AbstractOcimum gratissimum is a shrub that belongs to the Lamiaceae family of plants. Despite the known biological activities and ethnomedicinal applications, comparative evaluation of the effects of different extraction techniques on the chemical and bioactive properties of O. gratissimum extracts has not yet been performed. This study adopted different analytical techniques to determine the effect of extraction temperature and solvent type on the phytochemical and bioactive properties of O. gratissimum extracts. Chemical profiling showed increased concentrations of compounds for both the ethanolic and methanolic extracts compared to the water extracts. The results also revealed that the extraction temperature had an effect on the total phenolic content and radical-scavenging properties of the different extracts. The antioxidant kinetic modeling achieved the best fit when using the second-order kinetic model. Methanolic extracts had the highest levels of antibacterial activity against Escherichia coli, Bacillus cereus, Staphylococcus aureus, and Salmonella typhimurium. At high concentrations, all extracts lowered the viability of the breast cancer cell line MDA-MB-231. In conclusion, the chemical and bioactive properties of all extracts showed significant dependence on the extraction temperature and solvent type. With proper extraction methods, they boast a wide range of promising applications in the medical, pharmaceutical, and food industries.

Seseli L. is one of the largest genera of the Apiaceae family widely known for their traditional uses as herbal remedies. In the present study, the methanolic and water extracts of two Seseli species, S. gummiferum and S. transcaucasicum were evaluated for their bioactive contents and biological activities. The total phenolic and flavonoid contents in the extracts ranged from 19.09 to 24.33 mg GAE/g and from 0.45 to10.09 mg RE/g, respectively. Moreover, while narcissin was detected as the most abundant component in the methanolic extract of S. transcaucasicum (261.40 µg/g), chlorogenic acid was identified as the major component in all the other extracts, although a high amount was also present in the methanolic S. transcaucasicum extract (107.48–243.12 µg/g). The total antioxidant capacity was also determined by the phosphomolybdenum assay (0.66–1.18 mM TE/g). Other antioxidant assays such as the radical scavenging assays (DPPH: 5.51–11.45 mg TE/g; ABTS: 43.46–51.91 mg TE/g), reducing assays (CUPRAC: 41.67–53.20 mg TE/g; FRAP: 31.26–34.14 mg TE/g), as well as the metal chelating activity assay (14.38–38.57 mg EDTAE/g) were conducted. All the extracts showed inhibitory potential against the enzyme’s amylase (0.12–0.78 mM ACAE/g), acetyl- and butyryl-cholinesterase (0.15–9.71 mg GALAE/g), while only the methanolic extracts acted as inhibitors of tyrosinase (107.15 and 109.37 mg KAE/g) and only the water extract of S. gummiferum displayed anti-glucosidase activity (0.13 mM ACAE/g). Interestingly, the methanolic extracts of both Seseli species showed lower cytotoxicity towards HaCaT cells (IC50: >500 µg/mL), compared to the water extracts (IC50: 267.8 and 321.41 µg/mL). Besides, only the methanolic extracts showed a slight wound healing effect (28.21 and 31.23%). All extracts showed antibacterial action against Staphylococcus lugdunensis (minimum inhibitory and bactericidal concentrations: 0.025–2 mg/mL). S. gummiferum methanolic extract, which exhibited the highest antibacterial potency was found to inhibit adhesion and invasion of S. lugdunensis to HaCaT cells as well. Taken together, this study demonstrated the two Seseli species to harbour interesting bioactive components, in particular polyphenolics and to exhibit several biological properties that could be further investigated for their potential exploitation as healing agents as supported by various traditional medicinal uses.

