The aim of this study was to develop a protocol for production of polyploid M1-generation plants of Viola × wittrockiana Gams. Two variants of colchicine treatment were compared for their efficiency. Early detection of novel ploidy levels was achieved by screening of stomata size, leaf index value (leaf blade length/width), and other morphological characteristics of the M1-generation. Secondary screening for novel ploidy levels was performed by flow cytometry (FCM). Hexadecaploid, aneuploid, and mixoploid plants were successfully identified by FCM.

Abstract In vitro-derived plants with elevated ploidy levels can display distinguishing features from the plants they are derived from, especially owing to their indirect regeneration. Genome size affects cell size and, thus, affects plant morphological characteristics. Therefore, stomata traits may be used as a diagnostic feature allowing identification of ploidy. Actinidia chinensis var. deliciosa A. Chev. (A. Chev.), known as kiwifruit, is successfully cultured in vitro and redifferentiated into plants via endosperm-derived calli. To identify differences between hexaploids obtained from seeds and confirmed nonaploids obtained from endosperm-derived calli, we analyzed the stomata. Our results confirmed that ploidy coincides with mean stomatal length, width, and density. Despite this correlation, this method cannot be used to distinguish individual hexaploid kiwifruit plants from nonaploid ones because samples with different ploidy yielded overlapping measurements.

Urochloa (including Brachiaria, Megathyrus and some Panicum) tropical grasses are native to Africa and are now, after selection and breeding, planted worldwide, particularly in South America, as important forages with huge potential for further sustainable improvement and conservation of grasslands. We aimed to develop an optimized approach to determine ploidy of germplasm collection of this tropical forage grass group using dried leaf material, including approaches to collect, dry and preserve plant samples for flow cytometry analysis. Our methods enable robust identification of ploidy levels (coefficient of variation of G0/G1 peaks, CV, typically <5%). Ploidy of some 348 forage grass accessions (ploidy range from 2x to 9x), from international genetic resource collections, showing variation in basic chromosome numbers and reproduction modes (apomixis and sexual), were determined using our defined standard protocol. Two major Urochloa agamic complexes are used in the current breeding programs at CIAT and EMBRAPA: the ’brizantha’ and ’humidicola’ agamic complexes are variable, with multiple ploidy levels. Some U. brizantha accessions have odd level of ploidy (5x), and the relative differences in fluorescence values of the peak positions between adjacent cytotypes is reduced, thus more precise examination of this species is required. Ploidy measurement of U. humidicola revealed aneuploidy.

Satellite remote sensing is a valuable tool for coastal management, enabling the possibility to repeatedly observe nearshore sandbars. However, a lack of methodological approaches for sandbar detection prevents the wider use of satellite data in sandbar studies. In this paper, a novel fully automated approach to extract nearshore sandbars in high–medium-resolution satellite imagery using a GIS-based algorithm is proposed. The method is composed of a multi-step workflow providing a wide range of data with morphological nearshore characteristics, which include nearshore local relief, extracted sandbars, their crests and shoreline. The proposed processing chain involves a combination of spectral indices, ISODATA unsupervised classification, multi-scale Relative Bathymetric Position Index (RBPI), criteria-based selection operations, spatial statistics and filtering. The algorithm has been tested with 145 dates of PlanetScope and RapidEye imagery using a case study of the complex multiple sandbar system on the Curonian Spit coast, Baltic Sea. The comparison of results against 4 years of in situ bathymetric surveys shows a strong agreement between measured and derived sandbar crest positions (R2 = 0.999 and 0.997) with an average RMSE of 5.8 and 7 m for PlanetScope and RapidEye sensors, respectively. The accuracy of the proposed approach implies its feasibility to study inter-annual and seasonal sandbar behaviour and short-term changes related to high-impact events. Algorithm-provided outputs enable the possibility to evaluate a range of sandbar characteristics such as distance from shoreline, length, width, count or shape at a relevant spatiotemporal scale. The design of the method determines its compatibility with most sandbar morphologies and suitability to other sandy nearshores. Tests of the described technique with Sentinel-2 MSI and Landsat-8 OLI data show that it can be applied to publicly available medium resolution satellite imagery of other sensors.

