Background: A good collection of fungi culture can be maintained with good preservation methods. With appropriate methods, preservation can be performed in low-budget laboratories, such as with periodic subculture and preservation in either sterile water or in paraffin oil. The aim of this study was to assess the viability, morphology, contamination, and antifungal susceptibility profile of fungi preserved in sterile water or paraffin oil compared to a periodic subculture. Methods: This study was an experimental study using Aspergillus sp., Trichophyton mentagrophytes and Candida albicans. The fungi were preserved by a periodic subculture or in sterile water or in paraffin oil for six months. A comparison of viability, morphology, contamination, and antifungal susceptibility profile between methods were made before and after six months of study. Results: The viability of all the fungi was maintained in both periodic subculture and preservation with paraffin oil. However, the T. mentagrophytes was not viable in preservation using sterile water. All methods could maintain the morphology without contamination. There were reductions in the inhibition zone against antifungal drug in preservation with sterile water and paraffin oil. Conclusion: Preservation in paraffin oil is superior to preservation in sterile water in terms of maintaining fungal viability. This method is suitable for the use in a simple laboratory, albeit considerations should be given to the reduction of inhibition zone.

Microalgae present much usefulness for antimicrobial research because of its enormous biodiversity and rapid growth rate. From this study results it is reaveled that Chlamydomonas reinhardtii were isolated from a pond of water in the province of Diwaniyah. The culture supernatants were obtained when extracted with methanol solvent. Antimicrobial activity of extracts was tested for pathogens, and the best inhibition zone obtained was against Candida albicans (32mm), S.aureus (15mm), and to E.coli (9mm). While it showed no effect against both S.epidermidis and Klebsiella spp. Biofilm was formed by all tested isolates with differences in its strength formation. The C. reinhardtii Algal extract showed higher reduction of the existing Staphylococcus aureus biofilm and (10.9 %) was the remaining biofilm, while 55.21% and 54.98% were to Serratia spp. and Escherichia coli respectively, and it’s the lowest reduction. Klebsiella spp., Candida albicans, Acinetobacter spp., Staphylococcus epidermides, and Aspergillus sp. showed remaining biofilm as (16.7, 18.3, 19.6, 43.6, and 44.1) % respectively. The composition of the volatile compounds of the C. reinhardtii extract was determined by GC/MS. Different groups of compounds were identified   such as hydrocarbons, phenols, alcohols and esters, and two bioactive compounds;1-Heptacosanol and Octadecyl chloride which is used in medical and pharmaceutical fields.These results provide an indication of the existence of hopeful antibiofilm compounds in the algal species under study. Further chemical studies are required to illuminate these compounds, structures and activity.

Two hundred fifty mid-stream urine specimens were collected from Baqubah Teaching Hospital and Al-Batool Teaching Hospital from patients with urinary tract infections (UTI). Of these investigated urine specimens, 66 (26.4%) specimens showed positive growth culture of Gram-negative bacteria. From these, Escherichia coli was the most prevalent bacteria of the examined culture (41, 62.12%). Additionally, the cup assay was used to determine colicin producers while the most efficient colicin producers were estimated by the formation of larger inhibition zone. Approximately half of the investigated E. coli isolates (20, 49 %) was colicin producers. Colicins was extracted after induction by mitomycin-C showed a concentration of 3020 μg/ml,  as estimated utilizing the Lowry method, while its activity was 80 U/ml. Our study results showed that colicin had significant antibiofilm activity (P≤ 0.05) against Candida albicans and the effect seemed to be concentration dependent. . However, the values of biofilm inhibition varied depending on the different tested isolates. The biofilm of isolate 5 showed the most significant inhibition (P≤ 0.05) by colicin with a value of 46%, while isolate 3 was less affected with an inhibition rate of 19% at the concentration of 2500 μl/ml.

Plant extracts occupied a big place in diseases treatment and preserving human health because, they contain many active substances that can be exploited in the field of pharmaceutical manufacturing from natural materials. Therefore, this study was conducted to evaluate the effect of different concentrations of plant extracts for each of Nigella sativa, Alliumsativum and Allium cepa against the fungal growth of Candida albicans that cause many skin diseases and infections to humans as well as Trichophyton mentagrophytes, which affects the hair, skin and nails. These two fungi have been isolated and diagnosed from people who have skin infection. Both fungal isolates were treated with extracts of Nigella sativa, Alliumsativum and Allium cepa, which are prepared by using the cold method of extraction. The results showed the effectiveness of Nigella sativa extract against C. albicans and T. Mentagrophytes within two weeks of incubation period. On the other hand, it has been found that the extract of Nigella sativa was more effective on C. albicans, as the rate of inhibition zone was 3.0-4.2 in diameter and 4.5-5.0 on SDA and ESDA media respectively. The results of the fungus T. mentagrophytes indicated that the inhibition zone was 3.2 -3 cm in diameter on SDA medium and 3.0- 4.2 cm on ESDA medium compared with control, as there were no inhibition zones appearance for both fungi. Moreover, the results showed that Allium sativum extract comes in second place in their effectiveness against fangal growth, whereas, the Allium cepa extract did not show any significant impact