Background: Heteromorpha arborescens (Spreng.) Cham. and Schltdl (Apiaceae) is widely used traditionally for the treatment of a wide range of diseases in Southern and Eastern Africa. Although previous studies have reported the biological activities of hexane, ethyl acetate and methanol extracts of H. arborescens leaves, there is no scientific information on the phytochemical contents, antioxidant and antibacterial activities of acetone, ethanol, aqueous and blanched extracts. This study is therefore aimed to investigate and compare the phytochemical contents, antioxidant and antibacterial activities of acetone, ethanol, aqueous and blanched extracts of H. arborescens leaves. Methods: Phytochemical analysis for the total phenolic, flavonoid, proanthocyanidin, alkaloid and saponin contents of all the fractions were determined by spectroscopic methods, while the free radical scavenging potential of the extracts were evaluated using DPPH, ABTS radical scavenging and total antioxidant capacity assays. Micro dilution method was used to determine the Minimum Inhibitory Concentrations (MIC) of H. arborescens leaf extracts against Bacillus pumilus, Staphylococcus epidermidis, Staphylococcus aureus, Escherichia coli and Klebsiella pneumoniae. Results: Total phenol content of the extracts ranged between 15.10 mg GAE/g- 42.50 mg GAE/g, proanthocyanidin was 459-8402.1 mg QE/g, and flavonoid content of 109.24-235.79 mg QE/g. In addition, alkaloids (5.59%) and saponins (23.33%) were present in significant amounts. Based on the IC50 values, the ethanol extract exhibited the highest total antioxidant activity (0.013 mg/mL) with highest inhibition against DPPH and ABTS radicals (0.06 and 0.049 mg/mL respectively). Considerable antibacterial activities were observed in the acetone, ethanol and blanched extracts with MIC values ranging from 1.563-12.5 mg/mL; however, the aqueous extract was inactive against all the bacteria strains. Conclusion: The study suggests that H. arborescens leaves could be a valuable source of bioactive compounds. Although the blanching process significantly decreased polyphenolic contents and antioxidant activities of the extracts, it increased the antibacterial compounds.

Tea is the second most widely used drink all over the world after water. It has several health benefits. The aqueous and methanolic extracts of black, green and herbal teas for the treatment of hyperglycemia and its associated complications were investigated. Nine tea samples including black, green and herbal teas namely Lipton black tea (LBT), Supreme black tea (SBT), Tapal black tea (TBT), Jasmine green tea (JGT), Lemon grass green tea (LGrT), Lemon green tea (LGT), Berginia ciliata, Bistorta affinis and Paeonia emodi were selected for the study. Total phenolic contents (TPC), total flavonoid contents (TFC), antioxidant properties including inhibitory activity against 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) radical (ABTS•+), iron chelation and enzymes inhibitory potential for alpha glucosidase and alpha amylase were analyzed based on in vitro assays. The percentage yield of extracts was in the range of 7.2 to 47 %. Methanolic extracts showed greater TPC values (μg GAE/g) while aqueous extracts showed greater TFC values (μg Rutin E/g). The aqueous extracts of most of the studied tea samples showed strong biological activities with very low IC50 (50% inhibitory concentration) values (0.2-69.0 μg/ml) for ABTS•+ radical scavenging, Fe2+ chelation and alpha glucosidase inhibition assays. For amylase inhibition activity, some aqueous extracts including LBT, SBT, JGT and P. emodi exhibited greater IC50 values. Bangladesh J. Bot. 50(3): 491-498, 2021 (September)

Plant extracts are an important alternative to antibiotics, which are ever more restricted because of their developing microbial resistance and some adverse effects that have been observed following frequent application. The aim of the present study was to determine the antifungal and antioxidant activity of the methanolic extracts of Acorus calamus, Chlorella vulgaris, Lemna minuta and Scenedesmus dimorphus. The antifungal activity of the extracts against strains of Aspergillus flavus, Aspergillus parasiticus, Aspergillus ochraceus, Aspergillus niger, Aspergillus carbonarius, Fusarium graminearum, Fusarium oxysporum, Penicillium chrysogenum and Alternaria alternata was evaluated via the agar well diffusion method. The antioxidant activity of the extracts was measured through the determination of three parameters—total phenolic content, total flavonoid content and radical scavenging potential (determined through UV/Vis analysis). A. calamus extracts had the highest antimicrobial activity against eight fungal strains, followed by the C. vulgaris, L. minuta and S. dimorphus extracts, which were inhibitory against two to three strains. Among the extracts from the species studied, the extract from S. dimorphus showed the highest antioxidant potential, as determined via the DPPH (1,1’-diphenyl-2-picrylhydrazil-radical) method. This correlated to its high total phenolic and flavonoid content. From A. calamus and L. minuta, methanolic extracts were obtained that exhibited similar values of the aforementioned parameters, followed by C. vulgaris extracts, which showed the lowest antioxidant activity. Based on the Pearson correlation coefficients, the impacts of the total phenolic content and the total flavonoid content on radical scavenging capacity are similar, and flavonoids were a significant part of the total phenolic compounds extracted from the plant materials studied.

