Experimental cardiac necrosis in hypobaric and anemic hypoxia

Resistance to isoproterenol-induced cardiac necrosis (IPRO) was compared in rats exposed to two types of hypoxia (i.e., hypobaric and anemic). IPRO was induced by two consecutive, subcutaneous injections of isoproterenol (80 mg/kg) at 24-h intervals. The animals were killed on the third day and the severity of the lesion was evaluated on a 0 (no damage) to 4 (severely damaged) scale. White male rats (HA) were exposed in a barometric chamber to a simulated altitude of 7,000 m (307 mmHg) for 4 h/day for 24 days. Two groups of control rats were kept at sea level; one group (SLA) was the same age and one group (SLW) was the same weight as the altitude-exposed rats. The HA rats were significantly more resistant to IPRO with a mean necrogenic rating of 1.8 compared to 3.3 for the SLA and SLW rats. Infant rats (AA) were made anemic by feeding full-cream milk and glucose for 100 days after weaning. Two groups of control animals were fed a standard laboratory diet; one group (AC) was the same age and one group (AW) was the same weight as the AA rats. There was no significant difference in the necrogenic ratings of the AA (3.3), AC (3.5), or WC (3.7) hearts. Thus, hypobaric hypoxia affords some protection against IPRO which is not afforded by anemic hypoxia. Similarities and differences in the two hypoxias are discussed.

Background: The objective of this study was to evaluate the efficacy of subacromial injections of collagenase and corticosteroid in rats with experimentally induced adhesive capsulitis. Method: Thirty adult Wistar albino male rats were distributed into 3 groups of 10 rats each after stabilization of their shoulders for 3 weeks: the first group received a single dose of 0.002 mg (0.25 mL) subacromial collagenase; the second group received a single dose of 1.60 mg (0.25 mL) subacromial steroid, and the third group received a single dose of 0.25 mL subacromial saline solution. One week later, we investigated shoulder range of motions, collagen content of the shoulder, and joint cartilage structure. Results: There was no statistically significant difference in the cartilage damage between the groups (p > 0.05). Fibrosis measurements were significantly lower in the collagenase group than in the steroid and saline groups. There was no significant difference in fibrosis between the steroid and saline groups (p > 0.05). Abduction measurements were significantly higher in the collagenase group than in the steroid and saline groups (p < 0.001). No significant difference in the abduction measurements was observed between the saline and steroid groups (p > 0.05). Conclusion: We observed that subacromial injections of collagenase Clostridium histolyticum effectively treated adhesive capsulitis. The results suggest that this treatment could be considered for use in patients with an intact rotator cuff.

<p>Anemia is still a high prevalence disease in Indonesia, caused by a lack of iron in the body. The components from brown seaweed such as cobalamin (vitamin B12), pyridoxine (vitamin B6), chlorophyll and iron (Fe) can increase red blood cells in the body. The purpose of this research was to determine the effect of brown seaweed (<em>Sargassum duplicatum</em>) which can increase hematocrit levels in the blood of white male wistar strain rat (<em>Rattus norvegicus</em>) suffered from anemia in induction NaNO<sub>2</sub>.</p><p>            This research uses an experimental laboratory method with a post test only control group design. The samples were 30 male rats divided into 3 treatment groups, the negative control group (K<sub>0</sub>) without treatment, the positive control (K<sub>1</sub>) be treated anemia given NaNO<sub>2</sub> for 18 days without <em>Sargassum duplicatum </em>extract and treatment group (K<sub>2</sub>) treated with anemia given NaNO<sub>2</sub> for 18 days without <em>Sargassum duplicatum </em>extract at a dose of 2,45gr / kgBB. On the 40<sup>th</sup> day, a blood sample was taken, then measured the hematocrit level.</p><p>            The results showed that the mean number of blood hematocrit levels in K<sub>2</sub> was higher than K<sub>1</sub>, it showed a significant difference descriptively. Based on the Kruskal-Wallis test, a value of 0.260 was obtained that p &gt; 0,05 showed that there were no statistically significant differences between groups of experimental animals.</p><p>            From the results of this research, the conclusion is the administration of brown seaweed extract (<em>Sargassum duplicatum</em>) descriptively increased, but not statistically significant.</p><p> </p><p><strong>Keywords :</strong> Anemia, hematocrit, <em>Sargassum duplicatum</em>, NaNO<sub>2</sub></p>