Kerang bambu merupakan  salah  satu  jenis  Moluska  dari  famili  Solenidae  yang mempunyai nilai ekonomis. Potensi sumberdaya hayati kerang bambu ini menarik untuk diteliti lebih dalam mengingat permintaannya yang semakin meningkat. Upaya pengambilan kerang bambu jika tidak diimbangi dengan selektivitas ukuran dan dilakukan penangkapan secara terus-menerus maka dapat mengakibatkan hilangnya organisme ini. Mengingat masih minimnya informasi mengenai kerang bambu (Solen sp.) sehingga perlu dilakukan penelitian mengenai morfometri, hubungan panjang dan berat serta indeks kondisi kerang bambu. Tujuan dari penelitian ini adalah mengetahui hubungan panjang cangkang dan berat total serta nilai indeks kondisi dari kerang bambu (Solen sp.). Penelitian ini dilakukan dengan mengukur aspek morfometri seperti panjang, lebar dan berat total. Hasil penelitian menunjukkan hubungan antara panjang cangkang dan berat total memiliki nilai b = 3,99 dan R2=0.5742. Nilai indeks kondisi kerang bambu (Solen sp.) dari TPI Tasik Agung, Rembang, Jawa Tengah pada kategori kurus sebesar 1.9% dengan jumlah 1 ekor, kategori sedang sebesar 13,3% dengan jumlah 67 ekor dan kategori gemuk sebesar 86.4% dengan jumlah 433 ekor.Bamboo clams are type of mollusc from the Solenidae family that have economic value. The potential of bamboo clam is interesting to be investigated more deeply considering its increasing demand. Efforts to collect bamboo clams of it’s not balanced with size selectivity and continuous fishing can result in the loss of these organisms. Given the lack of information on bamboo clams (Solen sp.) it is necessary to conduct research on morphometry, length and weight relationship and condition index of bamboo clams. The purpose of this study was to determine condition index value of bamboo clams (Solen sp.). This research was conducted by measuring morphometric aspects such as length, width, and total weight. The result showed that the condition index value of bamboo clams (Solen sp.) in TPI Tasik Agung, R, Central Java in the thin category was 1.9% with 1 individuals, the moderate category was 13.3% with 67 individuals and the fat category was 86.4% with 433 individuals.

Eighty nine Artemisia samples treated with different concentrations of colchicine were used as breeding samples, with diploid Artemisia as the control. The ploidy levels of samples were determined by flow cytometry and confocal laser scanning microscopy (CLSM). An analysis of the flow cytometry results identified three suspected tetraploid plants and seven suspected triploid plants. The results of this study may be useful for breeding new Artemisia lines.

It is known that the typical for animals of different species the nature of the serrations of the free edges of the scales of the hair cuticle, the density of their location and proximity to each other can determine the species of animal hair samples. The aim of the study was to establish the value of the cuticular index in clinically healthy domestic cats and dogs. The area of the outer surface of the scales of the hair cuticle, or cuticular index, was determined in 20 clinically healthy domestic cats and 18 dogs. Examination of the cuticle was performed using prints on a colorless lacquer, which was applied to a glass slide, pressing the hair to the paint layer. The imprint was examined under a microscope (enlargement ×400) and the cuticular index value was established in the root zone, in the thickest part of the hair shaft, expressed in μM2. It was found that in domestic cats and dogs without clinical signs of any pathology, the values of the cuticular index are in the range of 80-170 and 100-210 μM2accordingly. The value of the cuticular index in domestic cats 60-70 μM2 and in dogs 60-90 μM2in 100 % accidents does not meet in healthy animals. The obtained results can be used as a control during diagnostic activities for numerous variants of internal pathology of domestic cats and dogs, as additional diagnostic tests.

<p><em>Salvia multicaulis</em> is a widespread species of Lamiaceae family in Iran. There are many discussions about its infraspecific variations. Although some varieties were definite for this species in various parts of the world, no infraspecific taxon was reported in Iran and all samples of this species were named as S. multicaulis. In this study, morphological characteristics of S. multicaulis populations, naturally growing in Iran, was examined. Twenty-two traits were examined in 94 individuals of this species to<br />identify their phenotypic difference. Most of the investigated features were showing a high degree of variability, but it was highly pronounced for some characteristics such as basal leaf shape, basal leaf width, basal leaf length/ width ratio and basal petiole length. Significant positive/negative correlations were observed between some morphological variables. Furthermore, significant negative correlations were found between the eastern distribution of populations with basal leaf petiole length and basal leaf length/ width ratio. Based on the UPGMA cluster analysis, populations were divided into two main branches. The first branch contained four populations, while the second branch was bigger and clustered in two sub-branches. In one of them,<br />three populations and in another one the rest populations arranged in two groups. CA joined plot confirmed that each of studied populations or group of populations had distinct morphological trait(s), which were useful in identification of them. Our findings supported population no. 13 had unique morphological traits such as the largest bracts and basal leaf petiole, highest flower number of each inflorescence cycle, widest and largest calyx. The conservation of the highly diverse populations of<br />Iranian S. multicaulis is recommended.</p>

