Experiments were designed to determine the effect of ovulation stimulus on the biochemical composition of oviduct fluid in alpacas (Vicugna pacos). Twenty-four adult females were used to determine profiles of protein, albumin, glucose, cholesterol, and triglycerides in oviduct fluid. The animals were stimulated with 1 mL of seminal plasma (SP), 0.042 mg of a gonadotropin-releasing hormone (GnRH) analogue (buserelin acetate; Conceptal, Intervet, Holland), or PBS as control group. The oviduct was removed inmediately after slaughter on Day 2 or 4 after ovulation induction and washed with 3 mL of PBS solution. Determination of the biochemical composition was carried out with specific kits from the FAR Diagnostics Italia laboratory using a semiautomatic biochemical analyzer of the Sinowa China brand, which is based on spectrophotometric evaluation. The results are presented in Table 1. Our results suggest that the method of induction of ovulation affected the biochemical composition of oviduct fluid, including concentrations of albumin, cholesterol, total protein, triglyceride, and glucose. Alpacas, like other camelids, are induced ovulators. A study was designed to determine the effect of the ovulation-inducing stimulus on the biochemical composition of follicular fluid. Adult female alpacas (n = 18) were examined daily for 3 days by transrectal ultrasonography using a 5-MHz linear-array transducer (Aloka SSD-500, Tokyo, Japan). When the largest growing ovarian follicle was ≥7 mm, alpacas were given 1.0 mL of seminal plasma intramuscularly (SP, n = 9) or 40 µg of busereline acetate intramuscularly (GnRH, n = 9). A transvaginal transducer with an attached needle guide (Aloka UST-945BP-5) was used for collection of follicular fluid 22 h post-induction. Follicular contents were then centrifuged at 800 × g for 20 min to separate the fluid from the cells. The follicular fluid was collected and stored at –20°C until analysis with a semi-automatic biochemical analyzer (SINOWA, China). The results were glucose 49.17 and 47.95 (mg/dL; P > 0.05), total protein 1.85 and 1.15 (g/dL; P < 0.05), albumin 1.11 and 1.13 (g/dL; P > 0.05), triglycerides 3.94 and 3.16 (mg/dL; P > 0.05), cholesterol 39.01 and 42.5 (mg/dL; P > 0.05), phosphatase 32.68 and 21.36 (IU/L; P < 0.05), alanine aminotransferase 3.66 and 5.07 (IU/L; P > 0.05), and lactate dehydrogenase 42.17 and 27.27 (IU/L; P > 0.05) for SP or GnRH treatments, respectively. Results suggest the need to continue research to explain the effect of possible differences in total protein, cholesterol, and phosphatase on oocyte-expressed genes and follicular development. The article gives an assessment of the effect of the new complex anthelmintic preparation "Prazinal F" on milk productivity and on the biochemical composition of milk of cows of black and white breeds with paramphistomatosis. Determination of the optimal dose after milking the cows of the black and white breed of the new preparation "Prazinal F" with paramphistomatosis showed that during the second, third, fourth and fifth months of lactation for milk production, the dose of 25 mg / kg of body weight has the most beneficial effect. During the second, third and subsequent months of lactation after prescribing "Prazinal F" in a dose of 25 mg / kg body weight, the milk productivity of the cows of the experimental groups and, especially, of the third group is increased. The cows of the 3rd group in terms of gross milk yield exceeded the cows of the control group infected with paramphistomatosis by 36,0%. In comparison with the control infected group, the difference for the third month of lactation, respectively, was 2.9-3.7-4.5 kg in favor of the experimental groups, 3.0-3.5-4.4 kg for the fourth month and the fifth month – 2,6-3,8-4,6 kg. The use of "Prazinal F" in the paramphistomatosis of cows of black and motley breed at a dose of 25 mg / kg of body weight, once allowed to increase the content of iodine, cobalt, magnesium and iron in milk by 31.7-45.6%. The appointment of the drug "Prazinal F" in the paramphistomatosis of cows of black and motley breed at a dose of 25 mg / kg of body weight, once allowed to increase the production of cow's milk, significantly enriching it with nutrient microelements.