Abstract Background There is a worldwide concern with respect to the antimicrobial resistance and the inappropriate use of antifungal agents, which had led to an ever-increasing antifungal resistance. This study aimed to identify the antifungal susceptibility of Candida species isolated from the pediatric patients with cancer and also to evaluate the clinical impact of antifungal stewardship (AFS) ‎interventions on antifungal susceptibility of Candida species. Methods In this study, Candida species colonization were evaluated among hospitalized children with cancer in a tertiary teaching hospital, Shiraz 2017-18. The broth microdilution method was used to determine the minimum inhibitory concentrations (MICs) for polyenes (amphotericin B), echinocandins (caspofungin), and azoles (voriconazole, fluconazole, posaconazole, and itraconazole). Antifungal susceptibility of Candida species was compared with our previous study results to determine the clinical impact of AFS interventions on the antifungal susceptibility. Results The prevalence of Candida albicans in the present study was significantly higher than other Candida species. Candida albicans species were completely susceptible to the azoles. The sensitivity rate of C. albicans to amphotericin B and caspofungin was 93.1% and 97.1%, respectively. The results confirm the positive effect of optimized antifungal usage and bedside intervention on the susceptibility of Candida species after the implementation of the AFS program. C. albicans and C. glabrata exhibited significant increase in sensitivity after the execution of AFS program. Conclusions Improving the antifungal agent usage can improve antifungal ‎susceptibility and reduce resistance. The AFS is recommended to be addressed, applied, and regularly assessed in centers.

Durian is a fruit that is in demand by the community, but during durian season environmental problems arise due to waste from skin of durian. The aimed of the study was to prove the antifungal activity of the ethanol fraction of durian skin against Trichophyton mentagrophytes and Candida albicans and to determine the active active compounds inhibiting T. mentagrophytes and C. albicans fungi. Durian skin was extracted by maceration method, then fractionated used ethanol, chloroform, and n-hexane solvents. Ethanol fraction was tested on T. mentagrophytes and C. albicans fungi used the disc method with a concentration of 10%, 20%, 40%, 60%, 80%, 100%, distilled water as a negative control, ketoconazole as a positive control. The best inhibition zone is fungi T. mentagrophytes concentration 100% ( 9.23 mm) negative control (0 mm), positive control (32.96 mm), fungi C. albicans the best inhibition zone is concentration of 100% (8.30 mm), positive control (17.17 mm) negative controls (0 mm).The ethanol fraction was screened by the Thin Layer Chromatography method and positively contained flavonoids (rf 0.7), tannins (rf 0.58), saponins (rf 0.76) and alkaloids (rf 0.64). Minimum Inhibitory Concentrations of T. mentagrophytes were 7% and C. albicans were 8%. The most effective active compound growht of T. mentagrophytes is tannins (rf 0.7) and for C. albicans is flavonoid (rf 0.6). The ethanol fraction of durian bark extract has antifungal activity against T. mentagrophytes and C. albicans. <p>“Kayu kuning” (<em>Arcangelisia flava </em>L.Merr) was used when someone has a skin problem caused by <em>Candida </em>albicans<em> </em>and <em>Trichophyton mentagrophytes</em>. Scientific based medicine on this traditional knowledge was necessary be  done. Stem powderwas extracted by  distilled water.The extract was then evaporated. Qualitative and quantitative analysis of the active substance e.g., Berberin chloride by Thin Layer Chromatography (TLC)  The antifungal activity againts <em>Candida albicans</em> and <em>Trichophyton mentagrophytes</em>were tested by using agar diffusion and microdilution methods.  The absorbance from microdilution were analized by One way ANOVA. The conclusion showed that the extract contained 1.55±0.12% w/walkaloid calculated as Berberine chloride. The inhibition zone for <em>Candida albicans </em>and <em>Trichophyton mentagrophytes </em>were 16.65±4.52 and 6.55±0.05 mm respectively. The MIC vallue for both fungi was 10 mg/mL.The MBC value for <em>Candida albicans</em> was 40 mg/mL and for <em>Trichophyton mentagrophytes </em>was 50 mg/mL. From the analysis with one-way ANOVA, shows that there are significant differences between the positive control group and the test solution with the negative control group with p=0.020 for <em>Candida albicans</em> and p=0.028 for <em>Trichophyton mentagrophytes</em> (p&lt;0.050). Post hoc Tukey analysis results showed  that both inter-group and between the concentration of the test solution to the control group did not differ significantly positive because the value of p&gt;0.050.</p>

