AbstractThe response of β-amylase in early stage germination of wheat seeds to Bacillus subtilis QM3 was mainly focused on to elucidate the promotion mechanism of B. subtilis QM3. The results showed that the changes in apparent activity of amylase and endosperm liquefaction after the strain QM3 treatment were much more obvious than that of the control group; the activity of β-amylase treated with the different concentrations of the strain QM3 increased significantly (P < 0.05) by 4% (107 CFU/mL) and 18.5% (106 CFU/mL) at the germination 6 h. Moreover, after presoaking with α-cyclodextrin, the activity of β-amylase increased significantly (P < 0.05) by 18.5% (107 CFU/mL) and 23.4% (106 CFU/mL) at the same stage of germination; the electrophoretogram of β-amylase isoenzymes showed that there is a growing trend in brightness and width of the band during the early germination from 3 to 6 h of wheat seed treated by the strain QM3 (106 CFU/mL). The increase in activity and isoenzyme expression of β-amylase may be one of the important reasons to promote the germination of wheat seeds after being treated by B. subtilis QM3.

Seed germination is the basis and initial stage in the process of plant growth and development. Bacillus subtilis QM3 is a plant growth promoting rhizobacterium (PGPR) with function of growth promoting, promoting, prevention of pathogens and pests attack and can improve plant resistance to different stress factors. Through the measurement of the early germination rate of wheat (Triticum aestivam L.). seeds under B. subtilis QM3 treatment, the results showed that B. subtilis QM3 can significantly promote the germination of wheat seeds, which has reached a significant level at 6h after seeds sowing or at 6 h of treatment, during imbibition. The β-amylase isoenzyme in early period of wheat seed germination was measured by denaturing polyacrylamide gel electrophoresis. The results showed that the band width and brightness of β-amylase isoenzyme of wheat seeds treated with B. subtilis QM3 increased during three to 6 h of imbibition and especially the effect was significant at 6 h. In the early period, the band of α-amylase isoenzyme could not be detected. It is suggested that the increase of β-amylase isoenzyme early band may be one of the main reasons for B. subtilis QM3 to promote wheat seed germination. Through the combination of B. subtilis QM3 with free and binding states of β-amylase, it was found that the former can increase the activity of β-amylase by either increasing free β-amylase or releasing binding β-amylaseisoenzyme. β-amylaseisoenzyme inhibitors can significantly inhibit β-amylase activity, nevertheless α-amylase activators and inhibitors have no significant effect on β-amylaseisoenzyme, which further proved that the β-amylase exerted the effects in the early period of wheat seed more precisely during the imbibition of the seeds.

Grafting is common in all cucurbits in Asia, and gourd (Lagenaria siceraria) is the most popular rootstock for watermelons. Since the grafting is practiced at very early stage (right after the cotyledon expansion), uniform germination of rootstocks as well as the scions is crucial for grafting efficiency. Seeds were divided into three groups; intact, dry-heat treated (75 °C for 72 h), and brushed (575 rpm for 5 min). In each group, various solid matrix priming (SMP) treatments were imposed. Microcel E was used for SMP treatment with water or chemical solutions (10 seed: 1 Microcel E: 3 water, by weight). SMP treatment promoted earlier seed germination in all tested cultivars, thus resulting in higher rate of graftable seedlings. Brushing before SMP further enhanced earlier and uniform seed germination. Dry heat treatment, which can eliminated the seed-borne Fusarium spp. and virus, significantly delayed the early germination although the final germination percentage was not influenced. The characteristics of seedlings will also be presented.