Abstract The aim of the study was to evaluate, the ability of a GnRH synthetic analogue [des-Gly10, D-Ala6]-LH-RH ethylamide to induce ovulation in rabbit does using intravaginal administration. A total of 138 primiparous lactating does were randomly divided into 4 groups that at the time of insemination received following treatments for ovulation induction: 1 μg of buserelin administered intramuscularly (control group); 5 μg of [des-Gly10, D-Ala6]-LH-RH ethylamide added to the semen dose (D5 group); 10 μg of [des-Gly10, D-Ala6]-LH-RH ethylamide added to the semen dose (D10 group); 15 μg of [des-Gly10, D-Ala6]-LH-RH ethylamide added to the semen dose (D15 group). Kindling rates were 68.8% in D10 and 66.7% in D15 groups and were comparable to that obtained in the control group (72.2%). The kindling rate in group D5 (29.4%) was significantly lower than those recorded in the other groups. The number of live born kits was not significantly affected by the ovulation induction treatment. The results of this study show that [des-Gly10, D-Ala6]-LH-RH ethylamide added directly into the semen dose can effectively stimulate ovulation in rabbits. The dose of 10 μg of [des-Gly10, D-Ala6]-LH-RH ethylamide per doe was sufficient to produce results comparable to those obtained by intramuscular administration of buserelin.

Alpacas are classified as induced ovulators, and an external stimulus is required for the occurrence of ovulation. The seminal plasma (SP) of camelids contains a protein identified as β nerve growth factor with the capacity to induce ovulation and corpus luteum formation. Alpacas have a poor reproductive efficiency and high embryo mortality rate. A study was designed to evaluate the use of SP to improve reproductive performance post natural mating (Exp. 1) and with different mounting time (MT; Exp. 2). Experiment 1: Nonpregnant alpacas (n = 117) exhibiting a dominant follicle = 7 mm, detected by transrectal ultrasonography, were bred by natural mating and then assigned randomly to 1 of 3 treatments: T1 (n = 40), 1 mL of SP IM; T2 (n = 39), 1 mL of GnRH analogue (0.0042 mg of buserelin acetate), IM; and T3 (n = 38), control. Experiment 2: Nonpregnant alpacas (n = 180) exhibiting a follicle = 7 mm were assigned randomly to the following treatments: T1 (n = 30): MT 5 min; T2 (n = 30): MT 5 min + 1 mL of SP; T3 (n = 30): MT 10 min; T4 (n = 30): MT 10 min + 1 mL of SP; T5 (n = 30): MT > 15 min; T6 (n = 30): MT > 15 min + 1 mL of SP. The same male was used for mating females in T1 and T2, T3 and T4, and T5 and T6, respectively. Animals were evaluated by ultrasound with an Aloka SSD 500 (Aloka, Tokyo, Japan) and 5.0-MHz linear transducer on Day 25 for pregnancy diagnosis. In Exp. 1 the conception rate was 67.5, 51.3, and 55.3% for T1, T2, and T3, respectively (P > 0.05). In Exp. 2 the conception rate was 46.2, 67.9, 57.1, 73.3, 61.8, and 72.7% for T1, T2, T3, T4, T5, and T6 (P < 0.05) and was significantly different between T1 v. T2 and T3 v. T4. The results suggest that the use of SP could be one method to improve reproductive performance in alpacas. Research was supported by Project N° 405-PNICP-PIAP-UNMSM.

