The present study was focused on the concentration-dependent changes in oral acute toxicity of leaf extracts of E. fluctuans in zebrafish. The study was also aimed at the details of histopathological changes in the gill, liver, brain, and intestine of zebrafish exposed to the leaf extracts of the plant E. fluctuans. Enydra fluctuans Lour is an edible semiaquatic herbaceous plant used widely for the alleviation of the different diseases. Since there were no toxicity studies conducted on this plant, the present study was an attempt to look into the elements of toxicity of the plants. Two types of experiments are conducted in the present study. First, the acute oral toxicity study was conducted as per the OECD guidelines 203. Second, histopathological changes were observed in the fishes exposed to the lethal concentrations of plant extract. The oral acute toxicity studies conducted on Zebrafish have revealed that the leave extracts of E. fluctuans were toxic to the tested fish at the concentration of 200 mg/kg body weight. The histopathological studies conducted on the intestine of treated fishes showed that treatment has induced rupturing of the villi structure and fusion of villi the membrane and detachment of the villi structure from the basal membrane of the intestine. The histology of the liver also showed severe vacuolization in the cells while it is not affected in control. The studies on gills showed the detachment of the basal epithelial membrane in the gills compared to control which might have led to death of the fish. The histopathological observations of brain tissues treated with test samples also revealed the marked impingement in the brain parenchyma while the control is normal without impingement of the brain.

Abstract Objectives Madhulai Manappagu – a well-known sastric and widely prescribed Siddha herbal syrup formulation indicated for treating Veluppu Noi (Anaemia especially Iron deficiency Anaemia) has been in day today practice in Tamil Nadu for a quite longer decades. The syrup is a herbal preparation which has a sweet pleasant odour and a palatable taste, contain the juice of pomegranate (Punica granatum L.) as the main ingredient. Though the formulation is a fruit juice, the safety profile of the syrup is not established and is being marketed without toxicological evaluation. The study is aimed at ascertaining the acute and sub-acute toxicity assessment of Madhulai Manappagu in Wistar Albino rats. Methods The acute and sub-acute (28day repeated oral) toxicity studies were performed as per the guidelines mentioned in the Organization for Economic Cooperation and Development (OECD) 423 (adopted on December 2001) and TG 407 (adopted on October 2008) with slight modifications respectively. For acute toxicity study, three female rats were randomly selected as control; three female rats were randomly selected and were administered a single dose of 5,000 mg/kg body weight per oral route. For sub-acute (28day repeated oral) toxicity studies, three doses of test drug MM of 500 mg/kg/day (low dose), 750 mg/kg/day (intermittent dose) and 1,000 mg/kg/day (high dose) were selected for administration. Both sexes of Wistar Albino rats were randomized into four groups of 10 animals each (five males, five females). Group I was kept as control group. Group II, III and IV served as low, intermittent and high doses of MM respectively. Animals were observed for mortality, morbidity, body weight changes, feed and water intake. Haematology, clinical biochemistry, electrolytes, gross pathology, relative organ weight and histopathological examination were performed. Results In the acute toxicity study, rats showed no toxicological signs on behavior, gross pathology and body weight of rats when treated with a single dose of 5,000 mg/kg body weight per oral route. In the subacute (28 days repeated oral) toxicity study, rats have showed no significant changes on behavior, gross pathology, body weight, and hematological and biochemical parameters when treated with Madhulai Manappagu in three different doses. Conclusions The toxicity studies which include both acute and 28 days repeated (subacute) oral toxicity studies, revealed no observed adverse effect level (NOAEL) of Madhulai Manappagu in animals. Thus the safety of the drug in human usage was ensured.

