AbstractBisphenol A (BPA) is an industrial chemical widely used as a monomer in polycarbonate, epoxy resin and thermal paper production, and as a stabiliser in polyvinyl chloride (PVC) production. Thus, BPA is present in a wide range of common consumer products. Due to contact with BPA-containing products, humans are regularly exposed via different routes. BPA is an endocrine active substance and can potentially lead to different adverse health outcomes. In the present study, total BPA levels were analysed by on-line SPE-HPLC-MS/MS in spontaneous urine samples in order to investigate the exposure to an Austrian population aged 6-15 and 18-81 years. In the total study population (n=594), total BPA were found in 15.7% of all samples, with concentrations up to 17 μg/l (median: n.d., 95th percentile: 3.7 μg/l). These results show generally low BPA levels among the investigated study population. However, children and adolescents exhibited higher exposure levels. Daily BPA intakes were estimated and compared to acceptable exposure levels, whereat no exceedances could be identified. Although BPA exposure was shown to be low in the Austrian population, the wide distribution and the ongoing discussion related to possible negative health effects even at low doses indicate need for further monitoring and investigations.

AbstractPerfluoroalkylated substances (PFASs) are a class of synthetic chemicals used in a wide range of processes and products due to their unique physicalchemical properties. Through intake of PFASs via food or several consumer products, humans can be exposed. Long-chain PFASs have been associated with adverse effects in laboratory animals, and there is also evidence for adverse health effects in humans. Although investigations of human exposure are mainly conducted in blood samples, some studies have shown that especially short-chain PFASs can be detected in human urine. In the present study, a sensitive analytical method was adapted for the measurement of 12 PFASs in human urine samples by HPLC-MS/MS. For verifying this method, concentrations in 11 male and female participants aged 25-46 years were analysed. In the study population, ranges of urinary PFASs concentrations were n.d.- 8.5 ng/l for perfluoropentanoic acid, <LOQ-3.0 ng/l for perfluorohexanoic acid, n.d.-1.8 ng/l for perfluorohexane sulphonate, n.d.-0.99 ng/l for perfluoroheptanoic acid, 0.79-5.1 ng/l for perfluorooctanoic acid, <LOQ-4.9 ng/l for perfluorooctane suphonate, and <LOQ-0.57 ng/l for perfluorononanoic acid. For the other investigated PFASs, no urinary exposure could be identified in any of the samples. The present study shows that several shortchain PFASs are detectable in human urine.

Abstract Background Phthalates and bisphenol A (BPA) are endocrine disrupting chemicals used in consumer products, building materials, and food processing and packaging materials. They are associated with adverse health outcomes, especially when exposure occurs during heightened windows of susceptibility. Objective We evaluated the relationship between housing and dietary characteristics and the concentration of several high-molecular-weight (HMW) phthalate metabolites and BPA in a cohort of Latina adolescents. Methods We collected information on recent food consumption and housing characteristics and quantified the concentration of HMW phthalate and BPA metabolites in urine collected at two different time points. We used generalized estimating equations (GEE) to assess predictors of each metabolite. Results No significant associations were observed between housing and dietary characteristics and metabolites of di(2-ethylhexyl) phthalate (DEHP) or BPA. In contrast, higher urinary monobenzyl phthalate (MBzP) concentration was associated with living in a home with vinyl or linoleum flooring (66.7% change, p-value <0.01), while higher urinary mono(3-carboxypropyl) phthalate (MCPP) concentration was associated with recent consumption of coffee (47.2% change, p-value <0.01), and fast food (30.3% change, p-value <0.05). Significance These findings may be useful in targeting interventions that reduce phthalate uptake in young adults.

AbstractNovel methods to characterize the plasma proteome has made it possible to examine a wide range of proteins in large longitudinal cohort studies, but the complexity of the human proteome makes it difficult to identify robust protein-disease associations. Nevertheless, identification of individuals at high risk of early mortality is a central issue in clinical decision making and novel biomarkers may be useful to improve risk stratification. With adjustment for established risk factors, we examined the associations between 138 plasma proteins measured using two proximity extension assays and long-term risk of all-cause mortality in 3,918 participants of the population-based Malmö Diet and Cancer Study. To examine the reproducibility of protein-mortality associations we used a two-step random-split approach to simulate a discovery and replication cohort and conducted analyses using four different methods: Cox regression, stepwise Cox regression, Lasso-Cox regression, and random survival forest (RSF). In the total study population, we identified eight proteins that associated with all-cause mortality after adjustment for established risk factors and with Bonferroni correction for multiple testing. In the two-step analyses, the number of proteins selected for model inclusion in both random samples ranged from 6 to 21 depending on the method used. However, only three proteins were consistently included in both samples across all four methods (growth/differentiation factor-15 (GDF-15), N-terminal pro-B-type natriuretic peptide, and epididymal secretory protein E4). Using the total study population, the C-statistic for a model including established risk factors was 0.7222 and increased to 0.7284 with inclusion of the most predictive protein (GDF-15; P < 0.0001). All multiple protein models showed additional improvement in the C-statistic compared to the single protein model (all P < 0.0001). We identified several plasma proteins associated with increased risk of all-cause mortality independently of established risk factors. Further investigation into the putatively causal role of these proteins for longevity is needed. In addition, the examined methods for identifying multiple proteins showed tendencies for overfitting by including several putatively false positive findings. Thus, the reproducibility of findings using such approaches may be limited.

