Abstract This study evaluated kampung chickens’ growth rate and production performance in extensive and semi-intensive rearing in Central Java, Indonesia. The survey method was employed for this study. The targets used in the study were kampung chicken farmers in the Banyumas and Kebumen areas. The survey method is carried out by collecting data, observing and measuring population, production performance, including meat and egg production. The data obtained, processed, and presented in the tables and graphs, calculated the population and production of chicken to evaluate its growth. The performance of chicken production, including egg production, egg weight, number of eggs hatched, and hatchability, was analyzed using general linear model (GLM) analysis. The result showed a 1.9% growth rate of the native chicken population, in which the hens outnumbered the roosters. The meat and egg production growth rates were 8.2% and 6.8%, respectively, between 2016 and 2020 in Central Java. Native chickens kept under semi-intensive maintenance produced more eggs than those in an extensive system. Conclusively, native chicken of Central Java experienced growth in population and production of meat and egg. The semi-intensive system, compared to the extensive system, had a higher rate of eggs production.

This study was conducted to compare the environmental conditions around farms which was located in Tegal for Tegal ducks and Magelang for Magelang ducks. This study was also aimed to compare the composition of both feed nutrient content and the egg production. The research was conducted by survey method with purposive random sampling based on duck populations and age (8-15 months) provided by the farmer. Each area was provided 10 farmers for every farm and the data were collected in July-September, 2017. Environmental condition measurement were the temperature of the duck housing, and the composition and nutrient content of feed and the egg production of ducks. The results showed that all farmers’ ducks on a dry system rearing (without water pool) with average temperature in Tegal area 30.53±1.38oC and Magelang 28.32±1.00oC. Feed compositions given for Tegal duck consist of: dried rice, rice bran, concentrate, trash fish, golden snail, vermicelli waste and shrimp waste. The average nutrient content was crude protein 19.25%, energy 2,913 kcal/kg, crude fiber 5.82%, crude fat 6.87%, Ca 3.06% and P 1.40%. Feed composition for Magelang ducks consisted of rice (nasi aking), rice bran and concentrate, with crude protein content of 17.99%, 2,801 kcal/kg, crude fiber 8.14%, crude fat 6.10%, Ca 2.29% and P 1.04%. Tegal duck egg production was lower than Magelang ducks (64.89 vs 75.44%), but the egg weight was relatively the same. It could be concluded that the temperature of Tegal duck housing in Tegal region was hotter than Magelang region. Although the food quality of Tegal ducks was better than that of Magelang ducks, Tegal duck had a lower egg production because of the high environmental temperature.

Background: Prolactin is a physiological candidate gene which has significant effects on egg production in poultry. Also, it plays a major role on incubation behaviour (broodiness) in birds. The main objective of the present study was identification of single nucleotide polymorphism (SNP) C-2161G in the promoter region of prolactin gene and its association with egg production in Tellicherry native chicken population. Methods: A total of 200 blood samples were collected from the randomly selected birds of Tellicherry native chicken in All India Co-ordinated Research Project on Poultry improvement (AICRP) farm, Mannuthy, Thrissur, Kerala. Isolation of Genomic DNA was done and the isolated samples were subjected to polymerase chain reaction (PCR) followed by restriction fragment length polymorphism (RFLP) analysis to identify the SNP C-2161G of prolactin gene. Result: On the basis of RFLP patterns, birds were designated with three different genotypes namely CC, CG and GG. The allelic and genotypic frequency was calculated. The observed genotypic frequency at the SNP site C-2161G was CC (0.61), CG (0.160) and GG (0.23) and the frequency of allele was 0.69 for C and 0.31 for G. The egg production was shown to be statistically similar for the genotypes of SNP C-2161G.

Abstract A total of 142 chicken blood samples were collected and a specific primer set was used to amplify a fragment of growth hormone locus using PCR. PCR products were digested with SacI and MspI restriction endonucleases. The amplified fragment digested with SacI enzyme revealed two “+” (wild type) and “-” (normal type) alleles with the frequency of 0.898 and 0.102, respectively. The amplified fragment digested with MspI enzyme revealed three A, B and C alleles with the frequency of 0.599, 0.102, and 0.299, respectively. Frequencies of +/+, +/- and -/- were 0.817, 0.162, and 0.021, respectively, and those of AA, AB, AC, BB, BC, and CC were 0.338, 0.113, 0.409, 0.007, 0.070, and 0.063, respectively, in the studied population. The results of 2 and likelihood ratio tests showed that this population was at Hardy-Weinberg equilibrium with respect to the marker locus. Marker-trait association analysis revealed statistically significant differences between “SacI-RFLP” genotypes for egg production and rate of laying eggs. The relationship between the molecular marker and these traits can be useful to improve the chicken breeding programmes.