According to the World Health Organization, cardiovascular diseases and diabetes mellitus occupy a significant niche in the structure of diseases with high disability and mortality impact and pose major healthcare and social issues. It should be stressed that 50-80% of fatal cases in patients with diabetes are associated with cardiovascular diseases. The purpose of this study is to determine the histostructural characteristics and to perform morphometric analysis of the components of the aortic wall and its hemomicrocirculatory bed of the aorta in 6 and 8 weeks of streptozotocin-induced diabetes mellitus. The material for the histological study included the sections of the wall of the ascending aorta, the aortic arch, and the descending aorta taken from 26 sexually mature white male rats weighing 100 - 160 g. For morphometric examination, a series of photos of the aortic wall was taken using a Meiji MT4300 LED microscope with an x40 objective, x10 ocular. The measurements were carried out using the Image J software. The development of micro - and macroangiopathies in experimental animals with 8-week streptozotocin-induced diabetes mellitus was histologically confirmed. Statistical analysis revealed a significant difference in all morphometric parameters of the components of the aortic wall and the vessels of its hemomicrocirculatory bed after 8 weeks of experimental diabetes as compared with the normal values, control values, and in values obtained at the 6-week period of the experiment. The study has demonstrated clear dependence between the severity of destructive changes in the aortic wall and sections of its hemomicrocirculatory bed and the duration of the experiment.

Background. Atherosclerotic cardiovascular diseases, as well as coronary heart disease, cerebrovascular disease, and peripheral artery disease (probably caused by atherosclerosis), are the leading cause of disability and mortality in people with diabetes. Objective. An explicit dependency of the severity of destructive changes in the wall of the aorta and links of its hemomicrocirculatory bed of vessels on the duration of the experiment was determined.

Objective(s):  Methylenedioxymethamphetamine (MDMA) is a hallucinogenic drug of abuse which is the most popular drugs in the world and has been shown to induce apoptosis in kidney cells. As Pentoxifylline (PTX) increases cAMP and reduces tumor necrosis factor-α, the present study aimed to investigate the effects of pentoxifylline on kidney damage induced by acute administration of MDMA in male rat.Materials & methods: Thirty male rats (250-300 g) were randomly divided into five groups: including control (without any intervention), MDMA group ,the group received 7.5 mg/kg MDMA three times at every two hours for one day ,first experimental group, received 100 mg/kg PTX a week before MDMA administration, second experimental group received 100 mg/kg PTX Just in the time of the third injection of MDMA and the third experimental group received 100 mg/kg PTX followed by one dose of MDMA. Two weeks later, kidneys were removed and prepared for H&E staining, TUNEL and western blot techniques. Results: histopathological studies showed significantly decrease in the kidney cellular damage, in experimental group 1 compared to MDMA group. The number of TUNEL-positive cells was increased significantly in MDMA group. A significant difference was revealed in the mean number of TUNEL-positive cells between the rats treated with PTX before MDMA administration and MDMA group. Expression of active caspase-3 was significantly increased in the MDMA group. While the PTX treatment could significantly decrease when administrated before MDMA injections.Conclusion: Pentoxifylline can significantly reduce the severity of lesions in the kidney following administration of MDMA.

Ciplukan (Physalis angulata L.) empirically has been used to treat sore throat, inflammatory of the airways, gingivitis, and other diseases. This research was to determine the inflammatory activity of water herb extract of P. angulata L. on white male rats edema Wistar strain induced carragenan. P. angulata L. herbs were extracted using infundation method and were dried with frezee drying. Parameter that would be observed of this research was the inhibition foot edema of rat after induction of 0.1 mL of λ-carragenan 2% for 360 minutes. Measurement of edema volume was using pletismometer. The results of this research showed water herb extract of P. angulata L. in dose 100, 200, and 400 mg/kgBW has potensial as an antiinflammatory drug by percent respectively were 20.13; 28.93; and 34.70%. The three doses hasn’t showed any significant difference (p>0.05) with the positive control of diclofenac sodium dose 4.5 mg/kgBW was 33.90%. Conclusion of this research states water herb extract of P. angulata L. has antiinflammatory activity with effective dose of 400 mg/kgBW.

