Pollution of the natural environment with fluorine compounds makes the continuous monitoring of the content of this element necessary, not only in the air, water and soil, but also in food. A high level of fluorine contamination of soils can affect quality of the obtained yields, affecting the amino acid composition of protein. It often becomes a cause of reducing nutrition and feed value of agricultural yields. In the undertaken studies, the influence of fluorine application to the soil on the amino acid content in proteins accumulated in the above-ground parts (aerial) of Zea mays L. and Lupinus luteus L. was studied. The lowest and medium doses of fluorine had a significant positive effect on the content of exogenous and endogenous amino acids in the protein of the aerial mass of maize, for which the increase of sum of all amino acids was 7% and 8%, respectively, in relation to controls. The above-ground parts of yellow lupine were characterised by a much higher content of the tested amino acids than maize. The subsequent influence of soil contamination with fluorine on the amino acid content in yellow lupine did not have the same effect as in the main plant, and the influence of fluorine was somewhat limited. With respect to yellow lupine, it was demonstrated that the lowest fluorine dose (100 mg F kg−1 of soil), did not have a beneficial influence on the sum of all amino acids. The medium and highest doses of fluorine depressed in a small degree the content of the sum of these compounds in the dry mass of yellow lupine.

Annotation Over the years of many years of research in the field, it was found that the creation of completely anthracnose-resistant varieties of yellow lupine (Lupinus Luteus L) is not possible. But at each stage of the pathological process, various defense mechanisms operate that determine the resistance or susceptibility of plants to diseases. There is a passive defense associated with morpho-biological features. A positive role in the control of anthracnose of early and mid-season forms with limited second-order shoots, fast-growing ones with anthocyanin coloration, xeromorphic leaf type, dark green color of vegetative organs, intensely colored seeds (2c, 4c, 2d), different corolla colors (numbers 52-87-2113 and 1477-1-17) and a flower (varieties Iputsky and Rodnik). Observations have shown that a waxy coating on a bean or a thick layer of hairs that helps water roll off, a more spreading bush that provides quick ventilation and moisture removal, reduces anthracnose infestation. It was found that the passive defense mechanism is largely due to the totality of morpho-biological characteristics and the nature of their interaction. Different infestation with anthracnose depending on the sowing density was shown: under conditions of sparse sowing (600 thousand seeds / ha), it averaged 4.6% during flowering and 8.0% at bean formation, and in a denser cenosis (1.2 mln. grain / ha), respectively, 18.7 and 62.0%. An active plant defense mechanism has been established and is being studied -hypersensitivity - this is an increased sensitivity of living cells of a plant - a host in places where a pathogen penetrates and its rapid death with the formation of necrosis. It was revealed that necrotic forms of yellow lupine are less affected by anthracnose during all phases of plant growth and development and, especially, during the formation of beans. At the site of penetration of the pathogen, an orange spot develops, around which a roller of dead tissue is formed, i.e., the pathogen’s access to living cells stops at the expense of which it feeds. The selected material with bean necrosis, without external sporulation, reduces the infectious load in the infected crops.

