Antimicrobial activity of endophytic fungi associated with Orchidaceae in Brazil

The purpose of this study was to examine antimicrobial activity of endophytic fungi isolated from the leaves, stems, and roots of 54 species of Orchidaceae collected in a Brazilian tropical ecosystem. In total, 382 filamentous fungi and 13 yeast isolates were obtained and cultured to examine the production of crude extracts. Thirty-three percent of the isolates displayed antimicrobial activity against at least one target microorganism. The multivariate statistical analyses conducted indicate that the extracts of endophytic fungi isolated from leaves of terrestrial orchids in semideciduous forest were more active against Escherichia coli , whereas extracts of endophytic fungi from roots of rupicolous orchids collected in rock fields were more active against Candida krusei and Candida albicans . Among the fungi that were screened in the study, 22 isolates held their antimicrobial activities after replication and were therefore selected for assessment of the minimum inhibitory concentration (MIC), which ranged from 62.5 to 250 µg/mL and 7.8 to 250 µg/mL against bacteria and fungi, respectively. One isolate of Alternaria sp. and one isolate of Fusarium oxysporum presented the strongest antibacterial activity. Three Fusarium isolates, Epicoccum nigrum , and Sclerostagonospora opuntiae showed the greatest MIC values against the pathogenic yeasts. This study is the first survey investigating the bioactive potential of endophytic fungi associated with tropical Orchidaceae species present in Brazilian ecosystems.

AbstractAntimicrobial peptides (AMPs) are a class of short, usually positively charged polypeptides that exist in humans, animals, and plants. Considering the increasing number of drug-resistant pathogens, the antimicrobial activity of AMPs has attracted much attention. AMPs with broad-spectrum antimicrobial activity against many gram-positive bacteria, gram-negative bacteria, and fungi are an important defensive barrier against pathogens for many organisms. With continuing research, many other physiological functions of plant AMPs have been found in addition to their antimicrobial roles, such as regulating plant growth and development and treating many diseases with high efficacy. The potential applicability of plant AMPs in agricultural production, as food additives and disease treatments, has garnered much interest. This review focuses on the types of plant AMPs, their mechanisms of action, the parameters affecting the antimicrobial activities of AMPs, and their potential applications in agricultural production, the food industry, breeding industry, and medical field.

Active components from natural sources are the current focus in most pharmacological research to provide new therapeutic agents for clinical use. Essential oils from the Pinus species have been traditionally used in medicine. This study aimed to investigate the chemical profile of two Pinus species, Pinus halepensis L. and Pinus pinea Mill, from different altitudes in Libya and study the effect of environmental conditions on the biological activities of essential oils. A clevenger apparatus was used to prepare the essential oils by hydrodistillation. Analyses were done using GC/MS. Anthelmintic and antimicrobial activities were tested against the earthworm Allolobophora caliginosa, gram-positive bacteria, gram-negative bacteria, and fungi. Different chemical profiles were observed among all tested essential oils, and terpenes were the most dominant class. All studied essential oils from the Pinus species exhibited a remarkable anthelmintic activity compared to the standard piperazine citrate drug. Pinus halepensis from both altitudes showed broad-spectrum antimicrobial activity against all tested microorganisms, while Pinus pinea was effective against only Escherichia coli. From these findings, one can conclude that there are variations between studied species. The essential oil compositions are affected by environmental factors, which consequently affect the anthelmintic and antimicrobial activity.

The present work reports the <i>in vitro</i> antimicrobial activities of the essential oil and methanol extract from <i>Satureja hortensis</i> as well as the content of its essential oil. The chemical composition of hydrodistilled essential oil of Satureja hortensis was analysed by means of GC-MS. Thirty constituents were identified. The main constituents of the oil were thymol (40.54%), &gamma;-terpinene (18.56%), carvacrol (13.98%), and <i>p</i>-cymene (8.97). The essential oil of <i>Satureja hortensis</i> exhibited the activity against 25 bacteria, 8 fungi, and a yeast, <i>C. albicans</i>; exerting the Minimum Inhibitory Concentration values (MIC) ranging from 15.62 to 250 &micro;l/ml. Similarly, methanol extract of the plant also showed antimicrobial activity.