Abstract Increasing prevalence of metabolic syndrome (MetS) is causing significant health burden among the European population. Current knowledge supports the notion that endocrine disrupting chemicals (EDCs) interfere with human metabolism and hormonal balance, contributing to the conventionally recognized life-style related risk factors for MetS. In relation to the Human biomonitoring initiative (HBM4EU) five priority substances (Bisphenol A, Per- and polyfluoroalkyl substances (PFASs), Phthalates, Cadmium and Arsenic) and their association with adverse metabolic health effects were examined. A methodological framework for scoping reviews was followed to increase consistency and transparency throughout the process. A literature review was conducted to identify epidemiological studies focusing on the association between MetS or its individual components and the five HBM4EU priority substances. Human biomonitoring studies have been able to present evidence supporting EDC exposure and development of individual MetS components; however the strength of the association varies between the components and EDCs. Most of the identified literature examined Bisphenol A and Phthalate exposure, usually targeting obesity, anthropometrics or glucose metabolism. Evidence suggests a positive association between Bisphenol A and Phthalate exposure and obesity-related components. The substance group of PFASs indicated weakest association, as the results were inconsistent and were suggestive only for a positive association with development of dyslipidaemia. Current evidence on metabolic disturbances and EDCs are inconclusive and fragmented, hence establishing harmonized and standardized human biomonitoring procedures among the European population are needed. Rigorous and ongoing human biomonitoring in combination with health monitoring could provide comprehensive information on EDC exposure and association of metabolic disturbances. Key messages EDC exposure is ubiquitous within European population, hence more human biomonitoring in combination with health surveys is needed to strengthen knowledge on human’s metabolic health. MetS is an increasing global health concern, which requires novel approaches to tackle the challenge.

Abstract The identification of early-life determinants of overweight is crucial to start early prevention. As weight gain accelerates between 2 and 6 years, we studied the association between diet quality in children aged 3 years and the change in BMI and overweight incidence in the following 7 years. From the Dutch GECKO Drenthe birth cohort, 1001 children born in 2006 or 2007 with complete data on diet (food frequency questionnaire at the age of 3 years) and growth at the age of 3 and 10 years were included. Diet quality was estimated with the evidence-based Lifelines Diet Score (LLDS). Measured height and weight at the age of 3 and 10 years were used to calculate BMI z-scores standardized for age and sex. The associations of the LLDS (in quintiles) with BMI-z change and overweight incidence were studied with linear and logistic regression analyses. Overweight prevalence in the total study population increased from 8.3% at the age of 3 years to 16.7% at the age of 10 years. The increase in overweight prevalence ranged from 14.7% in Q1 to 3.5% in Q5. Children with a better diet quality (higher quintiles of LLDS) increased significantly less in BMI-z (confounder adjusted βLLDS = −0.064 (−0.101; −0.026)). Children with a poor diet quality at the age of 3 years had a considerably higher risk for overweight at the age of 10 years (confounder adjusted OR for Q1 vs. Q5 was 2.86 (95% CI 1.34–6.13). These results show the importance of diet in healthy development in the early life following the first 1000 days when new habits for a mature diet composed of food groups with lifelong importance are developed, providing a relevant window for overweight prevention early in life.

Silver nanoparticles (Ag-NPs) have diverted the attention of the scientific community and industrialist itself due to their wide range of applications in industry for the preparation of consumer products and highly accepted application in biomedical fields (especially their efficacy against microbes, anti-inflammatory effects, and wound healing ability). The governing factor for their potent efficacy against microbes is considered to be the various mechanisms enabling it to prevent microbial proliferation and their infections. Furthermore a number of new techniques have been developed to synthesize Ag-NPs with controlled size and geometry. In this review, various synthetic routes adapted for the preparation of the Ag-NPs, the mechanisms involved in its antimicrobial activity, its importance/application in commercial as well as biomedical fields, and possible application in future have been discussed in detail.

Introduction: Various forms of malocclusion are a matter of serious concern in Nepalese population. This study was carried out to understand the prevalence of malocclusion among the school children of Biratnagar. The objective of this research is to find out the prevalence of malocclusion of children from different schools of Biratnagar visiting the Pedodontics and Orthodontics department of Nobel Medical College and Teaching Hospital, Biratnagar. Materials and Method: A descriptive cross sectional study method was used in this research. Data was collected by using direct observation of the subjects and occlusal assessment was done according to Angle’s classification and Dewey’s modification types of class I, class II and class III malocclusion. Result: Subjects with normal occlusion was found to be 39 % and with malocclusion was found to be 61%. Among them, class I malocclusion (60%) and angles class II div I subjects (88.33%) were in majority of the total study population. Conclusion: The present study helps to determine the prevalence of malocclusion and need of orthodontic treatment for the school children of Biratnagar

Background and Objectives: Increased concern over dental appearance has been observed during childhood and adolescence to early adulthood. The study was conducted with objectives to assess the self-perception of dental aesthetics, knowledge, attitude and management of dental trauma and halitosis.Materials and Methods: Self-administered questionnaire translated in Nepali language consisting of 32 questions was used. The completed questionnaires were analyzed using descriptive statistics.Results: Out of the total study population, 63% were satisfied with their overall tooth appearance and 71.7% with their tooth color. Twenty-nine percent had experienced dental trauma. More than half of the study population thought that immediate treatment was required after dental trauma. Fifty seven percent of the villagers had halitosis.Conclusion: The research clearly shows that the people of Aurabani VDC were satisfied with their tooth color and overall tooth appearance; however the knowledge regarding emergency management of dental trauma, the cause and management of halitosis was insufficient.JCMS Nepal. 2015;11(1):6-8