Clerodendrum is a genus of about 500 species belongs to the family Lamiaceae. Several species of this genus have been proved for the treatment of various diseases. Clerodendrum thomsoniae Balf.f were selected in this study; according to the literature available, there is no systematic toxicity studies for this plant were described. The current study was therefore carried out to evaluate the acute and sub-acute toxicity in mice and rats. The acute oral toxicity study was performed in mice following OECD guidelines 425, and the sub-acute toxicity was performed in male and female rats following OECD guidelines 407. The results showed that mice given a single dose of up to 2000 mg/kg orally did not show any toxicity signs or mortality. In the sub-acute toxicity analysis in rats, 3 specific daily doses of 150, 300, and 600 mg/kg for 28 days did not induce any major changes to the hematological and biochemical parameters. Histopathological studies revealed normal architecture that did not indicate any morphological disturbances. In our study, no deaths or any signs of toxicity were found in acute and subacute toxicity studies after oral administration according to OECD guidelines, which concluded that ethyl acetate extract of Clerodendrum thomsoniae Balf (EACT) could use for in vivo biological activity studies in laboratory animals to explore its various medicinal activity before study in human subjects.

This study was aimed to investigate the effect of high sub-chronic doses of the aqueous and methanol leaf extracts of Loranthus micranthus on biochemical parameters of albino rats. Acute toxicity studies were performed according to standard methods. The animals were divided into 5 groups (n = 5). Aqueous and methanol extracts of L. micranthus leaves were administered in doses of 1000 and 2000 mg/kg body weight to four groups of rats respectively for 30 days through the intraperitoneal route. The fifth group served as control and received saline (5 ml/kg b.w, i.p). Blood samples were collected by retrorbital puncture and analyzed for biochemical and haematological parameters using assay kits. Acute toxicity studies indicated that both extracts had an LD50 >           5000 mg/kg. The results indicated significant (p<0.001) increases in alkaline phosphatase serum levels in both extract treated groups. The extracts also produced significant elevation in serum bilirubin levels when compared with normal control (p<0.05). Both extracts did not affect the levels of alanine and aspartate transaminases significantly (p>0.05). There were significant increase in the serum levels of urea in the extracts treated rats (p<0.05; p<0.01). The 2000 mg/kg aqueous extract produced significant increases in mean serum chloride and bicarbonate levels of treated rats when compared with control (p<0.01). The extracts produced significant decrease in the serum creatine kinase levels of treated rats in a non-dose related manner when compared with control (p<0.05). There were no significant changes in sodium and potassium levels of treated rats. The methanol extract had no significant effect on the haematological indices studied. The aqueous extract produced significant reductions in the haemoglobin and PCV of treated rats (p< 0.01). The total and differential leucocyte counts were not affected by extract treatment (p>0.05). From these results, the extracts caused significant biochemical changes but were not cytotoxic to leucocyte cell lines. Therefore, there should be caution in the long term use of these extracts.

The aim of the study was to evaluate the acute oral and sub-acute toxicity of ethanolic root extract of Tetracera akara in Swiss albino mice and Wistar rats. Tetracera akara (Burm. f.) Merr. has been used as traditional medicine by the Kani tribe of Kerala to cure liver diseases. In acute toxicity studies, four groups of mice (n = 5/group/sex) were orally treated with doses of 0.625 g, 1.25 g, 2.5 g and 5.0 g/kg and mortality were recorded. In the sub-acute toxicity study, animals received T. akara extract at the doses of 0.1 g, 0.5 g and 2.5 g/kg/day (n = 5/group/sex) for 28 days, biochemical, hematological, morphological and histopathological parameters were determined. T. akara did not produce any mortality in the acute toxicity studies, showing LD50 higher than 5 g/kg. Sub-acute treatment with T. akara didn’t cause any changes in body weight gain, hematological, biochemical profiles when compared to normal control. In addition, no changes in morphological and histopathological aspect of organs were observed in the animals. Taking all factors into consideration, administration of Tetracera akara does not produce acute toxicity in Swiss albino mice or sub-acute toxicity in Wistar rats, suggesting it’s safe use by humans.

