Changes in endogenous hormones and seed-coat phenolics during seed storage of two Suaeda salsa populations

Changes in endogenous hormones and seed-coat phenolics during the storage of the dimorphic seeds of two Suaeda salsa populations were investigated. The results showed that, regardless of salinity, storage did not reduce the germination of brown or black seeds from an inland population, but it significantly decreased the germination of dimorphic seeds from an intertidal population. Abscisic acid (ABA) concentration was significantly higher in old seeds than in fresh seeds (freshly harvested dry seeds), whereas the opposite trend was evident for gibberellic acid (GA, including GA1 and GA3) concentration and GA : ABA ratio in both populations, and especially in the intertidal population. Concentration of seed-coat phenolics extracted from brown seeds was much higher than in black seeds in both populations, regardless of extracting time and storage. Storage increased the concentration of phenolics extracted from brown seeds in the inland population, whereas the opposite trend was evident in the intertidal population. Storage did not significantly change seed-coat phenolics extracted from black seeds relative to brown seeds in either population. The results indicated that changes in seed vigour during storage in dimorphic seeds of the two S. salsa populations are related to changes in endogenous hormones and seed-coat phenolics.

AbstractSuaeda salsa is an annual herbaceous euhalophyte in the family Chenopodiaceae that produces dimorphic seeds on the same plant under natural conditions. In order to determine the effect of salinity on seed quality traits during seed formation, seeds from plants grown under control conditions and on 200 mM NaCl were used to investigate the effect of NaCl on seed production and seed germination. Results showed that size and weight of both black and brown seeds generated from 200 mM NaCl-treated plants were markedly greater than those from controls. The germination percentage of brown seeds from both control and NaCl-treated plants was higher than that of black seeds. Furthermore, the germination percentage of the black seeds generated from 200 mM NaCl-treated plants was significantly higher than that of the control at different concentrations of NaCl, although germination percentage declined with the increase NaCl concentration. Surprisingly, NaCl did not affect germination of the brown seeds. The germination index and vigour index of both black and brown seeds from the control plants were significantly lower than those of seeds from the different NaCl treatments. Seed starch, soluble sugar, protein and lipid content of both black and brown seeds generated from the 200 mM NaCl-treated plants were significantly higher than those from the control. These results suggest that a certain concentration of NaCl plays a pivotal role in seed vitality of the euhalophyte S. salsa through increasing seed weight and contents of storage compounds such as protein, starch and fatty acids.

Seed vigour, viability, the contents of soluble carbohydrates, total protein, albumins, and globulins, as well as seed coat structure, were analysed in yellow lupin (<em>Lupinus luteus</em> L.) cv. Iryd seeds stored for 20 years at -14<sup>o</sup>C, 0<sup>o</sup>C or at room temperature (approx. +20<sup>o</sup>C). Seed storage at room temperature reduced viability (to 2%) and increased seed leachate electroconductivity. Determinations of total proteins showed that protein content was significantly reduced in seeds stored at +20<sup>o</sup>C compared to the other storage regimens. Raffinose family oligosaccharides were the main soluble carbohydrates in seeds stored at 0<sup>o</sup>C and -14<sup>o</sup>C, whereas sucrose dominated in seeds stored at room temperature. Scanning electron microscopy (SEM) of seed surface and seed coat sections revealed appearance of an amorphic layer on the surface of seeds stored at room temperature (not observed in other seeds) and distinct shrinking of macrosclereid layer in seeds stored at -14<sup>o</sup>C. Macrosclereids layer in all seeds was 100 um thick and accounted for 60% of seed coat thickness. The obtained results suggest that for long term storage of lupin seeds at 0<sup>o</sup>C is the most advisable temperature if both costs of storage and seed storability are considered.

The electrical conductivity test has been used to evaluate seed vigour in many species, especially legumes, such as peas, beans and soybeans. The objective of this research was to evaluate the effect of temperature and storage period on the results of the electrical conductivity test and the mineral composition of the imbibition solution of bean seeds (Phaseolus vulgaris L.). Three seed lots of the cultivar IAPAR - 81 and cultivar IPR - TIZIU were stored at 10, 25 and 25-10º C (six months at 10º C and six months at 25º C) The following evaluations were made every three months for a year: seed water content, germination and vigour (accelerated ageing and electrical conductivity) and the potassium, calcium and magnesium contents of the imbibition solution. The cultivars showed similar behavior in storage. The reduction in the vigour of bean seeds stored at 10º C was verified by accelerated ageing and electrical conductivity tests. The bean seed storage at low temperature (10º C) does not influence the results of the evaluation of seed vigour by electrical conductivity test and the quantification of calcium, magnesium and potassium ions. The evaluation of vigour by the electrical conductivity test is not recommended for bean seed stored by long periods (above 9 months).