Wheat seed samples were collected from ten unions of sadar upazilla of Thakurgaon district in wheat growing season of 2011. Seeds were tested by blotter method at Seed Pathology Center (SPC), Bangladesh Agricultural University, Mymensingh during the months April to November 2011 for recording and identifying the seed-borne fungi associated with wheat seeds. The health status of 20 seed samples were determined whereas five genera fungi were identified from a total of six fungus. The fungi were Bipolaris sorokiniana, Alternaria tenuis, Fusarium spp, Penicillium sp, Aspergillus flavus and Aspergillus niger. Prevalence of the total as well as the individual seed-borne fungal infections that were recorded varied significantly with respect to wheat varieties and sources of seed collection. Seed samples collected from Jagonathpur and Gorea unions of sadar upazilla showed highest percentage of seed-borne infection compared to the samples collection from other unions for both varieties. The seed-borne fungal infection in Hazar-8 and Satabdi collected from other unions showed lowest percentage of seed-borne fungal infection than that of Jagonathpur and Gorea unions. Seed germination also varied significantly depending on the varieties and the seed sources and a positive correlation between seed germination and seed-borne fungal infections were observed. Three seed treating agents viz., neem leaf extract 1:2, hot water and Provax were evaluated for controlling seed-borne fungi associated with wheat seeds. Among the seed treating agents, Provax was found superior to reduce the seed-borne infection of wheat. The results also showed that neem leaf extract at 1:2 dilutions was observed to be the most effective in reducing seed-borne fungi with highest percentage of germination followed by hot water treatment.

The results of studying the influence of a relatively new subclass of gas ? discharge sources of ultraviolet (UV) radiation-excilamp (XeCl, XeBr, KrCl) on the sowing qualities of seeds (germination energy, laboratory germination, seed morbidity) of soft spring wheat are presented. The objects of research are wheat seeds (Triticum aestivum L.) of the Irena variety (harvest 2016–2018), infected with the most common diseases in the Tomsk region. The studies have shown that among the three fungal diseases of wheat seeds, penicillosis was the best suppress using excilamps. Treatment of a batch of wheat seeds infected with penicillosis, XeBr or KrCl excilamp with a dose of 43.2 j/cm2 gives a disinfecting effect. In addition, this preserves the seed germination rate, which is crucial for their further use for sowing purposes. The same disinfecting effect is observe when the seeds are exposed to excilamp in relation to alternariosis, but the indicators of seed germination are reduced. None of the options impact of the excilamp on wheat seeds infected with Helminthosporium, showed no significant reduction of the infestation while preserving the germination level of control.

Liver disease or liver cancer is the sixth most common cancer and the third leading cause of cancer mortality in the world. Hepatitis viral infection, food additives, alcohol, fungal toxins (aflatoxins), toxic industrial chemicals, air and water pollutants are the major risk factors of liver cancer. Moreover, due to high tolerance of liver, HCC is seldom detected at an early stage and once detected treatment faces a poor prognosis in most cases.Fumaria indica possesses hepatoprotective activity as evidenced by the significant and dose dependent restoring the activities of entire liver cancer marker enzymes, diminution in tumor incidence, decrease in lipid peroxidation (LPO) and increase in the level of antioxidant enzymes (GSH, CAT, SOD, GPx and GST) through scavenging of free radicals, or by enhancing the activity of antioxidant, which then detoxify free radicals. These factors protect cells from ROS damage in NDEA and CCl4-induced hepatocarcinogenesis. Histopathological observations of liver tissues too correlated with the biochemical observations. Thus, present investigation suggested that the Fumaria indica would exert a chemoprotective effect by reversing the oxidant-antioxidant imbalance during hepatocarcinogenesis induced by NDEA and CCl4. Besides Fumaria indicais very much effective in preventing NDEA-induced multistage hepatocarcinogenesis possibly through antioxidant and antigenotoxic nature, which was confirmed by various liver injury and biochemical tumour markers enzymes. The hepatoprotective activity of a Fumaria indicaof 50 % ethanolic extract was studied using rats. The animals received a single intraperitoneal injection of N-nitrosodiethylamine 200mg/kg body wt followed by subcutaneous injection of CCl4 in a dose of 3 ml/kg body wt. Fumaria indica extract dose dependently and significantly the increase in serum hepatic enzyme levels after NDEAand CCl4 treatment compared to the toxin control group. The results of this study confirmed the antioxidant and hepatoprotective activity of the Fumaria indicaextract against carbon tetrachlorideand N-nitrosodiethylamine induced hepatotoxicity in rats. In addition to this, studies on molecular aspect of hepatoprotective therapy will give mechanistic information in hepatoprotective therapy and also critical balance should be there between the animal model and clinical research. The hepatoprotective properties of Fumaria indicashould provide useful information in the possible application in hepatic liver disease.

