Abstract Selenium (Se), a nutritionally essential mineral for human and animals, has a significant antagonistic effect on heavy metal cadmium (Cd) biotoxicity. Still, the impact of different Se source on alleviating Cd toxicity has received only limited attention. Therefore, the purpose of the current study was to assess the mitigation level of Cd-induced cardiotoxicity by different sources such as nanoparticles of selenium (Nano-Se), selenium-rich yeast (SY) and sodium selenite (SS). Two hundred of male, 1-day old Hy-Line Variety White chickens were randomly, equally divided into five groups (n = 25). These chicken groups were assigned to (1) basal diet (Con), (2) 140 mg/kg CdCl[2] of basal diet Cd (Cd), (3) 1 mg/kg Nano-Se plus 140 mg/kg CdCl[2] of basal diet (Nano-Se + Cd), (4) 3 mg/kg selenium-enriched yeast plus 140 mg/kg CdCl[2] of basal diet (SY + Cd group), and (5) 3 mg/kg sodium selenite plus 140 mg/kg CdCl[2] of basal diet (SS+ Cd group). The results evidenced that presence of Cd led to a significant increase in biochemical parameters such as lactate dehydrogenase (LDH) and creatine kinase (CK), as well as histopathological lesions in the heart of chickens. Cd exposure also resulted in more extensive effects on phase I metabolism enzymes and transcript CYP isoforms, elevated the levels of MDA and H[2]O[2] and depressed total superoxide dismutase (T-SOD), Cu-Zn SOD, total antioxidant capacity (T-AOC) and catalase (CAT) activities. The expression of nuclear receptors (NRs), AHR, CAR and PXR was declined, down-regulated Nrf2 and its downstream targets in the Cd-treat group. Notably, Se sources application alleviated Cd toxicity by triggering AHR/CAR/PXR/Nrf2 signaling pathway to promote restoring antioxidant defense system and phase I metabolism enzymes system. However, when compared the effectiveness of antagonism, the Nano-Se was superior in relieving Cd-induced cardiotoxicity via AHR/CAR/PXR/Nrf2 pathway activation than other Se- sources.

Intestinal infections are a major cause of morbidity and mortality in humans and agricultural animals, especially newborns and weaned animals. Preventive treatments that help weaned animals maintain homeostasis and balance the hindgut microbial populations are desirable. The present study aimed to explore the impact of bacitracin methylene disalicylate (BMD) on the intestinal health by analyzing the intestinal environment, morphology, expression of peptidoglycan recognition proteins (PGRPs), and flora of weaned rabbits. A total of 300 New Zealand weaned rabbits were randomly divided into the following five treatment groups for a 35-day feed trial: control group (basal diet), bacitracin zinc (BZ) group (50 mg/kg BZ), BMDa group (100 mg/kg BMD), BMDb group (50 mg/kg BMD), and BMDc group (rabbits fed a basal diet supplemented with 25 mg/kg BMD). In each treatment group, 28 rabbits were slaughtered for experimental analysis. The results showed that the supplementation of BMD increased the environmental acidity of the cecum of the weaned rabbits and reduced the ammonia-nitrogen concentration, which was beneficial to the survival of useful bacteria in the intestine. The morphology analysis of the duodenum using hematoxylin and eosin staining revealed that the villus length, villus/crypt ratio, and intestinal wall thickness increased in the BMD group, thereby improving the structure of the duodenum and the absorption capacity of the small intestine. Moreover, real-time polymerase chain reaction test showed that PGRPs (especially PGLYRP-1 and PGLYRP-2) in the intestinal had an antagonistic effect with BMD in the process of inhibiting pathogenic bacteria, resulting in their decreased expression (P < 0.05). Furthermore, through 16S rRNA sequencing in the cecal content, the abundance of the predominant phyla in the BMDa and BZ groups was found to be the closest. The abundance of the genera Lachnospira, Erysipelotrichaceae (p-75-a5), Paraprevotellaceae (YRC22), Mogibacterium, Peptococcaceae (rc4-4), Anaerovibrio, Succinivibrio, and Sphaerochaeta increased in the BMDa and BZ groups (P < 0.05). The relative abundance of Alistipes, Sedimentibacter, and Dorea significantly increased only in the BMDa group (P < 0.05). Conclusively, BMD, as well as microbes, improved the intestinal environment and structure to maintain the intestinal health of weaned rabbits.

