AbstractBackgroundL. (BV), commonly known as “MethodsSixty IOPS mice were divided into six groups and were treated as follows: group 1 (normal control) received double distilled water; group 2 (toxic control) received lead acetate (5 mg/kg body weight/day) in double distilled water for 40 days; groups 3–6 received BV aqueous extract at doses of 25, 50, 100 and 150 mg/kg body weight , respectively, once daily for 30 days from 11 day after beginning of lead acetate exposure to the end of the experiment.ResultsToxic control group showed a significant alteration of serum alanine-aminotransferase (ALT), aspartate-aminotransferase (AST), total cholesterol (TC), total bilirubin (TB), catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GPx) and reduced glutathione (GSH). Histological assessment of lead-intoxicated mice liver revealed alterations in hepatocytes and focal necrosis. BV treatment significantly prevented lead accumulation, increased ALT, AST, TC, and TB, inhibited lipid peroxidation and protein carbonyls(PCO) formation. Additionally, BV extract normalized the antioxidant enzymes (CAT, SOD and GPx), GSH and architecture of liver tissues.ConclusionsBV aqueous extract exerts significant hepatoprotective effects against lead-induced oxidative stress and liver dysfunction. The BV effect may be mediated through the enhancement of antioxidant status, lead-chelating abilities and free radicals quenching.

The effect of the aqueous extract of fig (Ficus carica) fruit was studied by using different concentrations of this fruit (15, 20 and 25 mg/kg of body weight) on some hematological parameters (hemoglobin concentration, packed cell volume, white blood cell count and platelets count) in female rabbits. Abstract Nephropathies and especially nephrotoxicity has become one of serious cause of life threatening condition, because of intensive exposure to xenobiotic either by environmental pollution or drug abuse. The present study was undertaken to assess the protective effects of Cinnamomum zeylanicum stem-bark aqueous extract (AECZ) on gentamicin-induced nephrotoxicity. AECZ was prepared by maceration in water and tested orally at the doses of 200 and 400 mg/kg/day to prevent gentamicin induced nephropathies in male Wistar rats. Gentamicin (100 mg/kg/day) was administered for 14 consecutive days by intraperitoneal route, concomitantly with AECZ or silymarin (50 mg/kg/day) used as reference drug. Animal body weight was monitored during the treatment. After the last treatment of the 14 th day, a 24h urine was collected and animals were sacrificed. Blood was collected for the evaluation of hematological and renal function biomarkers. The homogenate of one kidney was used to assess oxidative stress markers and pro-inflammatory cytokine, while the other one was fixed in formaldehyde for histopathological studies. Gentamicin decreased body weight, serum total proteins and calcemia, but increased kidneys relative weight, serum creatinine, urea and uric acid. Moreover, the levels of reduced glutathione, catalase and superoxide dismutase activities were decreased, while an increase in malondialdehyde, proinflammatory cytokines (TNFα, IL-1β, IL-6) and nitrites were observed in negative control group as compared to normal control. Histological analysis of the kidney revealed the presence of tubular necrosis, glomerular degeneration and macrophage infiltration in gentamicin treated group. All these impairment parameters were prevented by AECZ and silymarin treatments.AECZ has a protective effect against gentamicin-induced nephrotoxicity. The antioxidant and anti-inflammatory potentials of this extract may highly contribute to its nephroprotective activity.

Doxorubicin is an anthracycline drug which is believed to cause immediate damage to myocardial cells by free radical generation in the cause of treatment of cancer. This study was however aimed to investigate the effects of the aqueous extract of the seedlings of Vigna unguiculata on heart, kidney as well as lipid profile disorders caused by this drug on female Wistar albino rats. Fifteen female Wistar albino rats were divided into three groups. Group 1 animals served as normal positive control; Group 2 animals served as negative control which were treated with 0.5mL of (20mg/kg body weight doxorubicin) while Group 3 animals were treated with 0.5mL each of 20mg/kg body weight doxorubicin and 10% aqueous extract of seedlings of Vigna unguiculata. Some enzyme markers and lipid contents were determined. The results of the study showed an increase in the activities of plasma ALP and AST after treatment with the drug except in the heart for AST which showed a significant reduction, while treatment with the extract brought about a decrease in the plasma and the organs except for kidney AST. The ALT on the other hand showed slight increase in the plasma with a decrease in the kidney and heart after treatment with the drug as the treatment with the extract tend to restore it to the control in both cases, there was increased plasma and kidney but reduced heart HDL-C after treatment with the drug which was observed to be restored to control after treatment with extract in both situations. The triglyceride and total cholesterol did not show similar trend in the plasma and the studied organs. The LDL-C was also observed to be increased in both plasma and organs after the treatment with the drug which later reduced significantly towards the control after the administration of the sprout extract. The present study has shown that the doxorubicin has damaging effects on both kidney and heart tissues while the sprouts extract produce a restoration to the normal in both organs.