In this study, thein vitroantimicrobial activities of four plant essential oils (T. schimperi,E. globulus,R. officinalis, andM. Chamomilla) were evaluated against bacteria and fungi. The studies were carried out using agar diffusion method for screening the most effective essential oils and agar dilution to determine minimum inhibitory concentration of the essential oils. Results of this study revealed that essential oils ofT. schimperi,E. globulus, andR. officinaliswere active against bacteria and some fungi. The antimicrobial effect ofM. chamomillawas found to be weaker and did not show any antimicrobial activity. The minimum inhibitory concentration values ofT. schimperiwere<15.75 mg/mL for most of the bacteria and fungi used in this study. The minimum inhibitory concentration values of the other essential oils were in the range of 15.75–36.33 mg/mL against tested bacteria. This study highlighted the antimicrobial activity of the essential oil ofE. globulus,M. chamomilla,T. Schimperi, andR. officinalis. The results indicated thatT. schimperihave shown strong antimicrobial activity which could be potential candidates for preparation of antimicrobial drug preparation.

The major aim of this study was to determine the antimicrobial activity of the extracts of eight plant species which are endemic in Iran. The antimicrobial activities of the extracts of eight Iranian traditional plants, including Hypericum scabrum, Myrtus communis, Pistachia atlantica, Arnebia euchroma, Salvia hydrangea, Satureja bachtiarica, Thymus daenensis and Kelussia odoratissima, were investigated against Escherichia coli O157:H7, Bacillus cereus, Listeria monocytogenes and Candida albicans by agar disc diffusion and serial dilution assays. Most of the extracts showed a relatively high antimicrobial activity against all the tested bacteria and fungi. Of the plants studied, the most active extracts were those obtained from the essential oils of M. communis and T. daenensis. The MIC values for active extract and essential oil ranged between 0.039 and 10 mg/ml. It can be said that the extract and essential oil of some medicinal plants could be used as natural antimicrobial agents in food preservation. .

Costus spiralis(Costaceae) is a species native to the Amazon region and is used in traditional medicine. The endophytic fungi used in this study were obtained from leaves of this plant. 13 strains were selected to obtain hydroethanolic extracts and were submitted to hydroalcoholic extraction and evaluated for antioxidant activity by DPPH (2,2-difenil-1-picrilhidrazil) and FRAP (ferric reducing antioxidant power), and all of the fungi had positive results. The antimicrobial action of crude extracts had a good range of activities. All extracts had inhibitory activities against the yeasts ofCandida albicansandC. parapsilosis, with 125 to 500 μg/mL MIC. Eight extracts had antimicrobial activities againstBacillus subtilis(MIC from 62.4 to 125 μg/mL), 5 againstPseudomonas aeruginosa(MIC from 125 to 500μg/mL), 2 againstSalmonella enterica(MIC from 125 to 62.5 μg/mL), and 2 againstEnterococcus faecalis(MIC from 500 to 125 μg/mL). The presence of secondary metabolites, including coumarins, was observed during chemical evaluation by thin layer chromatography. Total phenol content was estimated, and a strong positive correlation to antioxidant activity was observed, according to its Pearson coefficient. This is the first report of the bioactive potential of endophytic fungi isolated from the Costaceae family in Brazilian ecosystems.

The fungal endophyte community associated with Baccharis trimera, a Brazilian medicinal plant, was characterized and screened for its ability to present antimicrobial activity. By using molecular methods, we identified and classified the endophytic fungi obtained into 25 different taxa from the phyla Ascomycota and Basidiomycota. The most abundant species were closely related to Diaporthe phaseolorum, Pestalotiopsis sp. 1, and Preussia pseudominima. The differences observed in endophytic assemblages from different B. trimera specimens might be associated with their crude extract activities. Plants that had higher α-biodiversity were also those that contributed more to the regional (γ) diversity. All fungal isolates were cultured and their crude extracts screened to examine the antimicrobial activities. Twenty-three extracts (12.8%) displayed antimicrobial activities against at least one target microorganism. Among these extracts, those obtained from Epicoccum sp., Pestalotiopsis sp. 1, Cochliobolus lunatus, and Nigrospora sp. presented the best minimum inhibitory concentration values. Our results show that the endophytic fungal community associated with the medicinal plant B. trimera included few dominant bioactive taxa, which may represent sources of compounds with antifungal activity. Additionally, the discovery of these bioactive fungi in association with B. trimera suggests that Brazilian plants used as folk medicine may shelter a rich fungal diversity as well as taxa able to produce bioactive metabolites with antimicrobial activities.

