<p>Colorectal carcinogenesis induced by cigarette smoke requires at least 30-40 years. This long time duration causes an animal research conducted becomes relevant. This research was carried out to observe colorectal carcinogenesis due to cigarette smoke exposure in Wistar Rat. The observations focused on changes in epithelial morphology and expression of APC, MSH2 and MLH1. Twenty male Wistar rats inbreed strain were randomly allocated into control group and experimental group exposure to cigarettes smoke for 14 weeks and 28 weeks sequentially.  Colorectal epithelial morphology was assessed on the histopathology examination, whereas the expression of APC, MSH2 and MLH1 was assessed on aspect of immunohistochemistry. The comparative analysis between the two groups was performed using non-parametric Mann-Whitney U test. Histology of colorectal epithelium showed pattern of colitis associated cancer that was significant both in 14 weeks and 28 weeks of treatment.  This research indicated negative expression of APC, MSH2 and MLH1 in the colorectal cancer that were significant at 28 weeks of exposure. This research implies that chronic exposure to cigarette smoke can induce colitis associated colorectal cancer through decreased expression of APC, MSH2 and MLH1. </p>

The exposure of cigarette smoke produce CO bond in hemoglobin. Melon (Cucumis melo) and gliadin contains of antioxidants that prevent tissue damage. The aim of this study was to analyze the difference of hemoglobin and carboxyhemoglobin concentration in male Wistar rats were exposed  to cigarette smoke and had been treated melon extract (Cucumis melo) and gliadin. The study was a laboratory experimental design, using Post Test Control Group Design Research and RAL method (Complete Randomize Design). The sample in this study were 25 male Wistar rats aged 3 months. The research was divided into 5 groups with 5 different treatment : control group, the treatment group were given exposure to cigarette smoke, the treatment group were given of cigarette smoke exposure and melon extract (Cucumis melo) and gliadin dose of 3 IU / day, 4.5 IU / day, and 9 IU / day.The subject were divided into 5 groups each treated for 28 day. The sample size used Federer formula. The collection of data was obtained from the results of laboratory tests to hemoglobin and Carboxyhemoglobin. Hb and HbCO data collected and analyzed by Manova test at 95% confidence level. The results showed significant difference in average Hb and HbCO concentration (p-value = 0.000) between male Wistar rats treated and not treated with melon extract (Cucumis melo) and gliadin. Melon extract has an effect on the decline of HbCO concentration due to exposure to cigarette smoke.Keywords:smoke, melon extract, Hb, HbCO

This study aims to look at the histopathological picture of the dorsal arteries of the penis of the hiperlipidemic wistar rats (Rattus novergicus) induction by high-fat diet that seen in terms of lumen diameter and thickness of the arterial endotelium wall. Hyperlipidemia is a risk factor for ateriosclerosis in the penile arteries causing erectile dysfunction in men. This study is an experimental study with a randomized posttest only control goup design. The sample are  10 individuals adult male wistar rats aged 3-4 months with a range of body weight 150-200 grams. Before treatment, adaptation was carried out for 7 days. After that the sample rats in the treatment group were made hyperlidemic by feeding lard for 50 days. Then surgery is performed for histopathological preparations at the posttest. To determine the differences in endotelium thickness and arterial lumen diameter in the two groups, an independent t-test was used. Thick diameter data of the endotelium and dorsal arteries of the penis of the wistar rat between the lower treatment group and the control group. The difference that occurred was statistically significant (p <0.05). So it can be concluded that the provision of high-fat diet (hyperlipidemia) decreases the lumen diameter and endotelium thickness of dorsal arteries penis. Cell proliferation and apoptosis in the remnant rat kidney after treatment with lowdose irradiation was investigated. Material and methods. In the first group (n=9), adult male Wistar rats underwent 5/6 nephrectomy (NPX); in the second group (n=9), NPX was combined with low-dose irradiation. Rats without surgery and irradiation formed the control group (n=9). Results. Hypertension and proteinuria induced by NPX were decreased by 3-Gy irradiation. The 5/6 NPX rats showed a dramatic increase in proliferating and apoptotic cells in the glomeruli and in the distal tubules at week 2, which was significantly decreased by low-dose irradiation. Conclusion. The data demonstrate that low-dose irradiation is a factor slowing the process of chronic renal injury.

