allotted to a 10 × 7 Youden square design with 10 diets and 7 periods. Ileal digesta were collected on days 6 and 7 of each 7-d period. Results from the experiments indicated that there were no effects of autoclaving at 110 °C on DE and ME or on AID and SID of AA in 00-RSM, but DE and ME, and AID and SID of AA were less (P &lt; 0.01) if 00-RSM was autoclaved at 150 °C compared with 110 °C. At 150 °C, there were decreases (quadratic, P &lt; 0.05) in DE and ME, and in AID and SID of AA as heating time increased. In conclusion, autoclaving at 110 °C did not affect ME or SID of AA in 00-RSM, but autoclaving at 150 °C had negative effects on ME and SID of AA and the negative effects increased as heating time increased.

AbstractTwo experiments were carried out with weaned piglets from 8 to 25 kg live weight in order to determine the dietary amino acid content necessary for maximum growth. Six diets based on wheat, soya-bean meal, soya-bean oil and free amino acids were compared in each experiment. Essential amino acids were in the same relative proportion for all diets: (methionine + cystine)/lysine = 0·60 to 0·65; threonine/lysine = 0·65; tryptophan/lysine = 0·19. The range of amino acid content was 9·53 to 12·52 g lysine per kg in the first experiment and 11·34 to 15·94 g lysine per kg in the second experiment. The number of piglets used per diet was 136 (20 pens) and 106 (16 pens) in respectively the first and the second experiment. The relationship between either dietary lysine content or daily lysine intake and growth rate was quadratic and significant. Dietary lysine content and daily lysine intake which enable maximum growth were calculated according to this model. Dietary lysine contents were 15·5 and 14·9 g/kg for the first 3 weeks (8 to 17 kg) and for the overall post-weaning period (8 to 25 kg) respectively. Daily lysine intakes were 10·6 and 13·3 g/day respectively for the same two periods. Reasons for these values being higher than those currently cited are discussed.

SUMMARYIn trials 1 and 2, supplements of urea and cottonseed meal increased the protein content of the basal diets from 12·2 to 14·0–14·6%, but had no significant effects on growth rate or carcass characteristics. In trial 3 the effect of cottonseed meal on growth rate was almost significant but urea had less effect. In trial 4, the basal diet contained 11·0% protein; soya bean meal increased this to 13·1% and had an almost significant effect on growth rate. Lysine and lysine + urea had no effect.

ABSTRACTTwo experiments have been carried out to examine the effects of the level of protein supplementation given with grass silage-based diets on the performance and carcass composition of bulls, and to compare diets based on silage and dried forage. The five treatments used consisted of grass silage offered ad libitum and supplemented with 2·5 kg dry matter (DM) of barley-based concentrates containing (1) zero (2) 200 (3) 400 and (4) 600 g soya-bean meal per kg and (5) artificially dried grass and hay supplemented with 3·2 kg concentrate DM. The silages used in both experiments were well preserved, containing on average 200 g DM per kg; 140 g crude protein (CP) per kg DM; 63 g ammonia-nitrogen per kg total N and 731 g digestible organic matter per kg DM. The bulls were of late-maturing breed type and were initially 12 months old and 412 and 405 kg live weight in experiments 1 and 2 respectively. For treatments 1, 2, 4 and 5 in experiment 1 respectively (treatment 3 was not used) total DM intakes were 8·3, 8·3, 81 and 110 (s.e. 0·21) kg/day; CP intakes 1063, 1271, 1664 and 1539g/day; metabolizable energy intakes (MEI) 98, 99, 96 and 87 MJ/day; carcass weights 317, 316, 317 and 316 (s.e. 3·2) kg; carcass saleable meat concentrations 714, 712, 718 and 716 (s.e. 5·8) g/kg and carcass fat trims 73, 81, 73 and 68 (s.e. 3·9) g/kg. In experiment 2 for treatments 1 to 5 respectively total DM intakes were 8·3, 8·5, 8·3, 8·4 and 11·2 (s.e. 0·26) kg/day; CP intakes were 1090, 1329, 1504, 1720 and 1561 g/day; MEI 102, 106, 103, 103 and 94 MJ/day; carcass weights 318, 331, 330, 327 and 321 (s.e. 3·3) kg; carcass saleable meat concentrations 726, 721, 725, 721 and 732 (s.e. 60) g/kg and fat trims 71, 77, 78, 80 and 64 (s.e. 4·5) g/kg. It is concluded that protein supplementation of a silage-based diet did not affect performance or carcass fatness in experiment 1 or carcass fatness in experiment 2, but including 200 or 400 g soya-bean meal per kg concentrate increased performance in experiment 2. Animals given silage produced fatter carcasses than those given dried forage in experiment 2 but not in experiment 1.