Abstract The study evaluated the hematological parameters and antibody titers against Newcastle Disease (ND) and Avian Influenza (AI) on native chickens under extensive and semi-intensive rearing in Central Java, Indonesia. The target of the study was native chickens in Banyumas and Kebumen areas. The chickens were reared extensive and semi-intensively. The survey method was carried out by drawing 324 chicken blood samples for hematological tests and measuring antibody titer to ND and AI. The data obtained were analyzed using a general linear model (GLM) with the Systat ver.13 program. The results showed that native chicken in extensive and semi-intensive systems had different physiological statuses (P<0.05). The number of erythrocytes, hemoglobin levels, hematocrit values, total plasma protein levels, and heterophil-lymphocyte ratios was higher on semi-intensive than extensive system. However, the number of leucocytes was relatively high the same (P>0.05). In extensive system rearing, the number of native chickens with effective antibody titers against ND and AI was higher than the semi-intensive. The study concluded that native chickens reared in semi-intensive systems had a healthier physiological status than chickens in extensive systems. However, the effective antibody titers against ND and AI in the extensive and semi-intensive systems were still very low.

Insuline-like growth factor binding protein 2 (IGFBP2) is one of the principal binding proteins that has biological functions involved in growth, development, and differentiation. Selection for rapid growth based on molecular Marker Assisted Selection is required to increase production performance. The present study was designed to analyze the associations of IGFBP2 gene polymorphisms with chicken growth and body composition traits. Kampong chicken, a native chicken in Indonesia, is slow-growing chicken. A total of 59 males were used in the current study. Growth and body composition were measured in 24 wk of age. Primers for intron 2 region were designed from genomic chicken sequence. A c.1032C>T SNP of the IGFBP2 gene intron 2 region was detected and PCR-RFLP method was then used to genotype Kampong chicken population. The result showed that IGFBP2 polymorphism was significantly associated with body, carcass, breast, breast muscle, pectoralis minor, leg, and wings weight in Kampong chicken population (P < 0.05). The research suggests that the IGFBP2 gene could be a candidate gene that affects growth and body composition traits in chicken.

Inbred lines generated from the native and exotic chickens were compared with their F1 and backcross populations for a range of short term egg production traits. The experimental birds were brooded for 8 weeks and raised on a deep litter pens from 8 to 40 weeks of age. Age at first eggs were longer in the crossbred groups and the delay was mostly caused by the dominant genes from the exotic parents. Highly significant (P>0.01) heterosis were obtained by the crossbred groups in hen day rate and total egg mass respectively. The heterosis observed in the crossbred groups for egg weight was low and mostly non significant. the backcross groups recorded residual heterosis in egg production traits ( hen day rate, total egg mass and egg weight) and the magnitude was higher in the exotic than the native backcrosses. The results further indicate that the egg production performance of the crossbred groups were mostly influenced by maternal, sex linked, dominance and epistatic gene effects. It was suggested that the egg production potential of the native chicken could be improved further by cross mating-selected native and exotic backcross groups. This should be followed by crisscrossing.

Background: Broodiness is a sex linked behavioural trait observed in most of the domestic fowls and it’s also known as incubation behaviour. Prolactin (PRL) is the principal gene which plays a crucial role to the onset and maintenance of broodiness in birds. The present study was aimed at identification of 24bp (insertion-deletion) indel polymorphism at the promoter region of prolactin gene and its association with broodiness in Telllicherry native chicken population. Methods: A total number of 200 birds of Tellicherry native chicken were randomly selected from All India Co-ordinated Research Project (AICRP) on poultry improvement, Mannuthy, Thrissur, Kerala. Blood samples were collected from the wing vein under aseptic condition and isolation of Genomic DNA was done. Isolated DNA samples were subjected to polymerase chain reaction (PCR) using specific set of forward and reverse primer to detect a 24bp indel polymorphism in the prolactin gene. PCR amplicons were subjected to further molecular analysis. Result: According to the polymorphic patterns birds were categorized to three different genotypes viz., II (insertion-insertion), ID (insertion-deletion) and DD (deletion-deletion). The genotype and allele frequency was calculated and the frequency of I allele (0.6975) was found to be higher than D allele (0.3025). Results of this study suggest that the incubation behaviour could be eliminated in Tellicherry native chicken population by increasing the frequency of I allele upon selective breeding which may enhance the egg production. Therefore, this 24bp indel polymorphism could be used as a molecular marker in poultry breeding.