Endophytic fungi have been emerged as fruitful resources for producing structurally fascinating and biologically active secondary metabolites. However, endophytic fungi from medicinal plants of Qinling Mountains–the most important natural climatic boundary between the subtropical and warm temperate zones of China with an astonishingly high level of biodiversity–have rarely been explored as potential sources of novel fungal species and active secondary metabolites. In this study, a total of 371 fungal colonies were successfully isolated from 510 tissue segments of the medicinal Tupistra chinensis Baker collected from Qinling Mountains, China. Roots of T. chinensis Baker are used as a folk medicine to ameliorate pharyngitis and treat rheumatic diseases. A total of 100 representative morphotype strains were identified according to ITS rDNA sequence analyses and were grouped into three phyla (Ascomycota, Basidiomycota, Mucoromycota), seven classes (Dothideomycetes, Sordariomycetes, Eurotiomycetes, Microbotryomycetes, Agaricomycetes, Leotiomycetes, Mortierellomycetes), and at least 35 genera. The genera of Collectotrichum (IF, 29.92%), Fusarium (IF, 8.36%), Aspergillus (IF, 8.09%), and Dactylonectria (IF, 5.39%) were most frequently isolated from the tissues of T. chinensis Baker. The Species Richness Index (S, 65) and the Shannon-Wiener Index (H′, 3.7914) indicated that T. chinensis Baker harbored abundant fungal resources. Moreover, five isolates were potential new taxa because of low similarity of ITS sequences ranged from 95.09%∼96.61%. Fifteen out of 100 endophytic fungal ethyl acetate extracts exhibited inhibitory activities against at least one pathogenic bacterium or fungus. Two important lead compounds produced by two stains (F8047 and F8075) with high antimicrobial activities were identified using high performance liquid chromatography (HPLC) and ultra-performance liquid chromatography-quadrupole-time of flight mass spectrometry (UPLC–QTOF MS) analyses. In addition, it was noteworthy that the strain F8001, which may be a potential new species, showed antimicrobial activity and should be investigated further. Overall, these results indicated that the endophytic fungi from T. chinensis Baker could be exploited as a novel source of bioactive compounds.

Abstract One of hidden mine of antibiotics is endophytic fungi especially that inhibited medicinal plants. In this regard, leaves, stems, fruits and bulbs of some commonly medicinal plants growing in Northwestern coast of Egypt were subjected for isolation of endophytic fungi with screening study of its antimicrobial activity. Practically, more than one hundred (101) endophytic fungal species isolated from Scorpiurus muricatus, Mellilotus indicus, Lotus polyphyllos, Ononis vaginalis, Nicotiana glauca, Lycium europaeum, Asphodelus aestivus, Echium angustifolium,Fagonia cretica, Pancratium maritimum,and Carduus getulus were tested in vitro for their antimicrobial activities against E. coli, Pseudomonas argenosa, Staphylococcus aureus, Bacillus subtilus, Candida albicanus, Candida glabrata, Penicillium expansum, Aspergillus flavus. The values of applied diversity indices revealed significant differences in presence, absence and abundance among endophytic fungal isolates. To the best of our knowledge, the present study is the first to report of Alternaria pluriseptata as endophytic species with most dominant and most active in its anitimicrobial activity among the isolated species. Seven distinctive groups were revealed from the two-way cluster analysis showing the intensity of antimicrobial activity against tested pathogens: Twenty-five percent of the isolates (26 strains assembled together in group V) exhibited no antimicrobial activity against all tested pathogens while six percent (6 isolates) assembled in group VII revealed high antimicrobial activity against five pathogens.

Recently, many scientists and polymer engineers have been working on eco-friendly materials for starch-based food packaging purposes, which are based on biopolymers, due to the health and environmental issues caused by the non-biodegradable food packaging. However, to maintain food freshness and quality, it is necessary to choose the correct materials and packaging technologies. On the other hand, the starch-based film’s biggest flaws are high permeability to water vapor transfer and the ease of spoilage by bacteria and fungi. One of the several possibilities that are being extensively studied is the incorporation of essential oils (EOs) into the packaging material. The EOs used in food packaging films actively prevent inhibition of bacteria and fungi and have a positive effect on food storage. This work intended to present their mechanical and barrier properties, as well as the antimicrobial activity of anti-microbacterial agent reinforced starch composites for extending product shelf life. A better inhibition of zone of antimicrobial activity was observed with higher content of essential oil. Besides that, the mechanical properties of starch-based polymer was slightly decreased for tensile strength as the increasing of essential oil while elongation at break was increased. The increasing of essential oil would cause the reduction of the cohesion forces of polymer chain, creating heterogeneous matrix and subsequently lowering the tensile strength and increasing the elongation (E%) of the films. The present review demonstrated that the use of essential oil represents an interesting alternative for the production of active packaging and for the development of eco-friendly technologies.