Background: Candida species have emerged as one of the most common causes of bloodstream infections (BSIs). There are limited data on the distribution of Candida spp. and susceptibility by year. Objectives: In this study, we analyzed changes in the distribution of Candida spp. and their antifungal susceptibility profiles from blood cultures. Methods: Records from January 2016 to December 2020 were obtained from the microbiology laboratory in Istanbul. Antifungal susceptibility tests were performed using the VITEK 2 compact system and evaluated according to EUCAST breakpoints. A total of 241 unique candidemia episodes were included in this study. Results: Candida albicans was the predominant pathogen (n = 95, 39.42%), followed by C. parapsilosis (n = 82, 34.02%), C. glabrata (n = 18, 7.47%), C. tropicalis (n = 17, 7.05%), C. krusei (n = 15, 6.22%), and other Candida spp. (n = 14, 5.79%). There was no statistically significant difference in the percentage of episodes of Candida spp. After data analysis, a tendency to shift from C. albicans to C. parapsilosis was observed in the period analyzed in this study. Candida albicans was the most common species in intensive care units (ICUs), hematology and hemopoietic stem cell transplantation units, and surgical clinics, with C. parapsilosis predominant in medical clinics. In general, micafungin susceptibility was the highest, and fluconazole was the lowest. There was reduced sensitivity to fluconazole and voriconazole for C. albicans and C. parapsilosis over 5 years. Conclusions: Detecting changes in the distribution of Candida spp. and antifungal susceptibility over time will lead to the selection of appropriate empirical therapy and monitor phenomena of antifungal resistance. Empirical treatment with antifungal agents is associated with high costs, toxicities, and risk of antifungal resistance. Therefore, it is mandatory to determine and monitor Candida spp. and antifungal susceptibility testing to select appropriate antifungal agents.

Background: Oral candidiasis is the most common clinical manifestation in a person infected with HIV which has been linked with the severity of the disease. Candida albicans responsible for most oral candidiasis. The increased resistance to the available antifungal drugs can reduce the efficacy of therapy. Therefore, it will be crucial to discover novel antifungal agents. One of them is tea tree oil (TTO) or Melaleuca alternifolia which has a potent antifungal effect to inhibit the growth of Candida sp.  Purpose: To evaluate the antifungal susceptibility of TTO 5% compared with nystatin against Candida sp. taken from HIV/AIDS patients. Methods: The study assessed the antifungal effect of TTO 5% and nystatin against 33 isolates of Candida sp. that was isolated from the oral cavity of 30 HIV/AIDS patients in Dr. Soetomo General Academic Teaching Hospital Surabaya, Indonesia. Antifungal susceptibility was evaluated by the disc diffusion method. The diameter of the inhibition zone was determined as the result of this study. Result: The most strains found in this study was Candida albicans (84.8%). The mean inhibition zone of nystatin against all strains was 23.24 mm whether it was 17.55 mm for TTO 5%. The inhibition zone of both TTO 5% and nystatin in Candida non-albicans were tended to be higher than in Candida albicans. It revealed that the inhibition zone of TTO 5% significantly lower than nystatin (P = 0.00; P < 0.05).Conclusion: Therefore, the antifungal susceptibility of nystatin was significantly higher than TTO 5% against Candida albicans and non-albicans.

Fungal infections that occur in the skin and nails were caused by fungi such as Candida albicans and Trichophyton mentagrophytes. Aloe vera contains saponins and acemannan that were effective as an antifungal. This study aims to determine the effect of carbopol concentration variation on aloe vera gel to antifungal activity of Candida albicans and Trichophyton mentagrophytes.Aloe vera powder was obtained from freeze drying aloe vera juice which had been frozen. Aloe vera powder was formulated in gel form using carbopol base with concentration 0.5; 1.0; 1.5; and 2.0 %w/w. The aloe vera gel was tested for its physical properties and then tested the antifungal activity using the diffusion method of wells.The result of evaluation of physical properties showed the high concentration of carbopol then pH, viscosity, adhesiveness increased, and spreading capacity decreased. The results of antifungal activity test showed that aloe vera powder had activity on Candida albicans and Trichophyton mentagrophytes with inhibition zone of 15 ± 3 mm and 13 ± 0,5 mm. Aloe vera gel test results with variation of carbopol concentration 0.5; 1.0; 1.5; and 2.0 %w/w did not affect the antifungal activity of Candida albicans and Trichophyton mentagrophytes.

Caring for the persons with first episode psychosis is challenging and demanding. It may lead to the increased burden, expressed emotions among the caregivers. The numerous studies have shown that high burden and negative expressed emotions among caregivers can lead to early relapse in the patients with first episode psychosis. To evaluate the effects of the brief psychoeducation on the caregivers burden and expressed emotions. A quasi experimental - before and after with control group research design was adopted for the study. A total of 60 caregivers have participated in the study, of which 30 caregivers in experimental group and 30 caregivers in the control group. Family Burden Interview Schedule (Pai and Kapoor, 1981) and Attitude Questionnaire (Sethi et al., 1981) was used to assess caregiver's burden and expressed emotions. At end of the psychoeducation intervention, burden among caregivers and negative expressed emotions of the caregivers have significantly reduced. The positive expressed emotions have been increased. Study results demonstrates the importance of psychoeducation intervention in reducing the burden and negative expressed emotions.