Typical reprographic toners consist of a thermoplastic polymer or polymers as the major component, a colorant or colorants (carbon black or color pigments), and small quantities of additives such as charge control and/or lubricating/release agents. Another type of toner contains iron oxides and polymers) as the major components. As a complement to the recently published Xerox chronic inhalation studies of toners, we are reporting the acute toxicity studies of some typical Xerox toners. The studies include acute oral toxicity in rats, acute dermal toxicity in rabbits, acute inhalation toxicity in rats, eye irritation in rabbits, skin irritation in rabbits, skin sensitization in guinea pigs, and the repeated-insult patch test in humans. These studies represent our acute toxicity testing using different protocols with various toners carried out during the period 1969–1984. In addition, we recently carried out acute dermal toxicity testing at 5 g/kg with two representative toners, for the purpose of classification of waste toners in the State of California. The test results consistently indicate that all toners were practically nontoxic: oral LD50 from <5 to <35 g/kg; dermal LD50 from <2 to <5 g/kg; and inhalation LC50 (4 h) from <0.17 to <10.2 g/m3. They were nonirritating to the eye and nonirritating/ nonsensitizing to the skin.

Ammi visnaga L (A.V.) is used in the traditional medicine for the treatment of kidney stones, diabetes and vitiligo. An in vivo anti-psoriasis, anti-inflammatory activities and oral toxicity studies of aqueous extract from seeds of Ammi visnaga L. was investigated. In the acute toxicity, the extract was administered orally in a single dose to rats (0- 2000-5000 mg/kg) and in the sub-acute toxicity daily for 28 days (0-300-600-1000 mg/kg/day). The symptoms of toxicity and mortality have been recorded daily and during 14 days of recovery with an examination of liver, kidney, hematologic, biochemical and histological analysis at the end of treatment. The anti-inflammatory activity was evaluated by induction of oedema and the anti-psoriasis by induction of a psoriasiform-like skin phenotype by UV-B radiations. No mortality was observed after single gavages by a dose up to 5000 mg/kg and no signs of toxicity noted. Clinical and biochemical examination during 28 days of gavages at all doses showed no significant difference compared to control group, while a significant reduction in MCV (mean corpuscular volume) and P-LCR (platelet large cell ratio) (p <0.05), PDW (platelet distribution width) and MPV (multi-purpose vehicle) (p <0.01) was observed and histopathological examinations showed slight inflammation in the liver and kidneys for the higher dose. Percentage of inhibition of the oedema was near the positive control 50% for all doses tested. Treatment with A.V. extract had decreased the thickness of the skin induced by UV-B irradiation.  In conclusion, the LD50 was estimated greater than 5000 mg/kg; therefore A.V. can be classified as non-toxic but if used in the long term can induce a slight toxicity dose dependant with high anti-psoriasis and anti-inflammatory activities.

Establishment of safety and toxicity profiles of metabolites of endophytic organisms from known medicinal plants are crucial in their pharmacological and biological applications. The aim of this study was to evaluate the oral acute toxicity (LD50) of crude extract of endophytic fungi isolated from Annona senegalensis Pers. The endophytic fungal metabolite was extracted with ethyl acetate. The LD50 was estimated following the method described by Lorke. Three dose levels (10, 100, and 1000 mg/kg) of the crude extract were administered to three mice each for the first phase using oral gavage needle in a single dose disposable syringe. The animals were observed for possible deaths or other side effects of the test substance in each group within 24 hours of the treatment. In the second phase, which was deduced from the first phase, eight mice were sub-divided into four groups of two mice each and they were treated with doses of 1200, 1600, 2900 and 5000 mg/kg orally. They were also observed within 24 hours and final LD50 value was determined. Results showed that the endophytic fungal extract exhibited no mortality or any histological defect in the liver tissues of the mice. More so, the immunological parameter tested showed significant increase in neutrophils and lymphocytes relative to the control in all the fungal isolates. Additionally, the LD50 for the crude metabolites was > 5000 mg/kg. This study has revealed that crude extract of endophytic fungi isolated from Annona senegalensis Pers did not show oral acute toxicity in mice. Further studies will evaluate long term-toxicity of the crude extract. Keywords: Endophytes, LD50, Annona senegalensis, Metabolites, Fungi