The data on the composition and amount of amino acids have been analyzed in the raw materials of five legume species. All of them grow in Southern Ukrainian flora (Securigera varia (L.) Lassen, Vicia cracca L., Lupinus luteus L., Melilotus officinalis (L.) Pall., Melilotus albus Medic.) and may be used as a source of amino acids, especially essential, whose resource has to be replenished from the outside. Aim. We have studied and compared the amino acid profile of some species of the Ukrainian South Legumes, and used the multidimensional statistical cluster analysis to construction of histograms based on the amino acids content and composition of such plants as: Securigera varia (L.) Lassen, Vicia cracca L., Lupinus luteus L., Melilotus officinalis (L.) Pall., Melilotus albus Medic. Materials and methods. The raw materials were harvested at the South of Ukraine and were investigated by gas-liquid chromatography. The amino acid analyzer has been used after hydrochloric acid hydrolysis at elevated temperature. Results. 19 amino acids have been identified, of which nine are essential or partially interchangeable. The non-polar amino acid proline is in the lead in terms of quantity among the essential amino acids. Its amount was 6932 mg/100 g and the ability to accumulate it was noted in Melilotus officinalis (2276 mg/100 g). The smallest proline amount was found in Lupinus luteus (388 mg/100 g). The sulfur-containing non-polar amino acid methionine is in the smallest amount in the selected plants (506 mg/100 g). Our attention was drawn to the absence of the polar amino acid glutamine among the non-essential amino acids in some plants. Securigera varia, Vicia cracca, and Melilotus officinalis did not contain glutamine. In this subgroup, the polar aspartic acid was found in the highest amount (6824 mg/100 g) with the highest content in Vicia cracca and the lowest – in Melilotus albus (2660 mg/100 g and 385 mg/100 g, respectively). Conclusions. The analysis of the presence and number of amino acids was the basis for our multidimensional statistical cluster analysis and histograms of the presentation of the amino acid profile of the studied plant members of the family Fabaceae L. In constructing the dendrogram, three clusters were identified, and representatives of one genus (Melilotus L.) were attributed to different clusters which is significant for further chemosystematic studies.

Lupin seeds can represent a valuable source of phenolics and other antioxidant compounds. In this work, a comprehensive analysis of the phytochemical profile was performed on seeds from three Lupinus species, including one cultivar (Lupinus albus) and two wild accessions (Lupinus cossentinii and Lupinus luteus), collected from the northern region of Tunisia. Untargeted metabolomic profiling allowed to identify 249 compounds, with a great abundance of phenolics and alkaloids. In this regard, the species L. cossentinii showed the highest phenolic content, being 6.54 mg/g DW, followed by L. luteus (1.60 mg/g DW) and L. albus (1.14 mg/g DW). The in vitro antioxidant capacity measured by the ABTS assay on seed extracts ranged from 4.67 to 17.58 mg trolox equivalents (TE)/g, recording the highest values for L. albus and the lowest for L. luteus. The DPPH radical scavenging activity ranged from 0.39 to 3.50 mg TE/g. FRAP values varied between 4.11 and 5.75 mg TE/g. CUPRAC values for lupin seeds ranged from 7.20 to 8.95 mg TE/g, recording the highest for L. cossentinii. The results of phosphomolybdenum assay and metal chelation showed similarity between the three species of Lupinus. The acetylcholinesterase (AChE) inhibition activity was detected in each methanolic extract analyzed with similar results. Regarding the butyrylcholinesterase (BChE) enzyme, it was weakly inhibited by the Lupinus extracts; in particular, the highest activity values were recorded for L. albus (1.74 mg GALAE/g). Overall, our results showed that L. cossentinii was the most abundant source of polyphenols, consisting mainly in tyrosol equivalents (5.82 mg/g DW). Finally, significant correlations were outlined between the phenolic compounds and the in vitro biological activity measured, particularly when considering flavones, phenolic acids and lower-molecular-weight phenolics.

This study aimed to investigate the impact of climate warming on hormonal traits of invasive and non-invasive plants at the early developmental stage. Two different lupine species—invasive Lupinus polyphyllus Lindl. and non-invasive Lupinus luteus L.—were used in this study. Plants were grown in climate chambers under optimal (25 °C) and simulated climate warming conditions (30 °C). The content of phytohormone indole-3-acetic acid (IAA), ethylene production and the adaptive growth of both species were studied in four-day-old seedlings. A higher content of total IAA, especially of IAA-amides and transportable IAA, as well as higher ethylene emission, was determined to be characteristic for invasive lupine both under optimal and simulated warming conditions. It should be noted that IAA-L-alanine was detected entirely in the invasive plants under both growth temperatures. Further, the ethylene emission values increased significantly in invasive lupine hypocotyls under 30 °C. Invasive plants showed plasticity in their response by reducing growth in a timely manner and adapting to the rise in temperature. Based on the data of the current study, it can be suggested that the invasiveness of both species may be altered under climate warming conditions.