Alpacas, like other camelids, are classified as induced ovulators because an external stimulus is required for the occurrence of ovulation. Recent studies have identified the β-nerve growth factor (β-NGF), a protein present in the seminal plasma (SP), as responsible for inducing ovulation in this species and having potent luteotropic function. We speculate that SP can be used in reproductive biotechnologies, such as embryo transfer (ET), to increase the number of genetically superior animals produced in breeding programs. The objective of this study was to evaluate the effect of inducing ovulation with SP or a gonadotrophin-releasing hormone (GnRH) analogue on pregnancy rate in recipients of an alpaca ET program. Semen from 5 adult male alpacas was collected with an artificial vagina and diluted 1:1 (v/v) with phosphate-buffered saline (PBS; Gibco-BRL). The diluted semen was centrifuged for 30min at 1200g, and the supernatant or SP was decanted and examined for absence of sperm. Then, the SP was centrifuged again for 20min at 1200×g. The SP was stored at −80°C until the use. Female alpacas (n=38; 6-8 years) with a body condition score of 2.5-3.5 (scale 1-5) were used for the experiment. Animals were evaluated daily by transrectal ultrasonography to determine the presence of a dominant follicle ≥7mm and randomly assigned to one of two groups: (1) GnRH (n=20), 0.04mg of Buserelin acetate IM, or (2) SP (n=18): 1.0mL of seminal plasma IM. Seven days after GnRH or SP treatment (Day 0) the recipients received a fresh embryo. Alpaca donors (n=18) were given GnRH (0.04mg IM) and treated 36h later with 700IU of equine chorionic gonadotropin (ECG). Donors were mated with fertile males 5 days after ECG (Day 0 of recipients), and embryos were recovered 7 days later. Embryos of similar quality were assigned to both groups and transferred nonsurgically to the uterine horn ipsilateral to the corpus luteum. Ultrasonography examinations were performed on Day 2 to confirm ovulation and Day 25 to determine pregnancy in all of the recipients. Data was analysed by chi-squared test. Ovulation rate was not different between groups (100% each). Pregnancy rate was 45% (9 out of 20) and 44% (8 of 18) in GnRH and SP groups, respectively (P=0.77). In conclusion, SP was effective to induce ovulation in alpacas and was able to produce a pregnancy rate similar to that of GnRH as an ovulation-inducing treatment. Thus, SP can be used as an alternative for ET programs in alpacas. ABSTRACT The fractional determination of 17-ketosteroids in the daily urine was performed in nine cases of hyperemesis gravidarum and in four control cases, in the first trimester of pregnancy both before and after corticotrophin administration. The excretion of total 17-KS is similar in the two groups. Only in the hyperemesis group does the excretion of total 17-KS increase significantly after corticotrophin administration. The fractional determination reveals no difference between the two groups of patients with regard to the values of the fractions U (unidentified 17-KS), A (androsterone) and Rest (11-oxygenated 17-KS). The excretion of dehydroepiandrosterone is significantly higher in the hyperemesis group than in the control group. The excretion of androstanolone seems to be lower in the hyperemesis group than in the control group, but the difference is not statistically significant. The differences in the correlation between dehydroepiandrosterone and androstanolone in the two groups is significant. The high excretion of dehydroepiandrosterone and low excretion of androstanolone in cases of hyperemesis gravidarum is a sign of adrenal dysfunction.

Aim. To study the influence of deproteinized dialysate from blood of milk-fed calves on the type of exudative discharge and dynamics of inflammatory reaction after hernia repair with plastics of the anterior abdominal wall with hernioendoprosthesis of polypropylene. Materials and Methods. The study involved 59 patients being on stationary treatment in the surgical department of Kursk Regional Clinical Hospital. Patients were hospitalized for the herniation of small or medium dimension. The patients were divided to two groups: the main (n=30) and control (n=29) groups. After endoprosthetics, the patients of the control group were given complex conservative treatment. The patients of the main group, besides standard treatment, were administered deproteinized dialysate from blood of milk-fed calves intravenously in drips 10 ml + 200 ml of 0.9% sodium chloride solution within 7 days. For cytological examination and determination of the type of cytograms, the traumatic discharge was collected and analyzed using the method of M.F. Kamaev and M.A. Palthsev. Results. Cytomorphometric examination was conducted on the third, fifth and seventh day after endoprosthetics with the aim of studying dynamic changes. Determination of cell composition and also of its changes characteristic of each studied period, was necessary for obtaining further information characterizing inflammatory process in the region of placement of the endoprosthesis. After endoprosthetics in patients who were administered hemodialysate, the inflammatory reaction was less pronounced than in patients who did not receive the preparation. This was associated with a more dynamic change of stages of the inflammatory process. In patients of the main group who received deproteinized dialysate, regenerative type of inflammation first appeared on the fifth day and made 6.9%; by the seventh day the share of patients with the regenerative type rose to 17.5%, while in the patients of the control group no regeneration stage was observed in both periods. Conclusion. Analysis of the efficiency of influence of hemodialysate on the inflammatory reaction in plastics of the anterior abdominal wall with polypropylene endoprosthesis evidences faster course of all stages of inflammation and reduction of its intensity by 10%.

Abstract This study describes changes in haematological parameters, cytokine profile, histopathology and cortisol levels in Swiss mice experimentally infected with Angiostrongylus costaricensis. Twenty-eight Swiss mice were divided into two groups (G1 and G2) of 14 animals each. In each group, eight animals were infected orally with ten third-stage larvae of A. costaricensis and six were used as a control group. The mice of groups G1 and G2 were sacrificed 14 and 24 days after infection, respectively. Samples were collected for histopathological and haematological analyses and determination of the cytokine profile and cortisol levels. Granulomatous reaction, eosinophilic infiltrate and vasculitis in the intestinal tract, pancreas, liver and spleen were observed with varying intensity in infected animals. Our results showed that the mice developed normocytic and hypochromic anaemia, and that the histopathological lesions caused by the experimental infection influenced increases in cortisol, neutrophil and monocyte levels. In addition to this, we detected increased interleukin-6 and tumour necrosis factor alpha levels in the infected animals.