The efficiency of introducing probiotics based on strains of Bacillus subtilis, Bacillus licheniformis and their consortium in the amount of 150 g/t of feed into the diets of laying quails has been studied. The experiment lasting 182 days has been carried out on four groups of quails with 30 heads in each. The quails have been housed in the broiler battery in compliance with the required microclimate conditions. Quails of all groups have been received the main diet (compound feed) developed taking into account their age and physiological characteristics. The quails of the 1st, 2nd and 3rd experimental groups in addition to the main diet received probiotics (150 g/t compound feed) based on strains Bacillus subtilis, Bacillus licheniformis and their consortium, respectively. It has been found that feeding the laying quails of the consortium of strains Bacillus subtilis and Bacillus licheniformis had the most significant positive impact on their productive performance, it allowed to increase egg production by 7,81 %, egg laying intensity by 5,0 %, egg mass yield by 9,77 %, while reducing feed expenditures for 10 eggs by 13,35 %. The yield of hatching eggs has been increased by 7,03 %, hatchability of chickens from laid and fertilized eggs by 8,33 and 8,35 %, brooding waste decreased by 21,74 %. Hematological parameters of quails during the whole experiment were within the physiological norm. The economic effect calculated on the basis of data on the cost of compound feed, probiotics and the cost of sold eggs of quail laying was 14,56 % in the 3rd experimental group (in relation to the control group).

Vitamins are essential for the health of all living organisms. Vitamins E, A, D and K are known as fat-soluble vitamins, and deprivation of vitamin E causes various disorders, especially in the reproduction and cardiovascular systems and in muscle functions. Vitamin A, on the other hand, has roles in various biological functions – like eyesight – and the growth, reproduction and differentiation of epithelial cells. Vitamin A deficiency leads to the keratinization of the epithelium, and disorders related to the metaplasies of the genital and genitourinary systems. Conversely, vitamin D is defined as a pro-hormone and is responsible for Cahomeostasis, and thus indirectly affects the bone metabolism, bone structure, and cellular and neural functions of Ca. White muscle disease (WMD) can occur in newborn lambs, but is more commonly seen in lambs of up to 3 months of age. In this study, 30 lambs of 3 to 50-days-old from different flocks diagnosed with White Muscle Disease (WMD) were selected as research material, while the control group consisted of 8 healthy lambs. With the aim of clarifying the cause of WMD, serum fat-soluble vitamins, retinol, α-tocopherol and vitamin D3 levels were determined in 16 lambs. Gluteal and heart musclet issue samples also were taken from 30 lambs with WMD. The vitamin levels of the samples were analysed by HPLC. The levels of serum α-tocopherol, retinols, and vitamin D3 were foundto be low in the diseased animals, but only retinol (p<0.001) and α-tocopherol (p<0.0011) level differences were statistically relevant. Macroscopically, Zenker’s necrosis was determined in the heart muscles of 17 lambs, and in the gluteal and chest muscles of 6 lambs. 7 lambs displayed necrosis in both their heart and in gluteal muscles. The samples were analyzed microscopically to reach similar findings: swollen homogeneous pink muscles, pycnotic nuclei, and hyperaemic and haemorrhagic blood vessels in gluteal, chest and heart muscles. Hyaline degeneration and Zenker's necrosis, dystrophic regions in necrotic areas, cc was detected as a severe disease in lambs at an early stage of life with advanced degeneration in different muscle tissues. Deficiency of fat-soluble vitamins was also detected in the sick animals. Control group lambs had higher levels of α tocopherol and retinol (p<0.001) compared to the sick lambs. .