Hypertriglyceridemia is one of the risk factors for cardiovascular disease and acute pancreatitis (Pejic and Lee, 2006; Brahm and Hegele, 2013; Afari et al., 2015). One way to treat it by using the apple vinegar (Malus sylvestris Mill) of manalagi varieties which contained high antioxidants and acetic acid as well as more durable than the fresh fruit. The study aimed to know the effect of the Apple vinegar of manalagi varieties (Malus sylvestris Mill) to blood triglyceride levels of white male rats (Rattus norvegicus) wistar strain with high-fat diet and induced by alloxan. The study used the post test only control group design, with 24 rats samples which were divided into three groups. The negative control group (K0) only received standard diet for 24 days, the positive control group (K1) received a high-fat diet for 7 days and induced by alloxan with doses of 90 mg/kgBW of rats, and treatment group (K2) was given a high-fat diet for 7 days, induced by alloxan and received the apple vinegar (Malus sylvestris Mill) of manalagi varieties with a dose of 0,27 ml/100 g BW of rats for 14 days. The results by using one way ANOVA showed the significance value of the data p = 0,974 (p>0,05) so that there is no significant difference of the mean of blood triglycerides levels between K0, K1 and K2. This study concluded that apple vinegar (Malus sylvestris Mill) of manalagi varieties can not significantly lower the blood triglyceride levels of white male rats (Rattus norvegicus) wistar strain with high-fat diet and induced by alloxan. Anti-inflammatory activity test of ethanolic extract of banana leaf (Musa Paradisiaca L.) on carrageenan-induced paw edema in white male rats (Rattus novergicus L.) has been conducted. It was aimed to investigate and to determine the anti-inflammatory activity and its effective dose. The extract was prepared by maceration method using ethanol 96%. Anti-inflammatory activity test was performed in five different groups. Each group consisted of 5 rats. The 1st group (negative control) was given 0.5% CMC-Na suspension; the 2nd group (positive control) was given diclofenac sodium 9 mg/KgBW; the 3rd, 4th, and 5th groups were successively given the banana leaf extract as much as 500, 750 and 1000 mg/KgBW. Each rat was then induced by 1% carrageenan and tested using subplantar method.  The inflamed paw diameter was measured using a calliper while the inflamed paw volume using pletysmometer. The measurements were done for 6 hours long with intervals of 60 minutes. The data was statistically analyzed using ANOVA (analysis of variance). The results showed that the negative control had a significant difference with the other treatment groups which did not show any anti-inflammatory effect. In conclusion, ethanolic extract of banana leaf has effective anti-inflammatory activity at a dose of 750 mg/KgBW

Background. Over the last 20 years, the quality of men’s sperm around the world has deteriorated twice. The causes are poor environment, stress, sedentary lifestyle, prevalence of sexually transmitted infections, obesity, alcohol abuse and various stimulants. The structure and function of the testes, when exposed to various drugs and chemicals, attracts the attention of present researchers. Objectives. The aim of the research was to study the structural changes of the spermatogenic epithelium in testicular damage caused by rubomycin hydrochloride. Methods. The spermatogenic epithelium of the testes of 30 white adult white male rats, which were divided into 2 groups, was morphometrically examined. The 1st group comprised 15 experimental intact animals, the 2nd – 15 rats, in which testicular damage with rubomycin hydrochloride was simulated. Euthanasia of rats was performed by bloodletting under thiopental anesthesia one month after the experiment beginning. Quantitative morphological parameters were statistically processed. In spermatocytes of the 1st P-order, spermatogonia and spermatids of testes, their height, diameter of nuclei, nuclear-cytoplasmic ratio and relative volumes of damaged cells of spermatogenic epithelium were evaluated. Results. It was established that spermatogenic epithelial cells were damaged in the simulated experimental conditions, which was morphometrically confirmed by changes in nuclear-cytoplasmic relations in the 1st, 2nd order spermatocytes, spermatogonia and spermatids. The relative volumes of damaged spermatogenic epithelial cells also increased significantly. The most significant morphometric parameters were altered in spermatids. Thus, the nuclear-cytoplasmic ratio in the studied cells was statistically significantly (p <0.001) increased by 7.4% compared with the similar control morphometric parameter. The relative volume of damaged spermatids in these experimental conditions reached (32.50±0.18) %, which with a high statistically significant difference (p<0.001) exceeded the same control value in almost 15.8 times. Optically, histological preparations of the testes showed severe vascular disorders, characterized by dilation and plethora mostly of venous vessels, which pointed to violation of venous drainage of the studied organs. Conclusions. According to the attained research results it has been established that nuclear-cytoplasmic relations are an objective and valuable informative indicator of a functional condition of cells and their structural changes in pathological conditions. When rubomycin hydrochloride is exposed to experimental animals, the most significant changes in nuclear-cytoplasmic relations are found in spermatic spermatozoa.

Authors searched actions of Cymbopogon citratus-Stapf aquous extract, 2% concentration, on montility of smooth muscle. It were performed 10 (ten) experiments, each of them consisted of the motility record of a segment of duodenum from male Wistar rats maintained in a Tyrode solution water-bath to isolated organ. Physiological parameters (temperature, pH, oxigen supply) were controled to obtain the functional nature of organ motility. It were realized three records to each experiment in this sequency:           First record: control record (standard) from motility of duodenum smooth muscle in Tyrode bath.           Second record: after addition of 0,5 ml physiological solution before the third record, the bath liquid was entirely substituted and the organ was washed tith Tyrode solution.            Third record: after 15 (fifteen) minutes from the addition of Cymbopogon citratus-Stapf tea 2% concentration, it was add 0,5ml acetylcholine solution (11,8165 Mg in 50 ml physiological solution).It was verified that it shows a significant difference between 2nd and third record groups compared whit control records (first record groups) on smooth muscle motility of isolated duodenum from Wistar male rats.