Chrysophyllum  albidum is a prominent, indigenous, economic fruit tree species found in compound farm agro-forestry systems . Investigation was conducted to overcome difficulty in germination and storability of C. albidum seeds in the forest nursery of Federal University of Agriculture, Abeokuta, Nigeria. The trial employed a 3x3 factorial experiment with three replications laid out in a completely randomized design to determine the effect of seed coat treatments including clipping at 2mm from distal, circumference and micropyle end; dipping in 10% sulphuric acid and no seed coat treatments and durations of seed storage 1, 5 and 9 days on the germination of C. albidum seeds. Pre-sowing and storage treatments significantly (p < 0.05) affected the germination of seeds. Percentage germination ranged from 17% to 61% for clipping to no seed coat treatment. Highest germination of 61% was recorded for seeds not pre-sowed. The percentage germination ranged from 16% to 60% for seeds stored for nine days to those stored for one day. Highest germination of 60% was recorded for seeds stored for one day. A significant (p < 0.05) decrease in percentage germination was recorded with increasing days of storage. The result of interaction effect revealed that highest germination of 86% was recorded for seeds stored for 5days without pre-sowing treatments. Storing of seeds for 5days is recommended for mass production of C. albidum seedlings in agro-forestry nursery.

Seed longevity of 76 spring barley gene pool samples (Hordeum vulgare L. subsp. distichon, convar. distichon: 56 nutans Schubl., two deficience (Steud.) Koern., two erectum Rode ex Shuebl., two medicum Koern.; convar. nudum (L.) A.Trof.: one nudum L. та subsp. vulgare: convar. vulgare: nine pallidum Ser., three rikotense Regel.; convar. coeleste (L.) A.Trof.: one coeleste (L.) A.Trof.) from 26 countries, 11 years and four places of reproduction was analyzed. Seeds with 5–8% moisture content were stored in chamber with unregulated and 4oC temperature. The possibility of seed storage under these conditions for at least 10 years without significant changes in germination has been established. The importance of meteorological conditions in the formation and ripening of seeds for their longevity is confirmed. The relationship between the decrease of barley seeds longevity and storage conditions, amount of rainfall, temperature regime during the growing season of plants is discussed.

Different seed storage methods, varying in storage temperature, moisture and/or oxygen content, were applied to commercial sugar beet seed lots from four breeding companies. After storage for 10–11 months, germination of the seed was tested in the laboratory (cold test, 10°C). In addition, the contents of active ingredients (fungicides and insecticide) were analyzed and compared with the initial contents before storage. Based on these results, a selection of the most promising storage methods was made to test plant emergence in a field experiment. This research was performed in 2015/16 and in 2016/17. In both years, two storage treatments outperformed the others: these were storage in a closed jar with the addition of moisture absorber (i.e. silica gel) at room temperature and storage at –18°C in a closed plastic bag. Using these two storage methods, seed vigour and contents of active ingredients were comparable to those in seed that had not been stored for one year. Based on the results from this study, the advice to growers for a successful storage of residual sugar beet seed was adjusted in 2017, after including some practical guidelines and considerations.

Seed quality may be affected by several factors, including permeability, color, and lignin content in the seed coat. This study aimed at evaluating influence of lignin content in the tegument of seed samples of six different soybean cultivars, in which half of each sample was inoculated with the fungus Aspergillus flavus, on the physical and physiological quality, and on the seed health, during 180 days storage period, under cold chamber with controlled conditions of temperature and RH. For that, at each interval of 60 days, samples were removed, and the physiological quality of these seeds was assessed by means of moisture and lignin contents; and by tests of seed health, germination, and electrical conductivity. The moisture content of seeds remained constant during all storage period. In the seed health test, it was found that inoculation was efficient, once the minimum incidence of the fungus in the inoculated seeds was 85%. In the germination test, there was a trend of reduction on percentage germination with the increase in storage period. However, there was an increase on electrical conductivity of seeds assessed. It was concluded that there is no interference of the lignin content in the seed coat on the resistance to infection by the fungus Aspergillus flavus, even after seed storage for a period of 180 days.

Seeds of native forest species for food and seedling production have a growing demand that impacts the need for studies involving seed storage in order to maintain good levels of germination quality. Thus, scientific analyzes that explore the physiological potential of the seeds of these species are necessary, especially for the pink pepper (Schinus terebinthifolia Raddi.), a species used as an alternative source of income for traditional communities and which has been arousing interest due to the consumption of its fruits as a condiment in the national and international market. This work aimed to assess the physiological quality of pink pepper seeds according to the storage period and seed size. The seeds came from a rural settlement, a pioneer in the extraction of pink pepper, in the state of Rio de Janeiro, Brazil. The seeds were evaluated for moisture content, germination, first count, length and dry weight of the germinated seedlings, during the period of time that they remained stored and the size of the seeds. The results allowed to conclude that: i) the germinative percentage of the stored pink pepper seeds decreased over the months; ii) the moisture content of the seeds decreased over the months evaluated iii) the different sizes of seeds did not show differences in germination and storage time and iv) values of length and dry weight of the seedlings did not differ according to the period of storage.

Seeds of Garcinia talbotii remained viable hardly for two weeks in open room conditions. As part of ex-situ conservation of Garcinia talbotii, seeds longevity was studied since seeds are the main propagule. For this, the relationship of seed viability with respect to different moisture content and storage temperature were analyzed. Seed storage behaviour is also investigated. Being recalcitrant, seeds are desiccation sensitive as well as chilling sensitive. During hermetic storage of seeds at 300C /70 % RH seeds retained viability about 6 months.