Abstract Background Endothelial glycocalyx (EG) abnormal degradation were widely found in critical illness. However, data of EG degradation in multiple traumas is limited. We performed a study to assess the EG degradation and the correlation between the degradation and organ functions in polytrauma patients. Methods A prospective observational study was conducted to enroll health participants (control group) and polytrauma patients (trauma group) at a University affiliated hospital between Feb 2020 and Oct 2020. Syndecan1 (SDC1) and heparin sulfate (HS) were detected in serum sample of both groups. In trauma group, injury severity scores (ISS) and sequential organ failure assessments (SOFA) were calculated. Occurrences of acute kidney injury (AKI), trauma-induced coagulopathy (TIC) within 48 h and 28-day all-cause mortality in trauma group were recorded. Serum SDC1 and HS levels were compared between two groups. Correlations between SDC1/HS and the indicators of organ systems in the trauma group were analyzed. ROC analyses were performed to assess the predictive value of SDC1 and HS for AKI, TIC within 48 h, and 28-day mortality in trauma group. Results There were 45 polytrauma patients and 15 healthy participants were collected, totally. SDC1 and HS were significantly higher in trauma group than in control group (69.39 [54.18–130.80] vs. 24.15 [13.89–32.36], 38.92 [30.47–67.96] vs. 15.55 [11.89–23.24], P <  0.001, respectively). Trauma group was divided into high degradation group and low degradation group according to SDC1 median. High degradation group had more severe ISS, SOFA scores, worse organ functions (respiratory, kidney, coagulation and metabolic system), and higher incidence of hypothermia, acidosis and shock. The area under the receiver operator characteristic curves (AUC) of SDC1 to predict AKI, TIC occurrence within 48 h and 28-day mortality were 0.838 (95%CI: 0.720–0.957), 0.700 (95%CI: 0.514–0.885) and 0.764 (95%CI: 0.543–0.984), respectively. Conclusions EG degradation was elevated significantly in polytrauma patients, and the degradation was correlated with impaired respiratory, kidney, coagulation and metabolic systems in early stage. Serum SDC1 is a valuable predictive indicator of early onset of AKI, TIC, and 28-day mortality in polytrauma patients.

AbstractThe present study was conducted to investigate effects of Bacillus subtilis on growth performance, serum parameters, digestive enzymes, intestinal morphology, and colonic microbiota in piglets. A total of 72 piglets were weighed and randomly allotted into three treatments (four replication pens per treatment with six piglets/pen) for a 28-day experiment. The dietary treatments were as follows: basal diet (control group, CTR), basal diet supplementation with antibiotic (antibiotic group, ABT), and basal diet supplementation with 0.1% Bacillus subtilis (probiotic group, PBT). The average daily gain of body weight increased in both the ABT and PBT groups, and dietary antibiotics decreased the feed:gain ratio (F:G), as compared to the CTR group (P < 0.05). Both ABT and PBT piglets had increased serum triglycerides and lipase, amylase, maltase activities and villus height:crypt depth ratio (V/C) in ileum (P < 0.05). The PBT group also showed an increase in serum glucose and villus height in the ileum (P < 0.05). Dietary antibiotics increased Lactobacillus johnsonii, as compared to the CTR group, but decreased bacterial diversity and increased Escherichia coli, as compared to the PBT group (P < 0.05). Piglets dietary with B. subtilis modulated the microbiota by increasing the abundance of Firmicutes (L. johnsonii, L. reuteri) and decreasing the abundance of E. coli, as compared to the control group (P < 0.05). These results indicate that dietary of B. subtilis improves growth performance and intestinal health and can be a promising alternative to antibiotics in piglets diet.