OBJECTIVE: To describe a new murine model of cigarette smoke-induced emphysema. METHODS: Twenty-four male Wistar rats were divided into two groups: the cigarette smoke group, comprising 12 rats exposed to smoke from 12 commercial filter cigarettes three times a day (a total of 36 cigarettes per day) every day for 30 weeks; and the control group, comprising 12 rats exposed to room air three times a day every day for 30 weeks. Lung function was assessed by mechanical ventilation, and emphysema was morphometrically assessed by measurement of the mean linear intercept (Lm). RESULTS: The mean weight gain was significantly (approximately ten times) lower in the cigarette smoke group than in the control group. The Lm was 25.0% higher in the cigarette smoke group. There was a trend toward worsening of lung function parameters in the cigarette smoke group. CONCLUSIONS: The new murine model of cigarette smoke-induced emphysema and the methodology employed in the present study are effective and reproducible, representing a promising and economically viable option for use in studies investigating the pathophysiology of and therapeutic approaches to COPD.

ABSTRACT Excessive energi consumption of high-fat diet increase the body’s adipose tissue which also affect the increased secretion of leptin, this is because leptin secretion is proportional to the mass of adipose tissue. Leptin activates the lateral hypothalamus as the center of hunger and feeding activity. The aim of this study was to obtain the relationship between high trans fat feeding on male Wistar rats and feeding activity. This method uses male Wistar rats (Rattus norvegicus) as an animal model. Sixteen rats were grouped into 4 treatment groups. The first group was given additional feed 20% trans fat, the second group was given additional feed 40% trans fat, a third group was given additional feed 60% trans fat, and the last as a control group. This feed was given once a day for 70 days. Observation of activities perform using Opto-varimex® auto-track system ver. 4.31. BACKGROUND: Cigarette smoke has a variety of dangerous chemicals and free radicals that can potentially cause infertility. One of the plants that are reported to have medicinal properties and have active compounds as antioxidants is Binahong (Anredera cordifolia (Ten.) Steenis). AIM: This study aims to investigate the effectiveness of A. cordifolia leaf extract (ACLE) in increasing the fertility of male Wistar rats after exposure to cigarette smoke. METHODS: Twenty-four adult male rats were divided into six groups (age 8–10 weeks, weight 200–250 g): Group 1 (normal control) received aquades (1 ml/day) without being given cigarette smoke, Group 2 received exposure to smoke without being given ACLE, Group 3 received cigarette smoke exposure + 25 mg/kg ACLE, Group 4 received cigarette smoke exposure + 50 mg/kg ACLE, Group 5 received cigarette smoke exposure + 75 mg/kg ACLE, and Group 6 received exposure to cigarette smoke + 100 mg/kg of ACLE. To produce smoke from cigarettes and expose rats to the smoke, a Smoke chamber is used. Rats in the treatment group were exposed to cigarette smoke for 2 weeks (40–60 s daily for 6 days each week). The evaluation of male rat fertility was carried out by measuring body weight, genital weight (testis and epididymis), and spermatozoa spermogram (velocity, motility, morphology, and total number). RESULTS: The least significance different test results showed no significant difference in the rate of weight gain between treatments, standard control, and negative control groups. The average testicular weight of rats exposed to cigarette smoke for 14 days was significantly different from normal controls and doses of 50, 75, and 100 mg/kg body weight. However, at a dose of 25 mg/kg of body weight had not shown a significant increase compared to control. The average spermatozoa velocity reached 12.43 mm2/s and 13.36 mm2/s. The spermatozoa velocity increased significantly at a dose of 100 mg/kg body weight. CONCLUSIONS: ACLE at a dose of 100 mg/kg body weight was effective in increasing the fertility of male Wistar rats exposed to cigarette smoke.