Sideritis niveotomentosa Hub. -Mor. is a local endemic species belonging to the Lamiaceae family. In this study, GC/MS analysis, total antioxidant capacity and anticancer effects of different extracts obtained from S. niveotomentosa were investigated comparatively. Total phenolic contents of extracts were determined by the Folin–Ciocalteu method, total flavonoid contents by aluminum chloride method, and also the free radical scavenging activities of the extracts by DPPH (2,2-diphenyl-1-picryl-hydrazyl-hydrate) assay. The cytotoxic effect of the extracts was studied via MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-Diphenyltetrazolium Bromide) assay on DLD1, HL60 and ARH77 cell lines. Pro-apoptotic gene expression levels were also tested in the most sensitive cell line ARH77 by Real-Time PCR. The expression levels of 4 pro-apoptotic genes, APAF, BAX, CASP3, and HRK were found to be upregulated in ARH77 cells that were treated extracts. Results showed that methanolic extracts contain more phenolic content than acetone extracts, consistent with DPPH results. As a result, Sideritis niveotomentosa extracts, especially methanolic extracts, are rich in phenolic content and have a strong radical scavenging effect. In addition, the extracts showed selective effects on cell lines. This study is pioneering in terms of future studies, and the findings provide hope for future experimentation.

Quercetin is a flavonoid that is found in many plant materials, including commonly eaten fruits and vegetables. The compound is well known for its wide range of biological activities. In this study, 5-O-acyl derivatives of quercetin were synthesised and assessed for their antiproliferative activity against the HCT116 colon cancer and MDA-MB-231 breast cancer cell lines; and their radical scavenging activity against the 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical cation and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical species. Four derivatives were found to have improved the antiproliferative activity compared to quercetin whilst retaining radical scavenging activity.

A dozen Iris species (Iridaceae) are considered traditional remedies in Kurdistan, especially for treating inflammations. Phytochemical studies are still scarce. The information reported in the literature about Iris species growing in Kurdistan has been summarized in the first part of this paper, although, except for Iris persica, investigations have been performed on vegetal samples collected in countries different from Kurdistan. In the second part of the work, we have investigated, for the first time, the contents of the methanolic extracts of Iris postii aerial parts and rhizomes that were collected in Kurdistan. Both extracts exhibited a significant dose-dependent free radical scavenging and total antioxidant activities, comparable to those of ascorbic acid. Medium-pressure liquid chromatographic separations of the two extracts afforded l-tryptophan, androsin, isovitexin, swertisin, and 2″-O-α-l-rhamnopyranosyl swertisin from the aerial parts, whereas ε-viniferin, trans-resveratrol 3,4′-O-di-β-d-glucopyranoside, and isotectorigenin were isolated from the rhizomes. This is the first finding of the last three metabolites from an Iris species. The various remarkable biological activities of isolated compounds scientifically sustain the traditional use of I. postii as a medicinal plant.