Abstract Background Herba Siegesbeckiae (HS), the dried aerial parts of Siegesbeckia orientalis L., S. pubescens Makino, or S. glabrescens Makino, is traditionally used for treating chronic diseases in China. However, there is no information about the chronic toxicity of HS. The objective of this study is to evaluate the 24-week oral dosing toxicities of HS aqueous extract (HSE) in rats. Methods S. orientalis-originated HS was reflux-extracted with distilled water. Sprague–Dawley rats were randomly divided into four groups, with 10 males and 10 females in each group. The rats were intragastrically administered with HSE at 5, 1.67 and 0.56 g/kg (experimental groups) or an equal volume of distilled water (control group), 6 days a week, for 24 weeks. The high dose of HSE (5 g/kg) was its maximum tolerated dose. Body weight was recorded every 2 days during the experimental period. Chemical, hematological and histopathological parameters, as well as organ weights, were measured at the end of the experiment. Results Decreased body weight gain; increased liver and lung relative weights; histopathological alterations in liver and lung tissues; elevated serum levels of alkaline phosphatase, aspartate aminotransferase, alanine aminotransferase and lactate dehydrogenase were found after HSE treatments. In liver tissues, HSE treatment upregulated levels of three pro-inflammatory cytokines: IL-6, IL-1β and TNF-α. In lung tissues, HSE treatment caused oxidative stress and activated mitogen-activated protein kinases (MAPKs). Conclusion Long-term oral administration of HSE caused toxicities in rats evidenced by decreased body weight gain, as well as liver and lung damage. Treatment-induced oxidative stress, inflammation and MAPK activation are involved in HSE’s toxicities. Caution should be taken when using HS to treat chronic diseases.

Mucuna pruriens (velvet beans) is an unconventional legume commonly found in the tropical regions of Africa with several ethnomedicinal uses. The present study was designed to investigate the effect of aqueous extract of Mucuna pruriens leaves in rats intoxicated with lead acetate. Thirty rats were divided into six groups (five rats per group). Group I (control group) was given distilled water only, group II was intoxicated with lead acetate (100 mg/kg) only, group III was administered M. pruriens extract (600 mg/kg) only daily for a period of 28 days, while groups IV, V and VI animals were co-administered M. pruriens extractat at different concentrations of 300, 600 and 1200 mg/kg body weight daily for 28 days. After 28 days of experiment, the animals where sacrificed. Blood samples were collected for some biochemical parameters and organs harvested for histology. The results showed that there were no significant differences in direct bilirubin, total protein, albumin and globulin, but significant increase (P˂0.05) in ALT after 28 days of intoxication with lead acetate when compared with the control. However there was no significant difference in ALP activities. Administration of aqueous extract of M. pruriens leaves at a dose of 600 mg/kg body weight for 28 days showed a significant increase (P˂0.05) in direct bilirubin, total protein, albumin, globulin, AST andALT, but significantly reduced ALP activities. Co-administration of the crude aqueous extract (300, 600 and 1200 mg/kg), did not show any ameliorating effect after 28 days when compared to rats intoxicated with lead acetate, but showed mild reduction in ALP activities with no significant changes in ALT and AST activities. Mucuna pruriens extract did not posses ameliorative effect in rats intoxicated with lead acetate, but showed potentials of resolving anaemia from the histopathological result.

Background: This investigation was aimed to evaluate the effect of Speman®, a well known ayurvedic proprietary preparation, in an experimental model of cyclophosphamide-(CP) induced oligospermia in rats.Materials and Methods: Thirty male rats were randomized in to five, equally-sized groups. Rats in group 1 served as a normal control; group 2 served as an untreated positive control; groups 3, 4, 5 received  Speman® granules  at doses of 300, 600, and 900mg/kg body weight p.o. respectively, once daily for 13 days. On day four, one hour after the respective treatment, oligospermia was induced by administering a single dose of CP (100mg/kg body weight p.o.)  to all the groups except group1. At the end of the study period the rats were euthanised and accessory reproductive organs were weighed and subjected to histopathological examination. The semen samples were subject to enumeration of sperms.  Weight of the reproductive organs, histopathological examination of the tissues, and sperm count were the parameters studied to understand the effect of Speman® on rats with CP-induced oligospermia.Results: Changes that occurred due to the administration of CP at a dose of 100 mg/kg body weight were dose dependently reversed with Speman® at a dose of 300, 600, and 900 mg/kg body weight. There was a statistically significant increase in sperm count and the weight of the seminal vesicle, epididymis, and prostate.Conclusion: Findings of this investigation indicate that Speman® dose dependently reversed the CP-induced derangement of various parameters pertaining to the reproductive system.  This could explain the total beneficial actions of Speman® reported in several other clinical trials.