Abstract: Herbal plant is often used to treat any kind of disease. There are many kinds of plants in Minahasa used as traditional medicine, inter alia daun sendok (Plantago major L.). Daun sendok is often used to treat bleeding. This study was aimed to obtain the effectiveness of daun sendok extract on bleeding time in male Wistar rats. This was a true experimental study with a posttest only group design. This study was conducted at the Laboratory of Pharmacology Faculty of Medicine University of Sam Ratulangi. Total samples were 14 rats divided into 2 groups: control group and treatment group. Wounds of 2 mm depth were perfomed on the rat tails of the treatment group followed by application of daun sendok extract. The results showed that the average bleeding time of the control group was 51.87 seconds meanwhile of the treatment group was 29.67 seconds. The unpaired t-test between the two groups showed a p value of 0.000. Conclusion: Daun sendok (Plantago major L.) extract could reduce the bleeding time in male Wistar rats. Di wilayah Minahasa terdapat banyak jenis tanaman yang digunakan sebagai obat tradisional, salah satunya ialah daun sendok (Plantago major L.) yang seringkali digunakan untuk mengobati perdarahan. Penelitian ini berrtujuan untuk mengetahui efektivitas ekstrak daun sendok terhadap waktu perdarahan tikus Wistar jantan. Jenis penelitian ialah true experimental dengan post test only control group design yang dilakukan di Laboratorium Farmakologi Fakultas Kedokteran Unsrat dengan total sampel berjumlah 14 ekor tikus Wistar yang dibagi menjadi 2 kelompok yaitu: kelompok kontrol dan kelompok perlakuan menggunakan ekstrak daun sendok. Pada kelompok uji diaplikasikan ekstrak daun sendok setelah dilakukan perlukaan di bagian ekor tikus sedalam 2 mm. Hasil penelitian memperlihatkan rerata waktu perdarahan pada kelompok kontrol ialah 51,87 detik sedangkan pada kelompok perlakuan 29,67 detik. hasil uji t tidak berpasangan antara kedua kelompok mendapatkan nilai p=0,000. Simpulan: Ekstrak daun sendok (Plantago major L.) efektif dalam memperpendek waktu perdarahan pada tikus Wistar.Kata kunci: waktu perdarahan, daun sendok, tikus Wistar

Background. The exposure of cigarette smoke impacts negatively on Leydig cell count and testosterone level. Astaxanthin is known for its ability to neutralize free radicals far more potent than any other kind of antioxidant. Research objectives. This research aims to prove the effect of Astaxanthin in inhibiting decrease of Leydig cell count and testosterone level in male Wistar rats exposed with cigarette smoke. Methodology. The posttest only control group study design was conducted on 36 male Wistar rats, 12-16 weeks in age, with 200-210 grams body weight. Samples were randomly divided into two groups, consisting of a control group exposed with cigarette smoke and 0.5 ml of aquadest and a study group exposed with cigarette smoke and 0.1 mg of astaxanthin/200 gr BW daily for 30 days. On day 31, blood samples were taken to measure the testosterone level. Both testes were taken for Leydig cells count assessment. Comparative analysis was done to see any significant difference between the study and control group. Research results. The results show that the mean number of Leydig cells and testosterone levels in the study group was significantly higher than the control group (p<0.01). Conclusion. Oral astaxanthin administration inhibited the decrease of Leydig cell count and testosterone level in male Wistar rats exposed with cigarette smoke. Background: Smoking is something that is usual for most Indonesian society, especially adult males. According to Riskesdas, smoking behavior of the population trends to increase. A previous study suggested that exposure to cigarette smoke produce CO bond to hemoglobin. Melon (Cucumis melo) contains antioxidants that prevent tissue damage.Objective: Analyze differences in levels HbCO Wistar male rats in treated and untreated extracts of melon.Method: The research was a laboratory experiment with the post test control group design and complete random design. The subjects were 25 male Wistar rats aged 3 months. Subjects were divided into 5 groups and each group received treatment for 28 days. Data of level HbCO was collected and analyzed by Anova One Way test at 95% confidence level.Results: The results showed a difference between the average levels of HbCO male Wistar rats (p value=0,000) treated and untreated extracts of melon.Conclusion: Melon extracts effect on decreasing the levels of HbCO due to exposure to cigarette smoke. There were differences in levels of male Wistar rats HbCO on treated and untreated extracts of melon and be given at a dose of 250 IU/day.