In this study, the optimal condition for the extraction of antioxidants from the fruit Buah Mahkota Dewa (Phaleria macrocarpa) was determined by using Response Surface Methodology (RSM). The optimization was applied using central composite design (CCD) to investigate the three independent variables, namely extraction temperature (oC), extraction time (minutes) and extraction solvent to-feed ratio (%v/v) on the responses of free radical scavenging activity (DPPH), ferric ion reducing power assay (FRAP), total phenolic content (TPC) and total flavonoid content (TFC).The optimal conditions for the antioxidants extraction were found to be extraction temperature (64oC), extraction time (66 minutes) and solvent to-feed ratio (75 %v/v) with the highest percentage yield of DPPH, FRAP, TPC and TFC were 86.85%, 7.47%, 292.86 mg/g and 3.22 mg/g respectively. Moreover, the data were subjected to response surface methodology (RSM) and the results showed that the polynomial equations for all models were significant, did not show lack of fit, and presented adjusted determination coefficients (R2) above 99%, proving the yield of phenolic, flavonoid and antioxidants activities obtained experimentally were close to the predicted values and the suitability of the model employed in RSM to optimize the extraction conditions. Hence, in this study, the fruit from P.macrocarpa could be considered to have the strong antioxidant ability and can be used in various cosmeceutical or medicinal applications.

This work aims to determine antioxidant, total phenolic content and Fourier transform infrared spectroscopy (FTIR) analysis of breadfruit leaves which are essential in management of diabetes. The methanolic extracts of breadfruit leaves was used to analyze for total phenolic content and antioxidant. Total phenolic content of the extracts was measured using the Folin–Ciocalteu assay while the antioxidant activity of plant extracts was measured by the 1, 1-diphenyl-2-picryhyradrazyl (DPPH) radical scavenging assay. FTIR analysis was used to determine the chemical components in the leaves. Total phenolic content in fresh breadfruit leaves (144.16 mg/g ± 17.98) was comparable to those of green tea. The results showed the breadfruit leaves extracts exhibited potent antioxidant activity. The presence of OH group also suggests antioxidant capacity of breadfruit leaves to deactivate free radicals as glucose itself could react with hydrogen peroxide in the presence of iron and copper ions to form hydroxyl radical.

Objective: The aim of this study is to evaluate the antioxidant potential, determination of total phenolic and flavonoid content in nine selected medicinal plants Spondias pinnata, Melia azedarach, Ageratina adenophora, Urtica dioica, Curcuma longa, Bauhinia variegata, Elaeocarpus angustifolius Blume, Achyranthes aspera, and Psidium guajava from Kavre district of Nepal using in vitro studies. Methods: Methanolic plant extracts were prepared by cold percolation method. The methanol extract of nine medicinal plants collected from Kavre district of Nepal, was screened for assessing bioactive phytoconstituents followed by antioxidant property, total phenolic, and flavonoid content. Different plants collected were powdered and extracted with methanol, concentrated by a rotatory evaporator and analyzed for the presence of phytochemicals. The antioxidant potential of the plant extracts was evaluated by 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay. Results: The phytochemical analysis of methanolic extracts of all nine medicinal plants displayed the presence of various secondary metabolites such as alkaloids, flavonoids, polyphenols, saponins, and quinones. The extract of S. pinnata showed the highest percentage of radical scavenging activity up to 87.94±1.88 with 50% inhibitory concentration (IC50) 17.51±1.27 μg/mL, followed by B. variegata, 80.63±1.06 with IC50 value 26.55±2.61 μg/mL. The standard, ascorbic acid has IC50 value of 20.13±1.17 μg/mL. Further, the ethyl acetate fraction of S. pinnata showed the maximum percentage of radical scavenging (85.92±1.37) with IC50 value of 46.95±1.17 μg/mL. Moreover, S. pinnata displayed the highest total phenolic content (TPC) 48.26±1.23 mg GAE/g (milligram gallic acid equivalent per gram) extract while the highest flavonoid content was displayed by Melia azedarach 41.07±1.53 mg QE/g (milligram quercetin equivalent per gram) extract measured by the Folin–Ciocalteu reagent method and aluminum chloride colorimetric method. Conclusions: The preliminary results of this study have put forward the extract of S. pinnata showed the highest percentage of radical scavenging activity and S. pinnata displayed the highest TPC while the highest flavonoid content was displayed by Melia azedarach methanolic extracts although the further studies are needed to assess its mechanism of action.