In this work, we analyzed the diversity of the nodule-forming bacteria associated with Lupinus luteus and Lupinus cosentinii grown in the Maamora Cork oak forest acidic soils in Morocco. The phenotypic analysis showed the high diversity of the strains nodulating the two lupine's species. The strains were not tolerant to acidity or high alkalinity. They do not tolerate salinity or high temperatures either. The strains isolated from L. luteus were more tolerant to antibiotics and salinity than those isolated from L. cosentinii. The plant growth promoting (PGP) activities of our strains are modest, as among the 28 tested isolates, only six produced auxins, six produced siderophores, whereas three solubilized phosphates. Only two strains possess the three activities. The rrs gene sequences from eight representative strains selected following ARDRA and REP-PCR results revealed that they were members of the genus Bradyrhizobium. Six strains were then retained for further molecular analysis. The glnII, recA, gyrB, dnaK, and rpoB housekeeping gene sequence phylogeny showed that some strains were close to B. lupini LMG28514T whereas others may constitute new genospecies in the genus Bradyrhizobium. The strains were unable to nodulate Glycine max and Phaseolus vulgaris and effectively nodulated L. luteus, L. cosentinii, L. angustifolius, Chamaecytisus albidus, and Retama monosperma. The nodC and nodA symbiotic gene phylogenies showed that the strains are members of the genistearum symbiovar.

Functional analysis of promoter sequences is important to understand the regulation of gene expression. This study aimed to investigate the promoter region of the Lupinus luteus cytoplasmic cyclophilin gene (LlCyP; AF178458). After bioinformatic analysis, four promoter deletion fragments were fused to the β-glucuronidase reporter gene. We used Lotus japonicus as a model plant. After Agrobacterium rhizogenes transformation of L. japonicus, only the longest promoter region (−1055 bp to ATG) supported the β-glucuronidase expression in root nodule parenchyma. Putative cis-elements located between −1055 and −846 bp were subjected to site-directed mutagenesis. Mutations incorporated in the TGATT and AGATT motifs (cytokinin response) abolished GUS expression in nodules, but the mutated AAAGAT motif (OSE, organ-specific element) still activated the GUS expression in root nodules, mainly in cells surrounding the vascular bundle. Promoter deletion and mutation experiments suggest that cis-elements responsible for gene expression in the nodule are located in the region spanning from −1055 to −846 bp. We constructed a deletion fragment, in which the DNA sequence located between −822 and −198 bp was removed (pCYPMG). The promoter region arranged in the pCYPMG supports the expression in the parenchyma of L. japonicus nodules, but it is lower than the whole promoter region. The obtained results may be useful for transgene expression in determinate and indeterminate root nodules.

Lupins are suitable candidates to replace soybean meal in livestock feeding in the Mediterranean area, presenting a solution for the European Union’s dependence on soybean importations. This study aimed to assess the effect of incorporating Lupinus albus and Lupinus luteus into Churra da Terra Quente lambs’ diets on growth performance and digestibility. Two trials were conducted over two years. In trial 1, two experimental diets containing 50 g/kg Lupinus albus and 50 g/kg Lupinus luteus were tested. In trial 2, lambs were fed with diets containing higher incorporations of Lupinus luteus (100, 150, and 200 g/kg: LL10, LL15, and LL20, respectively). Total dry matter, hay dry matter, and crude protein intake were calculated, as well as average daily gains. At the end of the growth trials, dry matter, organic matter, and NDF digestibility was determined. Incorporating 50 g/kg of lupins did not affect (p > 0.05) the performance. Lambs fed on LL20 diets presented the lowest HDMI and CPI values (p < 0.05). The highest intakes (p < 0.05) were observed from LL15 lambs. No differences were found in apparent digestibility coefficients between diets (p > 0.05), except for NDF digestibility which was highest (p < 0.05) for LL20. The optimum level of lupin inclusion in lambs’ diets seems to be 150 g/kg.