We report the production of hexaploid plants of interspecific hybrids of Pennisetum, with the ultimate aim to improve the biomass yield, drought tolerance and multicut behaviour of this genus. Chromosome doubling was achieved with the application of colchicine at three different concentrations (0.05, 0.1 and 0.2%) for two time durations (12 and 24 hours). The root slips and stem cuttings of interspecific hybrids were used for treatment and the root slips were found to be more efficient. The preliminary screening to select the putative hexaploid plant was done based on stomatal frequency and morphology. Plants containing significantly lower stomatal frequency and larger stomata size were selected for further analysis by chromosome counting. This experiment confirmed that 0.1% concentration of colchicine treatment to root slips for 24 hours was more effective to induce the amphiploids in Pennisetum.

The coffee growth is considered to depend, partly, on the environmental condition at which they develop to accomplish both vegetative and generative stages. An exploratory survey in Lampung aimed at investigating the morphological characteristics of 13 Robusta coffee accessions growing on different altitudes.Local superior coffee clones were selected from eight farms, and subjected to identifications. Three key issues were concerned for both quantitatively, i.e. plant components of leaves, branches, and fruits, and qualitatively such as leaves and fruit characteristics, i.e. colour and shape. The analysis shows a similarity of about 60% of total qualitative variable identified among different accessions. The remaining characters are suggested to be more varying such as flush colour, leaf surface, ripe fruit colour, and stipule shape. Furthermore, a quantitative analysis showed a higher level of similarity for fruit characters, i.e. fruit length, -width, and -thickness, and to a slightly lower level for leaf characteristics, i.e. leaf length and -width, and number of productive branches. It was revealed that some variables, i.e. fruit weight; leaf and seed; and canopy characteristics, explaining the morphological variation of coffee throughout the accessions. Furthermore, cluster analysis may indicated a possible similarity of coffee morphologies either from area with different or the same environmental conditions. A high heterogeneity related to environmental conditions, genotypic variations, plant nutritional status, and agronomic practices, which unable to confirm in the present study, may limit the specific conclusions.

The hardy Actinidia species represent a source of genetic diversity for improving A. deliciosa (kiwifruit) as well as for creating new economically important cultivars through intra- and interspecific crosses. Attempts at breeding in Actinidia have been complicated by the existence of intraspecific as well as interspecific variation in ploidy. The haploid chromosome number in Actinidia is 29 and diploid (2n=2x=58), tetraploid (2n=4x=116), and hexaploid (2n=6x=174) levels have been identified. Because of the problems encountered when crossing parents differing in ploidy level, it is desirable to know the ploidy levels of plants to be used in breeding. We determined the ploidy levels of 61 Actinidia accessions currently available in the U.S., including primarily accessions of relatively winter-hardy species. The 61 accessions, representing eight species and three interspecific hybrids, were screened for ploidy using flow cytometry. Mitotic root tip cells from one plant from each putative ploidy level were examined microscopically to confirm the ploidy level derived from flow cytometry. There were 17 diploids, 40 tetraploids, and 4 hexaploids. Intraspecific variation was not found among accessions of the species arguta, callosa, deliciosa, kolomikta, melanandra, polygama, or purpurea. All kolomikta and polygama accessions were diploid. All arguta, callosa, melanandra, and purpurea accessions were tetraploid. Actinidia deliciosa was hexaploid. One chinensis accession was tetraploid. Two accessions (NGPR 0021.14 and 0021.3), acquired as chinensis, were hexaploid and may, in fact, be A. deliciosa based on their morphology. `Issai' (arguta × polygama) was hexaploid and `Ken's Red' and `Red Princess' (both melanandra × arguta) were tetraploid.