AbstractCompanion animals, including bitches, may be exposed to zearalenone (ZEN) toxins that are often present in feed, and ZEN intoxication may lead to ovarian dysfunction. This study involved evaluation of the degree of ZEN-induced hypo stimulation of ovary by determination of proliferative and apoptotic indices and description of the ultra-structural organization of ovarian follicles in pre-pubertal bitches subjected to experimental, long-term exposure to low-dose ZEN mycotoxicosis. The experiment involved 30 clinically healthy, immature Beagle bitches aged approximately 70 days with initial average body weight of 8 kg, randomly divided into three groups of 10 animals each: two experimental groups (EI and EII) and a control group (C). Over a period of 42 days, ZEN was administered per os to EI animals at a dose of 50 μg/kg BW, and to EII bitches at a dose of 75 μg/kg BW. Control group animals were fed placebo containing no ZEN for 42 days. Analytical samples of the mycotoxin were administered daily in gelatin capsules before morning feeding. All the bitches were subjected to ovario-hysterectomy at the end of the experiment. Proliferation index (PCNA method) and apoptotic index (TUNEL test) values were determined by immune-histological analyses. The median for apoptotic index was determined at 13.45 for group EI, 17.84 for group EII, and 8.59 for group C. The median for proliferation index was determined at 35.25 for group EII, 42.44 for group EI, and 70.60 for group C. The results of ultra-structural examinations of the ovaries revealed that experimental, ZEN-induced hyperestrogenism enhanced apoptosis and lowered the proliferative ability of follicular cells what contributed to organelle destruction in pre-pubertal bitches. The changes observed were particularly advanced in EII animals, which were administered a higher dose of ZEN.

Abstract Background and Aims Antibodies to M-type phospholipase A2 receptor (PLA2R-Ab) are considered to be a promising biomarker for laboratory diagnosis of primary membranous nephropathy (PMN) and may be useful in the evaluation of the response to therapy and CKD prognosis. The aim of the study was to compare two immunoassay methods – indirect immunofluorescence (IIF) and enzyme immunoassay (ELISA) for the determination of circulating PLA2R-Ab in patients with PMN. Method The study included 54 patients aged 55 (40-63) yrs. (M: F [33:21]) with PMN before treatment (n=16) and treated with immunosuppressive therapy (IST) (n=38), and apparently healthy individuals of the corresponding gender and age (n=10). Proteinuria and estimated glomerular filtration rate (eGFR) were determined in all participants. The levels of PLA2R-Ab were determined by IIF and quantitative/ semi-quantitative ELISA (EURUIMMUN AG test, Germany). In 16 PMN patients without treatment and 28 PMN patients treated with IST the level of PLA2R-Ab was measured one time and in 10 PMN patients treated IST – in dynamic, from 2 to 5 times. Statistical comparisons among groups were performed using Mann–Whitney U-test and Kruskal-Wallis H tests. The association between variables was estimated using Spearman’s coefficient. Sensitivity and specificity of the methods were calculated. Results The correlation coefficient between IIF and ELISA was 0.82 (p &lt;0.005). There were more PLA2R-Ab-positive cases detected by ELISA, both before treatment (ELISA - 80%, IIF - 67%) and among patients treated with IST (ELISA - 63%, IIF - 50%). In control group, ELISA showed no positive results for PLA2R-Ab (specificity was 100%). The levels of proteinuria and eGFR were associated with autoantibodies determined by ELISA, both quantitative and semi-quantitative (proteinuria: r = 0.69, p = 0.001; eGFR: r = -0.38, p = 0.035) but not by IIF (proteinuria: r=0.33, p=0.061; eGFR: r=-0.26, p=0.082). The levels of PLA2R-Ab measured by ELISA correlated with the course of disease in patients treated with IST, while IIF did not show any dynamics is some cases. Conclusion Both quantitative and semi-quantitative ELISA were considered to be more preferable methods since the obtained results correlate with renal dysfunction and allow to assess the concentration of PLA2R-Ab in the course of disease more accurately, that may contribute to timely correction of treatment and improvement of outcome.