Background: Sodium nitroprusside (SNP) is an antihypertensive agent. It induces toxicity via the release of cyanide ions, nitric oxide (NO) and reactive oxygen species. Manniophyton fulvum (MF) is commonly used in Nigeria due to its therapeutic and nutritional potentials. This study evaluates the phytochemical composition of aqueous extract of MF root and influence in SNP induced oxidative stress in wistar rats.Methods: Gas chromatography was used for determination of the chemical composition of aqueous extract of MF root. Rat liver homogenate was used for determination of rhodanese, glutathione (GSH) and malondialdehyde (MDA). Twenty (20) rats were randomly divided into four different groups comprising five animals (n=5). Control (Group A), Groups B, C, D received 2.5 mg/kg body weight of SNP at intervals of three hours per day by intraperitoneal injection. In addition, Groups C and D received 200 mg/kg body weight of aqueous root extract of MF and 10 mg/kg body weight of Vitamin E respectively for a period of seven days.Results: Flavonoids had the highest composition while allicin had the lowest composition. Diallyl thiosulphinate>methyl allyl thiosulphinate >allyl methyl thiosulphinate. The activity of rhodanese, GSH and MDA concentrations showed that Group B had significant (p<0.05) increase in MDA concentration while GSH showed significant (p<0.05) decrease. Also, the activity of rhodanese showed significant (p<0.05) decrease compared to Group A. However, Groups C and D showed significant increase (p<0.05) in the activity of rhodanese enzyme compared to Group A and Group B. GSH levels of Group C and Group D showed no significant (p>0.05) difference while the MDA concentration showed significant (p<0.05) decrease. Correlation analysis between rhodanese and GSH showed strong significant (p=0.01, r=0.894) positive correlation.Conclusions: It can be deduced that the chemical components of aqueous extract of MF may serve as a pharmacological agent for molecular targets to up regulate detoxification and cytoprotective enzymes. Cellular localization of rhodanese enzyme can be induced by aqueous extract of MF root to collaborate with GSH and promote inhibition of lipid peroxidation, anti-oxidative reactions and up regulate cyanide detoxification in tissues.

The impact of lead toxicity was assessed in this study by analyzing the effects of the extracts on some electrolytes, urea and creatinine concentration. Thirty-five male wistar rats (150+50g) were distributed into five groups with seven rats in each group. Extraction and all biochemical analysis were carried out using standard laboratory techniques. Group one served as control, group two as test and were exposed to lead acetate only. Groups three, four and five were treatment groups, administered carrot and garden egg, carrot and tomato, garden egg, carrot and tomato respectively. All groups received feed and water ad libitum. Lead Acetate solution was administered orally at 50mg/kg body weight while 200mg/kg body weight received mixed juicy extract. On day 15, food was withdrawn, fasted overnight with free access to water. They were euthanized under chloroform vapor and sacrificed. Whole blood was collected via cardiac puncture for biochemical analysis. There was significant (P<0.05) decrease in potassium and chloride ion in group 2 (test group), compared to the control and treatment groups. Concentration of urea and creatinine in the test group was significantly higher compared with the other groups. Concentration of creatinine in group 3 and 5 significantly increased compared to control group. Effect of lead acetate was significantly reversed in group 5 compared to group 2. The results of the study indicate the potency of plants extracts against toxicity caused by the lead acetate. Flavonoids, saponins, tannins and alkaloids were present in the extracts.

Ancient literature alluded to the use of a number of plants/preparations as sex enhancer. One of such botanicals isMassularia acuminatain which the stem has been acclaimed to be used as an aphrodisiac. Documented experiments or clinical data are, however, lacking. Therefore, this study was undertaken to evaluate the acclaimed aphrodisiac activity ofM. acuminatastem. Sixty male rats were completely randomized into 4 groups (A–D) of 15 each. Rats in group A (control) were administered with 1 mL of distilled water (the vehicle) while those in groups B, C, and D were given same volume containing 250, 500, and 1000 mg/kg body weight of the extract, respectively. Sexual behaviour parameters were monitored in the male rats for day 1 (after a single dose), day 3 (after three doses, once daily), and day 5 (after five doses, once daily) by pairing with a receptive female (1 : 1). The male serum testosterone concentration was also determined. Cage side observation on the animals revealed proceptive behaviour (ear wiggling, darting, hopping, and lordosis) by the receptive female rats and precopulatory behaviour (chasing, anogenital sniffing and mounting) by the extract-treated male rats. The extract at 500, and 1000 mg/kg body weight significantly (P<.05) increased the frequencies of mount and intromission. In addition, the ejaculation latency was significantly prolonged (P<.05). The latencies of mount and intromission were reduced significantly whereas ejaculation frequency increased. The extract also reduced the postejaculatory interval of the animals. Computed percentages of index of libido, mounted, intromitted, ejaculated and copulatory efficiency were higher in the extract treated animals compared to the distilled water-administered control whereas the intercopulatory interval decreased significantly. The extract also significantly (P<.05) increased the serum testosterone content of the animals except in those administered with 250 mg/kg body weight on days 1 and 3. Data from this study identified that the aqueous extract ofMassularia acuminatastem enhanced sexual behaviour in male rats. The improved sexual appetitive behaviour in male rats at the doses of 500 and 1000 mg/kg body weight ofMassularia acuminatastem may be attributed, at least in part, to the alkaloids, saponins, and /or flavonoids since these phytochemicals has engorgement, androgen enhancing, and antioxidant properties.