Context Selenium (Se) is an essential trace element and plays pivotal roles in poultry nutrition. Aims The present study was designed to compare the impact of dietary supplementation of different sources of Se on growth performance, Se concentration of breast meat, and immune response of broiler chickens. Methods In total, 300 1-day-old as hatched broilers were randomly allocated to six dietary treatments, with five replicates of 10 birds per each. The experimental diets were as follows: (1) corn–soybean meal-based diet supplemented with 0.30 mg/kg of sodium selenite, (2) basal diet supplemented with 0.30 mg/kg of Se-enriched yeast, and (3–6) basal diet supplemented with 0.15, 0.30, 0.90 and 1.50 mg/kg of nano-Se respectively. Humoral immunity was assessed by antibody titer against a 5% sheep red blood-cell suspension and cellular immunity was measured by administration of phytohaemagglutinin-p at 38 days. Key results Results indicated that during the periods from 1 to 10 days, from 11 to 24 days, from 25 to 42 days and from 1 to 42 days, average daily gain, average daily feed intake, and feed conversion ratio did not differ among the treatments (P > 0.05). Assessment of orthogonal contrasts at the whole phase of feeding showed that the average daily gain in the broilers fed organic Se was significantly (P < 0.05) higher than that in those fed inorganic Se, as well as nano-Se in comparison to organic Se; such effects were not observed in earlier feeding stages. Se supplementation significantly increased the Se concentration of breast muscle (P < 0.0001). The birds that received 1.50 mg/kg of diet nano-Se showed higher (P < 0.05) total immunoglobulin and IgG titers in primary and secondary immune responses against sheep erythrocytes respectively. Toe web swelling after 24 h and 48 h of receiving phytohaemagglutinin-p was not affected by Se supplementation (P > 0.05). Conclusions In conclusion, the addition of nano-Se gave better results than did inorganic (sodium selenite) and organic (yeast selenium) forms of Se in performance traits, breast meat Se concentration and antibody response of broilers. Implications Novel elemental nanometer particulates, including nano-Se, exhibit new characteristics and a different mode of actions in comparison with organic and inorganic forms of Se sources in poultry diets. Inclusion of the nano form of Se in broiler diets and study of productive performance, immunity responses and meat quality leads to improve balanced broiler diets formulation in view of Se.

Abstract BackgroundGraphene Oxide (GO) is a promising candidate for nanomedicine applications. Due to the central role of liver in biotransformation of xenobiotics and drugs, the impact of GO on hepatic functional cells represents a crucial evaluation step for its potential implementation as drug. Primary human hepatocytes (PHH) are the election model for studying drug toxicity and metabolism, however current technical limitations may slow down the large-scale diffusion of this cellular tool in in vitro investigations. To assess the potential hepatotoxicity of GO, in this study, we propose an alternative approach employing second-generation upcyte® hepatocytes as cell model, which show metabolic and functional profiles akin to PHH. Cells have been acutely exposed to increasing GO concentrations for 24 hours. Upon sub-lethal concentrations of GO, stress-related cell responses to GO (such as apoptosis, oxidative stress and inflammatory response) have been evaluated, along with a broad investigation of GO impact on specialized hepatic functions.ResultsResults show an IC50 equal to 102.2 μg/mL, which is in line with recent data obtained by hepatocellular carcinoma-derived cells. However, at sub-lethal doses (≤ 80 μg/mL), it is detected a mild activation of early apoptosis, but not oxidative stress or inflammatory response. Importantly, we observed a clear impact of GO on phase-I drug metabolism enzymes (e.g., CYP3A4, CYP2C9) through the inhibition of gene expression and metabolic activity. Conversely, phase-II enzyme system and phase-III efflux transporters were not affected by GO. Finally, GO strongly downregulated the gene expression of Albumin.ConclusionThe presented model of upcyte® hepatocytes appears to be feasible for the assessment of hepatotoxicity of nanomaterials, specifically showing that sub-lethal doses of GO have a negative impact on the specialized hepatic functions of these cells. The impairment of cytochrome P450 system along with the alteration of Albumin gene expression by GO may suggest potential detrimental consequences for human health, as for instance, an altered detoxification from xenobiotics and drugs.

Recent evidences suggest that gut microbiota plays an important role in regulating physiological and metabolic activities of pregnant sows, and β-carotene has a potentially positive effect on reproduction, but the impact of β-carotene on gut microbiota in pregnant sows remains unknown. This study aimed to explore the effect and mechanisms of β-carotene on the reproductive performance of sows from the aspect of gut microbiota. A total of 48 hybrid pregnant sows (Landrace × Yorkshire) with similar parity were randomly allocated into three groups (n = 16) and fed with a basal diet or a diet containing 30 or 90 mg/kg of β-carotene from day 90 of gestation until parturition. Dietary supplementation of 30 or 90 mg/kg β-carotene increased the number of live birth to 11.82 ± 1.54 and 12.29 ± 2.09, respectively, while the control group was 11.00 ± 1.41 (P = 0.201). Moreover, β-carotene increased significantly the serum nitric oxide (NO) level and glutathione peroxidase (GSH-Px) activity (P < 0.05). Characterization of fecal microbiota revealed that 90 mg/kg β-carotene increased the diversity of the gut flora (P < 0.05). In particular, β-carotene decreased the relative abundance of Firmicutes including Lachnospiraceae AC2044 group, Lachnospiraceae NK4B4 group and Ruminococcaceae UCG-008, but enriched Proteobacteria including Bilophila and Sutterella, and Actinobacteria including Corynebacterium and Corynebacterium 1 which are related to NO synthesis. These data demonstrated that dietary supplementation of β-carotene may increase antioxidant enzyme activity and NO, an important vasodilator to promote the neonatal blood circulation, through regulating gut microbiota in sows.