Central Java is the region that produces the second largest duck egg production in Indonesia or reaches 13.55% of the total national duck egg production. Therefore, duck farming in Central Java requires an economic evaluation to identify the success of the business. The economic analysis in the duck farm industry will update the latest statistics on production costs, outputs and profitability. This study aims to assess the analysis of laying duck business, which includes: marginal production costs, total net income and input-output ratio of duck egg production in various regions in Central Java, Indonesia. The study was conducted using a survey method using multi-stage sampling involving 120 duck farmers from the regency of Tegal, Brebes, and Pemalang. Each district was taken 40 farms as respondents by purposive random sampling with criteria for the number of breeders who had a minimum duck population of 100 ducks and a production period of 8-12 months. The analytical tools used include descriptive statistics and farm budgetary analysis. This study showed that duck breeders incur large costs to obtain production inputs, especially from feed aspects. Although three regencies reported profit based on the estimated input-output ratios, it is only a marginal profit that amounted to1: 1.382, 1:1.658, and 1:1.433 in Tegal, Brebes, and Pemalang, respectively. The conclusion of the study is the performance of duck farms in Central Java are still profitable even in the smallholder duck farm industry, with the highest profits obtained by farmers in Brebes Regency.

The present study was conducted to assess the effect of supplementation of Multi-enzymes with lysophospholipids on production performance of pure line White Leghorn layers. Body weights before and after the experiment did not differ significantly across the experimental diets. Irrespective of the dietary treatments, the birds gained 3.83 per cent of live weight relative to its initial body weight. Mean per cent Hen housed egg production was significantly (Pis less than 0.05) higher in diet supplemented with 0.10 MEC-L than other groups. Mean egg weight and average daily feed consumption during 25 to 35 weeks of age indicated no significant effect of enzyme supplementation. Average daily feed consumption per bird in control, 0.05 % and 0.1% multi-enzyme supplemented groups was 108.13, 105.66 and 107.67 g respectively and birds offered control diet recorded numerically more feed intake than enzyme supplemented groups. Comparatively low feed per egg was observed in 0.10 per cent group followed by 0.05 per cent group which offers economic benefits than control diets. However, the egg quality traits between different dietary enzyme supplementation groups showed no significant difference.

This study was to investigate the effects of Bacillus subtilis on production performance and bone pathophysiological characteristics of layers. Twenty-four 48-week-old Lohmann Pink-shell laying hens were randomly divided into two groups: a basic diet (control) and the basic diet mixed with Bacillus subtilis (0.5 g/kg) for a 60-day trial. Statistically, independent-sample t-test was used to assess the treatment differences. The results showed that Bacillus subtilis supplementation improved the percent of marketable eggs (p < 0.05) with reduced numbers of broken and soft-shelled eggs but had no effects on egg weight, height of albumen, yolk color, and Haugh unit (p > 0.05). Bacillus subtilis supplement also elevated maximum load (p = 0.06), maximum stress (p = 0.01), stiffness (p < 0.01), and Young’s modulus (p < 0.01) but suppressed maximum strain (p = 0.06) in the femur. In addition, compared with control birds, phosphorous concentration (p < 0.01) was reduced in serum at day 61 but increased in the femur (p < 0.05) in Bacillus subtilis fed birds. Bacillus subtilis fed birds also had lower magnesium concentrations in both femur (p = 0.04) and feces (p = 0.09). Furthermore, Bacillus subtilis increased plasma estrogen concentration (p = 0.01) and femur TNF receptor superfamily member 11b (OPG) expression (p < 0.05) but reduced plasma IL-1 (p < 0.01) and TNF-α (p < 0.01) concentrations. These results indicate that Bacillus subtilis could be used as a health promotor to reduce overproduction-induced inflammation and associated bone damage and to increase marketable egg production. The data provide evidence for developing a management strategy to use Bacillus subtilis as a feed additive to improve marketable egg production and health and welfare status of laying hens.