The present study evaluated the effects of dietary supplementation of spirulina on laying hens’ performances: Eggs’ physical, chemical, and sensorial qualities. A total of 45 Lohman White hens, 44 weeks of age, were randomized into 3 groups of 15 birds. Hens were given 120 g/d of a basal diet containing 0% (control), 1.5%, and 2.5% of spirulina for 6 weeks. Albumen height and consequently Haugh unit were significantly affected by dietary supplementation of spirulina (p < 0.05) and by weeks on diet (p < 0.05). This supplement did not affect (p > 0.05) egg yolk weight or height. However, spirulina increased egg yolk redness (a*) from 1.33 (C) to 12.67 (D1) and 16.19 (D2) and reduced (p < 0.05) the yellowness (b*) parameter from 62.1(C) to 58.17 (D1) and 55.87 (D2). Egg yolks from hens fed spirulina were darker, more red, and less yellow in color than egg yolks from hens fed the control-diet (p < 0.0001). However, spirulina did not affect (p > 0.05) egg yolks’ total cholesterol concentration. In conclusion, a significant enhancement of egg yolk color was found in response to spirulina supplementation. Further investigations are needed to evaluate the impact of spirulina on egg yolks’ fatty acids profile.

Background The potential for machine learning to disrupt the medical profession is the subject of ongoing debate within biomedical informatics. Objective This study aimed to explore psychiatrists’ opinions about the potential impact innovations in artificial intelligence and machine learning on psychiatric practice Methods In Spring 2019, we conducted a web-based survey of 791 psychiatrists from 22 countries worldwide. The survey measured opinions about the likelihood future technology would fully replace physicians in performing ten key psychiatric tasks. This study involved qualitative descriptive analysis of written responses (“comments”) to three open-ended questions in the survey. Results Comments were classified into four major categories in relation to the impact of future technology on: (1) patient-psychiatrist interactions; (2) the quality of patient medical care; (3) the profession of psychiatry; and (4) health systems. Overwhelmingly, psychiatrists were skeptical that technology could replace human empathy. Many predicted that ‘man and machine’ would increasingly collaborate in undertaking clinical decisions, with mixed opinions about the benefits and harms of such an arrangement. Participants were optimistic that technology might improve efficiencies and access to care, and reduce costs. Ethical and regulatory considerations received limited attention. Conclusions This study presents timely information on psychiatrists’ views about the scope of artificial intelligence and machine learning on psychiatric practice. Psychiatrists expressed divergent views about the value and impact of future technology with worrying omissions about practice guidelines, and ethical and regulatory issues.

Limited attention has been directed towards understanding the impact of social media in the public sector, particularly in local government organisations. Although social media offer substantial benefits and opportunities to local government, research into the impact of social media remains scant. To address this gap, the authors draw on the technology, organisation, and environment (TOE) framework and propose a model of the determinants of social media impact in local government. The model is tested with data collected via a survey with 173 Australian local government organisations using social media. Data were analysed using the partial least squares-structural equation modelling (PLS-SEM) technique. The results indicate that TOE factors including perceived benefits, perceived security risks, compatibility, and degree of formalisation are important predictors of social media impact in local government.

Purpose – The purpose of this paper is to explore the influence of personal values on students’ learning behaviours. Hong Kong construction students are often criticized, by both practitioners and educators, as rote learners. To improve students’ learning processes, extensive research on a variety of aspects of construction education has been conducted. However, limited attention has been paid to the influence of personal values on students’ learning behaviours. The impact of personal value factors on the learning approaches of Chinese construction students in Hong Kong is investigated. Design/methodology/approach – A questionnaire survey was conducted to ethically Chinese construction students in four universities in Hong Kong. A total of 820 questionnaires were distributed and 431 valid returns were collected. Findings – The survey identified six influential values (namely, personal integrity, conservatism, determination, discipline, interpersonal relationships, and achievement), and shows that students who emphasize the virtues of personal integrity, determination, and a positive attitude toward achievement are better able to handle their interpersonal relationships, which will eventually lead them to engage in deep learning. Originality/value – Work to develop appropriate value systems is thus recommended in freshmen courses, as is the use of cooperative learning approaches. Such value systems will have a long-term influence on the learning approaches of construction students.
